The present study investigated the role of ERK in the onset of mechanical and cold allodynia in a rat model of compression of the trigeminal ganglion by examining changes in the air-puff thresholds and number of scratches following the intracisternal injection of PD98059, a MEK inhibitor. Male Sprague Dawley rats weighing between 250 and 260 g were used. Under anesthesia, the rats were mounted onto a stereotaxic frame and received 4% agar ($10\;{\mu}l$) solution to compress the trigeminal ganglion. In the control group, the animals were given a sham operation without the application of agar. Changes in behavior were examined at 3 days before and at 3, 7, 10, 14, 17, 21, 24, 30, and 40 days after surgery. Compression of the trigeminal ganglion significantly decreased the air-puff thresholds. Mechanical allodynia was established within 3 days and persisted over postoperative day 24. To evaluate cold allodynia, nociceptive scratching behavior was monitored after acetone application on the vibrissa pad of the rats. Compression of the trigeminal ganglion was found to produce significant cold allodynia, which persisted for more than 40 days after surgery. On postoperative day 14, the intracisternal administration of $1\;{\mu}g$ or $10\;{\mu}g$ of PD98059 in the rat model significantly decreased the air-puff thresholds on both the ipsilateral and contralateral side. The intracisternal administration of $10\;{\mu}g$ of PD98059 also significantly alleviated the cold allodynia, compared with the vehicle-treated group. These results suggest that central ERK plays an important role in the development of mechanical and cold allodynia in rats with compression of the trigeminal ganglion and that a targeted blockade of this pathway is a potential future treatment strategy for trigeminal neuralgia-like nociception.
Purpose: This study examined the effects of sagittal spinopelvic alignment on the clinical parameters, motor symptoms, and respiratory function in patients with mild to moderate Parkinson's disease (PD). Methods: This study was a prospective assessment of treated patients (n=28, Hoehn and Yahr (H&Y) stage 2-3) in a PD center. Twenty-eight subjects ($68.5{\pm}5.7yrs$) participated in this study. The clinical and demographic parameters, including age, sex, symptoms duration, treatment duration, and H&Y stage, were collected. Kinematic analysis was conducted in the upright standing posture with a motion capture system. A pulmonary function test (PFT) was performed in the sitting position using a spirometer. The motor symptoms were assessed on part III of the movement disorder society sponsored version of the unified Parkinson's disease rating scale (MDS-UPDRS). SPSS 18.0 was used to analyze the collected data. Results: The exceeding 12 degrees group of the lower trunk showed significantly higher on the clinical parameters than the below 12 degrees group. In addition, the exceeding 12 degrees group of the lower trunk showed a significantly lower forced expiratory volume at one second (FEV1) / forced vital capacity (FVC) (%) and 25-75% forced mid-expiratory flow (FEF) (L/s) than in the below group. On the other hand, there was no difference in the upper trunk and the cervical pelvis between the groups. Conclusion: These findings suggest that the sagittal balance in the lower trunk is related to the clinical parameters and respiratory function, but not the motor symptoms in patients with mild to moderate PD.
산화제로 질산 또는 하이포아염소산나트륨을 사용하는 염산침출에 의하여 자동차 폐촉매로부터 백금, 필라듐 및 로듐 등의 백금족 금속을 추출하는 연구를 수행하였다. 산화제의 종류 및 주입량, 반응시간, 광액농도가 백금족 금속의 추출에 미치는 영향을 조사하였다. 침출액으로부터 백금족 금속은 알루미늄을 환원제로 사용하는 세멘테이션법으로 회수하였다. 하이포아염소산나트륨을 산화제로 사용하였을 때 백금족 금속의 추출율이 더 높았으며 최적침출조건은 염산농도 8M, 하이포아염소산나트륨 첨가량 1.4mole, 침출온도 $90^{\circ}C$, 침출시간 180분, 광액농도 400 g/L 이었다. 이 조건에서 백금, 필라듐 그리고 로듐의 추출은 각각 96.1%, 93.6%, 77.3%이었다. 백금족 금속의 28당량인 2.0g의 알루미늄을 첨가하였을 때 백금, 필라듐 그리고 로듐은 각각 98%, 98.8% 그리고 65.3%가 환원되었다.
