한국재래닭 (오계) 원시생식세포의 완만동결과 급속동결의 비교 (Comparison of Vitrification and Slow Freezing for the Cryopreservation of Chicken Primordial Germ Cell (Ogye))

  • 동결 닭 PGCs의 생식계열 키메라를 이용한 생체에의 복원을 실용화 하기 위해서는, 닭 PGCs의 동결보존기술의 향상에 의해 동결 및 융해 후의 많은 생존세포를 확보 하는 것과, 생식계열 키메라의 제작효율을 높이는 것이 반드시 필요하다. 닭 PGCs는 배양 5.5일령의 닭 원시생식선으로부터 채취하고, ACS 방법에 의해서 순수 닭 PGCs를 분리했다. 닭 PGCs의 동결보존실험결과 다음의 4종류의 동결방법을 각각 비교 검토했다. 1. 플라스틱 스트로에 의한 완만동결법 (SF), 동결보호물질은 2M 에틸렌 글리콜 (EG), 2. 스트로에 의한 급속동결법 (RF), 8M EG + 7% PVP, 3. 동결용 Cryotube에 의한 SF, 2M EG, 4. 튜브에 의한 SF, 10% DMSO. 동결 및 융해 후의 PGCs의 생존율은 각각 76.4%, 70.6%, 80.5%, 78.1%로 관찰되었다.

자외선이 산개구리 (Rana dybowskii) embryo의 시원생식세포 수에 미치는 영향 (UV Effect on the Number of Primordial Germ Cells in the Embryo of Rana dybowskii)

  • 산개구리의 germ plasm 및 축결정 세포질에 미치는 자외선의 영향을 수량적, 시간적으로 조사하였다. 산개구리 난의 식물극을 수정 후 60분경에 1,600 $ergs/mm^2$이상의 자외선을 조사하였을때 40%이상의 시원생식세포의 수가 감소하였으나, 그 이하의 양에서는 전혀 영향이 없었다. 그러나 1,600 $ergs/mm^2$부터는 시원생식세포 수의 감소 정도는 자외선의 양에 비례하였다. 또 신경관 형성에 전혀 영향을 미치지 않으면서 시원생식세포의 형성을 완전히 억제시키는 자외선의 양은 4,800 $ergs/mm^2$이었다. 이에 4,800 $ergs/mm^2$의 자외선을 수정 후 일정한 시간간격으로 조사하여 germ plasm에 미치는 자외선의 영향을 조사하였다. 그 결과 자외선이 조사된 시기가 이를수록 시원생식세포수의 감소가 크며, 수정후 60분부터 차츰 감소율이 줄어 들다가 3분열 이후에는 자외선의 효과가 거의 없음을 관찰하였다. 또한 시원생식세포와 축형성에 미치는 자외선의 영향을 시기적으로 비교한 결과 산개구리난의 축결정이 다른 무미양서류에서와 같이 수정에서 제1분열 사이의 $0.7\\sim0.8$시기 이전에 이루어지므로 그 이후에는 자외선의 감응도가 급격히 저하되었다. 그러나 germ plasm의 자외선에 대한 감응도는 훨씬 늦게까지 지속되어 8구기에 이르기까지 그 영향이 나타났다.

AMPK/Sirt1/PGC-1α 신호 전달 경로의 조절을 통한 오미자 추출물의 비만 개선 효과 (Anti-Obesity Effect of Schizandrae Fructus Water Extract through Regulation of AMPK/Sirt1/PGC-1α signaling pathway)

  • Objectives : Although the anti-obesity effect of Schizandrae Fructus water extract has been demonstrated, its underlying mechanism is still unclear. Therefore, we aimed to evaluate the anti-obesity effect of Schizandrae Fructus water extract through the p-AMP-activated protein kinase (p-AMPK), sirtuin1 (Sirt1), and peroxisome proliferator-activated receptor-gamma coactivator 1α (PGC-1α) signaling in 60% high-fat diet (HFD)-induced obese mouse model. Methods : Male C57BL/6 mice were divided into four groups. The Normal group was fed a normal diet and the obese groups were fed 60% HFD. Except for the Control group, the GG group was supplemented with 0.5% Garcinia gummigutta and the SCW group was supplemented with 0.5% Schizandrae Fructus water extract. After 6 weeks, obesity-related biomarkers in serum were measured and the expressions of protein for lipid-related factors in liver tissue were analyzed by western blot. Results : Treatment with SCW significantly down-regulated body weight compared to the Control group. SCW down-regulated levels of triglyceride and total cholesterol in serum and significantly increased p-AMPK, Sirt1, and PGC-1α in liver tissue. In addition, the expressions of fatty acid oxidation-related proteins such as peroxisome proliferator-activated receptor α (PPARα), carnitine palmitoyltransferase 1A (CPT-1A), uncoupling protein 1 (UCP1), and uncoupling protein 3 (UCP3) were significantly up-regulated. However, fatty acid synthesis-related proteins including sterol regulatory element-binding protein-1 (SREBP-1), phospho-Acetyl-CoA Carboxylase (p-ACC), and fatty acid synthase (FAS) were significantly down-regulated. Conclusions : Taken together, SCW treatment showed anti-obesity effect by regulating both fatty acid oxidation-related and fatty acid synthesis-related proteins through AMPK/Sirt1/PGC-1α signaling in 60% HFD-induced obese mice.

