PCBs concentration patterns in Korean and Canadan cheese (한국과 캐나다산 치즈중 PCBs의 농도 특성)

  • All 209 PCB congeners were analyzed in cheese produced in Korea and Canada. 5 Korean and 10 Canadian cheeses were purchased in supermarket in their native countries. 93 PCB congeners were found in Korean cheese; 83 congeners were detected in Canadian cheese. Total PCB concentrations were 299.81 and 200.96 pg/g w.w. in Korean and Canadian cheeses respectively; PCB concentration was 1.5 times higher in Korean than Canadian cheese. Korean TEQ concentration (0.0067 pg-TEQ/g w.w.) in cheese was similar to in Canadian cheese (0.0064 pg-TEQ/g w.w.). These PCB concentrations range was low level compared with that of other cheeses globally. The correlation coefficient $(R^2)$ of regression was high (0.752) between each PCB congener concentration in both Korean and Canadian cheese.

Effects of NaOH and Humic Acid on the UV Photolysis of PCBs (PCBs의 광화학적 연구: NaOH 및 휴믹산 (humic acid, HA)에 의한 분해특성)

  • Objectives: This study was carried out to examine whether the apparent photolysis with or without sensitizers [NaOH and humic acid (HA)] was prompted photodegradation of polychlorinated biphenyl (PCB) in aqueous solution. Methods: PCBs photodegradation occurred using fluorescence black lamps at ${\lambda}_{max}=300nm$. PCB congeners were exposed in 10 ppm HA or 0.05N NaOH solutions, to investigate the decreasing profile of PCB concentration with time. The PCBs were then analyzed by gas chromatography/mass spectrometry (GC-MS). Reductive degradation profile of PCB congeners in the presence of both sensitizers under oxygen-saturated protic conditions was described using the wind-rose diagrams. Results: Use of HA or NaOH decreased PCB concentration with time in the dark and on irradiation, indicating that photolysis underwent through reductive dechlorination through energy transfer and possibly with reactive oxygens. The dechlorination was marked by a chromatographic shift, observed in the GC-MS plots. Therefore it is logical to assume that increasing the dose of sensitizers would increase the photodegradation rates of PCBs. The half-lives of pentachloro-PCB (penta-3) in 0.05N NaOH and 10 ppm HA were estimated at about 47 hours and 39 hours, respectively, under the same experimental conditions of photolysis. It was found that the rate of photolysis of pentachloro-PCB in aqueous solution followed apparent first-order kinetics compared to other congeners. Conclusion: Photochemical degradation (using 328 nm UV light) of penta- and hexa-PCBs in HA or alkaline solution is a viable method for pretreatment method. The results are helpful for the further comprehension of the reaction mechanism for photolytic dechlorination of PCBs in aquatic system.

Concentration Characteristics of PCBs in Pine Needle and Soil at Rural Area (교외지역에서 소나무 잎과 토양 중 PCBs의 농도 특성)

  • This study was to investigate concentration characteristics of atmospheric PCBs in soil sample and pine needle at rural area. Profiles of PCB congener detected in pine needle and soil sample have a difference that low molecular PCBs mainly existed in pine needle, whereas high molecular PCBs were mainly existed in soil sample because of their different vapor pressure and octanol-air partitioning coefficient($K_{OA}$) of each congeners. Correlation coefficients(r) of PCB congeners simultaneously detected in soil and pine needle were significant (r>0.71, p<0.01), which showed that PCB congeners patterns of atmosphere could be estimated in using pine needle and soil sample indirectly. Evaluation of human exposure to polychlorinated biphenyls using human serum by congener-specific analysis (혈액 중 PCBs 이성질체별 분석에 의한 인체 노출 평가)

