Protective Effect of Perilla frutescens Extract against Oxidative Stress-Induced Cell Death in a Staurosporine-Differentiated Retinal Ganglion Cell Line (Staurosporine에 의해 분화된 망막신경절세포에서 산화 스트레스 유도 세포사멸에 대한 차조기 추출물의 보호 효능)

  • In this study, we examined the effect of Perilla frutescens extract (PFE) on oxidative stress-induced cell death in RGC-5 cell lines. Staurosporine-differentiated RGC-5 (ssdRGC-5) cells obtained by treating RGC-5 cells with $1{\mu}M$ staurosporine were incubated with PFE for 30 min and then exposed to buthionine sulfoximine plus glutamate (B/G) for 20 h. Cell death was detected using lactate dehydrogenase release assay and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide reduction assay. To investigate the mechanism underlying cell death, we determined caspase-3 activity, level of reactive oxygen species (ROS) formation, and expression levels of cytoplasmic cytochrome c and mitochondrial Bax. Treatment of ssdRGC-5 cells with B/G increased intracellular ROS and induced apoptosis with increasing caspase-3 activity. PFE rescued ssdRGC-5 cells from oxidative stress-induced cell death by inhibiting intracellular ROS production and caspase-3 activation and regulating apoptosis-related proteins such as cytochrome c and Bax. These findings suggest that PFE may have a beneficial neuroprotective effect against oxidative stress-induced apoptotic death in ssdRGC-5 cells.

Effect of Dietary Polyunsaturated Fatty Acid and $\omega$-Tocopherol on Lipid Peroxidation in Rat Liver (식이 불포화지방산과 Vitamin E 함량이 흰쥐 간장내의 지질과산화에 미치는 영향)

  • To study effect of dietary polyunsaturated fatty acid and $\omega$-tocopherol content on lipid peroxidation in rat liver, rats were fed for 3, 6 and 9 weeks with normal tocopherol diet added 40mg of DL-$\alpha$-tocopherol/kg of diet (PF group), high tocopherol eit 200mg of DL-$\alpha$-tocopherol/kg of diet(PFE group), low tocopherol diet without addition of DL-$\alpha$-tocopherol to diet(PFO group), and control diet added 40mg of DL-$\alpha$-tocopherol/kg of diet(control group). Each diet group supplied 45% of total calorie from corn oil except control group which supplied 12% of total calorie from corn oil. After each feeding period, lipid peroxide and tocopherol contents were measured in the liver as well as activities of glutathione peroxidase and superoxide dismutase. PF group had almost the same contents of liver peroxide, tocopherol contents and activities of glutathione peroxidease and superoxide dimutase as control group, while PFE group had higher tocopherol content and glutathione peroxidase activity and lower lipid peroxide contents and superoxide dismutase activity than control group. On the other hand, changes in the values of PFO group were opposite to those of PFE group. Differences in the values among groups were more pronounced as feeding period became longer.

Simultaneous Determination of Organo-phosphorus and Organo-chlorinated Pesticides in Soil and Lawn Samples of Golf Courses (골프장 토양 및 잔디 시료 중 유기인 및 유기염소계 농약의 동시 분석)

  • Organo-chlorinated pesticides have been seldom measured in Korea because their use was banned for more than two decades. Nevertheless, the environmental persistency of these pesticides requires regular monitoring. Only organo-phosphorus pesticides are currently being monitored in Korea. Simultaneous determination of organo-chlorinated and organo-phosphorus pesticides was attempted using gas chromatography/mass spectrometer detector. Pressurized fluid extraction(PFE) was used in sample preparation. PFE showed equivalent extraction efficiency to traditional soxhlet extraction with less solvent use and less sample preparation time. Average recovery of spiked pesticides to soil and lawn samples was 62% ranging 52 to 154%. Method detection levels(MDLs) for these pesticides ranged from 3.4 to 10.5 ${\mu}g/kg$. Endosulfan, fenitrothion, and diazinon were found in the samples from 5 different golf courses out of 9 courses.

