Effect of Terephthalaldehyde to Facilitate Electron Transfer in Heme-mimic Catalyst and Its Use in Membraneless Hydrogen Peroxide Fuel Cell (테레프탈알데하이드의 전자전달 강화효과에 따른 헴 단백질 모방 촉매의 성능 향상 및 이를 이용한 비분리막형 과산화수소 연료전지)

  • Terephthalaldehyde (TPA) is introduced as a cross liker to enhance electron transfer of hemin-based cathodic catalyst consisting of polyethyleneimine (PEI), carbon nanotube (CNT) for hydrogen peroxide reduction reaction (HPRR). In the cyclic voltammetry (CV) test with 10 mM H2O2 in phosphate buffer solution (pH 7.4), the current density for HPRR of the suggested catalyst (CNT/PEI/hemin/PEI/TPA) shows 0.2813 mA cm-2 (at 0.2 V vs. Ag/AgCl), which is 2.43 and 1.87 times of non-cross-linked (CNT/PEI/hemin/PEI) and conventional cross liker (glutaraldehyde, GA) used catalyst (CNT/PEI/hemin/PEI/GA), respectively. In the case of onset potential for HPRR, that of CNT/PEI/hemin/PEI/TPA is observed at 0.544 V, while those of CNT/PEI/hemin/PEI and CNT/PEI/hemin/PEI/GA are 0.511 and 0.471 V, respectively. These results indicate that TPA plays a role in facilitating electron transfer between the electrodes and substrates due to the π-conjugated cross-linking bonds, whereas conventional GA cross-linker increases the overpotential by interrupting electron and mass transfer. Electrochemical impedance spectroscopy (EIS) results also display the same tendency. The charge transfer resistance (Rct) of CNT/PEI/hemin/PEI/TPA decreases about 6.2% from that of CNT/PEI/hemin/PEI, while CNT/PEI/hemin/PEI/GA shows the highest Rct. The polarization curve using each catalyst also supports the superiority of TPA cross liker. The maximum power density of CNT/PEI/hemin/PEI/TPA (36.34±1.41 μWcm-2) is significantly higher than those of CNT/PEI/hemin/PEI (27.87±0.95 μWcm-2) and CNT/PEI/hemin/PEI/GA (25.57±1.32 μWcm-2), demonstrating again that the cathode using TPA has the best performance in HPRR.

Adsorption Characteristics of Functionalized Activated Carbon for High Temperature CO2 Capture (고온 이산화탄소 포집을 위한 기능성 활성탄의 흡착특성)

  • Activated carbon impregnated with polyethyleneimine (PEI) was evaluated as a functionalized adsorbent for $CO_2$ capture. The $CO_2$ adsorption characteristics of the adsorbents was undertaken using GC/TCD, BET surface area and FT-IR. A series of adsorbents were synthesized by impregnating 10, 30, 50 wt% of PEI on activated carbons and were investigated $CO_2$ adsorption capacity at high and low adsorption temperature. The $CO_2$ adsorption capacity at $20^{\circ}C$ and $100^{\circ}C$ was as follow: AC > PEI(10)-AC > PEI(30)-AC > PEI(50)-AC at $20^{\circ}C$ and PEI(10)-AC > PEI(30)-AC > PEI(50)-AC > AC at $100^{\circ}C$. Adsorption capacities of amine functionalized AC was lager than virgin AC at high temperature due to chemisorption by amino-group content. From the results, the PEI(10)-AC showed one of the most promising adsorbents for $CO_2$ capture from flue gas at high temperature.

Immobilization of Glucose Oxidase using Branched Polyethyleneimines of Various Molecular Weights for Glucose Based Biofuel Cell (글루코스 기반 바이오연료전지를 위한 다양한 분자량의 폴리에틸렌이민을 이용한 글루코스 산화효소 고정화)

  • In this study, we fabricated the catalysts for enzymatic biofuel cell anode with carbon nanotube (CNT), glucose oxidase (GOx) and various molecular weights branched poly(ethyleneimine)(bPEI) and terephthalaldehyde (TPA) as cross-linker. In case of GOx/bPEI/CNT using only physical entrapments for immobilization, the molecular weights of bPEI didn't affect to electrochemical performances and long term stability. but that of the catalysts cross linked via TPA (TPA[GOx/bPEI/CNT]) improved and the mass transfer of glucose to FAD was interrupted as increasing of the bPEI's molecular weights. Furthermore, it was confirmed that the optimum molecular weight of PEI for TPA [GOx/bPEI/CNT]) structure is 750k that showed marvelous high performance (maximum power density of $0.995mW{\cdot}cm^{-2}$).