  • Blood serum concentrations of polychlorinated biphenyls (PCBs) were measured in employees who worked at a municipal solid waste incinerator (MSWI), members of residential community who lived near the MSWI (<0.3km) and members of residential community lived far from the MSWI (>10 km). Human blood serum samples were analyzed for all PCB congeners using high-resolution gas chromatography/high-resolution mass spectrometry (HRGC/HRMS). The mean levels of total PCBs and dioxin-like PCBs in 87 serum samples were 242.77 ng/g lipid and 8.83 TEQ pg/g lipid, respectively. The PCB homologue profiles showed that penta-, hexa-, hepta-chlorinated biphenyls contributed more than 80% of the total PCBs concentration. The most abundant congeners were PCB153, PCB138, PCB180, PCB187, PCB118. A statistical analysis was performed to determine whether there were significant correlations between PCB concentrations and specific variables such as age, gender, smoking habits, occupation, BMI (Body Mass Index) and time of residence. As a result, the age was found to be strongly correlated with serum PCB concentrations. In addition, there were strong correlations between total PCBs and PCB153 (r=0.93, p<.0001), dioxin-like PCBs and PCB118 (r=0.98, p<.0001). So these two congeners are satisfactory indicators for total PCB concentrations and dioxin-like PCBs in human blood respectively.

Detection of PCB77 by Indirect Competitive Enzyme-linked Immunosorbent Assay in Sea Sediment Samples

  • 3,3',4,4'-Tetrachlorobiphenyl (IUPAC PCB77) is one of seven indicative polychlorinated biphenyls (PCBs) in the surface sediments. The current study presents a novel polyclonal antibody for the determination of the PCB77 using indirect competitive enzyme-linked immunosorbent assay. Under optimum conditions, PCB77 was determined within the concentration range of 0.01-100 ${\mu}g\;L^{-1}$, with a detection limit of 0.057 ${\mu}g\;L^{-1}$. The assays were tested for their cross-reactivity profiles using 3 selected congeners and 4 Aroclor products. The assays were highly specific for coplanar PCB congeners, but less specific for Aroclor1248. The spiked recoveries from five sediment samples were 86%-114% for PCB77 from ELISA, which were satisfactory. The current study demonstrated that the developed antiserum and immunoassay procedure can be used to detect PCB77 in environmental samples. The results of the sediment analysis were confirmed by conventional GC/ECD.

The Pattern of PCBs Level in Adipose Tissue and Serum of Breast Cancer and Normal Women (유방암 환자와 정상여성의 혈액 및 지방조직 중 PCBs 형태에 관한 연구)

  • The purpose of this study was to identify the distribution of non-ortho and mono-ortho PCB congeners and homologues in adipose tissues and sera of women with breast cancer. The collected samples were 25 adipose tissues and 33 sera from women with breast cancer. The samples from the control group were 49 adipose tissues and 52 sera. The levels of three non-ortho and eight mono-ortho PCBs identified in adipose tissue and serum samples were determined by GC/MSD and GC/ECD analyses. Non-ortho and mono-ortho PCB congeners were more dominant in the control group than in the case group for serum samples. The Tetra-PCB and the Hexa-, Hepta-PCB were more dominant in tale and control groups, respectively. The level of PCB homologues in normal women was similar to that of the normal human milk samples. However, the levels of PCB homologues from breast cancer patients were almost same the level of sample from environment. As a result of this study, it is suggested that breast cancer could be related to environmental factors such as PCB level in stack gas and soil sample. More extended research should be to verify this result.

Effects of PCB Congeners in Rodent Neuronal Cells in Culture : Effects of Chitosan (PCB 이성질체가 설치류 신경세포에 미치는 영향: 키토산의 효과)

  • The present study attempted to analyze the mechanism of PCB-induced neurotoxicity with respect to the PKC signaling. Since the developing neuron is particularly sensitive to PCB-induced neurotoxicity, we isolated cerebellar granule cells derived from 7-day old SD rats and grew cells in culture for additional 7 days to mimic PND-14 conditions. Only non-coplanar PCBs at a high dose showed a significant increase of total PKC activity at $[^3H]PDBu$ binding assay, indicating that non-coplanar PCBs are more neuroactive than coplanar PCBs in neuronal cells. PKC isoforms were immunoblotted with respective monoclonal antibodies. PKC-alpha and-epsilon were activated with non-coplanar PCB exposure. The result suggests that coplanar PCBs have a PKC pathway different from non-coplanar PCBs. Activation of PKC with exposure was dampened with treatment of high molecular weight of chitosan. Chilean (M.W. > 1,000 kDa) inhibited the total activity of PKC induced by the non-coplanar PCBs. Translocation of PKC isoforms was also inhibited by the high molecular weight of chitosan. The study demonstrated that non-coplanar PCBs are more potent neurotoxic congeners than coplanar PCBs and the alteration of PKC activities by PCB exposure can be blocked with the treatment of chitosan. The results suggest a potential use of chitosan as a means of nutritional intervention to prevent the harmful effects of pollutant-derived diseases.

