Improvement Effect and Electrical Characteristics of Soft Ground with Plastic Electrode Spacing (전극간 거리에 따른 연약지반의 지반개량 효과와 전기적 특성)

  • Soft ground stabilization is needed to construct large civil facilities on the soft clay ground. Pre-loading method, which is accelerating consolidation method, is generally used to stabilize the soft ground. However, pre-loading method is required long construction period and quantities of fill material. Therefore, electro-osmosis method is used to replace pre-loading method for stabilizing the soft ground. Electro-osmosis method is disadvantageous in constructive and economic aspects because it is needed a metallic electrode. So, in order to solve the those disadvantages, plastic electrode was developed to replace metallic electrode. Plastic electrode, which is made by using nano-technology on existing Plastic Drain Board (PDB), was used to supply the electric power. In this study, therefore, the model test was conducted to confirm the effect of improvement and electrical characteristics of soft ground by spacing of plastic electrode. The result shows that the effect of improvement of soft ground was decreased up to 45% by increasing electrode spacing and electrical characteristics on the soft ground were influenced by consolidation settlement with electrode spacing.

Occurrence, Geochemistry and Origin of $Co_2$-rich Water from the Chungcheong Area, Korea (충청지역 탄산수의 산출양상, 지화학적 특성 및 생성기원)

  • Several $Co_2$-rich springs in the Chungcheong area, Le., the Angsung spring, the Chojung spring, the Myungam spring, the Bukang spring and the Daepyung spring have been long known for their particular water chemistry. The occurrences of these springs are closely related to the geologic structure of Mesozoic granite such as dyke swarms, fault zones and the geologic boundary between granite and its adjacent gneiss. The $Co_2$-rich water samples show a high $Co_2$ concentration ($P_{CO2}$ 0.25 atm to 0.99 atm), weak acidic pHs and the electrical conductivity ranging from 101 to 2,950 ${\mu}$S/cm. The $Co_2$-rich water samples are classified into the Ca-$HC0_3$ type and the Ca(Na)-$HCO_3$) type in chemical composition. Environmental isotopic data $^{2}H/^{1}H, ^{18}O/^{16}O$) indicated that $Co_2$-rich water was meteoric origin. The ${\delta}^{13}C$ values of $Co_2$-rich water range from -3.1$\textperthousand$ to -6.8$\textperthousand$ PDB. The values indicate that $H_2CO_3^0$ and $HC0_3^-$ of the water samples are mainly originated from a deep-seated source and partly contributed from carbonatc minerals. The major minerals determining the chemistry of $Co_2$-rich watcr arc probably the carbonate minerals which are present as veins and secondary minerals, and the plagiocalse in granite and gneiss.

Effects of Loess on the Mycellial Pellet Formation of Phosphate-solubilizing Fungus, Aspergillus sp. PS-104 in the Submerged Culture (Aspergillus sp. PS-104의 액침배양증 mycellial pellet 형성에 미치는 황토의 영향)

  • In order to investigate effects if loess on the mycellial pellet formation a phosphate-solubilizing fungus. Aspergillus sp. PS-104 was cultured in potato dextrose broth containing loess. The strain formed an amorphous pellet or loose aggregates agitated at a low speed (50 rpm) while spherical and regular pellets at a high speed (150 rpm) The higher concentration of loess was added, the smaller size of a pellet was formed during the submerged culture of the strain. As shown in results, being cultured in the PDB medium supplemented with 1.0% loess the pellet size was maximally reduced to a fourth compared to the control. Evaluating the addition effect of several components of loess such as $SiO_2$, $Fe_2O_3$, $Al_2O_3$, $CaCO_3$, $CaSO_4$ and $MgCO_3$ on the reduction of mycellial pellet size the higher concentration was supplied, the smaller size of pellet was formed except $Al_2O_3$. And the smallest pellet size was recorded at the concentration of 1.0% (W/V) magnesium carbonate.

