목적: 나노 산화철 입자 및 생고분자인 PCL(Polycarprolacton)로 표면 코팅한 T2 MR 조영제를 합성하고 동물 종양 모델을 이용하여 In vivo 특성을 조사하고자 하였다. 대상 및 방법: $FeC1_2$.$4H_2$O와 $FeC1_3$.$6H_2$O을 무게비를 1:2로 정량하여 첨가하고 NaOH 혹은 TMAOH로 pH를 조절한후 PCL를 첨가하여 magnetite가 생성되는 동시에 고분자로서 코팅을 한후 증류수로 여러번 씻어준다. TEM, SEM, DLS 및 IR spectroscopy와 SQUID등을 측정하여 최종 반응물의 입자크기, 자성, 코팅 상태등을 평가하였다. 최소의 입자크기를 형성하는데 필요한 실험 조건을 찾기 위해 반응온도, 코팅할 고분자의 함량, 교반속도별로 실험하여서 최적의 조건을 찾으려 하였다. 토끼의 간에 VX2 암종을 이식한 동물 모델을 이용하여 PCL로 표면 코팅된 나노자성체의 in vivo 영상 특성을 알아보았다.
최근 미세혈관 문합수술 시 봉합사 대신 기계식 문합커플러를 사용하는 수술례가 증가하고 있다. 그러나 기존의 비분해성 문합커플러는 염증반응 뿐만 아니라 수술 후 영구적으로 인체 내에 잔존한다는 한계성이 있다. 따라서 본 연구에서는 기존 비분해성 문합커플러의 한계성을 극복하기 위해 사출성형공정을 이용한 생분해성 PCL 문합커플러 제작을 채택하였다. 사출성형 공정조건 중 실린더온도와 사출압력에 따른 수축률을 계산하고 이에 따른 문합강도를 측정하였다. 그 결과 핀보다는 홀 파트의 수축률 변화가 크게 나타났다. 또한 수축률은 실린더온도가 상승할수록 증가하였으나 반대로 사출압력이 높아질수록 감소하는 경향이 나타났다. PCL 문합 커플러의 in-vitro 분해거동을 12주간 평가한 결과, 수분흡수는 증가하고 분자량은 감소하여 생체분해를 동반한 질량 및 문합강도의 감소를 확인할 수 있었다. 그러나 문합강도의 저하 수준이 전임상 요구조건을 충분히 상회하기에 PCL 문합커플러는 미세혈관수술에 적합한 후보재료임을 파악할 수 있었다.
생체 친화적이며 생분해성 고분자 소재인 poly($\varepsilon$-caprolactone)(PCL)을 rapid prototyping(RP) 공정인 바이오플로팅 시스템을 통해 세포 재생용 지지체(scaffold)를 제작하였다. 제작된 PCL 지지체는 DMA(dynamic mechanical analyzer)를 통해 동일한 재료로 제작된 기존 염침출법(salt-leaching)에 의한 지지체보다 월등히 향상된 기계적 강도를 갖고 있음을 확인하였고, 이는 기존 전통적인 세포지지체 제작에서 문제점중의 하나인 기계적인 강도적인 측면을 보완하여, 뼈조직 재생에 유용하게 활용될 수 있을 것으로 예상된다. 지지체 내부의 구조는 세포의 증식과 이동 및 영양분의 공급이 지속될 수 있도록 전체적으로 연결된 통로로 구성되어 있고, 다양한 세포의 증식이 가능하도록 지지체의 공극 크기와 strand의 굵기 등을 조절할 수 있으며, 이를 이용하여 대체하고자 하는 생체조직의 특성에 맞도록 기계적 강도를 조정할 수 있음을 확인하였다. 제조된 PCL지지체는 연골세포를 통하여 셀 컬쳐링 되었고, 3차원 세포 지지체로서의 충분한 가능성을 보여주었다.