Dechlorination of Individual Congeners in Aroclor 1248 as Enhanced by Chlorobenzoates, Chlorophenols, and Chlorobenzenes

  • Previous investigations showed that three classes of haloaromatic compounds (HACs; chlorobenzoates, chlorophenols, and chlorobenzenes) enhanced the reductive dechlorination of Aroclor 1248, judging from the overall extent of reduction in CI atoms on the biphenyl. In the present study, we further investigated the kind of polychlorinated biphenyl (PCB) congeners involved in the enhanced dechlorination by four isomers belonging to each class (2,3-, 2,5-, 2,3,5-, and 2,4,6-chlorobenzoates; 2,3-, 3,4-, 2,5-, and 2,3,6-chlorophenols; and 1,2-, 1,2,3-, 1,2,4-, and penta-chlorobenzenes). Although the PCB congeners involved in the enhanced dechlorination varied with the HACs, the enhancement primarily involved para-dechlorination of the same congeners (2,3,4'-, 2,3,4,2'-plus 2,3,6,4'-, 2,5,3',4'- plus 2,4,5,2',6'-, and 2,3,6,2',4'-chlorobiphenyls), regardless of the HACs. These congeners are known to have low threshold concentrations for dechlorination. To a lesser extent, the enhancement also involved meta dechlorination of certain congeners with high threshold concentrations. There was no or less accumulation of 2,4,4'- and 2,5,4'-chlorobiphenyls as final products under HAC amendment. Although the dechlorination products varied, the accumulation of ortho-substituted congeners, 2-, 2,2'-, and 2,6-chlorobiphenyls, was significantly higher with the HACs, indicating a more complete dechlorination of the highly chlorinated congeners. Therefore, the present results suggest that the enhanced dechlorination under HAC enrichment is carried out through multiple pathways, some of which may be universal, regardless of the kind of HACs, whereas others may be HAC-specific.

Analysis of Polychlorinated Biphenyls in the Sediment of the Kumho River (금호강 저니토 중 PCBs의 분석)

  • Concentrations of PCBs in the sediment of the Kumho River was determined. Samples were collected at 23 locations along the river. Total PCB levels ranged from 2.7 to 87.6 ng/g dry weight. The site near the Keomdan factory district had the highest concentrations. The levels of PCB congeners significantly correlated with the total organic carbons in the sediment. The major fractions of PCBs were congeners containing 4, 5, or 6 chlorine atoms per biphenyl molecule.

Only non-coplanar PCBs at a high dose showed a significant increase of total protein kinase C (PKC) activity at phobol 12,13-dibutyrate ([$^3M$]PDBu) binding assay, indicating that non-coplanar PCBs are more neuroactive than coplanar PCBs in neuronal cells. PKC isozymes were immunoblotted with the selected monoclonal antibodies. PKC-${\alpha}$, ${\delta}$, and ε were activated with non-coplanar PCB exposure. Receptor for activated C kinase-1 (RACK-1), anchoring protein for activated PKC, was more induced with exposure to coplanar PCBs than non-coplanar PCBs. Reverse transcription PCR (RT-PCR) analysis showed induction of neurogranin (RC-3) and growth associated protein-43 (GAP-43) mRNA with non-coplanar PCBs. The results indicate that these factors may be useful biomarkers for differentiating non-coplanar PCBs from coplanar PCBs. The present study demonstrated that non-coplanar PCBs are more neuroactive congeners than coplanar PCBs.
