황정(黃精)과 증숙 황정(蒸熟 黃精)의 항산화, 항피로 효능 비교 연구 (Antioxidant and Anti-physical fatigue Effects of Polygonati Rhizoma and steamed Polygonati Rhizoma)

  • Objectives: Polygonati Rhizoma (PR) has containing the bioactive compounds such as poly sccharide A,B,C, oligosaccharide, amino acid, it has reported to anti-diabetes and hypertension, atherosclerosis. In this study, we were evaluates antioxidant and anti-physical fatigue effects of PR and steamed PR.Methods : The sample was divided into 5 groups-PR0 (PR without steaming process), PR1 (PR with once steaming process), PR3 (PR with third steaming process), PR6 (PR with sixth steaming process), PR9 (PR with ninth steaming process). We measured anti-oxidant activity through contents of polyphenol, flavonoid and DPPH, ABTS free radical scavenging capacity. And, anti-physical fatigue effect was evaluated using the swimming test, and the AMPK protein expressions in soleus muscle.Results : As a result, polyphenol, flavonoid, DPPH, ABTS free radical scavenging capacity of PR were increased as steaming times. Anti-physical fatigue effects by swimming test, PR0 have significantly increased, but steamed PR groups were decreased. The AMPK protein expressions of PR0 and PR1 groups were increased comparing with PR3, PR6 and PR9. All groups had effects on decreasing TG, creatine in blood serum, but had no effects on TC in blood serum.Conclusions : In conclusion, PR with 9 steaming process was more excellent than not-processed PR in anti-oxidant effect such as DPPH, ABTS radical scavenging activity and contents of polyphenol, flavonoid, but, not-processed PR increased swimming times than processed PR. These results suggest that processed PR has anti-oxidant effect as steaming times, and not-processed PR may be a novel potential anti-physical fatigue agents than processed PR.

Developmental Gene Expression of Antimicrobial Peptide PR-39 and Effect of Zinc Oxide on Gene Regulation of PR-39 in Piglets

  • Two experiments were conducted to evaluate developmental gene expression of antimicrobial peptide PR-39 and effect of zinc oxide on gene regulation of PR-39 in piglets using semi-quantitative RT-PCR analysis. In experiment 1, fifteen female Tai-Hu pigs (a local breed in China) in five groups, each of three pigs at 1, 14, 28, 42 and 56 days of age were used to determine effect of age and weaning on mRNA expression of PR-39. In experiment 2, nine groups of pigs (total seventy-two female 36 days-age weanling Tai-Hu piglets) were assigned to three treatments (${ZnO}_0$, ${ZnO}_{100}$ and ${ZnO}_{3000}$). The feeding experimental period lasted 15 days. After feeding experiment, nine pigs with three animals in each treatment were chosen to determine the effect of ZnO on PR-39 mRNA expression of pigs. The results showed that PR-39 mRNA levels increased steadily in postnatal day 1-28 (preweaning), and weaning significantly decreased PR-39 mRNA expression of piglets (p<0.05). ${ZnO}_{3000}$ (3,000 mg zinc/kg diet) significantly increased PR-39 mRNA expression (p<0.05) when piglets were feed ${ZnO}_{3000}$ diet for 15 days. ${ZnO}_{100}$ (100 mg zinc/kg diet) also increased PR-39 gene expression, but the result was not statistically significant (p>0.05). The result was in accordance with the effect of ${ZnO}_{3000}$ and ${ZnO}_{100}$ on weight gain of piglets and prevention of diarrhea.

  • 희토류원소 Pr의 첨가가 Fe-B계 비정질합금의 자왜에 미치는 영향을 조사하기 위하여 비정질 $Fe_{86-x}B_{14}Pr_{x}(2{\leq}x{\leq}8\;at.%)$ 합금을 급속응고법을 이용하여 제조하였다. 급 속응고상태 비정질 $Fe_{86-x}B_{14}Pr_{x}$ 합금의 포화자왜는 Pr이 증가할 수록 증가하여 x=8에 서 약 50 ppm이 얻어졌다. 특히 $300^{\circ}C$에서 2시간 열처리된 비정질 $Fe_{84}B_{14}Pr_{2}$ 합금의 포화자왜는 70 ppm으로 증가하였다. $300^{\circ}C$에서 2시간 열처리된 비정질 $Fe_{84}B_{14}Pr_{2}$ 합금의 교류 자기이력손실 및 교류투자율$(f=50\;kHz,\;B_{m}=0.1\;T)$은 각각 15 W/kg, $5.5{\times}10^{3}$이었다.

