Carbohydrate antigens expressed on pig cells are considered to be major barriers in pig-to-human xenotransplantation. Even after ${\alpha}1,3$-galactosyltransferase gene knock-out (GalT-KO) pigs are generated, potential non-Gal antigens are still existed. However, to the best of our knowledge there is no extensive study analyzing N-glycans expressed on the GalT-KO pig tissues or cells. Here, we identified and quantified totally 47 N-glycans from wild-type (WT) and GalT-KO pig fibroblasts using mass spectrometry. First, our results confirmed the absence of galactose-alpha-1,3-galactose (${\alpha}$-Gal) residue in the GalT-KO pig cells. Interestingly, we showed that the level of overall fucosylated N-glycans from GalT-KO pig fibroblasts is much higher than from WT pig fibroblasts. Moreover, the relative quantity of the N-glycolylneuraminic acid (NeuGc) antigen is slightly higher in the GalT-KO pigs. Thus, this study will contribute to a better understanding of cellular glycan alterations on GalT-KO pigs for successful xenotransplantation.

This experiment was conducted to investigate the effects of pig skin collagen supplementation on growth performance, organ weight, blood characteristics, and intestinal microbiota in broilers. A total of 50 Ross 308 broilers were used for 2 weeks. The five dietary treatments were as follows: NC) basal diet, PC) NC + fish collagen powder 0.1%, T1) NC + pig skin collagen 0.1%, T2) NC + pig skin collagen 0.5%, and T3) NC + pig skin collagen 1.0%. The body weight gain (BWG), feed intake (FI), and feed conversion ratio (FCR) were not affected (p > 0.05) by the dietary treatments in this experiment. Additionally, there were no significant differences (p > 0.05) in the organ weights among the treatments. Broilers fed T1, T2 and T3 diets had higher (p < 0.05) white blood cell (WBC) counts than the broilers fed the NC and PC diets. The Lactobacillus counts in the excreta were improved (p < 0.05) in the broilers fed the T1 and T2 diets. Moreover, the Salmonella counts in the excreta were decreased (p < 0.05) in the broilers fed the PC and T1 diets. In conclusion, supplementation of pig skin collagen in diets improved the white blood cells (WBCs) in the blood and Lactobacillus counts in the excreta, and reduced the Salmonella counts in the excreta. However, when pig skin collagen was increased in the diets, there were no significant differences (p > 0.05). Therefore, the addition of 0.1% pig skin collagen in the feed provided beneficial effects on the blood characteristics and the intestinal microbiota environment.
The purpose of this study was to evaluate the effect of lower numbers of sperm $(3{\times}10^9)$ per dose liquid semen and type of semen used in artificial insemination (AI) on sow reproductive performance in subtropical area. Semen was supplied by two commercial AI centers. A total of 671 female pigs from seven farms were inseminated with either $3{\times}10^9$ or $5{\times}10^9$ sperm per dose. Two types of semen were used: heterospermic semen from two boars of the same breed and homospermic semen from a single boar. After insemination, conception rate, farrowing rate, total litter size, and number of dead piglets were recorded. The analysis of variance indicated that there was no significant effect of interactions between pig farm, type of semen, or number of sperm on any of the traits measured. There were significant differences in conception rate, farrowing rate, and total litter size among pig farms (p<0.05). The effect of number of sperm per dose liquid semen ($3{\times}10^9$ or $5{\times}10^9$) was not significant. Sows inseminated with homospermic semen showed significantly higher conception and farrowing rates but significantly lower total litter size (p<0.05). In conclusion, the number of sperm per dose liquid semen for AI could be lowered to $3{\times}10^9 $ without affecting reproductive performance in subtropical areas like Taiwan.
From among the NDT (nondestructive testing) methods, the MFL (magnetic flux leakage) method is specially suitable for testing pipelines because pipeline has high magnetic permeability. The system applied to MFL method is called the MFL PIG. The previous MFL PIG showed high performance in detecting the metal loss and corrosions. However, MFL PIG is highly unlikely to detect the cracks which occur by exterior-interior pressure difference in pipelines and the shape of crack is long and very narrow. In MFL PIG, the magnetic field is performed axially and there is no changes of cross-sectional area at cracks that the magnetic field passes through. Cracks occur frequently in the pipelines and the risk of the accident from the cracks is higher than that from the metal loss and corrosions. Therefore, the new PIG is needed to be researched and developed for detecting the cracks. The circumferential MFL (CMFL) PIG performs magnetic fields circumferentially and can maximize the magnetic flux leakage at the cracks. In this paper, CMFL PIG is designed and the distribution of the magnetic fields is analyzed by using 3 dimensional nonlinear finite element method (FEM). In CMFL PIG, cracks, standards of NACE, are detectable. To estimate the shape of crack, the leakage of magnetic fields for many kinds of cracks is analyzed and the method is developed by signal processing.

Objective: The objectives were to demonstrate that the nitrogen and energy in pig urine supplemented with hydrochloric acid (HCl) are not volatilized and to determine the minimum amount of HCl required for nitrogen preservation from pig urine. Methods: In Exp. 1, urine samples of 3.0 L each with 5 different nitrogen concentrations were divided into 2 groups: 1.5 L of urine added with i) 100 mL of distilled water or ii) 100 mL of 6 N HCl. The urine in open plastic containers was placed on a laboratory table at room temperature for 10 d. The weight, nitrogen concentration, and gross energy concentration of the urine samples were determined every 2 d. In Exp. 2, three urine samples with different nitrogen concentrations were added with different amounts of 6 N HCl to obtain varying pH values. All urine samples were placed on a laboratory table for 5 d followed by nitrogen analysis. Results: Nitrogen amounts in urine supplemented with distilled water decreased linearly with time, whereas those supplemented with 6 N HCl remained constant. Based on the linear broken-line analysis, nitrogen was not volatilized at a pH below 5.12 (standard error = 0.71 and p<0.01). In Exp. 3, an equation for determining the amount of 6 N HCl to preserve nitrogen in pig urine was developed: additional 6 N HCl (mL) to 100 mL of urine = 3.83×nitrogen in urine (g/100 mL)+0.71 with R2 = 0.96 and p<0.01. If 62.7 g/d of nitrogen is excreted, at least 240 mL of 6 N HCl should be added to the urine collection container. Conclusion: Nitrogen in pig urine is not volatilized at a pH below 5.12 at room temperature and the amount of 6 N HCl required for nitrogen preservation may be up to 240 mL per day for a 110-kg pig depending on urinary nitrogen excretion.

