  • DA-8159 is a new. highly selective. potent cyclic-GMP phosphodiesterase 5 inhibitor developed by Dong-A Pharmaceutical Company(Kyunggi, Korea) as an oral drug for the treatment of erectile dysfunction. NO- cGMP signal transduction pathway plays a key role for relaxation of corpus cavernosal smooth muscle.   • We conducted a sensitivity analysis of the simulated moving bed (SMB) chromatography with the case model of the separation of two amino acids phenylalanine and tryptophan. We consider a four-zone SMB chromatography where the triangle theory is used to determine the operating conditions. Competitive Langmuir isotherm model was used to determine the adsorption isotherm. The finite difference method is used to solve nonlinear partial differential equation (PDE) systems numerically. We examined the effects of alterations in the operating conditions(feed-extract, feed-raffinate, eluent-extract, eluent-raffinate, recycle, and switching time) and the adsorption isotherm parameters (Langmuir isotherm parameters a and b) on SMB efficiency. The variation range of operating conditions and Langmuir isotherm a was between -50 and 50% of original value and the variation range of the Langmuir isotherm b was between $2.25^{-5}$ and $2.25^5$ times of original value.

  • Vascular endothelial cells release proteinases that degrade the extracellular matrix (ECM), thus enabling cell migration during angiogenesis and vasculogenesis. Sildenafil citrate stimulates the nitric oxide-cyclic guanosine monophosphate pathway through inhibition of phosphodiesterase type V (PDE5). In this report, we examined the mechanisms underlying sildenafil citrate-induced cell migration using cultured mouse aortic endothelial cells (MAECs). Sildenafil citrate induced migration and proteinase secretion by murine endothelial cells. Sildenafil citrate induced the secretion of matrix metalloproteinase-2 (MMP-2) and MMP-9, which is inhibited by $NF-{\kappa}B$ inhibitors. Sildenafil citrate also induced the secretion of plasmin, which is inhibited by PI 3'-kinase inhibitors. It is suggested that sildenafil citrate-induced migrating activity in endothelial cells may be accomplished by increased secretion of proteinases.

  • In this paper, we develop a numerical method to solving an optimal control problem, which is governed by a parabolic partial differential equations(PDEs). Our approach is to approximate the PDE problem to initial value problem(IVP) in $\mathbb{R}$. Then, the homogeneous part of IVP is solved using semigroup theory. In the next step, the convergence of this approach is verified by properties of one-parameter semigroup theory. In the rest of paper, the original optimal control problem is solved by utilizing the solution of homogeneous part. Finally one numerical example is given.

Protective Effects of Ginkgolide B on Reperfusion of the Isolated Perfused Guinea Pig Heart (Ginkgolide B의 Guinea Pig 적출 심장에 대한 허혈 유발후 Reperfusion시의 보호 작용에 관한 연구)

  • The cardiac effects of PAF antagonist Ginkgolide B(BN 52051) have been investigated on the isolated perfused guinea pig hearts maintained at the constant hydrostatic perfusion pressure of 80 cm water. PDE(Phosphodiesterase) inhibitor KR-30289 was used as a positive control to see the positive inotropic effects on the perfused hearts. In this expriments, Ginkgolide $B(10^{-5}-SM)$ showed negative inotropic effects by decreasing of LVP, LVDP, LV dp/dt, HR and RPP(Rate Pressure Product). Ginkgolide B also decreased the number of extrasystole by $51.9\%(from\;23.75\pm9.22/min\;to\;11.43\pm435/min)$ induced by global ischemia and reperfusion. The rate, [-dp/dt]/[+dp/dt] increased in preischemia but decreased in postischemia. 1n the separated study the injection of 1ml of Ginkgolide B$(10^{-4M})$ on the isolated heart, increased coronary flow(CF) by $11.8\%(from\;7.5\pm7.65ml/min\;to\;8.5\pm0.29ml/min)$ and decreased the number of extrasystole by $47.6\%(from\;21\pm5.92/min\;to\;11\pm5.27/min)$. In conclusion, Ginkgolide B showed antiarrhythmic and protective effects by decreasing the number of extrasystole and by increasing the coronary flow, respectively.

