Enhancement of ATP-induced Currents by Phospholipase D1 Overexpressed in PC12 Cells

  • Using phospholipase D1 (PLD1)-overexpressing PC12 (PLD1-PC12) cells, the regulatory roles of PLD1 on ATP-induced currents were investigated. In control and PLD1-PC12 cells, ATP increased PLD activity in an external $Ca^{2+}$ dependent manner. PLD activity stimulated by ATP was substantially larger in PLD1-PC12 cells than in control cells. In whole-cell voltage-clamp mode, ATP induced transient inward and outward currents. The outward currents inhibited by TEA or charybdotoxin were significantly larger in PLD1-PC12 cells than in control cells. The inward currents known as $Ca^{2+}$ permeable nonselective cation currents were also larger in PLD1-PC12 cells than in control cells. However, the difference between the two groups of cells disappeared in $Ca^{2+}$-free external solution, where ATP did not activate PLD. Finally, ATP-induced $^{45}Ca$ uptakes were also larger in PLD1-PC12 cells than in control cells. These results suggest that PLD enhances ATP-induced $Ca^{2+}$ influx via $Ca^{2+}$ permeable nonselective cation channels and increases subsequent $Ca^{2+}$-activated $K^+$ currents in PC12 cells.

  • Open architecture controller (OAC) is well known technology in factory automation. To better understand the requirements of OAC, authors have discussed the OAC related topics with a number of control experts who represents different segments of the machining industry. There is no common concept that is accepted or used, however, the common ideas for OAC is the control system that is hardware independent, interchangeable, and easily scalable. This paper presents summary of the understaning and requirements of OAC. Based on the requirements of OAC, authors developed the software based PC-CNC. The main focus of the PC-CNC was on the user customization capability and open interface between control networks in manufacturing system. This paper introduces the developed PC-CNC briefly. In addition to introduction of the PC-CNC, to fill the gap between end users and vendors of OAC, this paper presents two applications using OAC. One is a remote monitoring system. The OPC (Ole for Process Control) standard interface was used to monitor the status of open architecture CNC across network. The other is the remote production management module for machine tools using standard database interface.

  • Recently As the performance of the personal computer has been improving rapidly lots of research for the pc-based numerical computer actively progress in an easy repair maintenance and improving the performance with less cost. This paper presents the design using complex programmable logic device(CPLD). The CPU of Motion Controller that function as the real time control of the independent multi-axis motion the error-detect module and external I/O control made use of 80C196KC, In this paper The PC-NC effectively distributed to the load of NCK(numerical computer kernel) and have the advantage of high speed and precision.

  • The greenhouse control system is designed by personal computer. This system should observe and control the growth conditions for crops in both plastic film and glass greenhouses. In this contribution puts emphasis on graphical user interface for greenhouse control system by personal computer under the aim of creating safer, more effective and more economical services. This system is developed for the requirements of the intelligent greenhouse control system and gives a lot of convenience for farmer who is not familiar with pc technique. The GUI and control system are programmed by Visual C++ and Visual Basic.

  • Due to the improvement of PC Performance and Windows-based OS, PC-based control system is gradually taking over industrial control in the industry site. In this paper, we suggest the method to configure PC-based fault diagnosis system of I/O cards and the method to generate test signals to find out the short conditions of adjacent channels of cables which connect I/O cards with terminal blocks.

Effect of Heparin, Chondroitin Sulfate A(CSA) and Phosphatidylcholine(PC12) on Motility and Acrosome Reaction of Bovine Sperm (Heparin, Chondroitin Sulfate A(CSA) 및 Dilauroylphosphatidyl-choline(PC12)이 소 정자의 활력과 첨모반응에 미치는 영향)

  • This study was carried out to investigate the effect of heparin, CSA and PC12 on sperm motility and acrosome reaction in bovine fresh and frozen semen which were washed and incubated in mTALP, and also the effect of heparin-pretreatment on motility and acrosome reaction in mTALP, and also the effect of heparin-pretreatment on motility and acrosome reaction of sperm treated with PC12, and the results obtained were as follows : 1. When fresh sperm was once washed and then incubated for 15 minutes in mTALP containing heparin 1, heparin 2, CSA and PC12, the percent of motile sperm of PC12 was significantly lower than that of control, heparin 1, heparin 2 and CSA. But the percent of acrosomereacted sperm of PC12 was signifciantly higher than that of control, heparin 1, heparin 2, and CSA. 2. When frozen sperm was once washed and then incubated for 15 minutes in mTALP containing heparin 1, heparin 2, CSA and PS12, there was no significant difference in the percent of motile sperm among treatments, but the percent of acrosome-reacted sperm of PC12 was signifciantly higher than that of heparin 2, and there was no significant difference in the percent of acrosome-reacted sperm among control, heparin and CSA. 3. When fresh sperm was twice washed and then incubated for 15 minutes in mTALP containing heparin and PC12, there was no significant differrence in the percent of motile sperm among treatments, but the percent of acrosome-reacted sperm of PC12 was significantly higher than that of control and heparin. When the sperm was incubated for 120 minutes, the percent of motile sperm of PC12 was significantly lower than that of control and heparin, but the percent of acrosome-reacted sperm of PC12 was significantly higher than that of control and heparin. 4. When fresh sperm was twice washed and preincubated in mTALP containing heparin for 0, 15, 120, and 240 minutes, and then incubated with PC12 for 15 minutes, there was no significant difference in the perce수 of motile sperm among treatments, but the percent of acrosome-reacted sperm of 120 and 240 minutes was significantly higher than that of 0 and 15 minutes.