Portulaca oleracea L. is known to have many biological benefits such as anti-oxidant, anti-inflammatory, anti-allergic and anti-tumor. The objective of this study is to explore the neuroprotective effect of P. oleracea L. against glutamate-induced oxidative stress in mouse hippocampal HT22 cells. P. oleracea L. 70% ethanol extract and solvent fractions have the potent neroprotective effects on glutamate-induced nerotoxicity by induced the expression of heme oxygenase (HO)-1 in HT22 cells. Especially, ethyl acetate fraction showed higher protective effect. In HT22 cell, P. oleracea L. treatment with ERK inhibitor (PD98059) and c-JUN N-terminal kinase (JNK) inhibitor (SP600125) reduced P. oleracea L. ethyl acetate fraction induced HO-1 expression and P. oleracea L. ethyl acetate fraction also increased ERK and JNK phosphorylation. Furthermore, we found that treatment of P. oleracea L. caused the nuclear accumulation of Nrf2. In conclusion, the ethyl acetate fraction of 70% ethanol extract of P. oleracea L. significantly protect glutamate-induced oxidative damage by induction of HO-1 via Nrf2, ERK and JNK pathway in mouse hippocampal HT22. Taken together these finding suggest that P. oleracea L. ethyl acetate fraction is good source for taking active compounds and may be a potential therapeutic agent for brain disorder that induced by oxidative stress and neuronal damage.
폼페병(PD, Pompe disease), 2형 당원축적병(Glycogen storage disease type II)은 보통염색체 열성 질환으로 용해소체 효소인 acid maltase (acid α-glucosidase, GAA) 결핍으로 인한 대사근육병이다. 한 종류의 효소 결핍에 의한 질환이지만, GAA의 결핍정도와 유전자형에 따라 임상양상이 다르게 나타난다. 발병 시기에 따라 크게 영아형 폼페병(infantile onset Pompe disease, IOPD), 성인형 폼페병(late onset Pompe disease, LOPD)으로 나눌 수 있다. 저자들은 신생아기 때 호흡곤란증후군을 진단받고 치료받은 후 잠시 혈중 CK 증가가 확인되었으나 다른 임상증상 없이 경과 관찰 후 호전되었고, LSD 스크리닝 검사 결과 상 GAA 수치가 0.58 umol/h/L로 감소되어 있음을 확인한 1례에 대해 보고하고자 한다. 해당 환아를 PD 의증으로 고려하여 시행한 GAA enzyme essay 상 total GAA level은 16.2 nmol/2hr/mg protein, GAA with acarbose level은 2.0 nmol/2hr/mg protein, acarbose/total level의 비율은 12.0%로 낮은 수치를 확인하였다. 환아의 유전자 검사 상 exon #4에서 두개의 likely pathogenic heterozygous mutation인 c.752C>T (p.Ser251Leu), c.761C>T (p.Ser254Leu), exon #12에서 heterozygous mutation인 c.1726G>A (p.Gly576Ser), exon #15에서 heterozygous mutation인 c.2065G>A (p.Glu689 Lys)이 확인되었다. 환아의 7세 오빠는 유전자 검사에서 정상으로 확인되었고, 아버지는 환아에서 동일하게 확인된 exon #12에서 heterozygous mutation인 c.1726G>A(p.Gly576Ser), exon #15에서 heterozygous mutation인 c.2065G>A (p.Glu689Lys)이, 어머니에서는 exon #4에서 두 개의 likely pathogenic heterozygous mutation인 c.752C>T (p.Ser251Leu), c.761C>T (p.Ser254Leu)가 확인되었다. Pathogenic한 유전자 두개가 있으면 보통 환자로 인식될 수 있으나 이 환아에서처럼 pathogenic한 유전자 두 개가 있더라도 cis 형태로 하나처럼 움직인 경우 PD 환자가 아니라 carrier 일 수 있다는 것을 경험한 증례였다. 이에 PD 환아의 유전검사 결과를 해석할 시 pathogenic variant 유전자가 두 개일지라도 혹시 cis 형태로 하나의 유전자인지를 확인하여, pseudodeficiency나 potential LOPD carrier일 수 있는 점을 고려하며 여러 임상 양상을 취합하여 진료를 시행하는 것이 필요하다.
