Protective Effects of Hemerocallis Fulva Extracts on Amyloid $\beta$-Protein-Induced Death in Neuronal Cells (아밀로이드 베타 단백질에 의해 유도된 신경세포 독성에 대한 원추리의 억제 효과 탐색)

  • Objectives : The amyloid $\beta$-protein ($A\beta$) is the principal component of the senile plaques characteristic of Alzheimer's disease (AD) and elicits a toxic effect on neurons in vitro and in vivo. Many environmental factors including antioxidants and proteoglycans modify $A{\beta}toxicity$. In this study, we have investigated the protective effects of water- and organic solvent-extracts of Hemerocallis fulva root fractions pre-extracted with methanol on $A\beta$-induced oxidative cell death in cultured rat pheochromocytoma (PC12) cells. Methods : For this study, we used MTT reduction assay for detection of protective effects of water- and organic solvent-extracts of Hemerocallis fulva root fractions pre-extracted with methanol on $A{\beta}_{25-35}$-induced cytotoxicity to PC12 cells. We also used cell-based $\beta$-secretase assay system to investigate the inhibitory effect of water- and organic solvent-extracts of Hemerocallis fulva root on $\beta$-secretase activity. Results : We previously reported that methanol extracts of Hemerocallis fulva root strongly attenuated cytotoxicity induced by the three $A\beta$ fragments ($A{\beta}_{25-35},\;A{\beta}_{1-42}\;A{\beta}_{1-43}$) to both SK-N-MC and PC12 cells. In the present study, we found that butanol-, ethylacetate-, chloroform-, and water-extracts of Hemerocallis fulva root fractions pre-extracted with methanol had strong protective effects against $A{\beta}_{25-35}$-induced cytotoxicity to PC12 cells and inhibitory potency to $\beta$-secretase activity. Conclusion : These results suggest that butanol-, ethylacetate-, chloroform-, and water-extracts of Hemerocallis fulva root fractions pre-extracted with methanol may contain the protective component(s) against $A\beta$-induced cell death in PC12 cells as well as inhibitory component(s) to $\beta$-secretase activity.

PKA-Mediated Regulation of B/K Gene Transcription in PC12 Cells

  • B/K protein is a novel protein containing double C2-like domains. We examined the specific signaling pathway that regulates the transcription of B/K in PC12 cells. When the cells were treated with forskolin ($50{\mu}M$), B/K mRNA and protein levels were time-dependently decreased, reaching the lowest level at 3 or 4 hr, and thereafter returning to the control level. Chemicals such as dibutyryl-cAMP, cellpermeable cyclic AMP (cAMP) analogue and CGS21680, adenosine receptor $A_{2A}$ agonist, also repressed the B/K transcription. However, 1,9-dideoxyforskolin did not show inhibitory effect on B/K transcription, suggesting direct involvement of cAMP in the forskolin-induced inhibition of B/K transcription. Effect of forskolin, dibutyryl cAMP and CGS21680 was significantly reduced in PKA-deficient PC12 cell line (PC12-123.7). One cAMP-response element (CRE)-like sequence (B/K CLS) was found in the promoter region of B/K DNA, and electrophoretic mobility shift assay indicated its binding to CREM and CREB. Forskolin significantly suppressed the promoter activity in CHO-K1 cells transfected with the constructs containing B/K CLS, but not with the construct in which B/K CLS was mutated (AC:TG). Taken together, we suggest that the transcription of B/K gene in PC12 cells may be regulated by PKA-dependent mechanism.

Protective Effects of Black Soybean Seed Coat Extracts against Oxidative Stress-induced Neurotoxicity (산화적 손상에 의해 유도된 신경세포독성에 대한 검정콩 껍질 추출물의 보호효과)

  • Rat pheochromocytoma cells (PC12) and mice were utilized as in vitro and in vivo models to determine the neuroprotective effects of a 70% acetone extract of black soybean seed coat (BSSCE). BSSCE showed higher total phenolic contents than other extracts. Intracellular reactive oxygen species accumulation from $H_2O_2$ treatment of PC12 cells was significantly reduced when BSSCE was present in the media compared to PC12 cells treated with $H_2O_2$ only. The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium-bromide (MTT) reduction assay and lactate dehydrogenase assay also showed significantly increased protective effects in PC12 cells. In addition, BSSCE improved the in vivo cognitive ability against amyloid beta peptide-induced neuronal deficits.

