Effects of Red Ginseng Saponin on Normal and Scopolamine-induced Memory Impairment of Mice in Passive Avoidance Task (정상 및 기억손상 유도 동물의 수동회피반응에 대한 홍삼 사포닌의 효과)

  • This study was performed to examine the effect of red ginseng total saponin and extract on memory in mice using one trial step-down type passive avoidance method. Red ginseng total saponins (No. 1: PD/PT ratio=1.24, No. 2: PD/PT ratio=1.47) were prepared with the different mixing ratio by using the parts of red ginseng. In single administration of total saponin No. 1 (100 mg/ kg, bw) or No. 2 (50 mg/kg, bw) increased the latency time as compared with control group but was not statistically significant. Treatment of total saponin No. 1 (50 mg/kg, bw) for 10 days produced an increase of latency time but was not statistically significant. In scopolamine-induced memory deficient mice total saponin No. 1 (50 mg/kg, bw) and No. 2 (100 mg/kg, bw) significantly improved the latency time. These results show that red ginseng total saponin may improve the memory of sco-polamine-induced memory deficient mice and have nootropic activity.

Pseudoprogression and Pseudoresponse in the Management of High-Grade Glioma : Optimal Decision Timing According to the Response Assessment of the Neuro-Oncology Working Group

  • Objective : We evaluated pseudoprogression (PsPD) following radiation therapy combined with concurrent temozolomide (TMZ), and we assessed pseudoresponse following anti-angiogenic therapy for patients with recurrent disease using the Response Assessment of the Neuro-Oncology Working Group. Methods : Patients who were pathologically confirmed as having high-grade glioma received radiotherapy with concurrent TMZ followed by adjuvant TMZ. Bevacizumab (Avastin) with CPT-11 were used as a salvage option for cases of radiologic progression. Magnetic resonance imaging (MRI) was routinely performed 1 month after concurrent radiochemotherapy (CRT) and every 3 months thereafter. For cases treated with the bevacizumab-containing regimen for progressive disease, MRI was performed every 2 months. Results : Of 55 patients, 21 (38%) showed radiologic progression within 4 weeks after CRT. Of these patients, 16 (29%) showed progression at second post-CRT MRI (etPD) and five (9%) showed improvement (PsPD). Seven of thirty-four initially non-progressed patients showed progression at the second post-CRT MRI (ltPD). No difference in survival was observed between the etPD and ltPD groups (p=0.595). Five (50%) of ten patients showed a radiological response after salvage bevacizumab therapy. Four of those patients exhibited rapid progression immediately after discontinuation of the drug (drug holiday). Conclusion : Twelve weeks following treatment could be the optimal timing to determine PsPD or true progression. MRI with gadolinium enhancement alone is not sufficient to characterize tumor response or growth. Clinical correlation with adequate follow-up duration and histopathologic validation may be helpful in discriminating PsPD from true progression.

TNF-α Inhibitor Reduces Odontoclast Formation in Diabetes Rats with Ligature-Induced Periodontitis

  • To determine the effect of the tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) in odontoclast formation, we administrated a $TNF-{\alpha}$ inhibitor in rats with diabetes rats with periodontitis. The rats included in the study were divided into three groups: control rats without diabetes or periodontitis (the C group), rats with periodontitis and diabetes (the PD group), and rats with periodontitis and diabetes treated by infliximab, the TNF inhibitor (the PD+infliximab group). The PD and PD+infliximab groups received intravenous administrations of streptozotocin (STZ, 50 mg/kg) to induce diabetes. After 7 days of STZ injections, the mandibular first molars were ligatured to induce periodontitis. The PD+infliximab group was intrapenitoneally administrated by infliximab (5 mg/kg). On days 3 and 20 after the ligature administration, odontoclast formation along root surfaces was evaluated by tartrate resistant acid phosphatase (TRAP) staining and cathepsin K immunohistochemistry. On day 3, the number of TRAP- and cathepsin K-positive cells increased more so in the PD group than in the C group. The PD+infliximab group showed a lower number of positive cells than the PD group. There was no difference in all the groups on day 20. On day 3, the cathepsin-K positive multinucleated and mononucleated cells were higher in the PD group than in the C group. The number of cathepsin-K positive multinucleated cells was lower in the PD+infliximab group than in the PD group. The PD group showed more cathepsin K-positive cells in the furcation and distal surfaces than the c group. The Cathepsin K-positive cells of the PD+infliximab group were lower than that of the PD group in furcation. These results suggest that $TNF-{\alpha}$ stimulates odontoclast formation in diabetes with periodontitis.

