The present study was conducted for the production of transgenic cloned cows by somatic cell nuclear transfer (SCNT) that secrete human prourokinase into milk. To establish an efficient production system for bovine transgenic SCNT embryos, the offset was examined of various conditions of donor cells including cell type, size, and passage number on the developmental competence of transgenic SCNT embryos. An expression plasmid far human prourokinase (pbeta-ProU) was constructed by inserting a bovine beta-casein promoter, a green fluorescent protein (GFP) marker gene, and a human prourokinase target gene into a pcDNA3 plasmid. Three types of bovine somatic cells including two adult cells (cumulus cells and ear fibroblasts) and fetal fibroblasts were prepared and transfected using a lipid-meidated method. In Experiment 1, developmental competence and rates of GFP expression in bovine transgenic SCNT embryos reconstructed with cumulus cells were significantly higher than those from fetal and ear fibroblasts. In Experiment 2, the effect of cellular senescence in early (2 to 4) and late (8 to 12) passages was investigated. No significant differences in the development of transgenic SCNT embryos were observed. In Experient 3, different sizes of GFP-expressing transfected cumulus cells [large (>30 ${\mu}{\textrm}{m}$) or small cell (<30 ${\mu}{\textrm}{m}$)] were used for SCNT. A significant improvement in embryo development and GFP expression was observed when small cumulus cells were used for SCNT. Taken together, these results demonstrate that (1) adult somatic cells could serve as donor cells in transgenic SCNT embryo production and cumulus cells with small size at early passage were the optimal cell type, and (2) transgenic SCNT embryos derived from adult somatic cells have embryonic development potential.
Epidemiologic studies have shown important relationships between oxidative stress and Alzheimer's disease (AD) brain. In this study, free radical scavenging activity and neuronal cell protection effect of aqueous methanol extracts of Acanthopanax senticosus (A. senticosus) were examined. $H_2O_2$-induced oxidative stress was measured using 2',7'-dichlorofluorescein diacetate (DCF-DA) assay. Pretreatment with the phenolics of A. senticosus prevented oxidative injury against $H_2O_2$ toxicity. Since oxidative stress is known to increase neuronal cell membrane breakdown, leading to cell death, lactic dehydrogenase release, and trypan blue exclusion assays were utilized. We found that phenolics of A. senticosus have neuronal cell protection effects. It suggests that the phenolics of A. senticosus inhibited $H_2O_2$-induced oxidative stress and A. senticosus may be beneficial against the oxidative stress-induced risk in AD.
Amyloid $\beta$-peptide (A$\beta$), which is a product of the proteolytic effect of $\beta$-secretase (BACE) on an amyloid precursor protein, is closely associated with Alzheimer's disease (AD) pathogenesis. There is sufficient evidence to suggest that a BACE inhibitor may reduce A$\beta$ levels, thus decreasing the risk of AD. In a previous study, an extract of Clavicorona pyxidata DGUM 29005 mycelia was found to inhibit the production of a soluble $\beta$-amyloid precursor protein (s$\beta$APP), A$\beta$, and BACE in neuronal cell lines. We sought to determine whether this mycelial extract exerts the same effect in human rhabdomyosarcoma A-204 and rat pheochromocytoma PC-12 cells. We found that the production of A$\beta$ decreased in a dose-dependent manner in the presence of the mycelial extract and that the concentration of A$\beta$ never exceeded $50{\mu}g/ml$. The presence of sAPP was detected in every culture medium to which the mycelial extract had been added and its concentration remained the same, regardless of the concentration of the extract used. Endogenous $\beta$-secretase activity in A-204 and PC-12 cellular homogenates also decreased in the presence of this extract. These cells, in culture, were not susceptible to the cytotoxic activity of the mycelial extract.