The Antioxidative and Neuroprotective Effect of Guibi-tang(Guipitang) and Guibi-tang gamibang(Guipitang jiaweijang) on PC12 cells. (귀비탕(歸脾湯)과 귀비탕가미방(歸脾湯加味方)의 항산화 효과 및 6-Hydroxydopamine에 대한 PC12 세포 보호효과 비교연구)

  • Objectives : This Study was performed to assess the antioxidative and neuroprotective effect of Guibi-tang(Guipi-tang) and Guibi-tang gamibang(Guipitang jiaweifang) on PC12 cells. Methods : The antioxidative effect was investigated through the DPPH radical and ABTS cation scavenging methods and total polyphenol amount of Guibi-tang(Guipitang) and Guibi-tang gamibang(Guipitang jiaweifang). The neuroprotective effect of Guibi-tang(Guipitang) and Guibi-tang gamibang(Guipitang jiaweifang) was assessed using MTT assay in PC12 cells. The scavenging effect of Guibi-tang(Guipitang) and Guibi-tang gamibang(Guipitang jiaweifang) on NO and ROS production induced by 6-OHDA in PC12 cells was evaluated, as well as the attenuating effect of Guibi-tang gamibang(Guipitang jiaweifang) on GSH reduction. Results : 1. Guibi-tang(Guipitang) and Guibi-tang gamibang(Guipitang jiaweifang) had concentration-dependent scavenging activities of DPPH radical 2. Guibi-tang(Guipitang) and Guibi-tang gamibang(Guipitang jiaweifang) had concentration-dependent scavenging activities of ABTS cation. 3. Total polyphenol amount of Guibi-tang(Guipitang) and Guibi-tang gamibang(Guipitang jiaweifang) was calculated 79.10${\pm}$2.20 pg/IO mg and 121.03${\pm}$1.11 pg/IO mg, respectively. 4. Cell viability of Guibi-tang(Guipitang) was increased in a dose dependent manner. Guibi-tang gamibang(Guipitang jiaweifang) was increased at low concentrations, but decreased at high concentrations. 5. In Guibi-tang(Guipitang), cell viability of PC12 cell treated with 6-OHDA was decreased by pre-treatment, and increased by post- and co- treatment. Cell viability of Guibi-tang gamibang(Guipitang jiaweifang) showed variable effects by pre-treatment, but increased by post- and co- treatment. 6. NO production rate of Guibi-tang(Guipitang) didn't show a significant effect, but that of Guibi-tang gamibang(Guipitang jiaweifang) was decreased in a dose dependent manner. 7. ROS production rate of Guibi-tang(Guipitang) was decreased at some concentrations. In Guibi-tang gamibang(Guipitang jiaweifang), ROS production rate was decreased at high concentrations. 8. Guibi-tang gamibang(Guipitang jiaweifang) protected the 6-OHDA-induced GSH reduction. Conclusions : These results demonstrate that both Guibi-tang(Guipitang) and GBTGMB have antioxidative and neuroprotective effect, but Guibi-tang gamibang(Guipitang jiaweifang) has more antioxidative and neuroprotective effect than Guibi-tang.