Objective : Post-traumatic stress disorder (PTSD) is a group of diseases that are observed in patients who had experienced a serious trauma or accident. However, some experienced it even after only a mild traumatic brain injury (TBI), and they are easily ignored due to the relatively favorable course of mild TBI. Herein, the authors investigated the incidence of PTSD in mild TBI using brief neuropsychological screening test (PTSD checklist, PCL). Methods : This study was conducted on patients with mild TBI (Glasgow coma scale ${\geq}13$) who were admitted from January 2012 to December 2012. As for PCL, it was done on patients who showed no difficulties in communication upon admission and agreed to participate in this study. By using sum of PCL, the patients were divided into high-risk group and low-risk group. PTSD was diagnosed as the three major symptoms of PTSD according to the Diagnostic and Statistical Manual of Mental Disorders, fourth-edifion. Results : A total of 314 TBI patients were admitted and 71 of them met the criteria and were included in this study. The mean age was 52.9 years-old (range : 15--94). The mean PCL score was 28.8 (range : 17--68), and 10 patients were classified as high-risk group. During follow-up, 2 patients (2.7%) of high risk group, were confirmed as PTSD and there was no patient who was suspected of PTSD in the low-risk group (p=0.017). Conclusion : PTSD is observed 2.8% in mild TBI. Although PTSD after mild TBI is rare, PCL could be considered as a useful tool for screening of PTSD after mild TBI.

The 14k, 45k, and 70k Mw PCL have different crystallization temperatures and therefore have slightly different characteristics affecting the fiber diameter. To observe these behaviors, the fiber was produced at every step of $10^{\circ}C$ for each molecular weights and the diameter was measured. Moreover, the fiber was fabricated over the cooled ground plate to observe the change in fiber diameter in comparison to the normal ground plate. In case of molecular Mw 14k PCL, the diameter increased as the temperature increased. For Mw 45k PCL, the fiber diameter decreased as the temperature increased. As of Mw 70k PCL, the fiber diameter decreased with increasing temperature as well. When the experiment was conducted over the cooled collector plate, the data did not change significantly from the previous lexperiments.
최근 20년간 다양한 세포에서 파크리탁셀(PTX)의 효과에 관한 연구는 많이 있지만, 세포증식을 억제하기 위한 약물방출 동역학에 관한 연구는 거의 보고되지 않고 있다. 본 연구에서는 약물방출스텐트 (DES)에 적용하기 위해서 생분해성 고분자로부터 파크리탁셀의 방출거동을 고찰하였다. 다양한 생분해성 고분자인 poly(lactic acid-co-glycolic acid) (PLGA), poly-L-lactide(PLLA) 및 polycaprolactone(PCL)에 파크리탁셀의 함유량을 달리하여 필름을 제조한 후 약물방출 거동을 평가하였다. 약물방출은 8주 동안 이루어졌으며 FE-SEM을 통해 고분자의 분해정도를 관찰하였다. PCL의 생분해 속도는 가장 느리지만 파크리탁셀의 함량이 같을 경우 PCL로부터의 파크리탁셀 방출속도가 가장 빨랐으며 PLGA 그리고 PLLA 순서를 보였다. 이와 같은 결과를 바탕으로 PCL과 같이 유리전이온도($T_g$)가 낮은 고분자의 경우 체내에서 파크리탁셀과 같은 소수성 약물의 움직임이 용이하기 때문에 약물방출 속도가 빠를 수 있음을 제시하고 있다.
조직공학의 중요한 요소로 작용하는 scaffold는 여러 가지 필수적인 조건들을 만족시켜야 한다. 대표적인 특징들로는 (1)생분해성 및 비독성, (2)넓은 표면적을 갖는 상호 연결된 내부 다공성 구조, (3)구조적 안정성, (4)세포부착 기질의 제공, (5)낮은 면역 반응성, (6)혈전 형성 억제, (7)친수성, (8)생체 기능성 등을 들 수 있다. 이러한 scaffold가 갖추어야 할 특성 중에서 세포 부착 기질 제공을 위하여 scaffold에 표면 개질을 통한 기능기를 도입하였다. 본 연구에서는 BCP scaffold의 구조적 안정성 부여를 위하여 PCL(polycaprolactone)을 infiltration 하였다. PCL은 소수성의 특징을 갖고 있어 세포와 상호작용 할 수 있는 생물학적 반응기가 없기 때문에 세포와의 친화성이 떨어진다. 세포의 친화성을 높여주기 위해 실리콘의 전구체인 TEOS(tetraethly orthosilicate)를 코팅하고, 그 위에 카복실기(carboxylic acid group)를 도입하였다. 또한 세포의 고정화를 높여주기 위해 fibronectin을 코팅하여 BCP/PCL scaffold의 세포 친화성을 높여주었다. 이와 같이 제조된 고기능성 BCP/PCL scaffold의 내부 구조와 특성을 Micro-CT로 확인하였고, 또한 실리콘 코팅 여부를 확인하기 위하여 SEM-EDS를 통해 관찰하였으며, FT-IR 관찰을 통해 카복실기 도입 여부를 확인 하였다. 또한 생체적합성 평가를 위해 MTT assay, 조골세포의 부착에 미치는 영향을 관찰하기 위해 SEM, 조골세포의 유전자 발현에 미치는 영향을 관찰하기 위해 RT-PCR을 통해 확인 하였다.