Neurodegenerative disorders in the elderly are characterized by gradual loss of memory and cognitive function. Oxidative stress caused by reactive oxygen species is associated with progressive neuronal cell damage and death in Alzheimer's disease, one of the most common neurodegenerative disorders. An edible brown seaweed, Ecklonia cava, contains a variety of biologically active compounds such as phlorotannins. In this study, we comparatively evaluated the total phenolic content, antioxidant capacity, and neuroprotective effects of the phlorotannin-rich extract from E. cava (PEEC). The total phenolic content of PEEC and dieckol was 810.8 mg gallic acid equivalents (GAE)/g and 996.6 mg GAE/g, respectively. Antioxidant capacity of PEEC was 1,233.8 mg vitamin C equivalents (VCE)/g and 392.1 mg VCE/g determined using ABTS and DPPH assays, respectively, while those of dieckol were 2,238.4 mg VCE/g and 817.7 mg VCE/g. High-performance liquid chromatography results revealed 48.08 ± 0.67 mg dieckol/g of PEEC. PEEC had neuroprotective effects in pheochromocytoma (PC-12) and human neuroblastoma (SH-SY5Y) cells against H2O2- and AAPH-induced oxidative damage, partly due to reduced intracellular oxidative stress. PEEC treatment inhibited acetylcholinesterase and butyrylcholinesterase in a dose-dependent manner. Taken together, these findings suggest that PEEC is a good source of antioxidants and neuroprotective materials.
The antioxidative activity of various enzymatic extracts from Sarcodon aspratus (S. aspratus) was evaluated by measuring 1,1-diphenyl-2-picrylhydrazyl (DPPH), and alkyl radical scavenging activity using an electron spin resonance (ESR) spectrometer. For this study, the S. aspratus were enzymatically hydrolyzed by seven carbohydrases (Viscozyme, Celluclast, Dextrozyme, AMG, Promozyme, Maltogenase, and Termamyl) and eight proteases (${\alpha}$-chymotrypsin, Alcalase, Flavourzyme, Neutrase, papain, pepsin, Protamax, and trypsin). The DPPH radical scavenging activities of Viscozyme and pepsin extracts were the highest, and the half maximal inhibitory concentration ($IC_{50}$) values were 0.896 and 0.734mg/mL, respectively. The Celluclast and trypsin extracts showed the highest scavenging activities on alkyl radical, and their $IC_{50}$ values were 0.278 and 0.575mg/mL, respectively. The Celluclast extracts was decreased cell apoptosis in PC-12 cells against $H_2O_2$-induced oxidative damage. The findings of the present study suggest that enzymatic extracts of S. aspratus exhibit antioxidative activity against oxidative stress on PC-12 cells.
Objectives : In this study, we compared the anti-oxidant and anti-inflammatory activities of Curcumae longae Radix (CLRa) and Curcumae longae Rhizoma (CLRh). Methods : We performed 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and 2,2-azinobis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) cation scavenging assays, and determined total polyphenolic content to examine the anti-oxidant effects of CLRa and CLRh. We also evaluated the anti-oxidant effects of CLRa and CLRh against hydrogen peroxide ($H_2O_2$)-induced toxicity in PC12 cells using thiazolyl blue tetrazolium bromide (MTT) and reactive oxygen species (ROS) assays. Next, to compare the anti-inflammatory effects of CLRa and CLRh against lipopolysaccharide (LPS)-induced inflammation in microglia BV2 cells, we measured nitric oxide (NO) assay and inducible nitrite synthase (iNOS) using Western blotting analysis. Results : CLRa showed higher activity in DPPH and ABTS assays and lower total polyphenolic contents compared with CLRh. In PC12 cells, CLRa and CLRh showed no difference in H2O2-induced cell toxicity and ROS overproduction. In BV2 cells, CLRa showed higher effect than CLRh in NO and iNOS production induced by LPS. Conclusions : These results demonstrate that CLRa has higher radical scavenging activities and anti-inflammatory effect in BV2 cells comparing CLRh. However, CLRa and CLRh have no effect and no difference in $H_2O_2$-induced toxicity.