  • 대부분의 항공우주 추진은 정압 Brayton 사이클에 기초하고 있으나 성능 향상을 위한 압력비 증가는 기계적 한계에 직면하고 있다. 지난 십여 년간 고속 추진에 적합한 연소방식으로 기대 받은 데토네이션 추진이, 최근에는 추진기관과 동력 장치의 열효율을 획기적으로 증대시켜 줄 수 있는 "game-changer"로 연구되고 있다. 즉, 데토네이션 파에 수반하는 강한 충격파의 압축 효과는 기존의 압축 방식에서 얻기 힘든 열효율을 증가시키는 것으로 여겨진다. 본 논문에서는 펄스데토네이션엔진의 최신 기술 동향과 더불어 정적연소에 기초한 압력증가연소 체계의 연구 동향에 대하여 소개할 것이다.

PGC-Enriched miRNAs Control Germ Cell Development

  • Non-coding microRNAs (miRNAs) regulate the translation of target messenger RNAs (mRNAs) involved in the growth and development of a variety of cells, including primordial germ cells (PGCs) which play an essential role in germ cell development. However, the target mRNAs and the regulatory networks influenced by miRNAs in PGCs remain unclear. Here, we demonstrate a novel miRNAs control PGC development through targeting mRNAs involved in various cellular pathways. We reveal the PGC-enriched expression patterns of nine miRNAs, including miR-10b, -18a, -93, -106b, -126-3p, -127, -181a, -181b, and -301, using miRNA expression analysis along with mRNA microarray analysis in PGCs, embryonic gonads, and postnatal testes. These miRNAs are highly expressed in PGCs, as demonstrated by Northern blotting, miRNA in situ hybridization assay, and miRNA qPCR analysis. This integrative study utilizing mRNA microarray analysis and miRNA target prediction demonstrates the regulatory networks through which these miRNAs regulate their potential target genes during PGC development. The elucidated networks of miRNAs disclose a coordinated molecular mechanism by which these miRNAs regulate distinct cellular pathways in PGCs that determine germ cell development.

Effects of exercise on AKT/PGC1-α/FOXO3a pathway and muscle atrophy in cisplatin-administered rat skeletal muscle

  • Cisplatin has been reported to cause side effects such as muscle wasting in humans and rodents. The physiological mechanisms involved in preventing muscle wasting, such as the regulation of AKT, PGC1-α, and autophagy-related factor FOXO3a by MuRF 1 and Atrogin-1, remain unclear following different types of exercise and in various skeletal muscle types. Eight-week-old male Wistar rats (n = 34) were assigned to one of four groups: control (CON, n = 6), cisplatin injection (1 mg/kg) without exercise (CC, n = 8), cisplatin (1 mg/kg) + resistance exercise (CRE, n = 9) group, and cisplatin (1 mg/kg) + aerobic exercise (CAE, n = 11). The CRE group performed progressive ladder exercise (starting with 10% of body weight on a 1-m ladder with 2-cm-interval grids, at 85°) for 8 weeks. The CAE group exercised by treadmill running (20 m/min for 60 min daily, 4 times/week) for 8 weeks. Compared with the CC group, the levels of the autophagy-related factors BNIP3, Beclin 1, LC3-II/I ratio, p62, and FOXO3a in the gastrocnemius and soleus muscles were significantly decreased in the CRE and CAE groups. The CRE and CAE groups further showed significantly decreased MuRF 1 and Atrogin-1 levels and increased phosphorylation of AKT, FOXO3a, and PGC1-α. These results suggest that both ladder and aerobic exercise directly affected muscle wasting by modulating the AKT/PGC1-α/FOXO3a signaling pathways regardless of the skeletal muscle type.

