Interspecific relationship between two food organisms in the combination culture tank of rotifer, Brachionus rotundiformis and copepod, Tigriopus japonicus (로티퍼, Brachionus rotundiformis와 코페포다, Tigsiopus japonicus의 혼합 배양조에 있어서 두 종간의 상호 관계)

  • The common harpacticoida copepod species Tigriopus japnnicus was often obseued in the mass culture of marine rotifer tanks of sea farming stations. In this study, we investigated the co-existing effect of T. iaponicus to the rotifer Brachionus rotundiformis population growth. The culture conditions as temperature, salinity, culture volume, photo period, culture period and observation interval were 25{\circ}C, 22ppt, 40ml, 12L:12D, 16 deys$ and every two days during the experimental period, respectively. Nannochloropsis oculata was used as the food for the two testing organisms. After counting the growth of rotifer and copepod, they were transferred to new culture tank with N. oculata at the density of $7{times}10^5\;cells/ml$. The population growth of B. rotundiformis was suppressed by T. japonicus when these two organisms were mix-cultured comparing to single culture of B. rotundiformis. But population growth of T, japomcus was not affected by the presence of B. rotundiformis. The interspecific relationships of prey-predation and food competition were not observed in the combination culture of rotifer B. rotundiformis and copepod T japonicus.

  • The radio communications by SSB and VHF transceivers are still used very efficiently in coast stations. The SSB and VHF equipments are very important to transmit and receive informations in the sea and the land. The communication system by the conventional SSB and VHF transceivers between a coast station and a terminal is an one-to-one system. In this dissertation, however, the conventional one-to-one system is expanded to one-to-multiple systems. Then, frequencies can be used effectively for distress, urgency, safety traffic. In addition, one to multi-number systems can be used to interrupt. When the ICS equipments are set up to the VHF transceiver. It is possible to communicate with ship in far distance the communication range can be enlarged. The line switching system by the ICS is to be remote-controlled by ASK modulated PTT signals and audio signals. An ICS can change a connection between terminal and transceiver through a circuit switching system at any time. For this purpose, the author has researched and developed a ASK transmission system, ICS system, control algorithm, multiprocessor system, and monitoring system. As a conclusion, the developed line switching control systems and equipments can be used effectively for maritime communication, military communication, fishery communication, etc.

Egg Development and Morphology of Larvae and Juveniles of Spotted Knifejaw, Oplegnathus punctatus (강담돔 (Oplegnathus punctatus)의 난 발생 및 자치어 형태발달)

  • Morphological changes in eggs and larvae of spotted knifejaw, Oplegnathus punctatus were observed by cultivation of fertilized eggs to juvenile stage. Fertilized egg was demersal and transparent. The egg has one oil glouble and its size ranged 1.01~1.15 mm ($1.08{\pm}0.10mm$; $mean{\pm}SD$, n=20). Culture chambers were maintained $22.5{\sim}24.5^{\circ}C$ in water temperature and 33.5~34.5ppt in salinity. It took 25 mins from fertilization to the blastoderm stage, 55 mins to 2 cell stage and 20 hrs 50 mins to hatching. Newly hatched yolksac larvae ranged 2.59~3.02 mm ($2.81{\pm}0.25mm$, n=5) in total length, and mouth and anus were not still open. Yolk and oil globule were absorbed 2 days after hatching (DAH). Flexion larvae in 12 DAH ranged 3.02~3.07 mm TL ($3.04{\pm}0.04mm$), and the caudal fin grew to fan-shaped, melanophores appeared on top of digestive canal.

