  • Purpose To investigate the physico-chemical properties of CB-ph [2-benzoyloxycinnamaldehyde], an anticancer drug obtained from Cinnamomum cassia using methylenechloride. and its stability in various aqueous solutions. Results CB-ph was rarely soluble in water but soluble in methanol and very soluble in ether. Kinetic salt effect on degradation of CB-ph in buffer solutions at pH 4.0 and 6$0^{\circ}C$ showed a linear relationship having a positive slope that means reactions between hydronium ions and protonated substrates. Anti-inflammatory Effect of Portulacae Herba Water Extract on Lipopolysaccharide-activated RAW 264.7 Macrophages (마치현(馬齒莧) 물추출물의 항염효능에 관한 연구)

  • Objectives : The present study aimed to investigate the anti-inflammatory effects of the water extracts of Portulacae Herba (PH).Methods : We measured the effects of the water extracts of Portulacae Herba (PH) on the cell viability of mouse macrophage RAW 264.7 cells, the intracellular calcium production, and the proinflammatory mediators including nitric oxide (NO), vascular endothelial growth factor (VEGF), platelet-derived growth factor (PDGF)-BB, which are induced by the lipopolysaccharides (LPS), and obtained the results shown below.Results : After the cultivation of the PH extracts along with the mouse macrophages, the cell survival rate did not decrease with the MTT assay. However, the PH extracts did significantly suppress the production of NO by the mouse macrophages induced by LPS at the concentrations of 25, 50 and 100 ㎍/mL. The PH extract also significantly suppressed the VEGF, PDGF-BB and intracellular calcium production of the mouse macrophages by LPS at concentrations of 25, 50 and 100 ㎍/mL. As shown in the results above, the PH extracts do not have a toxic effect on the macrophages, but still have an anti-inflammatory effect that significantly reduces the intracellular calcium production as well as the production of NO, VEGF and PDGF-BB at concentrations above 25 ㎍/mL.Conclusions : In conclusion, the inhibitory anti-inflammatory effects of the PH extract can be used for a new treatment of anti-inflammatory diseases.

Effects of Different Dietary Fats and Fibers on the Lipid Concentrations of Liver and Serum and Biochemical Index in Rats (흰쥐의 간장 및 혈청 지질농도와 임상생화학적 지표에 미치는 식이섬유 및 식이지방 급원을 달리한 식이 투여의 영향)

  • This study investigated the effects of different types of dietary fats and fibers on the concentrations of serum and liver lipids and biochemical index in male Sprague-Dawley rats for 6 weeks. The experimental groups were divided into three groups; 5% corn oil +5% cellulose (CO group),10% corn oil +10% psyllium husk (CO +PH group) and 10% lard +10% psyllium husk (LA+ PH group). Body weight gain and FER (food efficiency ratio%) decreased significantly in the LA+PH group and decreased slightly in the CO+PH group compared with the CO group. Food intake and tissue weights were not significantly different among the dietary groups. Concentrations of serum total cholesterol decreased significantly in the CO + PH group compared with the CO group, but no significant different in the LA + PH group. Concentrations of serum triglyceride and phospholipid decreased significantly in the CO+PH and the LA+PH groups compared with the CO group. Liver cholesterol concentration was significantly decreased in the LA+PH group compared with the CO and the CO+PH groups. Glutamic pyruvic transaminase activity was significantly increased in the LA + PH group compared with the CO and the CO + PH groups. However, lactate dehydrogenase and blood urea nitrogen was significantly decreased in the LA+ PH group compared with the CO and the CO +PH groups. The others biochemical parameters, total protein, albumin, glucose, glutamic oxaloacetic transaminase, alkaline phosphatase were not significantly different among the dietary groups. These results showed that the lowering effect of dietary psyllium husk on serum lipid concentrations was more pronounced in the corn oil -feeding group than in the lard-feeding group as dietary fat source.

  • Cruciferous vegetables including diindolylmethane (DIM) have been shown to have anticancer activity. Especially, DIM-pPhBr and DIM-pPhF used in this study was reported to have more effective and less toxic effects than DIM. However, there is no report presenting their anti-tumorigenic activity in oral cancer. In the present study, we examined the effects of DIM-pPhBr and DIM-pPhF on the cell proliferation and apoptosis in KB human oral cancer cells. DIM-pPhBr and DIM-pPhF decreased cell proliferation and induced apoptosis evidenced by western blot analysis, DAPI staining and sub-$G_1$ population. This provides the first evidence that DIM-pPhBr and DIM-pPhF originating from Cruciferous vegetables induce apoptotic cell death in human oral cancer cells to inhibit cancer cell proliferation.

