Enhanced Anti-tumor Reactivity of Cytotoxic T Lymphocytes Expressing PD-1 Decoy

  • Programmed death-1 (PD-1) is a strong negative regulator of T lymphocytes in tumor-microenvironment. By engaging PD-1 ligand (PD-L1) on tumor cells, PD-1 on T cell surface inhibits anti-tumor reactivity of tumor-infiltrating T cells. Systemic blockade of PD-1 function using blocking antibodies has shown significant therapeutic efficacy in clinical trials. However, approximately 10 to 15% of treated patients exhibited serious autoimmune responses due to the activation of self-reactive lymphocytes. To achieve selective activation of tumor-specific T cells, we generated T cells expressing a dominant-negative deletion mutant of PD-1 (PD-1 decoy) via retroviral transduction. PD-1 decoy increased IFN-γ secretion of antigen-specific T cells in response to tumor cells expressing the cognate antigen. Adoptive transfer of PD-1 decoy-expressing T cells into tumor-bearing mice potentiated T cell-mediated tumor regression. Thus, T cell-specific blockade of PD-1 could be a useful strategy for enhancing both efficacy and safety of anti-tumor T cell therapy.

PD-L1 Targeted Immunoliposomes with PD-L1 siRNA and HDAC Inhibitor for Anti-Lung Cancer Immunotherapy

  • Immunotherapy, which uses an immune mechanism in the body, has received considerable attention for cancer treatment. Suberoylanilide hydroxamic acid (SAHA), also known as a histone deacetylase inhibitor (HDACi), is used as a cancer treatment to induce active immunity by increasing the expression of T cell-induced chemokines. However, this SAHA treatment has the disadvantage of causing PD-L1 overexpression in tumor cells. In this study, we prevented PD-L1 overexpression by blocking the PD-1/PD-L1 pathway using PD-L1 siRNA. We designed two types of liposomes, the neutral lipid 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholin (POPC) for SAHA, and 1,2-dioleoyl-3-trimethylammoniumpropane (DOTAP) for siRNA. To effectively target PD-L1 in cancer cells, we conjugated PD-L1 antibody with liposomes containing SAHA or PD-L1 siRNA. These immunoliposomes were also evaluated for cytotoxicity, gene silencing, and T-cell-induced chemokine expression in human non-small cell lung cancer A549 cells. It was confirmed that the combination of the two immunoliposomes increased the cancer cell suppression efficacy through Jurkat T cell induction more than twice compared to SAHA alone treatment. In conclusion, this combination of immunoliposomes containing a drug and nucleic acid has promising therapeutic potential for non-small-cell lung carcinoma (NSCLC).

Synthesis of Pd and Pt Based Low Cost Bimetallic Anode Electrocatalyst for Glycerol Electrooxidation in Membraneless Air Breathing Microfluidic Fuel Cell

  • The different weight ratios of Pd to Pt, i.e., 16:4, 10:10, 4:16 in Pd-Pt/C and Pd (20 wt. %) /C electrocatalysts with low metal loading were synthesized for glycerol electrooxidation in an air breathing microfluidic fuel cell (MFC). The cell performance on Pd-Pt (16:4)/C anode electrocatalyst was found best among all the electrocatalysts tested. The single cell when tested at a temperature of 35℃ using Pd-Pt (16:4)/C, showed maximum open circuit voltage (OCV) of 0.70 V and maximum power density of 2.77 mW/㎠ at a current density of 7.71 mA/㎠. The power density increased 1.45 times when cell temperature was raised from 35℃ to 75℃. The maximum OCV of 0.78 V and the maximum power density of 4.03 mW/㎠ at a current density of 10.47 mA/㎠ were observed at the temperature of 75℃. The results of CV substantiate the single cell performance for various operating parameters.