Global pig production has increased by 140% since the 1960s. The increase in global population, coupled with improving socioeconomic conditions of many countries has led to an increased consumption of meat globally, including pork. To keep up with demand and capitalize on economic opportunities, the countries of China, the United States (US), and the European Union (EU) have become the top 3 pork producers globally. China is of particular interest, as it is the both the largest country in pork production and pig numbers, as well as being the largest importer of pork from other countries. Globally, the efficiency of pork production has improved, in relation to the integration of pig production and the dramatic increase in research efforts in pig nutrition and production. Through integration, large producers can consolidate resources and maximize profits and efficiency. The increased research interest and efforts in pig production have given scientists and producers the opportunity to collaborate to adapt to challenges and identify possible solutions to issues brought on by a volatile global market. Intestinal health (23%), general nutrition and growth (23%), and amino acid nutrition (15%) were the top 3 areas (61%) leading research trends in pig nutrition and production. Major dietary interventions with feed additives evaluated include functional amino acids, feed enzymes, pre-/pro-/post-biotics, and phytobiotics with a common goal to improve the growth efficiency by enhancing nutrient utilization and intestinal health. With increasing global issues with environment, pig producers and the supporting scientists should continue their efforts to improve the production efficiency and to reduce the environmental footprint from pig production.
Cytogenetic and hematological analysis was performed in peripheral blood of pig in the vicinity of Yeonggwang nuclear power station and control area. The frequency of micronuclei (MN) in peripheral blood lymphocytes from pig was used as a biomarker of radiobiological effects resulting from exposure to environmental radiation. An estimated dose of radiation was calculated by a best fitting linear-quadratic model based on the radiation-induced MN formation from the swine lymphocytes exposed in vitro to radiation over the range from 0 Gy to 4 Gy. MN rates in lymphocytes of pig from Yeonggwang nuclear power station and control area were 10.60/1,000 and 11.10/1,000, respectively. There were no significant differences in MN frequencies and hematological values in pig between Yeonggwang and control area. The study indicates that the MN assay in lymphocyte of pig is a rapid, sensitive and accurate method that can be used to monitor a large population exposed to radiation.
The mammalian glycosylphosphatidylinositol (GPI) anchor consists of three mannoses attached to acylated GlcN-(acyl)PI to form $Man_3$-GlcN-(acyl)PI. The first of the three mannose groups is attached to an intermediate to generate Man-GlcN-(acyl)PI by the first mannosyltransferase (GPI-MT-I). Mammalian and protozoan GPI-MT-I have different substrate specificities. PIG-M encodes the mammalial GPI-MT-I which has 423 amino acids and multiple transmembrane domains. In this work we cloned PIG-M homologues from humans, Plasmodium falciparum (PfPIG-M), and Saccharomyces cerevisiae (GPI14), to test whether they could complement GPI-MT-I-deficient mammalian cells, since this biosynthetic step is likely to be a good target for selective screening of inhibitors against many pathogenic organisms. PfPIG-M partially restored cell surface expression of the GPI-anchored protein CD59 in PIG-M deficient mammalian cells, and first mannose transfer activity in vitro; however, this was not the case for GPI14.
This survey was conducted to collect basic data about the effect of buffering zone installation in weaned pig house. Buffering zone was installed either inside or outside of pig house to compare changes of temperature, humidity and air velocity of pig room. The body weight and mortality of weaned pigs in house with buffering zone was also measured. There was no difference in temperature, humidity and air velocity between inside and outside buffering zone. Mortality of weaned pig in house with buffering zone was drastically decreased compare to pigs in house without buffering zone that could be useful to maintain constant temperature and decrease mortality.
The purpose of this study is to develop the strain-specific PCR primers for the identification of prevotella inter-media ATCC 49046 which is frequently used in the pathogenesis studies of periodontitis. The Hind III-digested genomic DNA of P. intermedia ATCC 49046 were cloned by random cloning method. The specificity of cloned DNA fragments were determined by Southern blot analysis. The nucleotide sequence of cloned DNA probes was determined by chain termination method. The PCR primers were designed based on the nucleotide sequence of cloned DNA fragment. The data showed that Pig6 DNA probe were hybridized with the genomic DNA from P. intermedia strains (ATCC $25611^T$ and 49046) isolated from the Westerns, not the strains isolated from Koreans. The Pig6 DNA probe were consisted of 813 bp. Pig6-F3 and Pig6-R3 primers, designed base on the nucleotide Sequences Of Pig6 DNA Probe, were 3150 specific to the only both P. intermedia ATCC $25611^T$ and P. intermedia ATCC 49046. In the other hand, Pig6-60F and Pig6-770R primers were specific to the only P. intermedia ATCC 49046. The two PCR primer sets could detect as little as 4 pg of chromosomal DNA of P. intermedia. These results indicate that Pig6-60F and Pig6-770R primers have proven useful for the identification of P. intermedia ATCC 49046, especially with regard to the maintenance of the strain.