Modified Atmosphere Packaging of Minimally Processed Cut Garlic (최소가공된 절단 마늘의 환경기체조절포장)

  • There is a need in food industry to store minimally processed garlic for long time to have it be used just at the time of demand for final product processing. Optimal modified atmosphere packaging is expected to slow down the quality change extending its storage life. In order to find optimal packaging conditions, plastic films of different gas permeability properties (low density polyethylene (LDPE) $30{\mu}m$, polyolefin $50{\mu}m$ (PD 900), polyolefin $20{\mu}m$ (PD 941)) were used for packaging 400 g of minimally processed garlic. Perforated LDPE packages were prepared as control. The packaged products were stored at $1{\pm}1^{\circ}C$ for 52 days. Package treatments were compared in weight loss, decay, surface color, hardness and soluble solid content. While control package had normal atmosphere of air, LDPE, PD 900 and PD 941 packages attained internal concentration of $O_2$ 4.6% / $CO_2$ 12%, $O_2$ 0.9% / $CO_2$ 21% and $O_2$ 0.5% / $CO_2$ 13% after 45 days, respectively. Control packaging had rapid weight loss with high mold decay and great surface color change in 45 days. In PD 900 film packages of lowest gas permeability, the fresh-cut garlic could be stored without mold decay for 52 days. Except control packaging, there were no significant differences in surface color, hardness and soluble solid content among package treatments.

Ridge Formation by Dry-Etching of Pd and AlGaN/GaN Superlattice for the Fabrication of GaN Blue Laser Diodes

  • In these days, the desire for the precise and tiny displays in mobile application has been increased strongly. Currently, laser displays ranging from large-size laser TV to mobile projectors, are commercially available or due to appear on the market [1]. In order to achieve a mobile projectors, the semiconductor laser diodes should be used as a laser source due to their size and weight. In this presentation, the continuous etch characteristics of Pd and AlGaN/GaN superlattice for the fabrication of blue laser diodes were investigated by using inductively coupled $CHF_3$ and $Cl_2$ -based plasma. The GaN laser diode samples were grown on the sapphire (0001) substrate using a metal organic chemical vapor deposition system. A Si-doped GaN layer was grown on the substrate, followed by growth of LD structures, including the active layers of InGaN/GaN quantum well and barriers layer, as shown in other literature [2], and the palladium was used as a p-type ohmic contact metal. The etch rate of AlGaN/GaN superlattice (2.5/2.5 nm for 100 periods) and n-GaN by using $Cl_2$ (90%)/Ar (10%) and $Cl_2$ (50%)/$CHF_3$ (50%) plasma chemistry, respectively. While when the $Cl_2$/Ar plasma were used, the etch rate of AlGaN/GaN superlattice shows a similar etch rate as that of n-GaN, the $Cl_2/CHF_3$ plasma shows decreased etch rate, compared with that of $Cl_2$/Ar plasma, especially for AlGaN/GaN superlattice. Furthermore, it was also found that the Pd which is deposited on top of the superlattice couldn't be etched with $Cl_2$/Ar plasma. It was indicating that the etching step should be separated into 2 steps for the Pd etching and the superlattice etching, respectively. The etched surface of stacked Pd/superlattice as a result of 2-step etching process including Pd etching ($Cl_2/CHF_3$) and SLs ($Cl_2$/Ar) etching, respectively. EDX results shows that the etched surface is a GaN waveguide free from the Al, indicating the SLs were fully removed by etching. Furthermore, the optical and electrical properties will be also investigated in this presentation. In summary, Pd/AlGaN/GaN SLs were successfully etched exploiting noble 2-step etching processes.

Purification and enzymatic properties of a peroxidase from leaves of Phytolacca dioica L. (Ombú tree)

  • A peroxidase (PD-cP; 0.47 mg/100 g leaves) was purified from autumn leaves of Phytolacca dioica L. and characterized. PD-cP was obtained by acid precipitation followed by gel-filtration and cation exchange chromatography. Amino acid composition and N-terminal sequence of PD-cP up to residue 15 were similar to that of Spinacia oleracea (N-terminal pairwise comparison showing four amino acid differences). PD-cP showed a molecular mass of approx. 36 kDa by SDS-PAGE, pH and temperature optima at 3.0 and $50.0^{\circ}C$, respectively and seasonal variation. The Michaelis-Menten constant ($K_M$) for $H_2O_2$ was 5.27 mM, and the velocity maximum ($V_{max}$) $1.31\;nmol\;min^{-1}$, while the enzyme turnover was $0.148\;s^{-1}$. Finally, the presence of $Ca^{2+}$ and $Mg^{2+}$ enhanced the PD-cP activity, with $Mg^{2+}$ 1.4-fold more effective than $Ca^{2+}$.

Voice Handicap Index and Voice-Related Quality of Life in Idiopathic Parkinson's Disease (파킨슨병 환자의 음성장애지수 및 음성관련 삶의 질 연구)

  • Objectives : The purpose of this study is to evaluate the voice handicaps of the idiopathic Parkinson's Diseases (PD) and their voice-related quality of life. Methods : Voice handicap index-10 (VHI-10) and Voice related Quality of Life were completed by 17 idiopathic PD patients, and Unified Parkinson's Disease Rating Scale (UPDRS) part I, II, III were assessed. The relations between VHI-10, VRQOL and UPDRS scores were analysed. Results : VHI-10 score of PD patients was $14.35{\pm}8.07$ and VRQOL total score of PD patients was $59.12{\pm}20.25$, social-emotional $59.93{\pm}20.50$, physical function $58.58{\pm}21.77$. There were significant relations between VHI-10, VRQOL score and UPDRS II (activities of daily living). Conclusions : These results suggest that voice impairments affect the daily living of PD patients and their quality of lives.