  • Phospholipase D2 (PLD2) has been implicated in the tyrosine kinase-mediated signaling pathways, but the regulation events are yet to be identified. Herein, we demonstrate that pleckstrin homology (PH) domain of PLD2 (PLD2-PH) exerts an antitumorigenic effect via the suppression of PLD2 and focal adhesion kinase (FAK). The kinase domain of FAK interacts with PLD2-PH and induces tyrosine phosphorylation and activation of PLD2. Furthermore, PLD2 increased tyrosine phosphorylation of FAK. However, ectopic expression of the PLD2-PH competes for binding to FAK and reduces the interaction between PLD2 and FAK, thereby suppressing FAK-induced PLD activation and tyrosine phosphorylation of FAK. The PLD2-PH suppressed the migration and invasion of glioblastoma cells, as well as tumor formation in a xenograft mouse model. This study uncovers a novel role of PLD2-PH as a negative regulator of PLD2 and FAK.

  • The chemical shifts in the $^{13}C$ NMR spectra of 2,3,4,5-tetraphenyl-1-silacyclopentdienide dianion $[SiC_4Ph_4]^{2-}{\cdot}2[K^+]$ (3) and 2,3,4,5-tetraphenyl-1-germacyclopentdienide dianion $[GeC_4Ph_4]^{2-}{\cdot}2[K^+]$ (4) were compared to those of $[SiC_4Ph_4]^{2-}{\cdot}2[Li^+]$ (5), $[SiC_4Ph_4]^{2-}{\cdot}2[Na^+]$ (6), and $[GeC_4Ph_4]^{2-}{\cdot}2[Li^+]$ (7). The average polarizations in two phenyl groups of two potassium salts are decreased over 15% to 20% comparing to those of the lithium salts and sodium salt {$[EC_4Ph_4]^{2-}{\cdot}2[M^+]$ (E=Si, Ge, M=Li, Na) due to the effect of the counter potassium cation.

Effect of Laser Beam Diameter on the Microstructure and Hardness of 17-4 PH Stainless Steel Additively Manufactured by Direct Energy Deposition (레이저 빔 직경 변화에 따른 17-4 PH 스테인리스 강 DED 적층 조형체의 미세조직 및 경도 변화)

  • The effect of the laser beam diameter on the microstructure and hardness of 17-4 PH stainless steel manufactured via the directed energy deposition process is investigated. The pore size and area fraction are much lower using a laser beam diameter of 1.0 mm compared with those observed using a laser beam diameter of 1.8 mm. Additionally, using a relatively larger beam diameter results in pores in the form of incomplete melting. Martensite and retained austenite are observed under both conditions. A smaller width of the weld track and overlapping area are observed in the sample fabricated with a 1.0 mm beam diameter. This difference appears to be mainly caused by the energy density based on the variation in the beam diameter. The sample prepared with a beam diameter of 1.0 mm had a higher hardness near the substrate than that prepared with a 1.8 mm beam diameter, which may be influenced by the degree of melt mixing between the 17-4 PH metal powder and carbon steel substrate.

Comparison of Ablation Characteristics of Carbon Nanotube reinforced Hybrid Al2O3 by using Ultrashort Pulse Laser (순수 알루미나와 탄소나노튜브 강화 알루미나 복합체의 극초단 펄스레이저 가공특성 비교)

  • In this paper, pure $I_{ph}$ and hybrid carbon nanotube reinforced $I_{ph}$ were sintered using the SPS(spark plasma sintering) method for high densification. A nanosecond laser (${\lambda}=1063nm$, ${\tau}P=10ns$) and a femtosecond laser (${\lambda}=1027nm$, ${\tau}P=380fs$) were installed on an optical system for the micromachining test. The ablation characteristics of the pure $I_{ph}$ and CNT/$I_{ph}$ composites, such as thermal effect and ablation depth, were investigated using FE-SEM and a confocal microscope device. Laser machining results for the two mating materials showed improved performances: CNT/$I_{ph}$ composites showed good surface morphology of hole drilling without a melting zone due to the composites' high thermal properties; also, the ablated depth of CNT/$I_{ph}$ was higher than that of pure $I_{ph}$.

The Effect of Repetitive Compression with Constant Stress on the Compressive Properties of Foams (일정 응력 반복압축이 발포체의 압축 특성에 미치는 영향)

  • To study the compressive stress, recovery force and permanent strain of foams for footwear midsole, polyurethane(PU), phylon(PH) and injection phylon(IP) foams were repetitively compressed with constant compressive stress. Maximum compressive stress of PU did not decrease with repetitive compression on the constant compressive stress, but that of IP largely decreased. Engineering strain of foams were formed by repetitively compressing the three types of foam. The engineering strain of PU was smaller than that of IP and PH. Compressive stress and recovery force of IP and PH at certain strain were decreased with repetitive compression, but that of PU was not noticeably changed.