Fundamental Cultural Characteristics of Chlorociboria spp. Native to Korea (한국에 자생하는 녹청균류의 기초 배양 특성)

  • Members of Chlorociboria are soft-rot ascomycetes that produce blue-green pigment. We investigated the growth characteristics of two Korean species of Chlorociboria, eight strains of Chlorociboria aeruginascens and Chlorociboria poutoensis, under various culture conditions (solid media, temperature, pH) and screened them for extracellular enzyme activity. Although the growth rate was slow, all tested strains of Chlorociboria spp. grew well on potato dextrose agar (PDA; 16.3~42.6 mm after 60 days) or Sabouraud dextrose agar (SDA), but not on malt extract agar (MEA). Compared with C. aeruginascens strains, C. poutoensis strains exhibited higher expression of blue-green pigments on both PDA and SDA media. The optimal temperature for mycelial growth was $20{\sim}25^{\circ}C$, and mycelial growth was lower at $30^{\circ}C$ than at $10^{\circ}C$. All strains tended to have increased mycelial growth as the incubation temperature increased in the range of 10 to $20^{\circ}C$. The optimal pH of potato dextrose broth (PDB) for mycelial growth varied according to the strain under static culture conditions. Maximum biomass production was obtained at pH 6.0 for NIFoS 579 ($114.3{\pm}5.1mg/60days$), but it maintained a stable pigment expression under a broad pH spectrum. The activities of both cellulase and laccase were observed in all tested strains of Chlorociboria spp. Enzyme activities of NIFoS 579 were remarkably higher than those of the other strains. From these results, we suggest that C. poutoensis NIFoS 579 is a potential candidate for use as a source of natural blue-green dye.

Inhibitory Effect of Phorbol 12-Myristate 13-Acetate on NO Production Induced by Interleukin-1 beta in Aortic Vascular Smooth Muscle Cells of Rats (혈관평활근세포에서 Phorbol 12-Myristate 13-Acetate의 전처리가 Interleukin-1β에 의한 Nitrite생성에 미치는 영향)

  • To examine the role of protein kinase C (PKC) in regulation of interleukin-1 beta (IL-1$\beta$)-induced iNOS expression, IL-1$\beta$-induced nitrite production was observed in cultured vascular smooth muscle (VSM) cells pretreated with phorbol 12-myristate 13-acetate (PMA) and phorbol 12,13-butyrate (PDB) as PKC activator; 4$\alpha$-phorbol-didecanoate (PDD) as PKC non-activator. Nitrite production induced by IL-1$\beta$ was increased by the presence of increasing concentration of PMA ranging from 2 to 200 nM. However, in VSM cells pretreated with PMA and PDB, IL-1$\beta$-induced $NO_2$ production was decreased in proportion to the duration of pretreatment, and most significantly decreased in pretreatment time of 24 hours. Using RT-PCR method, the expression of iNOS mRNA induced by IL-1$\beta$ was decreased in VSM cells pretreated with PMA 200 nM for 24 hours. These results suggest that decrease in IL-I$\beta$-induced nitrite production by the pretreatment of PMA result from inhibition of iNOS expression and the inhibition related to PMA-induced PKC down-regulation.

Molecular Cloning of cDNA Encoding a Putative Eugenol Synthase in Tomato (Solanum lycopersicum 'Micro-Tom') and Prediction of 3D Structure and Physiochemical Properties (토마토 'Micro-Tom' 과실의 eugenol synthase 유전자 클로닝, 단백질의 3차 구조 및 생리화학적 특성 예측)

  • Eugenol is a volatile compound synthesized by eugenol synthase in various plants and belongs to phenylpropene compounds. However, characteristics of eugenol synthase in tomato has not been known. Therefore, we cloned a full length cDNA of a putative eugenol synthase from tomato 'Micro-Tom' using rapid amplification of cDNA ends (RACE) technique and named a clone SlEGS. Open reading frame of SlEGS was 921bp long and its deduced amino acid sequence was 307bp. The BLAST analysis indicated that SlEGS shared high similarity with PhEGS1 (67.1%) and CbEGS2 (69.4%). Amino acid composition of SlEGS was determined by CLC genomics workbench tool and 3D structure of SlEGS was constructed by homology modeling using Swiss-PDB viewer and validated using PROCHECK and ProSA-web tool. In addition, the physiochemical properties of SlEGS was evaluated using ExPASy's ProtParam tool. Molecular weight was 33.93kDa and isoelectric point was 5.85 showing acidic nature. Other properties such as extinction coefficient, instability index, aliphatic index, and grand average hydropathy was also analyzed.