SIF AND FINITE ELEMENT SOLUTIONS FOR CORNER SINGULARITIES

  • In [7, 8] they introduced a new finite element method for accurate numerical solutions of Poisson equations with corner singularities. They consider the Poisson equations with homogeneous boundary conditions, compute the finite element solutions using standard FEM and use the extraction formula to compute the stress intensity factor(s), then they posed new PDE with a regular solution by imposing the nonhomogeneous boundary condition using the computed stress intensity factor(s), which converges with optimal speed. From the solution they could get an accurate solution just by adding the singular part. Their algorithm involves an iteration and the iteration number depends on the acuracy of stress intensity factors, which is usually obtained by extraction formula which use the finite element solutions computed by standard Finite Element Method. In this paper we investigate the dependence of the iteration number on the convergence of stress intensity factors and give a way to reduce the iteration number, together with some numerical experiments.

  • Fowler-Nordheim의 전자 방출과 열전자 방출 메카니즘을 이용하여 절연유체 내 전계에 의한 도체의 음극에서 전자 방출현상과 열에 의한 열전자 방출현상을 고려하고 유한요소법(Finite Element Method)을 이용하여 해석하였다. 절연유체 내 공간전하에 대한 해석기법으로 푸아송 방정식, 양이온, 음이온, 전자에 대한 전하연속 방정식, 온도에 대한 열 확산 방정식으로 이루어진 5개의 지배방정식에 Fowler-Nordheim의 전계 방출과 Richardson-Dushman의 열전자 방출을 경계조건으로 부여하였다. 단자 전류는 유한요소법과 잘 부합하는 에너지법으로 계산되었다. 쌍 곡선형 PDE의 공간전하 전파에 대한 지배 방정식은 일반적으로 수치적인 불안정성을 가지므로 인공 확산 항을 고려하여 이를 해결하였다. 제안된 해석법은 세 개의 캐리어를 가진 x-y 좌표축의 2차원 평판 모델에 적용하여 그 유효성을 확인하였다.

Shape and Appearance Repair for Incomplete Point Surfaces (결함이 있는 점집합 곡면의 형상 및 외관 수정)

  • In this paper, we present a new surface content completion system that can effectively repair both shape and appearance from scanned, incomplete point set inputs. First, geometric holes can be robustly identified from noisy and defective data sets without the need for any normal or orientation information. The geometry and texture information of the holes can then be determined either automatically from the models' context, or manually from users' selection. After identifying the patch that most resembles each hole region, the geometry and texture information can be completed by warping the candidate region and gluing it onto the hole area. The displacement vector field for the exact alignment process is computed by solving a Poisson equation with boundary conditions. Out experiments show that the unified framework, founded upon the techniques of deformable models and PDE modeling, can provide a robust and elegant solution for content completion of defective, complex point surfaces.

Pharmacokinetics of DA-8159, a new PDE5 inhibitor, after single and 1-week repeated oral administrations in mice

  • DA-8159 is a new PDEV (Phosphodiesterase V) inhibitor, synthesized by Dong-A Pharm., as an oral agent to treat male erectile dysfunction. To make a selection of the dosage of oral administration in carcinogenic studies, we studied preliminarily the pharmacokinetics of DA-8159 after single (at the 1$\^$st/ day) and 1-week (at the 7$\^$th/ day) oral administrations of the drug at doses of 15, 50 and 150 mg/kg/day, to male ICR mice. In 15mg/kg single and 1-week repeated oral administration groups, the concentrations of DA-8159 and DA-8164(the main metabolite of DA-8159) were below the limit of quantitation(LOQ:50ng/ml).