  • This study proposed an expanded technology acceptance model (TAM) incorporating perceived fee (PFE) and perceived enjoyment into the original TAM We examined that the extended TAM provides supplementary information about behavior intentions (BI) toward mobile data services (MDS). This study also investigated some significant differences in the relative influence of the determinants of BI toward MDS depending on experience. The extended TAM was empirically evaluated by using survey data collected from 149 inexperienced users and 393 experienced users. The findings indicated that PFE is the strongest determinant of BI toward MDS, and the antecedents leading to users' BI vary over time. The results may provide further insights into MDS practitioners and marketing managers.

Inhibition of Proinflammatory Cytokine Generation in Lung Inflammation by the Leaves of Perilla frutescens and Its Constituents

  • This study was designed to find some potential natural products and/or constituents inhibiting proinflammatory cytokine generation in lung inflammation, since cytokines such as tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and interleukin-6 (IL-6) are pivotal for provoking airway inflammation. In our preliminary screening procedure, the 70% ethanol extract of the leaves of Perilla frutescens (PFE) was found to clearly inhibit TNF-${\alpha}$ production in the lung at 100 mg/kg, after intranasal lipopolysaccharide treatment of mice. Based on this result, ten constituents including phenylpropanoids (allyltetramethoxybenzene, caffeic acid, dillapiole, elemicin, myristicin, nothoapiole, rosmarinic acid methyl ester, rosmarinic acid) and monoterpenes (perilla aldehyde and perilla ketone) were successfully isolated from the extract. Among them, elemicin and myristicin were found for the first time to concentration-dependently inhibit IL-$1{\beta}$-treated IL-6 production from lung alveolar epithelial cells (A549) at concentrations of $10-100{\mu}M$. These findings suggest that the phenylpropanoids including elemicin and myristicin have the potential to be new inhibitory agents against lung inflammation and they may contribute, at least in part, to the inhibitory activity of PFE on the lung inflammatory response.

  • On this study, as a Human Factor(HF)'s point of view, typical PVI(Pilot Vehicle Interface) evaluation for small airplane cockpit design, PVI evaluation items and procedures to show compliance to KAS Part 23 "Airworthiness Standards : Normal, Utility, Acrobatic, And Commuter Category Airplanes" are introduced and reviewed. This study is based on the experience of Korean small civil airplane development and certification program.

Anti-Adipogenic Effects of Ethanol Extracts Prepared from Selected Medicinal Herbs in 3T3-L1 Cells

  • Obesity is a major risk factor for various metabolic diseases such as cardiovascular disease, hypertension, and type 2 diabetes mellitus. In this study, we prepared ethanol extracts from Agastache rugosa (ARE), Chrysanthemum zawadskii (CZE), Mentha arvensis (MAE), Perilla frutescens (PFE), Leonurus sibiricus (LSE), Gardenia jasminoides (GJE), and Lycopus coreanus (LCE). The anti-oxidant and anti-adipogenic effects were evaluated. The $IC_{50}$ values for ascorbic acid and LCE against 2,2-diphenyl-1-picrylhydrazyl radicals were $246.2{\mu}g/mL$ and $166.2{\mu}g/mL$, respectively, followed by ARE ($186.6{\mu}g/mL$), CZE ($198.6{\mu}g/mL$), MAE ($337.1{\mu}g/mL$), PFE ($415.3{\mu}g/mL$), LSE ($548.2{\mu}g/mL$), and GJE ($626.3{\mu}g/mL$). In non-toxic concentration ranges, CZE had a strong inhibitory effect against 3T3-L1 adipogenes (84.5%) than those of the other extracts. Furthermore, the anti-adipogenic effect of CZE is largely limited in the early stage of adipogenesis, and we revealed that the inhibitory role of CZE in adipogenesis is required for the activation of Wnt signaling. Our results provide scientific evidence that the anti-adipogenic effect of CZE can be applied as an ingredient for the development of functional foods and nutri-cosmetics for obesity prevention.