Protective effect of ethanolic extract of antler-shaped Ganoderma lingzhi against oxidative stress in PC12 neuronal cell line (PC12 신경세포주에서 녹각영지버섯 주정 추출물의 산화 스트레스 개선 효과)

  • This study was carried out to identify medicinal mushrooms with protective effects against oxidative stress in PC12 neuronal cell line, followed by evaluation of their antioxidant property. Extracts of medicinal mushrooms, including Ganoderma lucidum extract (GLE), antler-shaped Ganoderma lingzhi extract (AGLE), Hericium erinaceus extract (HEE), and Sanghuangporus baumii extract (SBE), were screened for cytotoxicity using MTT assay. None of the extracts up to $10{\mu}g/ml$ concentration affected cell viability. These extracts were further checked for their protective effect against oxidative stress-induced reactive oxygen species (ROS) production. Exposure to $50{\mu}M$ $H_2O_2$ induced ROS generation in PC12 cells, which was inhibited only by treatment with AGLE. In addition, inhibition of $H_2O_2-induced$ ROS generation by AGLE was found to be in a dose-dependent manner (2.5, 5, and $10{\mu}g/ml$). Microscopic examination of DCF fluorescence for detection of ROS showed a similar pattern. Further, antioxidant activity of AGLE was determined by ABTS radical cation assay, and its $IC_{50}$ was found to be $46.90{\pm}0.31{\mu}g/ml$. Taken together, these results suggest that AGLE may help to alleviate oxidative stress in PC12 neuronal cells.

Antioxidant Effects of Gagam-jangwon-hwan(jiajianzhuangyuanwan) on Hydrogen Peroxide-Induced Oxidative Stress in PC 12 Cell Lines ($H_2O_2$로 유도된 산화적 스트레스에 대한 장원환가감방(壯元丸加減方)의 PC 12 cell 에서의 항산화 효과)

  • Objectives : Antioxidant effects of Gagam-jangwonhwan(LMK01 and 02) water extract against $H_2O_2$-induced oxidative damage and cell death were investigated in rat pheochromocytoma line PC 12. Methods : The cells were treated with LMK01 and 02 water extract and $H_2O_2$, oxidative damage-inducing materials for 24 h. The cellular viability was assessed by WST-1 assay, oxidative damages of the cells by 8-OHdG quantitation, apoptosis by Hoechst 33342 staining assay and activity of antioxidant enzymes by catalase and glutathione peroxidase assay. Results : 1. LMK01 and LMK02 water extracts improved significantly cell viability in $H_2O_2$-treated groups than $H_2O_2$-alone treated cells 2, LMK02 suppressed significantly oxidative damage in $H_2O_2$-treated groups than $H_2O_2$-alone treated cells but LMK01 didn't. Meanwhile, difference of oxidative damages in conditions treated with LMK01 or LMK02 was not significant, 3. The $H_2O_2$ induced-apoptosis in PC 12 cell lines was inhibited effectively by LMK01 and LMK02, and especially the features of apoptosis were obviously reduced in LMK02-treated cells. 4. LMK01 and LMK02 increased significantly activities of both catalase and glutathione peroxidase than those of $H_2O_2$-alone treated group and moreover, LMK02 showed significantly higher activities than those of LMK01. Conclusions : As shown, LMK01 and LMK02 suppressed $H_2O_2$-induced oxidative damage and cell death in PC 12 cell effectively. And they increased activity of major antioxidant enzymes in PC 12 cell line. Therefore, this study suggests the possibility of clinical usage over oxidative stress-induced neurodegenerative disease such as Alzheimer's disease.

Chemical Composition and Protective Effect of Essential Oils Derived from Medicinal Plant on PC12 Neuro-cells Induced by Oxidative Stress (약용식물 유래 정유성분 분석 및 산화 스트레스로부터 PC12 신경세포 보호 효과)