Perilla transformation using selection markers containing antibiotics and basta (항생제와 제초제 이중 선발 마커를 이용한 들깨 형질전환)

  • A modified method of Agrobacterium-mediated perilla transformation was developed using two selection markers of an antibiotics (either hpt or nptll) and an herbicidal (bar) gene. Perilla hypocotyl explants were cocultured with Agrobacterium tumefaciens EHA 105 strain harboring plasmid vector (either pMOG6-Bar or pCK-Bar) for three days, respectively. Primary shoots were selected with antibiotics of hygromycin (15 mg/L) or kanamycin (125 mg/L) and regenerated shoots were further selected with herbicide phosphinothricin (ppt,1.2 mg/L) to obtain authentic transformants. Roots were induced for the regenerated shoots on the MS medium without hormone and 80 putative transgenic plants were obtained. Transgene integration into perilla genome was confirmed by Southern blot and their expression was analyzed by Northern blot. T1 perilla seeds drived from To plants were tested 0.3% basta spray for identification of stable gene delivery to next generation.

Factors Influencing Agrobacterium-Mediated Transformation Efficiency in Perennial Ryegrass (Agrobacterium 매개에 의한 페레니얼 라이그라스의 형질전환에 영향을 미치는 요인)

  • A system for the production of transgenic plants has been developed for perennial ryegrass (Lolium perenne L.) via Agrobacterium-mediated transformation. Included in this study were two factors which may affect the gene transfer efficiency: concentrations of acetosyringone (AS, 0 to 300 ${\mu}M$), and co-culture period (1 to 7 days). Both factors were very important to achieve high efficiency gene transformation in the perennial ryegrass. The highest transformation efficiency was obtained when embryogenic calli were inoculated with Agrobacterium in the presence of 100 ${\mu}M$ AS with the culture medium for 5 days. Phosphinothricin resistant calli were developed with into complete plants. GUS histochemical assay, polymerase chain reaction (PCR) and Northern blot analysis of transgenic plants demonstrated that transgenes were integrated into the genome of perennial ryegrass. Using this protocol, it was possible to obtain transformants efficiently for further study.

Changes of Saponin and β-Glucan Content on the Cultured Ginseng with Mushroom Mycelia (버섯균사체로 배양된 인삼 Saponin과 β-Glucan 함량 변화)

  • This study investigated the changes of saponin and $\beta$-glucan content on the cultured ginseng with mushroom mycelia of Phellinus linteus (PL), Ganoderma lucidum (GL), and Hericium erinaceum (HE). Cultured ginsengs with mushroom mycelia were extracted with 80% ethanol, fractionated with n-buthanol, and analysed for ginsenosides by high performance liquid chromatography (HPLC). Crude saponin content of raw ginseng was 4.11% (d.b) but cultured ginseng with mushroom mycelia of PL, GL, and HE were increased to 6.74, 6.77 and 6.23% (d.b), respectively. Ginsenoside-Rd, among the 12 ginsenosides which were available for analysis, was remarkably increased to 13.61, 24.26, and 32.69 mg/g, respectively (raw ginseng: 0.80 mg/g). The $\beta$-glucan content of cultured ginseng with mushroom mycelia of PL, GL, and HE were decreased to 8.85, 5.51 and 5.46% rather than mushroom mycelia of 29.14, 19.44, and 23.39% (d.b), respectively.

Development of Instructional materials using various convex lenses 'VR glasses' for area of Communication technology of the 2015 Revision Curriculum in middle school (2015 개정 중학교 교육과정 통신기술 영역에서 다양한 볼록렌즈를 사용한 'VR 글라스' 수업자료 개발)

  • The purpose of this study is to develop a using various convex lenses 'VR glasses' instructional materials for area of communication technology of the 2015 Revision Curriculum in middle school. This study was progressed depend on ADDIE model. In the analysis step, a necessity of developing the instructional materials was confirmed through social needs and leaner's needs and literature researches were reviewed about the 2015 Revision Curriculum and virtual reality and confirmed the area of communication technology related to VR glasses. The communication process of information was selected as a communication technology area related to VR glass. In the design step, Based on the analysis stage, instructional objectives were set and instructional method and medium were selected. In the development step was developed instructional materials, what are teaching-learning guidance, power-point presentations, the paper for group learning activities, Individual portfolio, and using various convex lenses VR glasses example. As a result of 'VR glasses' using various convex lenses, the viewing angle was enlarged when the diameter of the convex lens was large, but it was difficult to quantify it. In the implementation step, the instructional materials developed for middle school students was applied. In the evaluation step, instructional materials were modified and improved through a students evaluation, experts evaluation.