Analytical Validation of Rosmarinic Acid in Water Extract of Perilla frutescens Britton var. acuta Kudo as Functional Health Ingredient (건강기능식품 기능성 원료로써 장흥 차조기 열수 추출물의 지표성분인 로즈마린산 분석법 검증)

  • This study attempted to establish an HPLC analysis method for determination of marker compounds as a part of material standardization for the development of health functional food materials from Perilla frutescens Britton var. acuta Kudo. The quantitative determination method of rosmarinic acid as a marker compound of P. frutescens Britton var. acuta Kudo extract (PFE) was optimized by HPLC analysis using a C18 column ($4.6{\times}150mm$, $5{\mu}m$) with 0.1% acetic acid as the elution gradient and methanol as the mobile phase at a flow rate of 1 mL/min and detection wavelength of 280 nm. The HPLC/UV method was applied successfully to quantification of the marker compound in PFE after validation of the method with linearity, accuracy, and precision. The method showed high linearity in the calibration curve at a coefficient of correlation ($R^2$) of 0.9995, and the limit of detection and limit of quantitation were $0.36{\mu}g/mL$ and $1.2{\mu}g/mL$, respectively. Relative standard deviation (RSD) values of data from intra- and inter-day precision were less than 3.21% and 1.43%, respectively. Recovery rate test at rosmarinic acid concentrations of 12.5, 25 and $50{\mu}g/mL$ scored between 97.04~98.98% with RSD values from 0.25~1.97%. These results indicate that the established HPLC method is very useful for the determination of marker compound in PFE to develop a health functional material.

Effects of Perilla frutescens Extract on Anti-allergic Reactions in a Mouse Model (소엽 추출물이 마우스모델에서 항알레르기 반응에 미치는 영향)

  • We investigated the effect of Perilla frutescens (PF) ethanol extract powder (PF-E30) on the local allergic reaction activated by anti-DNP IgE and the mast cell-mediated immediate-type allergic reactions induced by compound 48/80 in a mouse model. One gram of PF powder extracted with 30% ethanol at $80^{\circ}C$ contained 12.3 mg of rosmarinic acid. Oral administration of PF-E30 (0.1 to 0.5 mg/kg body weight) significantly reduced plasma histamine levels and inhibited histamine release from peritoneal mast cells in mice activated by compound 48/80 or anti-DNP IgE. Moreover PF-E30 dose-dependently inhibited the production of antigen-induced IgE. These results indicate that the PF ethanol extract inhibits mast cell-mediated immediate-type allergic reactions in vivo and in vitro.

Geophysical Logging of Frequency-domain Induced Polarization for Mineral Exploration (광물탐사를 위한 진동수영역 유도분극 물리검층)

  • Induced polarization (IP) is useful for mineral exploration and hydrogeological studies by visualizing the electrochemical reactions at the interface between polarized minerals and groundwater. Frequency-domain IP (FDIP) is not actively applied to field surveys because it takes longer to acquire data, despite its higher data quality than conventional time-domain IP. However, data quality is more important in current mineral exploration as the targets gradually shift to deep or low-grade ore bodies. In addition, the measurement time reduced by automated instrumentation increases the potential for FDIP field applications. Therefore, we demonstrate that FDIP can detect mineral exploration targets by performing geophysical logging in the boreholes of a skarn deposit, in South Korea. Alternating current (AC) resistivity, percent frequency effect (PFE) and metal factor (MF) were calculated from impedance values obtained at two different frequencies. Skarn zones containing magnetite or pyrite showed relatively low AC resistivity, high PFE, and high MF compared to other zones. Therefore, FDIP surveys are considered to be useful for mineral exploration.