  • The purpose of this study was to investigate the protective effect on oxidative stress induced PC12 cells, and volatile flavor composition of essential oils derived from medicinal plant seeds- Gossypium hirsutum L. (G. hirsutum), Coix lachryma-jobi (C. lachryma-jobi) and Oenothera biennis (O. biennis). The essential oils were obtained by the solvent (hexane) extraction method from the seeds. The essential oils of the seeds were analyzed by the solid-phase micro-extraction gas chromatography mass spectrometry (SPME-GC/MS). The major compounds of G. hirsutum, C. lachryma-jobi and O. biennis were cyclonexanol (16.65%), β-asarone (14.29%) and ylangene (50.01%). The DPPH radical scavenging activity (IC50) was the highest value of 8.52 mg/mL in the O. biennis. Additionally, IC50 values of G. hirsutum and C. lachryma-jobi were 26.76 mg/mL and 36.81 mg/mL. For the oxidative stress on PC12 cells, we treated with hydrogen peroxide (H2O2). The pretreatment of oxidative stress induced PC12 cells with all the essential oils preserved or increased their cell viability and G. hirsutum and O. biennis attenuated the ROS generation (by 68.75% and 56.25% vs. H2O2 control). The results of this study suggest that the essential oils derived from medicinal plant seeds could be used as valuable back data as a natural essential oil material to prevent neurodegenerative diseases by protecting neuro-cells.

PC12 Cell Protective Effects of Broccoli (Brassica oleracea var. italica) Leaf Fraction against H2O2-induced Oxidative Stress (H2O2로 유발된 산화적 스트레스에 대한 브로콜리(Brassica oleracea var. italica) 잎 분획물의 PC12 cell 보호 효과)

  • To examine the physiological effects of broccoli (Brassica oleracea var. italica) leaf, the bioavailable compounds in broccoli leaf extract, and its in vitro neuroprotective effects against $H_2O_2$-induced oxidative stress were examined in this study. The chloroform fraction of broccoli leaf extract had the highest total phenolic content of all the fraction than others, and the highest 2,2"-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical-scavenging activity and malondialdehyde (MDA) inhibitory effect. Intracellular reactive oxygen species (ROS) accumulation resulting in $H_2O_2$-treated in PC12 cells was significantly lower when the chloroform fraction was present in the medium compared to that in PC12 cells treated with $H_2O_2$ alone. In a cell viability assay performed using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), the chloroform fraction showed protective effects against $H_2O_2$-induced neurotoxicity and inhibited lactate dehydrogenase (LDH) release into the medium. High-performance liquid chromatography (HPLC) analysis showed that ferulic acid was the predominant phenolic compound in chloroform fraction of broccoli leaf.

Neuroprotective Effects of Korean Kiwifruit against t-BHP-induced Cell Damage in PC12 Cells (국내산 참다래 추출물의 신경독성 방어효과)

  • Oxidative stress induced by reactive oxygen intermediates has been implicated in a variety of human diseases including neurodegenerative disorders, cancer, cardiovascular and respiratory diseases, and mode of action of environmental toxicants. Tert-butylhydroperoxide (t-BHP) is an organic lipid hydroperoxide analogue, which is commonly used as a pro-oxidant for evaluating mechanisms involving oxidative stress in cells and tissues. In this study, the underlying mechanisms involved in the protective effects of Hwabuk 94 kiwifruit (Actinidia deliciosa cv. 'Hwabuk 94'), which is cultivated in Jeju, on the t-BHP-induced cytotoxicity in PC12 cell. The pretreatment of rat pheochromocytoma cell line PC12 with Hwabuk 94 extract ($1-100\;{\mu}g/ml$) resulted in a significant recovery from t-BHP-induced cell death and increased Bcl-2 and procaspase-3 expression, whereas the expression of Bax and cleaved PARP were decreased in a dose-dependent manner compared to the control. Furthermore, Hwabuk 94 inhibited the t-BHP-induced p38 MAP kinase and extracellular signal-regulated kinase 1/2, but not c-Jun N-terminal kinase activations. Finally, these findings suggest that Hwabuk 94 kiwifruit might attenuate t-BHP-induced PC12 cell cytotoxicity, at least in part, through the inhibition of signaling pathways mediated by the ERK1/2 and p38 MAP kinase.

  • In this paper Wifi robot car working as web sever on wifi network was implemented using NodeMCU ESP-12E board and its operation was verified using web browser of smart phone and PC. Current research result can be applicable to Wifi home automation applications with ESP-12E board.