Objective: To evaluate the effects of curcumin on matrixmetalloproteinase-9 (MMP-9) and invasion ability induced by transforming growth factor-${\beta}1$ (TGF-${\beta}1$) in MDA-MB-231 cells and potential mechanisms. Methods: Human breast cancer MDA-MB-231 cells were used with the CCK-8 assay to measure the cytotoxicity of curcumin. After treatment with 10 ng/ml TGF-${\beta}1$, with or without curcumin (${\leq}10{\mu}M$), cell invasion was checked by transwell chamber. The effects of curcumin on TGF-${\beta}1$-stimulated MMP-9 and phosphorylation of Smad2, extracellular-regulated kinase (ERK), and p38 mitogen activated protein kinases (p38MAPK) were examined by Western blotting. Supernatant liquid were collected to analyze the activity of MMP-9 via zymography. Following treatment with PD98059, a specific inhibitor of ERK, and SB203580, a specific inhibitor of p38MAPK, Western blotting and zymography were employed to examine MMP-9 expression and activity, respectively. Results: Low dose curcumin (${\leq}10{\mu}M$) did not show any obvious toxicity to the cells, while $0{\sim}10{\mu}mol/L$ caused a concentration-dependent reduction in cell invasion provoked by TGF-${\beta}1$. Curcumin also markedly inhibited TGF-${\beta}1$-regulated MMP-9 and activation of Smad2, ERK1/2 and p38 in a dose- and time-dependent manner. Additionally, PD98059, but not SB203580, showed a similar pattern of inhibition of MMP-9 expression. Conclusion: Curcumin inhibited TGF-${\beta}1$-stimulated MMP-9 and the invasive phenotype in MDA-MB-231 cells, possibly associated with TGF-${\beta}$/Smad and TGF-${\beta}$/ERK signaling.
본 연구에서는 지점빈도분석과 지역빈도분석을 이용하여 확률홍수량을 산정 하였다. 지점빈도 분석은 Annual Maximum Series(AMS) 및 Partial Duration Series(PDS)를 이용하여 자료를 추출하고 각 자료에 적합한 확률분포를 이용하여 확률홍수량을 산정하였다. 그러나 AMS를 이용한 확률홍수량의 산정은 표본의 개수가 부족하면 이에 따른 변동성(variability)이 커지게 되는 단점이 존재하며, PDS를 사용하면 임계값(threshold)에 따른 주관적 영향이 결과에 반영되는 단점이 존재하는 것으로 알려져 있다. 따라서 본 연구에서는 PDS를 사용하는 경우의 단점을 해결하기 위해 연 1.7회의 발생횟수를 갖는 자료를 추출하고 몬테카를로 모의시험을 통하여 주관적 영향을 제거하였다. 또한 두 가지 방법에 의해 산정된 확률홍수량의 비교검토를 위해 지역빈도분석을 수행하였다. 유역의 면적과 일평균강우량으로부터 확률홍수량을 산정할 수 있는 것으로 알려진 Bayesian-Generalized Least Square(B-GLS) 방법을 이용하여 확률홍수량을 산정하였다. 최종적으로 안양천 유역의 13개 소유역에 대한 세 가지 방법에 의해 산정된 확률홍수량을 비교 검토한 결과, 특정한 방법이 항상 우수하다는 결론은 얻을 수 없었으나 각 유역별로 AMS가 가장 크고 B-GLS가 가장 작은 확률홍수량을 갖는 경향을 나타내었다.