  • Injection molding has been widely used for the mass production of a plastic product. With the development of the relative technique, various injection molding techniques have been developed and we could get more precise plastic product. The temperature of a melting resin is an important factor in the injection molding and this temperature has direct influence on the quality of a plastic product. In the present injection molding machine, the deriation of a temperature controlled by PID control method is within 2$^{\circ}C$ in the injection molding machine but PID control method takes too much time to stabilize after preheating and its overshoot is so big. We applied fuzzy control to alleriate the problem. In this research, we experimented the fuzzy temperature control with the usage of PC based PLC.

Precise Air-Fuel Ratio Control on Transient Conditions with the PC-ECU in SI Engine (PC-ECU를 이용한 SI 기관의 비정상상태 정밀공연비 제어)

  • In a SI engine, three-way catalyst converter has the best efficiency when A/F ratio is near the stoichiometry. The feedback control using oxygen sensors in the commercial engine has limits caused by the system delays. So it is necessary to control fuel quantity in accordance with intake air amount in order to reduce exhaust emission and improve the specific fuel consumption. Precise A/F ratio control requires measurement of air amount with respect to the cylinder and injection fuel according to the air amount In this paper, we applied nonlinear fuel injection model and developed the algorithm of A/F ratio control. This algorithm includes the methods of measurement of transient air mass flowing into each cylinder, of calculation of injection pulse width for measured air mass, and the method of feedback and engine control by using lambda sensor. Also we developed control program for IBM-PC by using C++ Builder, and tested it in the commercial engine.

The Effects of Perpendicular Needling Laogong ($PC_8$) on the Improvement of Cerebral Hemodynamics in Rats (노궁(勞宮)($PC_8$) 직자(直刺)가 백서(白鼠)의 뇌혈류력학(腦血流力學)에 미치는 영향)

  • Objectives : This study was designed to investigate the effects of acupuncturing $PC_8$ used perpendicular needling method determine the mechanism of action of acupuncturing $PC_8$ by measuring the changes of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in normal rats. Methods : This study also investigated the effects of acupuncturing $PC_8$ on the change of rCBF in cerebral ischemic rats, and revealed the mechanism of its action. In addition, the effects of acupuncturing $PC_8$ on focal ischemic brain injury was studied in cerebral ischemic rats. Results : 1. Acupuncturing $PC_8$ significantly increase rCBF but decreased MABP in normal rats. 2. Acupuncturing $PC_8$ increased of rCBF was significantly inhibited by pretreatment with indomethacin (1mg/kg, i.p.), an inhibitor of cyclooxygenase in normal rats. 3. Acupuncturing $PC_8$ increased of rCBF was significantly inhibited by pretreatment methylene blue (10 ${\mu}g$/kg, i.p.), an inhibitor of guanylate cyclase in normal rats. 4. Acupuncturing $PC_8$ was significantly improved the rCBF than control group increased unstable in cerebral ischemic rats. 5. Acupuncturing $PC_8$ was not significantly improved the rCBF than control group by pretreatment with indomethacin (1mg/kg, i.p.), an inhibitor of cyclooxygenase in cerebral ischemic rats. 6. Acupuncturing $PC_8$ was significantly increased the rCBF than control group by pretreatment methylene blue ($10{\mu}g$/kg, i.p.), an inhibitor of guanylate cyclase in cerebral ischemic rats. Conclusions : In conclusion, our study suggested that acupuncturing $PC_8$ can increase rCBF in normal state, and improve stability of rCBF in ischemic state. In addition, we suggested that mechanisms related with acupuncturing $PC_8$ was involved in the guanylate cyclase pathway.

Coordinated Control Strategies with and without Circulating Current in Unified Power Quality

  • Under traditional unified power quality conditioner (UPQC) control, a UPQC series converter (SC) is mainly used to handle grid-side power quality problems while its parallel converter (PC) is mainly used to handle load-side power quality problems. The SC and PC are relatively independent. The SC is usually in standby mode and it only runs when the grid voltage abruptly changes. In this paper, novel UPQC coordinated control strategies are proposed which use the SC to share the reactive power compensation function of the PC especially without grid-side power quality problems. However, in some cases, there will be a circulating current between the SC and the PC, which will probably influence the compensation fashion, the compensation capacity, or the normal work of the UPQC. Through an active power circulation analysis, strategies with and without a circulating current are presented which fuses the reactive power allocation strategy of the SC and the PC, the composite control strategy of the SC and the compensation strategy of the DC storage unit. Both of the strategies effectively solve the SC long term idle problem, limit the influence of the circulating current, optimize all of the UPQC units and reduce the production cost. An analysis, along with simulation andexperimental results, is presented to verify the feasibility and effectiveness of the proposed control strategies.