Background: Autogenous bone grafts have several limitations including donor-site problems and insufficient bone volume. To address these limitations, research on bone regeneration is being conducted actively. In this study, we investigate the effects of a three-dimensionally (3D) printed polycaprolactone (PCL)/tricalcium phosphate (TCP) scaffold on the osteogenic differentiation potential of adipose tissue-derived stem cells (ADSCs) and bone marrow-derived stem cells (BMSCs). Methods: We investigated the extent of osteogenic differentiation on the first and tenth day and fourth week after cell culture. Cytotoxicity of the 3D printed $PCL/{\beta}-TCP$ scaffold was evaluated by 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium assay, prior to osteogenic differentiation analysis. ADSCs and BMSCs were divided into three groups: C, only cultured cells; M, cells cultured in the 3D printed $PCL/{\beta}-TCP$ scaffold; D, cells cultured in the 3D printed $PCL/{\beta}-TCP$ scaffold with a bone differentiation medium. Alkaline phosphatase (ALP) activity assay, von Kossa staining, reverse transcription-polymerase chain reaction (RT-PCR), and Western blotting were performed for comparative analysis. Results: ALP assay and von Kossa staining revealed that group M had higher levels of osteogenic differentiation compared to group C. RT-PCR showed that gene expression was higher in group M than in group C, indicating that, compared to group C, osteogenic differentiation was more extensive in group M. Expression levels of proteins involved in ossification were higher in group M, as per the Western blotting results. Conclusion: Osteogenic differentiation was increased in mesenchymal stromal cells (MSCs) cultured in the 3D printed PCL/TCP scaffold compared to the control group. Osteogenic differentiation activity of MSCs cultured in the 3D printed PCL/TCP scaffold was lower than that of cells cultured on the scaffold in bone differentiation medium. Collectively, these results indicate that the 3D printed PCL/TCP scaffold promoted osteogenic differentiation of MSCs and may be widely used for bone tissue engineering.
생체 면역조직에서는 면역세포의 성장, 분화에 있어서 매우 중요한 역할을 수행하는 면역조직 기질세포가 존재하며, 이들은 서로 연결된 3차원적인 그물구조를 형성하면서 그 사이의 공간에 위치한 면역세포와의 상호작용을 통해 다양한 면역반응을 수행한다. 따라서 생체환경을 모사한 면역세포의 배양이 이루어지기 위해서는 면역세포들이 상호작용할 수 있는 3차원적 면역조직 기질세포 뼈대의 구축이 매우 중요한 의의를 지닌다. 특히 면역반응에서 핵심적인 기능을 수행하는 T세포의 생존, 성장 및 분화에 있어서 필수적인 역할을 하는 흉선상피세포에 대한 3차원적 배양은 T세포의 연구에 필수적으로 요구되지만, 아직 이에 관한 연구가 거의 이루어지지 않은 실정이다. 본 연구에서 흉선상피세포는 폴리도파민으로 코팅된 PCL 및 PCL/PLGA 지지체에서 비코팅군에 비해 부착 및 성장이 촉진되었다. 또한 폴리도파민으로 코팅된 지지체에서 흉선상피세포를 배양하였을 때 2차원 배양군에 비해 흉선세포형성촉진인자의 유전자 발현이 더 증가하였다. 따라서 본 연구는 면역조직 기질세포의 3차원 배양 기술의 개발에 크게 기여할 수 있을 것으로 사료된다.