Apoptosis Induced by Polyethylenimine/DNA Complex in Polymer Mediated Gene Delivery

  • Polyethylenimine (PEI) has been widely investigated for delivery of DNA into cells. It was previously reported that there were at least two types of cytotoxicity in PEI-mediated gene delivery, immediate and delayed toxicities. PEI-mediated gene delivery protocols use net cationic complexes with an excess of PEI to maintain equilibrium between the complexed and dissociated forms in solution. In this study, toxicity of free PEI or PEI/ DNA complex was investigated. Human embryonic kidney 293 cells were incubated with free PEI or PEI/DNA complex for 4 hrs. Then, the cells were analyzed at 6, 24, 48, and 96 hrs after the incubation. In MTT assay, the viability of the cells incubated with PEI/DNA complex was continuously decreased with time, while that of the cells incubated with free PEI was not. On the contrary, the expression level of the luciferase gene increased gradually along with time. Release of DNAs from the complexes for transcription produces free PEIs in the cells. This process may proceed slowly due to high charge density of PEI and may be related to delayed toxicity. In addition, apoptotic cells were observed only in the cells incubated with the PEI/DNA complex from 24 hrs after the incubation. The results suggest that PEI/DNA complex contributes to the delayed toxicity by inducing apoptosis and that the delayed toxicity may be related to decomplexation of the complexes in the cells.

Low Molecular Weight Polyethylenimine-Mitochondrial Leader Peptide Conjugate for DNA Delivery to Mitochondria

  • It has been found that a number of diseases are associated with mutations in the mitochondrial DNA. Therapeutic gene delivery to mitochondria has been suggested as a clinical option for these diseases. In this study, we developed a gene carrier to mitochondria by the conjugation of mitochondrial leader peptide (LP) to polyethylenimine (PEI). Mitochondrial LP conjugated PEI (PEI-LP) was synthesized with low molecular weight PEI (2,000 Da, PEI2K). Gel retardation assay showed that PEI2K-LP formed complexes at a 1.0/1 weight ratio. In addition, PEI2K-LP protected DNA from the enzymatic degradation for at least 60 min, while naked DNA was completely degraded within 20 min. PEI2K-LP was compared with LP conjugated high molecular weight PEI (25,000 Da, PEI25K) in terms of toxicity and delivery efficiency. MTT assay showed that PEI2K-LP had much lower cytotoxicity than PEI25K-LP to 293 cells. In addition, cell-free DNA delivery assay showed that PEI2K-LP delivered more DNA to mitochondria at a 1.8/1 weight ratio than naked DNA or PEI. This result suggests that PEI2K-LP may be useful for the development of mitochondrial gene therapy system with lower cytotoxicity.

Polyethylenimine Mediated Gene Delivery with Various Liposomal Formulations (폴리에틸렌이민 및 그들의 리포좀이 중재된 Plasmid DNA의 운반)

  • The transfection efficiency of plasmid DNA was inspected using multi-cationic polymer, 5, 10, 25 and 50KD polyethylenimine (PEI). The optimal neutralization ratio of PEI/DNA complexes by agarose assay was 1.5-2.0 (nmol/nmol) without much difference in molecular weight of PEI.In vitro transfection assay, most of PEI-mediated plasmid delivery was better compared to the naked DNA. Especially, 25KD PEI at optimal condition gave higher transfection rather than the standard assay of DEAE-dextran or Lipofectin. To enhance the cell targeting delivery, the liposome formulations were introduced using phospholipids. As a result, PC/PE liposomes increased 2-2.5 times of the transfection efficiency of PEI single or PC/PE single delivery, but not the case of 25KD PEI. Moreover, the DOTAP/PE-introduced PEI delivery reduced the transfection of DOTAP/PE single delivery. All these results proved that the PEI can be used not only good transfectants and but also good DNA condensing agents in neutral/anionic liposome for cell targeting delivery.