스마트폰을 이용하는 게임 산업과 관련 시장은 매년 급성장하고 있으며 플레이어는 다양한 환경에 게임요소와 즐거운 게임의 그래픽적 시각개선을 기대하고 있다. 게임애플리케이션의 설계에서 시각요소의 사용자 관점의 환경은 PC게임등과 달리 매우 간단한 조작이지만 보다 정교하며 게임의 질적 향상을 위한 계획적 접근이 요구된다. 따라서 본 연구는 최근 다운로드 순위 상위랭킹에 게임을 사례로 기존의 연구를 바탕으로 구조화된 설문지를 작성하여 설문 조사를 실시하였다. 조사대상은 2억명 이상 다운로드된 음양사게임으로 중국의 유저들을 대상으로 한정하여 각 GUI의 선호도를 조사하여 분석하였다. 선호도에서 나타난 UI/UX 디자인의 그래픽 기본요소의 성공요인 개선해야 할 점등을 파악하고 게임 어플리케이션을 설계할 시 고려되어야 할 계획요소에 대한 고찰을 연구목적으로 하여 시각 요소, 정보 요소, 조작 및 신속 대응이 얼마나 중요한지를 검증해 본다.
Essential oils and aromatherapy have traditionally been used for the treatment of anxiety and depression with few side effects. This study aimed to investigate the effects of essential oils from six herbal prescriptions known to be effective in treating anxiety and depression in Korean medicine. The neuroprotective and anti-neuroinflammatory effects of six essential oils, including Gamisachil-tang (GMSCT), Guibi-tang (GBT), Sihogayonggolmoryeo-tang (SYM), Danchisoyosan (DCSYS), Sihosogansan (SHSGS), and Soyosan (SYS), were examined in PC12 and BV2 cells. In corticosterone (CORT)-stimulated PC12 cells, all six essential oils ameliorated the CORT-induced decrease in cell viability at a concentration of 10 ㎍/ml. GMSCT, GBT, and SHSGS recovered CORT-induced cytotoxicity at concentrations of 1 ㎍/ml and 10 ㎍/ml. In lipopolysaccharide (LPS)-stimulated BV2 cells, GBT (10 ㎍/ml) decreased interleukin (IL)-1β production, whereas SHSGS (1 ㎍/ml) inhibited tumor necrosis factor (TNF)-α production. In the MK-801-induced anxiety in zebrafish, electroencephalogram (EEG) assessment indicated that GMSCT and SHSGS induced recovery in the delta and beta power densities and reduced theta/beta and delta/beta ratios. DCSYS and SYS decreased theta power density and theta/beta ratio, whereas GBT and SYM showed no effects on EEG signals. In the tail suspension test (TST) in mice, GBT, DCSYS, SHSGS, and SYS exhibited antidepressant-like effects by decreasing immobility time. These results suggest that the essential oils from the six herbal prescriptions, except SYM, may have beneficial effects on anxiety and/or depression. Further studies should be conducted to investigate the molecular signaling pathways that mediate the effects of these essential oils on anxiety and depression.