Production of Transgenic Plants in Brassica napus Winter Cultivar 'Youngsan' (영산 유채를 이용한 형질전환체 생산)

  • To improve genetic transformation of Brassica napus winter cultivar 'Youngsan', factors influencing shoot regeneration and transformation from cotyledonary petioles were investigated. Shoot induction was enhanced in the combination of 0.5 mg/L NAA and 2~4 mg/L kinetin. Silver nitrate was essential for successful shoot regeneration, ranging from 5 to 9 mg/L. The addition of $GA_3$ promoted plant regeneration. Among the tested Agrobacterium strains, co-cultivation times, and antibiotic selection regimes, choice of appropriate Agrobacterium strain was the most critical factor for efficient transformation of B. napus cv. 'Youngsan'. The EHA105 succinamopine strain was the most efficient and the maximum transformation efficiency was 26.8%. Transgenic shoots were selected on 10 mg/L phosphinothricin (PPT) containing media. The transgenic plants expressing bar and gus genes were resistant for commercial herbicide "Basta" and stained with X-Gluc. Southern blot hybridization indicated that the presence of one to three gus gene copies per genome and inheritance of the gus gene into the $T_1$ generation.

  • The interest of ACT (Automated Container Terminal) is increasing because of expansion of container's transportation quantity, appearance of large-sized and. high-speed vessel, high labor cost of container terminal and the change of technical level. Therefore, the ACT had been developed in several ports in the world, such as ECT (Europe Combined Terminals) and CTA (Container Terminal Altenwerder). They have studied detailed technique for the operation of ACT. In Korea, it also has increased not only expansion and improvement of container terminal, but also necessity of ACT. Now, many projects related to the development of ACT are working in Korea. And the technical improvement of operating system, automated equipments and information system has accomplished by the projects. The ACT consists of the basic equipment and the integrated information system for operating and controlling automated equipments. The productivity of ACT is maximized through efficient connection between them. Thus, the automated degree of container terminal is dependent on the developing time of operating system, reliability, transportation quantity of container, investment amount and technical level. In this paper, we propose important strategy in developing ACT through analyzing the property of each ACT.

Synthesis and Biological Activity Test of the Sex Pheromone of the Summer Fruit Tortrix Moth (애모무늬잎말이나방의 성 페로몬 합성과 생물활성 시험)

  • Synthesis and biological activity test are described for the (Z)-9-tetradecen-1-yl acetate(1) and (Z)-11-tetradecen-1-yl acetate(1) and (Z)-11-tetradecen-1-yl acetate(2), the sex pheromone of the summer fruit tortrix moth, Adoxophyes orana. 8-Bromoctan-l-ol THP ether was prepared from 8-bromoctan-l-ol. The lithium anion of 1-hexyne was alkylated with 8-bromoctan-l-ol THP ether gave 9-tetradecyn-l-ol THP ether. Catalytic hydrogenation over Pd/BaSO4 followed by deprotection afforded (Z)-9-tetradecen-l-ol. Acetylation gave (Z)-9-tetradecen-1-yl acetate(1). l0-Bromodecan-l-ol THP ether was obtained from l0-bromodecan-l-ol. In liquid ammonia with THF and HMPA as cosolvents, sodium acetylide could be alkylated with 10-bromodecan-l-ol THP ether to give 11-dodecyn-l-ol THP ether. 11-Dodecyn-l-ol THP ether was then treated with n-BuLi in THF to give the lithium acetylide, which was alkylated with bromoethane to afford 11-tetradecyn-l-ol THP ether. Catalytic hydrogenation, deprotection, and acetylative gave (Z)-11-tetradecen-1-yl acetate(2). The synthetic pheromone thus obtained was attractive to the males of the summer fruit tortrix in the field.