세르톨리세포 관찰을 위한 PA-TCH-SP-PD 염색의 적용 (Application of Periodic Acid Thiocarbohydrazide Silver Proteinate Physical Development ( PA-TCH-SP-PD) Stain to Observation of Sertoli Cell)

  • 본 연구는 Sertoli 세포 column 과 Sertoli 세포돌기 미세구조의 연구를 위해 당질검출을 위한 PA-TCH-SP, -PD 투과전자현미경용 특수염색을 정세관에 적용한 후 각 정세포와 Sertoli 세포를 관찰하고 비교하여 이 분야 연구에 본염색법의 적용 가능성을 검토하였다. PA-TCH-SP, -PD 특수염색을 개의 정세관에 적용하여 투과전자현미경으로 관찰하였던 바, Sertoli 세포 column 과 돌기 부위에 수많은 반응과립이 관찰되었으며 정세관 기저부의 정조세포질에서도 약간의 반응과립이 관찰되었다. 이와 같은 반응과립은 각 정세포의 핵, 정모세포질, 정자세포질 및 잔류체에는 존재하지 않아 반응과립이 존재하는 Sertoli 세포 colunm 과 돌기 및 정자세포질에 침투된 돌기들을 명확히 구분할 수 있었다. 따라서 PA-TCH-SP, -PD 염색법은 투과전자현미경하에서 Sertoli 세포 colunm 과 돌기의 형태학적 및 기능학적 연구에 대단히 유용한 염색법으로 사료된다.

PD98059 Induces the Apoptosis of Human Cervical Cancer Cells by Regulating the Expression of Bcl2 and ERK2

  • PD98059 is the specific inhibitor of extracellular signaling-regulated kinase (ERK) kinase (MEK). ERK is involved in a mitogen-activated protein kinase (MAPK) cascade controlling cell growth and differentiation. Although the inhibition of ERK is known to induce cell death in various cell lines, this effect is still controversial and the role of PD98059 on the death of HeLa $S_3$ cells, a subclone of the cervical cancer cell line, is not well understood. The apoptosis of HeLa $S_3$ cells increased after the treatment of 50 ${\mu}M$ PD98059. The induction of apoptosis by PD98059 was occurred in a time- and a dose-dependent manners. The expression of Bcl-2 was reduced in accordance with decrease of ERK2 expression. Taken together, these results indicate that PD98059 has a cytotoxicity in HeLa $S_3$ cells and it may be used as a potential target for the treatment of cervical cancer.

Systemic and Cell-Type Specific Profiling of Molecular Changes in Parkinson's Disease

  • Parkinson's disease (PD) is a complicated neurodegenerative disorder although it is oftentimes defined by clinical motor symptoms originated from age dependent and progressive loss of dopaminergic neurons in the midbrain. The pathogenesis of PD involves dopaminergic and nondopaminergic neurons in many brain regions and the molecular mechanisms underlying the death of different cell types still remain to be elucidated. There are indications that PD causing disease processes occur in a global scale ranging from DNA to RNA, and proteins. Several PD-associated genes have been reported to play diverse roles in controlling cellular functions in different levels, such as chromatin structure, transcription, processing of mRNA, translational modulation, and posttranslational modification of proteins. The advent of quantitative high throughput screening (HTS) tools makes it possible to monitor systemic changes in DNA, RNA and proteins in PD models. Combined with dopamine neuron isolation or derivation of dopamine neurons from PD patient specific induced pluripotent stem cells (PD iPSCs), HTS techonologies will provide opportunities to draw PD causing sequences of molecular events in pathologically relevant PD samples. Here I discuss previous studies that identified molecular functions in which PD genes are involved, especially those signaling pathways that can be efficiently studied using HTS methodologies. Brief descriptions of quantitative and systemic tools looking at DNA, RNA and proteins will be followed. Finally, I will emphasize the use and potential benefits of PD iPSCs-derived dopaminergic neurons to screen signaling pathways that are initiated by PD linked gene mutations and thus causative for dopaminergic neurodegneration in PD.

The Effect of Different Light Quality on the Change of Membrane PD of the Guard Cell in Tradescantia virginiana L.

  • The effects of different light quality on the change of membrane potential difference (PD) of the guard cell in the intact leaf have been investigated. The mombrane PD was about -5.5 mV by white light of 600 $\mu$moles $m^{-2}\; s^{-1}$. The mean PD of change caused by red light was about -5.2 mV at the light intensity of 80 $\mu$moles $m^{-2}\; s^{-1}$. Membrane PD of guard cells in response to blue light was saturated at low light intensity. However, red and green light enhanced the change of membrane PD of guard cells with increasing intensity. In green light the biggest change of memrane PD was around -4 mV, whereas, with blue light the change of of memrane PD was around -2 mV. Accordingly, the membrane PD of guard cell showed the different degree of hyper-polarization by each wavelength.

