RAPD를 이용한 Pecan 품종의 유전적 관계 분석 (Analysis of Genetic Relatedness by Random Amplified Polymorphic DNA (RAPD) in Pecan Taxa)

  • 재배되고 있는 22 페칸종간의 RAPD분석을 한 결과, Agarose gel상에서 primer당 평균 6개의 DNA단편이 증폭되었다. 증폭된 DNA 단편의 크기 는 100bp에서 1500bp의 범위에 위치하였다. 페칸종간의 유연 관계를 분석하기에 적합한 primer를 선발한 결과 OPA-02, OPA-10, OPA-12, OPB-01이 최적이었다. 4종의 primer를 이용하여 증폭한 PCR산물을 1.5% Agarose gel상에 분획하여 단편의 종류를 분석한 결과 22의 페칸종은 5개의 그룹으로 구분 할 수 있었다. 40종 primer로부터 증폭된 466 DNA단편을 기초로하여 유전적 근연계수를 계산하였던 결과 유사도 값이 가장큰 것은 C. flacra와 Black walnut로 0.90를 나타났으며, Kiowa와 Red hickory, C. tomentosa, C. flacra, Black walnut간에는 유사도 값이 0.31로 가장 작았다. 선발된 4종의 primer를 이용하여 증폭시킨 PCR산물을 Polyacrylamide gel상에 분획한 후 검출된 DNA단편을 분석하였다. 유사도 값이 가장 큰 것은 그룹 V의 Red hickory, C. tomentosa, C. flacra, Black walnut간으로 1.0이었고, Farley와 Pawnee간과 Sturya와 Clarke간은 공히 0.98를 나타내었다. Polyacrylamide gel분획 후 은염색하여 단편을 검출하는 것은 Agarose gel에 비하여 시간, 기술, 경비가 요구되지만 보다 정확한 유전적 배경을 설명할 수 있다고 생각되었다. 또한 Agarose gel 분석, Polyacrylamide gel 분석 및 주성분 분석법에서 Farley, C. tomentosa, C. flacra, Black walnut, C. corpiformis만이 동일 그룹에서 이탈되지 않았다.

Fat replacement by pecan nut and oregano oil and their impact on the physicochemical properties and consumer acceptability of frankfurters

  • Objective: A study was conducted to determine the physicochemical quality and consumer acceptability of beef frankfurter-type sausages formulated with pecan nut paste and Mexican oregano oil (MO) of two varieties, Poliomintha longiflora Gray (Mexican oregano Poliomintha, MOP) or Lippia berlandieri (Mexican oregano Lippia, MOL). Methods: Frankfurters were processed under six treatment conditions: control (10.67% pork fat), MOP (control + 0.01% MOP), MOL (control + 0.01% MOL), MOP-N (control + 0.01% MOP + 6% pecan nut paste), MOL-N (control + 0.01% MOL + 6% pecan nut paste), and C-N (control + 6% pecan nut paste). The physicochemical properties and the consumer acceptability were determined. Results: The addition of MO and nut did not influence the water holding capacity, drip loss, and luminosity of frankfurters, but those ingredients increased pH and shear force (p<0.05) and decreased redness (p<0.05) of frankfurters. Frankfurters were generally well accepted by consumers. However, general acceptability of sausages decreased with the addition of MO. Control sausages showed the highest acceptability. Conclusion: The MO and pecan nut paste do not affect drastically the quality of frankfurters. These results provide evidence that partial replacement of pork fat by pecan nut in frankfurters maintain a consistent physicochemical quality and its commercialization looks promising given consumers' acceptance.

Varietal Classification by Multivariate Analysis on Quantitative Traits in Pecan

  • Twenty two varieties of pecan including wild types were classified based on 6 characters measured by principal component analysis score distance. The results are summarized as fellow. Twenty two varieties were classified into 5 groups based in PCA score distance. Five groups were distinctly characterized by many morphological characters. Total variation could be explained by 51%, 95%, 99% with first, third and fifth principal components respectively. Varimax rotation of the factor loading of the first factors indicated that the first component was highly loaded with leaf characters, the second component with fruit characters, but fruit length was negative loaded. The second, the third and the fourths groups of cultivars had very close genetic parentage similarity.