원지(遠志)가 NMDA로 유발된 선경세포 손상에 미치는 효과 (A Study on the Protective Effects of Polygalae Radix on Neurotoxicity Induced by N-methyl-D-aspartic acid(NMDA))

  • Objectives : Polygalae Radix (PR) from Polygalae tenuifolia (Polygalaceae) has been clinically used as a sedative, anti-inflammatory, and anti-bacterial agent. To extend pharmacological effects of PR in the central nervous system (CNS) on the basis of its CNS protective effect, the present study was conducted to identify the effect of PR, whether it shows the neuroprotective action against excitatory neurotoxicity. Methods : To identify the protective effect of PR to excitatory neuro-toxic agent, the present study was focused on the PR effect on cell death, that was caused by applying NMDA to nerve cell, elevation of $(Ca^{2+})_i$, releasement of glutamate, and ROS generation. Result : 1. PR methanol extract, at the concentration range of 0.05 to 5 g/ml, significantly inhibited NMDA (1 mM)-induced neuronal cell death as well as MK-801 (non competitive NMDA antagonist). 2. PR methanol extract $(0.5\;{\mu}g/ml)$ inhibited NMDA (1 mM)-induced elevation of cytosolic calcium concentration $[Ca^{2+}]_i$. NMDA application in the presence of MK-801 $(10\;{\mu}M)$ failed to produce the increase of $[Ca^{2+}]_i$ through all the measurement time. 3. PR methanol extract $(0.5\;{\mu}g/ml)$ inhibited the NMDA-induced elevation of glutamate release. Also, MK-801 showed similar protective effects. 4. PR methanol extract $(0.5\;{\mu}g/ml)$ inhibited the NMDA-induced elevation of ROS generation. Also, MK-801 showed similar protective effects. Conclusion : The present study provides the availability of PR to exert its protective effect on the neuronal cell death in various neurodegenerative pathophysiological conditions.

PR실무자의 직무특성이 번영에 미치는 영향 : 일의 의미와 직무열의의 이중매개효과 연구 (The Effect of Job Stability on Senior Citizen's Quality of Life : Mediated Effect of Job Satisfaction)

  • 본 연구의 목적은 PR실무자의 직무특성이 번영에 미치는 영향과 일의 의미와 직무열의의 단순 매개 및 이중 매개 역할을 확인하고자 하였다. 분석을 위하여 총 353명의 PR기업에 종사하는 PR실무자를 대상으로 설문조사를 실시하여, 직무특성, 일의 의미, 직무열의, 번영에 대한 데이터를 수집하고 다중회귀분석과 SPSS Process Macro 부트스트래핑 방법을 통해 연구가설을 검증하였으며, 결과는 다음과 같다. PR실무자의 직무특성이 번영에 영향을 미치는 관계에 대해서 일의 의미과 직무열의가 각각 단순매개효과를 보였으며, 일의 의미와 직무열의는 순차적으로 영향을 미치는 이중매개효과를 보이는 것으로 나타났다. 세부적으로는 자신의 일이 주변에 미치는 영향력이 크다고 생각할수록, 직무 수행에 필요한 기술과 지식이 다양할수록, 자율성이 높을수록, 피드백을 많이 받을수록 일할 만한 가치를 높게 여겨 직무열의가 높아지고, 이에 따른 번영감을 더 잘 경험하는 것으로 나타났다. 또한 일할 만한 가치가 높다고 느낄수록 더 잘 집중하고 직무에 빠져서 몰두하여 일하면서도 번영감이 높아지는 결과가 나타났다. 이러한 결과는 직무특성과 직무열의와 무관하게 일의 의미가 번영감에 영향을 미치는 중요한 요인이며, PR기업은 PR실무자들의 일의 의미감을 높이기 위한 노력이 중요하다는 것을 의미한다. 본 연구의 의의와 한계점은 결론에서 다루었다.

  • The photorefractive (PR) effect in liquid crystals sandwiched between photoconductive polymer layers was first studied by Ono et al. They reported that the PR effect vanished at steady state If there were not insulating layers because no charge trapping occurred in the photoconductive poly(N-vinylcarbazole) (PVK) layers. However we observed a significant PR effect in the polymer layered liquid crystal (PLLC) system where a liquid crystal layer doped with fullerene is sandwiched between two photoconductive PVK layers. 칡의 부위별 골다공증 치료효과 (Pueraria lobata Ohwi as an Osteoporosis Therapeutics)