연구배경 : 기관지천식은 일종의 염증성 질환으로서 다형핵구가 병태생리에 중요한 역할을 하는 것으로 알려져 있다. 특히 다형핵구로부터 생성되는 독성산화물들은 기도 및 폐조직 손상을 야기시켜 기관지 천식의 병인에 기여하는 것으로 알려져 있으나, 이에 대한 실질적인 증거가 아직 부족할 뿐아니라 이들이 폐조직손상을 일으키는 기전 또한 불분명하다. 이에 저자들은 기관지천식환자들의 다형핵구로부터 생성되는 과산화 음이온이 실제로 폐포세포손상에 중요한 역할을 하는지 여부와 과산화 음이온이 폐포세포 손상을 일으키는 기전을 보다 명확히 하고자 본 연구를 시작하였다. 방법 : 임상적으로 안정된 12명의 기관지천식환자와 5명의 정상인들을 대상으로 하였으며, 말초혈액 다형핵구는 말초혈액으로부터 dextran 침강법 및 Ficoll-Hypaque 밀도차 분리법을 이용하여 분리하였다. 다형핵구의 과산화 음이온 생성능 및 혈장 SOD 활성도는, 과산화 음이온이 cytochrome c를 환원시켜 나타나는 발색반응을 이용하여 분광측정법으로 측정하였으며, 다형핵구의 A549 폐포세포에 대한 세포독성작용은 $^{51}Cr$ release assay로 측정하였으며, A549 lysis 및 A549 detachment 두가지 측면에서 관찰하였다. 결과 1) 다형핵구의 과산화 음이온 생성능은 천식환자군이 정상군에 비해 현저히 높았으며 ($2.81{\pm}0.95$대 $6.65{\pm}0.58\;mmol/1{\times}10^6$ cells, p<0.05), 각 환자의 $FEV_1$과는 역상관관계를 보였으나 (R=-0.63, p<0.05), $PC_{20}$ histamine과는 상관관계가 없었다. 2) 혈장 SOD 활성도는 천식환자군이 정상군에 비해 다소 감소되어 있는 경향을 보였으나 통계학적 의미는 없었으며, 각 환자의 $FEV_1$과는 상관관계를 보였으나(R=0.63, p<0.05) $PC_{20}$ histamine과는 상관관계가 없었다. 3) 정상군에서는 혈장 SOD 활성도와 다형핵구의 과산화음이온 생성능간에 의미있는 상관관계를 보였으나(R=0.88, p<0.05), 천식환자군에서는 상관관계가 없었다. 4) 두군 모두에서 다형핵구에 의한 A549 폐포세포의 손상형태가 lysis 보다는 detachment가 더욱 현저한 경향을 보였으며, 두가지 모두 천식 환자군에서 다소 높은 경향을 보였다. 또한 두군 모두에서 SOD 전처치에 의해 A549 detachment는 현저하게 억제되었으나 A549 lysis는 억제되지 않았으며, 두가지 모두가 혈창 전처치에 의해서는 억제되지 않았다. 5) 두군 모두에서 다형핵구의 과산화 음이온 생성능과 A549 detachement와는 유의한 상관관계가 있었으나(정상군 : R=0.90, 환자군 : R=0.82), A549 lysis와는 별로 상관관계가 없었으며, 혈장 SOD 활성도는 상관관계를 보이지 않았다. 또한 천식환자군에서 A549 lysis나 A549 detachment는 환자의 $FEV_1$ 도는 $PC_{20}$ histamine과는 의미있는 상관관계를 보이지 않았다.6) 다형핵구에 의한 폐포세포손상의 양상에 있어서 알레르기성 천식환자와 비알레르기성 천식환자 사이에는 의미있는 차이가 없었다. 결론 : 이상의 연구결과 다형핵구로부터 생성되는 과산화 음이온에 의한 기도 및 폐조직 손상이 기관지천식의 병인에 부분적으로 영향을 미칠 것으로 생각되며, 과산화 음이온은 주로 폐포세포의 탈락을 야기시킴으로써 폐포세포 손상을 일으키는 것으로 추측된다.

Oxidative stress is believed to contribute to the neuronal damage induced by cerebral ischemia/reperfusion injury. The present study was undertaken to evaluate the possible antioxidant neuroprotective effect of hydroxyfullerene (a radical absorbing cage molecule) against neuronal death in hippocampal CA1 neurons following transient global cerebral ischemia in the rat. Transient global cerebral ischemia was induced in male Wistar rats by four vessel- occlusion (4VO) for 10 min. Lipid peroxidation in brain tissues was determined by measuring the concentrations of thiobarbituric acid-reactive substances (TBARS). Furthermore, the apoptotic effects of ${H_2}{O_2}$ on PC12 cells were also investigated. Cell viabilities were measured using MTT [3-(4,5-dimethylthiazolyl-2)-2,-5-diphenyltetrazolium bromide] assays. Hydroxyfullerene, when administered to rats at 0.3-3 mg/kg i.p. at 0 and 90 minutes after 4-VO was found to significantly reduce CA1 neuron death by 72.4% on hippocampal CA1 neurons. Our findings suggest that hydroxyfullerene protects neurons from transient global cerebral injury in the rat hippocampus by reducing oxidative stress and lipid peroxidation levels, which contribute to apoptotic cell death.
