Toward Complete Bacterial Genome Sequencing Through the Combined Use of Multiple Next-Generation Sequencing Platforms

  • PacBio's long-read sequencing technologies can be successfully used for a complete bacterial genome assembly using recently developed non-hybrid assemblers in the absence of second-generation, high-quality short reads. However, standardized procedures that take into account multiple pre-existing second-generation sequencing platforms are scarce. In addition to Illumina HiSeq and Ion Torrent PGM-based genome sequencing results derived from previous studies, we generated further sequencing data, including from the PacBio RS II platform, and applied various bioinformatics tools to obtain complete genome assemblies for five bacterial strains. Our approach revealed that the hierarchical genome assembly process (HGAP) non-hybrid assembler resulted in nearly complete assemblies at a moderate coverage of ~75x, but that different versions produced non-compatible results requiring post processing. The other two platforms further improved the PacBio assembly through scaffolding and a final error correction.

A Study of Laboratory Measurement of EO GRD Resolution for Airborne EO/IR Sensor (항공용 EO/IR 센서의 EO GRD 분해능 실험실 측정 연구)

  • EO GRD(Ground Resolved Distance) resolution of airborne EO/IR(Electro-Optical/Infrared) sensor is a critical factor in test and evaluation for EO sensor performance. We propose the laboratory measurement set-up for EO GRD by constructing optical collimator which includes integrated sphere, blackbody, equivalent 3-bar target and 6 DOF motion simulator. GRD is measured in the photographic imagery of bar targets by 3 different distances for 3 EO/IR sensors and the measured results were analyzed statistically. We found that at least 7 sheets of imagery are needed in order to obtain meaningful EO GRD. The result of statistical analysis shows that the distribution of the measured GRD is nearly symmetric about the average GRD, and the better imagery ratio above the average GRD is about 40~70%. Also from the best GRD analysis, it is estimated that the design goal for EO GRD should be 30% superior to the required GRD.

Effect of the Hole on the Tensile Fatigue Properties of CFRP Laminates

  • The current study assessed the effect of a bolt hole on tensile fatigue properties of CFRP laminates. Two specimens, i.e. $[(0/90)_3]S$, $[(0/45/90/-45)_2]_S$, were analyzed using a finite element method and were experimentally tested for cases, both with and without a hole, whose diameter corresponded to 0.12 times the specimen width. Delamination positions predicted by a 3-dimensional static finite element analysis were matched well to those observed by an ultrasonic imaging system in the middle of fatigue test. A hole whose diameter corresponds to 0.12 times the specimen width caused the fatigue strength to decrease by 9% and 11% under 5 Hz loading frequency, and by 22% and 25% under 10 Hz loading frequency for $[(0/90)_3]_S$ and $[(0/45/90/-45)_2]_S$, respectively. Because the decrease in sectional area due to the hole was normalized in calculation of the tensile strength, a stress concentration around the hole is believed to induce the strength degradation of fatigue specimens. From the finite element analyses, the stress concentration factor around a hole was expected as 8.8 and 9.5 for $[(0/90)_3]_S$ and $[(0/45/90/-45)_2]_S$, respectively.

  • Optimized conditions for Agrobacterium-mediated lisianthus pollen transformation were adjusted using various factors such as temperature, pH and sucrose concentration. Pollen tube growth was successfully achieved in a medium (pollen germination medium; PGM) containing $7-15\%$ sucrose with pH in the range of 5.5-7.0 at temperature of $20-27^{\circ}C$. Lisianthus pollen was vacuum-infiltrated with Agrobacterium cell suspension for 20 min, and transformed pollen was confirmed by GUS histochemistry and Southern hybridization following RT-PCR. Transgenic pollen system may be utilized for establishing an area of plant transient expression systems based on the convenient pollen transformation procedure presented in here.

Miniaturization of SIW-Based Linearly Polarized Slot Antennas for Software-Defined Radar

  • Two substrate integrated waveguide (SIW)-based antennas for the application of software-defined radar are proposed and investigated herein. It is usually well known that SIWs are easily integrated, lightweight, have low insertion loss, and low interference levels compared to conventional microstrip structures. The primary function of the proposed antennas is to transmit continuous waves for indoor motion detection, with the lowest amount of loss and an appropriate amount of gain. Moreover, the results of this study show that the size of the antenna can be reduced significantly (i.e., by about 40%) by applying a meander line structure. The operating frequencies of the proposed antennas are both within the industrial, scientific, and medical band (i.e., 2.4-2.4835 GHz). Measured results of return loss are -16 dB and -20 dB at 2.435 GHz and 2.43 GHz, respectively, and the measured gain is 8.2 dBi and 5.5 dBi, respectively. Antenna design and verification are undertaken through commercially available full electromagnetic software.