개미산 분해 반응에서 수소 생산성 증대를 위한 Pd/Pd3Fe 합금 촉매: 범밀도 함수 이론 연구 (Pd/Pd3Fe Alloy Catalyst for Enhancing Hydrogen Production Rate from Formic Acid Decomposition: Density Functional Theory Study)

  • 본 연구에서는 양자 역학 계산 이론 중 하나인 Density Functional Theory (DFT)를 사용하여 $Pd/Pd_3Fe$ 촉매 표면에서 개미산(HCOOH) 분해 반응으로부터 수소를 생산하는 반응 메커니즘을 분석하였다. 기존 연구에 따르면, 단일 원자 촉매 중에서 개미산 분해 반응에 가장 높은 수소 생산성을 기록하는 원자는 Pd 촉매이지만, 부 반응으로 생산되는 CO가 Pd에 독성을 띄우기 때문에 Pd 촉매의 성능을 저하시킨다. 이러한 단점을 극복하고자, Pd를 기반으로 Pd와 Fe를 3:1로 합금하여 $Pd_3Fe$가 코어(core) 형태로 존재하고 Pd가 표면에 위치한 core-shell $Pd/Pd_3Fe$ 촉매를 설계하여 개미산 분해 반응에 의한 수소 생산 속도를 계산하였다. 순수 Pd촉매 보다 $Pd/Pd_3Fe$ 촉매의 수소 생산 반응의 활성 에너지가 감소하였다. 그 이유는 Pd와 Fe가 합금화 되면서 $Pd_3Fe$의 격자 상수가 $2.76{\AA}$로 줄어 들어 HCOO의 흡착에너지를 0.03 eV 감소시켰고, Fe에서 표면 Pd로 전자가 이동하면서 표면 전자 구조가 변화하여 HCOO의 흡착에너지를 0.29 eV 낮추었기 때문이다. 본 연구에서 제안하는 결과를 바탕으로 추후 개미산으로부터 수소 생산이 더 용이한 새로운 촉매 설계 메커니즘을 제안하고자 한다.

  • To design the practical core-shell electrocatalysts, combination of core and shell materials is important to meet catalytic activity and durability target. In general, Pd is considered as a good core material due to its best activity caused by strain/ligand effect. Preparing Pd nanoparticles can be a starting point in fabricating core-shell type electrocatalysts, much simplified Pd preparing process is suggested by using carbon monoxide (CO) as a reducing agent and/or capping agent. The solvent composition and reaction temperature can control to nanosheet, tetrahedron, and sphere without using additional stabilizer. Among them, Pd nanosheet which has mainly (111) plane showed about 3 times higher electrocatalytic activity for oxygen reduction reaction (ORR) to the spherical Pd nanoparticles. The enhanced ORR activity of Pd nanosheets can be attributed to the exposure of Pd (111) surface and the high electrochemical surface area. Therefore, we demonstrated that the shape of Pd nanomaterials is easily controlled via a facile reduction method using CO, and (111) plane-oriented Pd nanosheets can be a promising ORR catalysts and core material for polymer electrolyte fuel cells (PEFCs).

Hydrogen Transport through Palladium Foil Placed in Nafion Electrolyte of H2/O2 Fuel Cellsorption

  • Placing a hydrogen conducting, methanol impermeable metallic barrier like palladium (Pd) is a well-known method for preventing methanol crossover through solid polymer electrolyte for direct methanol fuel cells (DMFC). Applying a bias potential between the anode and the barrier can further develop this concept so that the hydrogen transfer rate is enhanced. Since hydrogen diffuses in Pd as atomic form while it moves through nafion electrolyte as ion, it has to be reduced or oxidized whenever it passes the interface formed by Pd and the electrolyte. We performed experiments to measure the hydrogen transport through the Pd membrane placed in Nafion electrolyte of hydrogen/oxygen fuel cell (PEMFC). Applying a bias potential between the hydrogen electrode of the cell and the Pd membrane facilitated the hydrogen passage through the Pd membrane. The results show that the cell current measured with the Pd membrane placed reached almost 40 % the value measured with the cell without Pd membrane. It was found that the current flown through the bias path is only a few percent of the cell current.