A Big Data Based Random Motif Frequency Method for Analyzing Human Proteins (인간 단백질 분석을 위한 빅 데이타 기반 RMF 방법)

  • Due to the technical difficulties and high cost for obtaining 3-dimensional structure data, sequence-based approaches in proteins have not been widely acknowledged. A motif can be defined as any segments in protein or gene sequences. With this simplicity, motifs have been actively and widely used in various areas. However, the motif itself has not been studied comprehensively. The value of this study can be categorized in three fields in order to analyze the human proteins using artificial intelligence method: (1) Based on our best knowledge, this research is the first comprehensive motif analysis by analyzing motifs with all human proteins in Protein Data Bank (PDB) associated with the database of Enzyme Commission (EC) number and Structural Classification of Proteins (SCOP). (2) We deeply analyze the motif in three different categories: pattern, statistical, and functional analysis of clusters. (3) At the last and most importantly, we proposed random motif frequency(RMF) matric that can efficiently distinct the characteristics of proteins by identifying interface residues from non-interface residues and clustering protein functions based on big data while varying the size of random motif.

  • In recent industrialization and development due to information and communication technology, modern women in modern society are exposed to physical and mental health due to numerous stresses. Popular inflammations are attributable to a decrease in lactic acid bacteria, frequent antibiotic use, and a decrease in immunity. It is necessary to develop products that are helpful and reflected. The inner care gel currently introduced on the market can increase beneficial bacteria and maintain a healthy y-zone. The inner gel contains a hydrogel component. 90% is made up of water, and other components act as support for supporting water and are formed through crosslinking between polymer chains. Hydroxyethyl cellulose (HEC) is a hydroxyethyl ethylenetel of cellulose. The purpose of use is to act as a binder, an emulsion stabilizer, a viscosity enhancer (water-soluble), and a film forming agent. CA (crosslinker) is a crosslinking agent and serves to bind. Hydrogel in the beauty field acts as a film forming agent that gently wraps around the skin by forming a thin film and serves as an emulsion stabilizer that helps to prevent separation of other raw materials. It also acts as a thickener by increasing viscosity in cosmetics. In addition, it is used for glucose monitoring, nursing care, cell transplantation, and wound treatment in the bio field. Currently, it is understood that no products using functional hydrogel have been released, so in this study, a Y zone care hydrogel solution was manufactured to find out the antibacterial properties of the functional hydrogel, and a new solution was developed. As a result, it was confirmed that the appropriate Ph was applied to the Y zone, and after culturing Candida albicans in PDB medium, all three products of the Y zone care hydrogel solution showed an antibacterial effect of 0.5-1.0mm

Web Services Based Biological Data Analysis Tool

  • Biological data and analysis tools are accumulated in distributed databases and web servers. For this reason, biologists who want to find information from the web should be aware of the various kinds of resources where it is located and how it is retrieved. Integrating the data from heterogeneous biological resources will enable biologists to discover new knowledge across the specific domain boundaries from sequences to expression, structure, and pathway. And inevitably biological databases contain noisy data. Therefore, consensus among databases will confirm the reliability of its contents. We have developed WeSAT that integrates distributed and heterogeneous biological databases and analysis tools, providing through Web Services protocols. In WeSAT, biologists are retrieved specific entries in SWISS-PROT/EMBL, PDB, and KEGG, which have annotated information about sequence, structure, and pathway. And further analysis is carried by integrated services for example homology search and multiple alignments. WeSAT makes it possible to retrieve real time updated data and analysis from the scattered databases in a single platform through Web Services.

Isolation and Morphological Characterization of Monilinia sp. KV-27 Associated with Apple Anthracnose of Fuji Apples in Korea

  • This study was undertaken to isolate and to identify a fungal pathogen Monilinia sp. KV-27 associated with apple anthracnose. Rotted Fuji apples were used for the isolation of the fungus. The infected tissues were sterilized with 70% ethanol, washed with sterilized distilled water and were transferred to 50 ml containing potato dextrose broth (PDB) flasks. The peripheral hyphae of the fungal colony which developed from the infected tissues were isolated on to potato dextrose agar (PDA). On PDA plates the fungus grew well at $25^{\circ}C$ and occupied more than half of a 9 cm petri dish within 5 days. The fungal cultures on PDA were used for morphological observation and identification of the fungus. Conidiophores were produced on the gray to whitish sporodochial structures scattered on PDA plates. These conidiophores gave rise to chains of conidia, which were branched and easily detached in water. These structures were dark brown to black and consisted of hyphal masses. Conidia produced on PDA plates were hyline or light colored, lemon shaped or ellipsoidal ($10-13{\times}8.5-11{\mu}m$) in size.