  • The ESD(electrostatic discharge) protection performance of PPS(PMOS pass structure) embedded N-type silicon controlled rectifier(NSCR_PPS) device with different partial p-well(PPW) structure was discussed for high voltage I/O applications. A conventional NSCR_PPS standard device shows typical SCR-like characteristics with low on-resistance, low snapback holding voltage and low thermal breakdown voltage, which may cause latch-up problem during normal operation. However, our proposed NSCR_PPS devices with modified PPW_PGM(primary gate middle) and optimal CPS(counter pocket source) implant demonstrate the stable ESD protection performance with high latch-up immunity.

  • Enemy information has significant value when it comes to the process of military actions in battle field. Our Army now uses Battlefield Management Systems(BMSs) equipped in Ground Fighting Vehicles(GFVs) and we need to make research on what kind of role enemy information can play in such systems. Also, enemy information can be shared among GFVs and target information shall be extracted from it in view of KVMF scheme. Because KVMF becomes requisite standard in modern BMSs, we need to implement target information handling process in KVMF standard. In this article, we will focus on how target information and its sharing process can be managed efficiently without information conflicts. Also, situation map produced by it will be noted.

  • Recently, the implementation & modern policy for Global Navigation Satellite System have actively been performed by USA, RUSSIA, EU & CHINA. Therefore 100+ navigation satellites will be in orbit by 2015, and the user of military and civil will benefit from the use of a total constellation of 100+ satellites. It means that the deepest dependence to GPS would be declined. In the paper, the latest technology development & implementation policy of GNSS have been analyzed. Specially, we focused on circumstance change of GNSS & application of navigation technology for modem weapon system. Finally, the application strategy of the integrated GNSS is suggested for military and civil as well.

Changes of CO, $CO_2$, TVOC and Aerosol of Tobacco Smoke in a Poorly-Ventilated Indoor (환기가 불량한 실내공간에서, 담배연기에 의한 CO, $CO_2$, TVOC 및 에어로졸의 변화)

  • Number of aerosol, CO, $CO_2$ and TVOC after one-, two-, three-cigarettes smoking were monitored with time every 10 minute for 180 minutes in the seminar room (volume $51.1m^3$) when poorly-ventilated. IAQ monitor (IAQRAE, model PGM-5210) and PortCount (TSI, model 8020) were used for monitoring. Aerosol was decreased with exponential decay equation and it was estimated that number of aerosol would be long suspended (one cigarette 75/cc. two cigarettes 66/cc, three cigarettes 141/cc by 8hrs after smoking). While CO was also decreased with exponential or linear decay equation and correlated with number of aerosol strongly, TVOC and $CO_2$ were increased with linear equation in accordance with time lag. Most of TVOC and $CO_2$ were above standard levels of Korean Indoor Air Quality (Ministry of Environment) without regarding number of cigarettes. When naturally ventilated, all of CO, $CO_2$ and TVOC concentrations were dramatically decreased below standard levels of Korean Indoor Air Quality.

BLOOD PROTEIN POLYMORPHISMS OF NATIVE AND JUNGLE FOWLS IN INDONESIA

  • In an attempt to reveal the interrelationship between fowls of jungle and native origin, their gene constitutions were compared using gene frequencies at the 16 loci controlling blood protein variations. Of the 16 loci analysed by electrophoresis, polymorphism was detected at following seven loci: Es-1, Amy-1, Akp-akp, Akp-2, Alb, Tf and 6-PGD. The other nine loci: Amy-3, Es-D, PGM, PHI, MDH, To, LDH, Hb-1 and Hb-2, were noted to be monomorphic. Genetic distance between pairs of native fowl and jungle fowls was estimated by a numerical taxonomic method. The Indonesian native fowl was genetically close to the Indonesian red jungle fowl, and the grey jungle fowl was genetically similar to the Ceylonese jungle fowl. It was also suggested that the green jungle fowl was genetically remote from the other jungle fowls and from the Indonesian native flow. The proportion of polymorphic loci (Ppoly), the expected average heterozygosity per individual $\bar{H}$, and the effective number of alleles per locus (Ne) were calculated to evaluate the genetic variabilities in the native and jungle fowls. The Indonesian native fowl exhibited slightly higher the proportion of polymorphic loci than the jungle fowls.