PECAN: Peer Cache Adaptation for Peer-to-Peer Video-on-Demand Streaming

  • To meet the increased demand of video-on-demand (VoD) services, peer-to-peer (P2P) mesh-based multiple video approaches have been recently proposed, where each peer is able to find a video segment interested without resort to the video server. However, they have not considered the constraint of the server's upload bandwidth and the fairness between upload and download amounts at each peer. In this paper, we propose a novel P2P VoD streaming system, named peer cache adaptation (PECAN) where each peer adjusts its cache capacity adaptively to meet the server's upload bandwidth constraint and achieve the fairness. For doing so, we first propose a new cache replacement algorithm that designs the number of caches for a segment to be proportional to its popularity. Second, we mathematically prove that if the cache capacity of a peer is proportional to its segment request rate, the fairness between upload and download amounts at each peer can be achieved. Third, we propose a method that determines each peer's cache capacity adaptively according to the constraint of the server's upload bandwidth. Against the proposed design objective, some selfish peers may not follow our protocol to increase their payoff. To detect such peers, we design a simple distributed reputation and monitoring system. Through simulations, we show that PECAN meets the server upload bandwidth constraint, and achieves the fairness well at each peer. We finally verify that the control overhead in PECAN caused by the search, reputation, and monitoring systems is very small, which is an important factor for real deployment.

Removal of Pb(II) and Cd(II) From Aqueous solution Using Oxidized Activated Carbons Developed From Pecan Shells.

  • Oxidized activated carbons were prepared by reacting steam-activated carbon developed from pecan shells with nitric acid of varying strength (15, 30, 45 and 60%). The textural properties and the chemistry of the surface of the non-oxidized and of the oxidized carbons were determined from nitrogen adsorption and base neutralization capacities. The uptake of Pb(II) and Cd(II) from aqueous solution by these carbons was determined by kinetic and equilibrium experiments as well as by the column method. Treatment with nitric acid brought about drastic decrease in surface area and remarkable increase in the pore size of the carbon with these changes depending on the strength of nitric acid. Nitric acid increased the surface acidity by developing new surface oxygen functional groups of acidic nature. $HNO_3$-oxidized carbons exhibited high adsorption capacities for Pb(II) and Cd(II). The adsorption of these ions increased with the decrease of the surface pH of the carbon and with the increase of the solution pH from 2.5 to 6 and 7. The amount adsorbed from lead and cadmium was also related to the amount of surface acidity, the pH of the point of zero charge and on some metal ion parameters. Cadmium and lead uptake by the investigated carbons followed pseudo-second order model and the equilibrium sorption data fitted Langmuir adsorption model.

견과류의 지방산, 트리아실글리세롤, 토코페롤 및 파이토스테롤의 조성 연구 (Studies on the Content of Triacylglycerol Species, Tocopherols, and Phytosterols from the Selected Nuts)

  • 견과류 중 sunflower seed, cashew nut, walnut, pistachio, pumpkin seed, ginkgo, hazel nut, pecan을 원료로 하여 지방산 조성 분석 및 sn-2 position 분석, triacylglycerol species의 분석, tocopherol, phytosterol 함량을 분석하였다. 조지방 함량은 hazel nut에서 약 39.60 wt%로 가장 높았으며, walnut 35.25 wt%, pistachio 30.22 wt% 등의 순이었다. GC에 의한 sn-2 위치의 지방산 조성 분석 결과, oleic acid의 함량이 많은 견과류는 cashew nut, pistachio, hazel nut, pecan 이였으며, 특히 ginkgo의 경우 특이적으로 oleic acid의 이성체인 vaccenic acid가 10.72 wt% 나타냄을 확인할 수 있었다. 반면, linoleic acid 등이 높은 견과류는 sunflower seed, walnut, pumpkin seed 이었다. 견과류들의 TAG species의 PN값은 40∼52 사이에 존재하였다. Tocopherol의 함량은 ginkgo, sunflower seed, pumpkin seed 등에서 각각 48.57, 38.35, 31.43 mg/100 g 순으로 높은 함량을 나타내었다. 또한 총 phytosterol의 경우 88.60∼947.20 mg/100 g 사이에 함량을 나타내었다.

