  • The study was conducted in order to get basic information on seed germination and seedling growth through the experiments such as basal medium, concns of auxin, sucrose agar and pH values. Seed germination was very excellent in $3g/{\ell}$ hyponex and $5g/{\ell}$ peptone containing medium. Multiple shoots were formed in $1-3g/{\ell}$ hyponex and $5g/{\ell}$ pepton containg medium supplemented with $1.0g/{\ell}$ IBA, $30g/{\ell}$ sucrose and $8g/{\ell}$ agar. Growth of seedlings was more stimulated in the same amount of hyponex and peptone as above medium substituted with $40g/{\ell}$ sucrose and $6g/{\ell}$ agar without IBA.

  • Nutritional characteristics and cultural conditions for trehalose synthesis and mycelial yield of Pleurotus sajor-caju and Pleurotus ostreatus 201 in submerged culture were investigated. The results were as follows: Among the carbon sources, glucose was most excellent for trehalose synthesis and mycelial yield. The optimal concentration of glucose was 1%. Among the nitrogen sources, peptone was most excellent for trehalose synthesis and mycelial yield. The optimal concentration of peptone was 0.05%. The optimal concentration of $KH_2PO_4$ and $MgSO_47H_2O$ for trehalose synthesis and mycelial yield was 0.1%,0.04% and 0.2%,0.04-0.08%, respective­ly. The optimal temperature and pH for trehalose synthesis were $25^{\circ}C$ and pH 5.5. but optimal temperature and pH for mycelial yield were $30^{\circ}C$ and pH 5.5. The maximum yield of trehalose was obtanined after 10 day cultivation.

  • This experiment was carried out to study the practical utility of Mucor rennet in making Blue cheese and to investigate the changes in the level of various nitrogen compounds. 1. The Mucor rennet cheese, the calf rennet cheese and the mixed rennet cheese did not show any significant difference in their yields. 2. The dry matter contents of Blue cheese was increased during the ripening and the levels of Mucor rennet did not have any influence in these respect. 3. The water soluble nitrogen contents of Blue cheese increased during ripening. On 0 day of ripening the Mucor rennet cheese contained water soluble nitrogen than the calf rennet cheese. On 40 days of ripening the mixed cheese contained less water soluble nitrogen than the calf rennet cheese. 4. Non protein nitrogen, peptone amino nitrogen, water soluble protein nitrogen, proteose nitrogen and peptone nitrogen appeared to contain similar levels of water soluble nitrogen. 5. The results of electrophoresis indicated a greater degredation on as-casein and ${\beta}$-casein in the Blue cheese made with Mucor rennet than in those made with calf rennet. On 60 days of ripening the mixed cheese more casein than did the Mucor rennet cheese. 6. The free amino acid contents of the Mucor rennet cheese was higher than the calf rennet cheese at 60 days of ripening.

Optimal condition for mycelial growth of Sparassis crispa (꽃송이버섯(Sparassis crispa)의 균사생장 최적화)

  • This study was carried out to obtain the basic data on artificial culture of Sparassis crispa. The mycelial growth and density were good in the PBA, PDA and PDM media, and especially prominent in the PBA medium. The optimum condition for the mycelial growth was $25^{\circ}C$ and pH 5.0, respectively. The carbon sources such as glucose, fructose, mannose, xylose(monosaccharide) and maltose(disaccharide) were favorable to mycelial growth of S crispa. The optimal concentration of glucose is 1.0~1.5%. As nitrogen sources, threonine, peptone, glycine, glutamine and valine appeared to be favorable to the mycelial growth and density. The optimal concentration of peptone is 0.3%.

  • Nutritional characteristics and conditions for mycelial yield of Pleurotus ostreatus and Auricularia auricula-judae in shaking culture were investigated. Among the sugar substances, glucose and mannitol showed the good effect for mycelial yield of P. ostreatus, and mannitol and fructose were good for the mycelial yield of A. auricula-judae. Among the various organic acids, fumaric acid were good for the mycelial yield of P. ostreatus and A. auricula-judae. Among the nitrogen sources, peptone and urea showed the good result for mycelial yield of P. ostreatus, and peptone and casamino acid were good for mycelial yield of A. auricula-judae. Among the various amino acids, asparagin and threonine showed the good result for mycelial yield of P. ostreatus, and serine and threonine were good for mycelial yield of A. auricula-judae. Among the various vitamins, folic acid and thiamine were suitable for mycelial yield of P. ostreatus, and folic acid, inositol and riboflavin were suitable for mycelial yield of A. auricula-judae. Mycelial yield of P. ostreatus and A. auricula-judae were enhanced by the addition of $MgSO_4\;and\;KH_2PO_4$ at the concentration of 0.08 and 0.2% respectively. The optimum temperature and pH for mycelial yield were from $25\;to\;30^{\circ}C$ and pH 5.5 to 6.5 in P. ostreatus, and from $25\;to\;30^{\circ}C$ and pH 6.0 to 7.0 in A. auricula-judae.

Production and Characterization of Thermostable Protease from Bacillus licheniformis Isolated from Korean Traditional Soybean Paste (재래식 된장에서 분리된 Bacillus licheniformis의 내열성 Protease 특성과 생산성)

  • Among 63 Bacillus strains grown at $60^{\circ}C$ from sixteen samples of homemade Korean soybean paste, one strain was selected for producing the thermostable protease. The isolate has been identified as Bacillus licheniformis on the basis of its 16S rDNA sequence, morphology and biochemical properties. Culture filtrate of the isolate showed maximal protease activity at the reaction condition of $60-65^{\circ}C$ and pH 11. The culture filtrate retained more than 87% of initial protease activity after incubation for 30 min at $60^{\circ}C$ without substrate. In order to develop the medium composition, effects of ingredients including nitrogen sources, carbon sources, metal ions and phosphate were examined for protease production of the isolate. Lactose and soytone peptone were the most effective carbon and nitrogen source for the enzyme production. After the late logarithmic growth phase the isolate began to produce the protease, and the maximum protease productivity was reached to 550 unit/ml in the optimized medium consisting of lactose (3%), soytone peptone (1.5%), $MgSO_4$ (0.1%), $K_2HPO_4$ (0.03%), and $KH_2PO_4$ (0.03%) at 28 h of incubation.