Ammonium pyrrolidine dithiocarbamate(APDC)를 착화제로 사용하여 해수중 흔적량 비스무트, 카드뮴, 인듐을 동시에 용매추출하여 정량하는 방법을 연구하였다. 최적조건을 찾기위하여, 시료용액의 pH, 착화제의 양, 흔들어주는 시간, 유기용매의 종류 등에 대하여 조사하였고 역추출효율도 검토하였다. 해수시료 200mL를 취하여, pH를 4.0으로 조정하고 1% APDC 5.0mL를 첨가하고, MIBK 10.0mL로 35분 동안 흔들어 추출한다. 그리고 0.05M 수산화나트륨용액 10mL로 유기층의 HPDC를 세척한다. 유기층을 150.mu.g/mL Pd(II)를 포함하는 4M 질산으로 5분동안 흔들어 분석원소를 역추출한다. 팔라윰이 포함된 4M질산은 역추출효율을 증가시키고 흡광도 측정에서 매트릭스를 개선하여 감도를 증가시켰다. 바탕 흡광도에 대한 표준편차의 3배에 해당하는 농도로서의 검출한계는 Bi(III) :0.038, Cd(II) : 0.0057, In(III) : 0.023 ng/mL이었다. 동해 시료2 가지를 취하여 본방법으로 정량한 결과 Bi(III), In(III)은 검출한계 이하였고, Cd(II)는 0.018과 0.016 ng/mL이었다. 해수시료에 일정량 원소를 가해서 구한 회수율은 모두 90% 이상으로 아칼리금속 매트릭스에서 흔적량 원소의 동시분석에 이 방법이 응용될수 있음을 알 수 있었다.
바이오디젤과 바이오디젤을 기반으로 생산된 neopentyl polyol ester (NPE)계 윤활유 베이스, 디젤유 윤활유와 석유디젤을 분석대상으로 하여 독성을 평가하였다 조류를 이용하여 수용액 상에서 화학물질의 독성시험을 조사하는 OECD 201에 근거하여 Chlorella vulgaris를 96시간 동안 배양하면서 독성시험 분석한 결과, 바이오디젤과 NPE계 윤활유 베이스의 반수저해농도 $(EC_{50})$ 값은 각각 84와 69mg/L로서 두 물질이 독성 이 적은 경독성 (slightly toxic)물질인 것으로 판명되었다. 디젤유 윤활유와 석유디젤의 경우 반수저해농도 $(EC_{50})$ 값이 각각 42와 24 mg/L로서 중등독성 (moderately toxic) 물질로 판정 되었다.
The purpose of the present study was to investigate the pharmacokinetics of PEG-hemoglobin SB 1, a modified bovine hemoglobin with polyethylene glycol, after its single and multiple administration in beagle dogs. For this purpose, the analytical method of free hemoglobin in the plasma was developed and validated. Excellent linearity ($r^2$=0.999) was observed in the calibration curve data, with the limit of quantification of 0.005 g/dL. The precision and the deviation of the theoretical values for accuracy were always within $\pm$15% in both the between-and the within-day results. The method was tested by measuring the plasma concentrations following intravenous administration to beagle dogs and was shown to be suitable for pharmacokinetic studies. In a single dose study, the plasma half-life (t$_{1}$2/) increased and the total body clearance (Cl$_{t}$) decreased with the dose (i.e., 0.017 to 0.75 gHb/kg as PEG-hemoglobin SB1) in both sexes. The volume of distribution at steady-state (Vd$_{ss}$ ) showed no difference with the dose. In contrast, the values of t$_{1}$2/, CL$_{t}$ and the area under the plasma concentration-time curve (AUC) after the multiple dose were significantly different from those of the single dose administration. The values of t$_{1}$2/ in the multiple administration were about two times higher-than that of the single dose. As a result, t$_{1}$2/ of hemoglobin after the administration of PEG-hemoglobin SB1 was about 15-30 h, indicating the PEG modification of the hemoglobin lead to a prolongation of plasma concentration of the protein. Therefore, these observations suggested that the PEG modification of hemoglobin is potentially applicable in the hemoglobin-based therapeutics.tics.