Root Barrier and Fertilizer Effects on Soil CO2 Efflux and Cotton Yield in a Pecan-Cotton Alley Cropping System in the Southern United States

  • Little information is available on soil $CO_2$ efflux and crop yield under agroforestry systems. Soil $CO_2$ efflux, microbial biomass C, live fine root biomass, and cotton yield were measured under a pecan (Carya illinoinensis K. Koch)-cotton (Gossypium hirsutum L.) alley cropping system in southern USA. A belowground polyethylene root barrier was used to isolate tree roots from cotton which is to provide barrier and non-barrier treatments. The barrier and non-barrier treatment was randomly divided into three plots for conventional inorganic fertilizer application and the other three plots for organic poultry litter application. The rate of soil $CO_2$ efflux and the soil microbial biomass C were affected significantly (P < 0.05) by the fertilizer treatment while no significant effect of the barrier treatment was occurred. Cotton lint yield was significantly (P < 0.0 I) affected by the root barrier treatment while no effect was occurred by the fertilizer treatment with the yields being greatest ($521.2kg\;ha^{-1}$) in the root barrier ${\times}$ inorganic fertilizer treatment and lowest ($159.8kg\;ha^{-1}$) in the non-barrier ${\times}$ inorganic fertilizer treatment. The results suggest that the separation of tree-crop root systems with the application of inorganic fertilizer influence the soil moisture and soil N availability, which in tum will affect the magnitude of crop yield.

Molecular Dynamics of the C-Terminal Domain Mouse CDT1 Protein

  • The backbone molecular dynamics of the C-terminal part of the mouse Cdt1 protein (tCdt1, residues 420-557) was studied by high field NMR spectroscopy. The Secondary structure of this protein was suggested by analyzing of chemical shift of backbone atoms with programs TALOS and PECAN, together with NOE connectivities from 3D $^{15}N-HSQC-NOESY$ data. Measurement of dynamic parameters $T_1,\;T_2$ and NOE and limited proteolysis experiment provided information for domain organization of tCdt1(420-557). Analysis of the experimental data showed that the C-terminal part of the tCdt1 has well folded domain for residues 455-553. The residues 420-453 including ${\alpha}-helix$ (432-441) are flexible and probably belong to other functional domain in intact full length Cdt1 protein.

Prediction of Energy Consumption in a Smart Home Using Coherent Weighted K-Means Clustering ARIMA Model

  • Technology is progressing with every passing day and the enormous usage of electricity is becoming a necessity. One of the techniques to enjoy the assistances in a smart home is the efficiency to manage the electric energy. When electric energy is managed in an appropriate way, it drastically saves sufficient power even to be spent during hard time as when hit by natural calamities. To accomplish this, prediction of energy consumption plays a very important role. This proposed prediction model Coherent Weighted K-Means Clustering ARIMA (CWKMCA) enhances the weighted k-means clustering technique by adding weights to the cluster points. Forecasting is done using the ARIMA model based on the centroid of the clusters produced. The dataset for this proposed work is taken from the Pecan Project in Texas, USA. The level of accuracy of this model is compared with the traditional ARIMA model and the Weighted K-Means Clustering ARIMA Model. When predicting,errors such as RMSE, MAPE, AIC and AICC are analysed, the results of this suggested work reveal lower values than the ARIMA and Weighted K-Means Clustering ARIMA models. This model also has a greater loglikelihood, demonstrating that this model outperforms the ARIMA model for time series forecasting.

Xylella fastidiosa in Europe: From the Introduction to the Current Status

  • Xylella fastidiosa is xylem-limited bacterium capable of infecting a wide range of host plants, resulting in Pierce's disease in grapevine, citrus variegated chlorosis, olive quick decline syndrome, peach phony disease, plum leaf scald, alfalfa dwarf, margin necrosis and leaf scorch affecting oleander, coffee, almond, pecan, mulberry, red maple, oak, and other types of cultivated and ornamental plants and forest trees. In the European Union, X. fastidiosa is listed as a quarantine organism. Since its first outbreak in the Apulia region of southern Italy in 2013 where it caused devastating disease on Olea europaea (called olive leaf scorch and quick decline), X. fastidiosa continued to spread and successfully established in some European countries (Corsica and PACA in France, Balearic Islands, Madrid and Comunitat Valenciana in Spain, and Porto in Portugal). The most recent data for Europe indicates that X. fastidiosa is present on 174 hosts, 25 of which were newly identified in 2021 (with further five hosts discovered in other parts of the world in the same year). From the six reported subspecies of X. fastidiosa worldwide, four have been recorded in European countries (fastidiosa, multiplex, pauca, and sandyi). Currently confirmed X. fastidiosa vector species are Philaenus spumarius, Neophilaenus campestris, and Philaenus italosignus, whereby only P. spumarius (which has been identified as the key vector in Apulia, Italy) is also present in Americas. X. fastidiosa control is currently based on pathogen-free propagation plant material, eradication, territory demarcation, and vector control, as well as use of resistant plant cultivars and bactericidal treatments.