Identification of characterization and statistical optimization of medium constituent for Bacillus subtilis SCJ4 isolated from Korean traditional fermented food (전통 장류 유래 Bacillus subtilis SCJ4의 특성확인 및 통계학적 방법을 이용한 배양조건 최적화)

  • 612 strains isolated from Korean traditional fermented food in Sunchang and their investigated biochemical characterization and ability of biogenic amines non-producing. We selected the SCJ4 having various activity by measurement of extracellular enzyme, antioxidant and antimicrobial activities. Selected strain SCJ4 by 16S rRNA sequencing and biochemical characterization was named Bacillus subtilis SCJ4. And then, we investigated cell growth of SCJ4, and optimized of culture medium constituents using response surface methodology as statistically method. Response surface methodology used Plackett-Burman experimental design for screening of medium constituent. Tryptone, peptone and $MgSO_4$ as medium constituent improving cell growth selected. In order to find out optimal concentration on each constituent, we carried out central composite design. Consequently, optimized concentrations of tryptone, peptone and $MgSO_4$ were predicted to be 15.35 g/L, 12.235 g/L, and 3.5 g/L respectively. Through the model verification, we confirmed about 1.28-fold improvement of the dried cell weight from 0.8767 g/L to 1.1222 g/L when compared to basal medium.

Fruit-body Formation of Pleurotus florida on the Synthetic Medium (합성배지(合成培地)를 이용(利用)한 고온성(高溫性) 느타리버섯의 자실체(子實體) 형성(形成)에 관한 연구(硏究))

  • Nutritional and physio-chemical conditions for mycelial growth and fruit-body formation of Pleurotus florida were determined in synthetic media. Mannitol and sucrose were good sugar substances for the mycelial growth and fruit-body formation whereas less mycelial growth and no fruit-body formation was obtained with arabinose, lactose and inulin. The optimum concentration of mannitol was about 2%. Peptone as a nitrogen sources resulted in a rapid mycelial growth and fruit-body formation with higher yield, but nitrite-nitrogens inhibited the mycelial growth. The higher yield was obtained with 0.2% peptone. Among the vitamins used, the greatest mycelial growth and fruit-body formation brought about by thiamine and folic acid. $KH_2PO_4$ and $MgSO_4$ at 0.2% and 0.02%, respectively, were effective for the mycelial growth and fruit-body formation, but other inorganic salts used were ineffective. The optimum temperature for mycelial growth and fruit-body formation were $25^{\circ}C$ and $20^{\circ}C$, respectively, and light intensity of $100{\sim}500\;lux$ and pH 6.0 appeared to be effective.

Optimizing Culture Conditions to Maximize the Production of Laccase from Pholiota highlandensis (Pholiota highlandensis 유래 laccase 생산을 위한 배양조건의 최적화)

  • The culture conditions needed to maximize the production of laccase from Pholiota highlandensis mycelia were investigated. Among the tested media for laccase production, Coriolus versicolor medium (CVM; 2% dextrose, 0.4% peptone, 0.6% yeast extract, 0.046% KH2PO4, 0.1% K2HPO4, 0.05% MgSO4·7H2O) showed the highest activity for the enzyme. Then, to optimize culture conditions for laccase activity, the influences of various carbon, nitrogen, phosphorus, and inorganic salt sources in CVM were investigated. The optimum culture medium was 2% fructose, 0.4% peptone with 0.6% yeast extract, 0.05% NaH2PO4, and 0.05% MgSO4·7H2O as carbon, nitrogen, phosphorus, and inorganic salt sources, respectively. Several aromatic compounds in the medium enhanced laccase activity to varying degrees. Guaiacol induced maximum laccase production, yielding 114.1 U/ml laccase activity after cultivation for 11 days at 25℃. The optimum pH and temperature for laccase production were 8.0 and 35℃, respectively. Native polyacrylamide-gel electrophoresis (PAGE) followed by laccase-activity staining with 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) as the substrate was performed to identify the presence of laccase under the optimum conditions studied. Zymogram analysis of the supernatant culture showed an enzymatic band with a molecular mass of about 90 kDa.

Optimal Culture Conditions for Production of Subtilisin-like Protease Inhibitor from Streptomyces thermocarboxydus C12 (Streptomyces thermocarboxydus C12에서 Subtilisin-like Protease Inhibitor 생산을 위한 최적배양조건)

  • The objective of this paper was to investigate optimal culture conditions for the production of an inhibitor against subtilisin-like protease from Streptomyces thermocarboxydus (S. thermocarboxydus) C12 isolated from sediments of Gwangyang coast. The optimal temperature and initial pH for the production of subtilisin-like protease inhibitor were $40^{\circ}C$ and pH 8.0, respectively. Inhibition activities were high for galactose, glucose and fructose. The best carbon source and its concentration were galactose and 1.6% (w/v), respectively. Inhibition activities were also high in medium containing polypeptone, proteose and peptone. Optimal nitrogen source and concentration were protease peptone and 0.5% (w/v), respectively. Optimal concentrations for inhibitor production were 1% (w/v) for NaCl and 1 mM LiCl for metal salts. The subtilisin-like protease inhibitor from S. thermocarboxydus C12 showed a maximum inhibitor activity after cultivation for 84 h under the optimized medium.