Effect of the Bifunctional Chelate on the Biodistribution of 99mTc-labeled Cyclic RGD Peptide

  • A novel $N_3S_1$ chelate, Pro-Lys-Cys (PKC) to cyclic RGD to radiolabel with $^{99m}Tc$ was conjugated in an effort to decrease the high intestinal accumulation observed for $^{99m}Tc$-labeled PGC-RGD. The target specificity of the resulting PKC-RGD was similar to that of PGC-RGD as determined by a cell binding assay and a competition binding assay. The $^{99m}Tc$ radiolabeling of PKC-RGD resulted in radiochemical yields of 98% under mild conditions at high specific activities. Biodistribution data in normal mice clearly showed a significant decrease in intestinal uptake at 2 h postinjection for the $^{99m}Tc-PKC-c$ (RGDyK) compared to the $^{99m}Tc-GC-c$ (RGDyK) (from $19.65%ID{\cdot}g^{-1}$ to $7.31%ID{\cdot}g^{-1}$ for the GI tract). The $^{99m}Tc-PKC-c$ (RGDyK) biodistribution was also shown by a higher retention of radioactivity in the whole body, but with kidney accumulation over 8-fold higher than observed with $^{99m}Tc-PGC-c$ (RGDyK) at 2 h ($12.62%ID{\cdot}g^{-1}$ for PKC-RGD and $1.54%ID{\cdot}g^{-1}$ for PGC-RGD, respectively). These results show that the biodistribution may be altered especially concerning lipophilicity resulting in renal rather than hepatobiliary excretion. This comparative study made it possible to explore the effects of lipophilicity on the biodistribution of $^{99m}Tc$-labeled c (RGDyK) through the use of different tripeptide $N_3S_1$ chelators. Therefore, $^{99m}Tc-PKC-c$ (RGDyK) may be an attractive alternative for the in vivo imaging of integrin receptors.

C2C12 골격근세포에서 황기의 미토콘드리아 조절 작용 (The Effects of Astragali Radix Extracts on Mitochondrial Function in C2C12 Myotubes)

  • Objective: The prevalence of metabolic syndrome and type 2 diabetes is increasing worldwide. Mitochondrial dysfunction is known to be involved in insulin resistance and obesity, researches have been increasing highly. Astragali Radix extract (ARE) or its main components have been shown to perform comparably to insulin by significantly reducing blood glucose levels in animal models however, the influence on mitochondrial dysfunction are not well understood. Methods: ARE (0.2, 0.5 and 1.0 mg/ml) or metformin (2.5 mM) were treated in C2C12 after 6 day-differentiation. The expressions of adenosine monophosphate (AMP)-activated protein kinase (AMPK) and phosphorylation AMPK, peroxisome proliferators-activated receptror ${\gamma}$ coactivator $1{\alpha}$ ($PGC1{\alpha}$), nuclear respiratory factors 1 (NRF1), mitochondrial transcription factor (Tfam) and myosin heavy chain were detected with western blotting or polymerase chain reaction analysis. The morphological changes were also investigated. Results: ARE dose dependently increased phosphorylation of AMPK and respectively activated mRNA expressions of $PGC1{\alpha}$, NRF1 and Tfam which are mitochondrial biogenesis regulators. Furthermore, there were some morphologic differences of differentiated cells between ARE treatment and control. Conclusions: This study suggests that ARE has the potential to increase muscle mitochondrial function by activating AMPK and $PGC1{\alpha}$.

RDE 기반 PGC 추진기관 연구 동향, Part I: 기초연구 (Research Activities on PGC Propulsion based on RDE, Part I: Basic Studies)

  • 데토네이션을 이용한 유체역학적 정적연소 기술은 현재의 정압연소 시스템의 효율 및 성능 한계를 극복할 수 있는 획기적 미래 추진 기술로 주목받고 있다. 지난 수년간 RDE의 기초적인 작동 실험과 CFD를 이용한 이론 분석 등 수많은 연구가 진행되었다. 본 논문에서는 아직 부족한 국내 RDE 연구에 참고가 되고자하는 목적으로 근래의 세계 각지에서 수행되고 있는 RDE 연구의 동향 및 핵심 기술에 대한 포괄적 고찰을 수행하였다.

THE WARPED DISK OF INTEGRAL-SIGN GALAXY PGC 20348

  • We examine the morphology and luminosity distribution of a strongly warped spiral galaxy PGC 20348 by conducting a detailed BVI CCD surface photometry using BOAO 1.8m telescope. The radial surface brightness shows a break at warp radius $(r_{\omega})$ with a shallow gradient in the inner disk and a steeper gradient in the outer disk. The luminosity of east side of the disk is ${\sim}0.5$ mag fainter than the west side at r > $r_{\omega}$. The reason for the asymmetric luminosity distribution is thought to be the asymmetric flarings that result in the formation of a large diffuse region at the edge of the east disk and a smaller diffuse region at the west disk. The vertical luminosity profiles show a thick disk component whose scale heights increase with increasing galactocentric distances. The warp of PGC 20348 seems to be made by the tidal interactions with the two massive companion galaxies since the flarings and radial increase of disk scale heights are thought to be general properties of tidally perturbed disks. According to the colors of the two clumps inside the diffuse region at the edge of the east disk, they seem to be sites of active star formation triggered by tidal forces from the companion galaxies.