  • 콩과 식물인 갈근에는 genistein과 daidzein이 함유된 것으로 알려졌으므로 본 연구는 갈근(PR), 갈화(PF) 및 어린순인 갈용(PL)에 함유된 phytoestrogen의 함량을 HPLC법으로 정량 분석하였다. 동시에 phytoestrogen의 골다공증 치료제에 대한 효능 검색의 일환으로 조골세포의 증식작용(MTT법 및 ALP분석)과 피골세포의 활성(TRAP법)을 측정하였고 난소절제 흰쥐에 PR 1 g/kg/day(PR-1) 및 PR 5 g/kg/day(PR-5)을 9주 동안 투여한 후 혈액분석, 혈장 Alkaline phospatase(ALP), 칼슘, 무기성 인산염, 총콜레스테롤, LDL- 및 HDL-콜레스테롤을 측정하였고 적출된 경골 및 요추골의 소주골면적을 측정하였다. PR과 PF의 총 daidzein의 함량은 $10436{\pm}2144\;mg/kg$$1003{\pm}206\;mg/kg$이었고 genistein과 formononetin은 PR에만 존재하고 PF 및 PL에는 함유되어 있지 않았다. 인간 유사 조골 세포주인 Saos-2에 PR은 대조군의 158% 정도(5 mg/mL)의 세포증식효과와 ALP 활성을 증가시켰으나 전처리한 PR, PF 및 PL의 세포증식효과는 없었고 PL만이 ALP 활성을 증가시켰다. 파골세포의 증식억제실험에서 genistein의 $IC_{50}$$1.57{\times}10^{-4}\;mg/mL$ $(5.81{\times}10^{-7}\;M)$이었고, daidzein과 PR은 효과가 없었다. 난소절제 흰쥐에 대한 in vivo 실험에서 대조군이 sham군보다 난소적출 1주 후부터 체중의 증가가 급격하게 나타났으나 PR의 투여로 둔화되었다. PR 투약에 의한 자궁의 무게는 PR-5군은 대조군에 비해 증가되었고(p<0.05), PR-1군은 대조군과 유의성이 없었다. 혈장 ALP의 활성은 주령의 증가에 EK라 감소하는 경향을 보였고, HDL-콜레스테롤의 농도는 모든 군에서 주령에 따라 감소되었으나(p<0.01), PR-5군의 LDL-콜레스테롤의 농도는 대조군에 비해 감소되었다(p<0.01). 대조군에 비해 sham군의 경골 및 요추골의 면적은 증가되었고(p<0.01), PR 투여군(PR-1 및 PR-5)도 sham보다 적으나 대조군에 비해 소주골 면적이 증가되었다(각 p<0.01). 특히 고용량의 PR 투여는 PR의 골다공증 치료효과가 우수함을 증명하였으며(p<0.01), PL과 PF는 효능이 미비하나 PR은 골다공증의 예방 및 치료제로는 상당히 우수한 한약재임이 확인되었다.

백작약 추출물이 항산화활성, LDL 산화 억제 및 혈전용해에 미치는 영향 (Preventive Effects of Peony Root Extracts on Oxidative Stress, Thrombosis and Atherosclerosis)

  • Objectives: There is currently increased interest in the identification of natural antioxidant compounds derived from various plants. Peony Root (PR) is used worldwide for the treatment of many types of cardiovascular disease including atherosclerosis and hypertension. It has been used in Korean traditional medicine for the treatment of glycosuria, hypertension and cancer. However, to date, no studies concerning the antioxidant properties of PR have been conducted. Therefore, this study was conducted to evaluate the in vitro scavenging activity, inhibitory effect of LDL oxidation of pro-oxidant reactive species and anti-thrombosis effect in response to treatment with PR using various screening methods including biological and non-biological oxidants. Methods: In this study, the antioxidant activity of extract from PR was studied with in vitro methods by measuring the antioxidant activity by TEAC, measuring the scavenging effects on reactive oxygen species (ROS) [superoxide anion, hydroxyl radical] and on reactive nitrogen species (RNS) [nitric oxide and peroxynitrite] as well as measuring the inhibitory effect on $Cu^{2+}$-induced human LDL oxidation and the inhibitory effect on collagen-induced platelet aggregation. Results: The PR extracts were found to have a potent scavenging activity of oxidative stress [DPPH, superoxide anion, hydroxyl radical, nitric oxide and peroxynitrite, etc.] as well as an inhibitory effect on LDL oxidation and on platelet aggregation. Conclusions: The PR extracts have anti-oxidative and anti-atherosclerotic effects in vitro system, which can be used for developing pharmaceutical drugs against oxidative stress and atherosclerosis.

Protective Effect of Puerariae Radix on Ovariectomy-induced Bone Loss in Rats

  • To examine the bone-protective effects of Puerariae Radix (PR) in an estrogen-deficient animal model, we treated ovariectomized rats with PR at a dose of 10.0 mg/kg 3 times a week for 16 weeks. Our results showed that PR increased mineral content and density of the trabecular bone at the neck of the left femur. Moreover, biochemical data indicated that PR had a positive effect on bone turnover. No endometrial hyperplasia was detected in the PR group. The present data suggest that PR should be considered for use in the treatment of bone loss in women with postmenopausal osteoporosis.

Recycling of Sintered Nd-Fe-B Magnets Doped with PrNd Nanoparticles

  • The waste of sintered Nd-Fe-B magnets was recycled using the method of dopingPrNd nanoparticles. The effect of PrNd nanoparticle doping on the magnetic properties of the regenerated magnets has been studied. As the content of the PrNd nanoparticles increases, the coercivity increases monotonically, whereas both the remanence and the maximum energy products reach the maximum values for 4 wt% PrNd doping. Microstructural observation reveals that the appropriate addition of PrNd nanoparticles improves the magnetic properties and refines the grain. Domain investigation shows that the self-pinning effect of the rare earth (Re)-rich phase is enhanced by PrNd nano-particle doping. Compared to the magnet with 4 wt% PrNd alloy prepared using the dual-alloy method, the regenerated magnet doped with the same number of PrNd nanoparticles exhibits better magnetic properties and a more homogeneous microstructure. Therefore, it is concluded that PrNd nanoparticle doping is an efficient method for recycling the leftover scraps of Nd-Fe-B magnets.