  • Cancer growth and progression are associated with immune suppression. Cancer cells have the ability to activate different immune checkpoint pathways that harbor immunosuppressive functions. Monoclonal antibodies that target immune checkpoints provided an immense breakthrough in cancer therapeutics. Among the immune checkpoint inhibitors, PD-1/PD-L1 and CTLA-4 inhibitors showed promising therapeutic outcomes, and some have been approved for certain cancer treatments, while others are under clinical trials. Recent reports have shown that patients with various malignancies benefit from immune checkpoint inhibitor treatment. However, mainstream initiation of immune checkpoint therapy to treat cancers is obstructed by the low response rate and immune-related adverse events in some cancer patients. This has given rise to the need for developing sets of biomarkers that predict the response to immune checkpoint blockade and immune-related adverse events. In this review, we discuss different predictive biomarkers for anti-PD-1/PD-L1 and anti-CTLA-4 inhibitors, including immune cells, PD-L1 overexpression, neoantigens, and genetic and epigenetic signatures. Potential approaches for further developing highly reliable predictive biomarkers should facilitate patient selection for and decision-making related to immune checkpoint inhibitor-based therapies.

A Study on the Shear Bond Strength of the Reinforced Composite Resin to Dental Alloys (강화형 복합레진과 수종의 치과용 합금간의 전단결합강도에 관한 연구)

  • The reinforced composte resin as the esthetic operative material continuously has been studied because the porcelain fused metal prosthesis is widely used for its excellent esthetics, rigidity and marginal integrity, but it has low fracture resistance against the tensile strength and stress, attrition of the opposite teeth. The reinforced composite resin is well adapt with the dental alloy but it is low the shear bond strength with the dental alloy vs the porcelain fused metal prosthesis, and then has been studied continuously. The purpose of the study was to examine how metal was the higher shear bond strength among the dental alloy was used to the reinforced composite resin and to find the effect that the particle size of sandblasting influenced the shear bond strength. We built up the reinforced composite resin with 4 mm in diameter, 3 mm in height on circular alloy with 5 mm in diameter, 2 mm in height. Type II gold, type IV gold, and Ag-Pd alloy was used as alloys and $50{\mu}m$, $110{\mu}m$, $250{\mu}m$ of the particle size was sandblasted at each alloy in bonding between alloy and resin. We made 90 secimens of 10 per each group and we measured the shear bond strength using the Instron($M100EC^{(R)}$, Mecmesin Co., England). The obtained results were as follows : 1. In comparison among each alloys, Ag-Pd alloy had the highest shear bond strength and the shear bond strength was decreased significantly in the sequence of the type II gold and type IV gold(P<0.001). 2. In comparison according to the size of sandblasting particle, (1) In Ag-Pd alloy, shear bond strength was decreased in the sequence of $110{\mu}m$, $250{\mu}m$, $50{\mu}m$ and there were significant difference in all the group. (P<0.05) (2) In type II gold, it was decreased in the sequence of $250{\mu}m$, $50{\mu}m$, $110{\mu}m$ and there were significant difference. (P<0.05) (3) In type IV gold, it was decreased in the sequence of $110{\mu}m$, $50{\mu}m$, $250{\mu}m$. There were significant difference between the group of $110{\mu}m$ and $50{\mu}m$, the group of $110{\mu}m$ and 250, but there were no significant difference in the group of $50{\mu}m$ and $250{\mu}m$. 3. The highest shear bond strength according to the size of sandblasting particle was $110{\mu}m$ in Ag-Pd alloy and type IV gold, $250{\mu}m$ in type II gold.

  • Transmission equipment is mainly used for the XLPE (cross-linked polyethylene) insulation cable for ultra high voltage power to minimize power loss. The experiment examined the partial discharge characteristics according to the insertion of the bar electrode and needle electrode into the XLPE specimen and the air voids. XLPE insulation cable manufactured by T. company and tungsten electrode material by K. company were used for specimens, by adhering conductive tape on the semi-conductive material of the lower electrode of XLPE specimen with the dimension of $16{\times}40{\times}30$ [mm] was used as negative electrode. In order to investigate the PD with ${\phi}$-q-n of XLPE specimen according to the electrode shape and the size of air voids. we examined the PD by varying the voltage after applying voltage of 3~20 kV on the electrode. Therefore, it was confirmed from the result of PD characteristics of specimen that the larger the air void than the gap between electrode (+) and electrode(-), the larger effect on the discharge when the bar electrode and needle electrode inserted into XLPE, and the closer the distance between the insulation and the needle electrode, the faster insulation breakdown.