A Study on Dyeability of PEI-treated Cotton Fabric with Polychromatic Natural Dyes (PEI를 처리한 면직물의 다색성 천연염료에 대한 염색성 변화)

  • This study examined the change in the dyeability of natural dyes on cotton fabrics by a PEI (polyethyleneimine) pretreatment instead of mordants. Cotton fabrics were treated with PEI and the changes in the dyeability were shown by measuring the amount of PEI on cotton fabrics. Samples treated with PEI were dyed with two natural polychromatic dyes with a different affinity to cotton fibers: Alizarin Red S and Curcumin. The changes in dyeability by three variables (time, temperature and concentration of dyes) on cotton fabrics were analyzed by the K/S value to define optimum dyeing conditions. Subsequently, the PEI treatment improved the dyeability of cotton fabrics with both dyes of low and high affinity to cotton fibers. Thus, PEI could be a suitable heavy metal mordant replacement.

  • Polyethylenimine (PEI) is a cationic polymer that can bind to negatively charged biomaterials such as nucleic acids through strong electrostatic interactions. Based on these properties, PEI has been used as an efficient drug delivery system for a long time. However, the strong cationic nature of PEI has the problem of causing cytotoxicity by non-specific interaction with anionic biological materials in the cells. In order to overcome these problems, many researchers have developed various types of biocompatible PEI-based materials. In this review, we would like to introduce the recent developments of functional PEI and their applications in biomedical research.

Separation of Hydrogen-Nitrogen Gas Mixture by PTMSP-Silica-PEI Composite Membranes (PTMSP-Silica-PEI 복합막에 의한 수소-질소 혼합기체 분리)

  • The poly(1-trimethylsilyl-1-propyne) (PTMSP) and silica-filled PTMSP membranes were prepared by casting from a toluene solution on porous polyetherimide (PEI). FT-IR spectrum, GPC and SEM pictures have been taken to characterize the membranes. The particle size of membrane decreases as silica content of the membrane increases from 23 to 60 wt%, and a uniform distribution of the silica is observed. The separation properties of the gas mixture (32 mol% $H_2$/ 68 mol% $N_2$) through the composite membranes were studies as a function of pressure and percentage of silica. Selectivity values of $H_2$/$N_2$ increased as the pressure of permeation cell and silica content of the membrane increased. The real separation factor($\alpha$), head separation factor($\beta$), and tail separation factor((equation omitted)) of PTMSP-PEI composite membrane were 2.28, 1.58, and 1.44 respectively at $\Delta$P 30 psi and $25^{\circ}C$. $\alpha$, $\beta$, and (equation omitted) of PTMSP-Silica-PEI composite membrane for 60 wt% silica were 3.34, 1.95, 1.72 at $\Delta$P 30 psi and $25^{\circ}C$.

Evaluation of Interfacial Properties on the Electrodeposited Carbon Fiber Reinforced Polyetherimide Toughened Epoxy Composites using Micromechanical Test (Micromechanical 시험법을 이용한 전기증착된 탄소섬유 강화 Polyetherimide로 강인화된 에폭시 복합재료의 계면물성 평가)

  • Interfacial properties and microfailure modes or electrodeposition(ED) treated carbon fiber reinforced polyetherimide(PEI) toughened epoxy composites were investigated using microdroplet test. ED was performed to improve the interfacial shear strength(IFSS). As PEI content increased, IFSS increased due to enhanced toughness and plastic deformation of PEI. In the untreated cafe, IFSS Increased with adding PEI content, and IFSS of pure PEI matrix showed the highest. On the other hand, thor ED-treated case IFSS increased with PEI content with rather low improvement rate. In the untreated case, neat epoxy resin appeared brittle microfailure mode, whereas pure PEI matrix exhibited more likely ductile microfailure mode. In the ED-treated case, neat epoxy exhibited more ductile fracture compared to the untreated case. Interfacial properties of epoxy-PEI composite can be affected efficiently by both the control of matrix toughness and ED treatment.
