Relative Effectiveness of Some Antioxidants in A Dark and A Sunlight-irradiated Condition (암소저장(暗所貯藏) 및 일사광선조사(日射光線照射) 조건하(條件下)에서의 일부(一部) 항산화제(抗酸化劑)의 항산화효과에 대하여)

  • Relative retarding effect of BHA, BHT, PG, ${\alpha}-tocopherol$, ascorbic acid, pyrogallol, and antage 3C on the peroxide value and the free fatty acid value development of two groups of edible soybean oils was studied. The antioxidants were added respectively to the oils at a level of 0.05%, and one group of the oils was irradiated, 4 hours daily, with direct sunlight and the other group was stored in a dak place at $45{\pm}0.5^{\circ}C$. The retarding effect of the antioxidants on the P. V. development was, in general, more pronounced in case of the oils stored in the dark place than in case of the irradiated oils. BHT, PG, and antage 3C exhibited, in both cases, strong retarding effect on the P. V. development of the oils. In both cases, ${\alpha}-tocopherol$ showed some retarding effect, but the effect decreased rapidly as storage time increased. The inhibitory effect of the antioxidants on the free fatty acid value development was much more pronounced in case of the irradiated oils than in case of the oils stored in the dark place. The inhibitory effect of pyrogallol on the free fatty acid value development of the oils was, in both cases, especially strong and lasting.

Anti-inflammatory and Anti-allergic Effects of Herbal Extracts on Atopic Dermatitis(Part I) (약용식물추출물의 아토피성피부염에 대한 항염증 및 항알레르기효과(제 1보))

  • Atopic dermatitis is a chronic inflammatory skin disease associated with cutaneous hyper-reactivity to environmental triggers. In order to develop effective therapeutic herbal extracts for atopic dermatitis, cytotoxicity, antioxidant, anti-inflammatory and anti-allergic activities were investigated for various herbal extracts. Among candidate extracts, we selected Aloe vera L. (AV), Viola mandshurica W. Becker (VM), Punica granatum L. (PG), Dendrobium nobile L. (DN) and mixture of the above extracts (MX) for further investigations. All of them did not show cytotoxic activities to macrophage RAW264.7 cells below the concentration of 100 ppm. All showed antioxidant, anti-inflammatory and anti-allergic effects, although to various extents. In antioxidant effects, AV showed the highest effect, followed by PG and VM, while DN did the lowest. In evaluation for anti-inflammatory activities in macrophage RAW264.7 cells, AV and DN inhibited almost completely the LPS-induced production of nitric oxide, while AV, DN and VM showed strong inhibitory activities on the LPS-induced production of TNF-${\alpha}$. In anti-allergy effect in mast cell HMC-1, DN showed the highest effect, followed by AV and PG, while VM did the lowest. In the topical allergy reaction induced by compound 48/80 in Sprague-Dawley rat, DN exhibited significant anti-allergic effect, while PG, VM and AV did slight effect. These results suggest that AV, VM, PG and DN have antioxidant, anti-inflammatory and anti-allergic activities, and thus have the potential to reduce and alleviate the symptoms of atopic dermatitis.

Changes in Pectic Enzyme Activity during Maturation of Tomato Fruit and Purification, Some Properties of Polygalacturonase Isozymes (Tomato 성숙중(成熟中) Pectin 효소(酵素)의 활성변화(活性變化)와 Polygalacturonase의 분리(分離) 및 그 성질(性質))

  • The changes in pectinesterase (PE) and polygalacturonase (PG) activities were investigated during maturation and ripening of tomato fruits. PG was isolated from ripe tomatoes and purified by using DEAE-sephadex A-50 column chromatography. Its property was examined by general methods. PE activity was increased during maturation and then peaked at the on set of color change. PG activity was not detectable in mature green tomato fruits but increased during softening. Slab electrophoresis of the crude enzyme isolated from ripe tomatoes shoed 6 bands. Two polygalacturonase (PGI, PGII) were separated from crude enzyme of ripe tomatoes by chromatography on DEAE-sephadex A-50. PGI and PGII were purified by 61 fold and 98 fold by the present procedure, respectively. The Rm values of partially purified PGI and PGII were estimated to be 0.25 and 0.31, respectively. The optimum temp, and pH of PGI activity were $37^{\circ}C$ and pH 4.5, while those of PGII activity were $40^{\circ}C$ and pH 4.7, respectively. Isozyme PGI activity was the most stable at pH 4.3 and retained 50% of its activity when exposed at $78^{\circ}C$ for 5 min.
