Ultrastructural studies on the neurosecretory cells in the pars intercerebralis of larvae of tasar silkworm, Antheraea mylitta (D) eco-race Bhandara (Lepidoptera: Saturniidae)

  • The cerebral neurosecretory cells (NSC) constitute four paired groups, medial (MNC), lateral (LNC-I, LNC-II) and posterior (PNC) in the brain of larvae of tasar silkworm Antheraea mylitta (D) Eco-race Bhandara. The MNC is the largest groups of peptidergic neurosecretory cells and are located in the pars intercerebralis region. The transmission electron microscopic (TEM), ultrastructure of the NSC confirmed the presence of mitochondria, endoplasmic reticulum, Golgi bodies, lysosomes and neurosecretory granules. The median neurosecretory cells shows secretory activity and release of secretory products, the neurosecretory granules (NSG) in the axons of NSC as well as the blood sinus.

Lesion of Subthalamic Nucleus in Parkinsonian Rats : Effects of Dopamine $D_1$ and $D_2$ Receptor Agonists on the Neuronal Activities of the Substantia Nigra Pars Reticulata

  • Objective : It was hypothesized that dopamine agonist administration and subthalamic nucleus (STN) lesion in the rat might have a synergistic effect on the neuronal activities of substantia nigra pars reticulata (SNpr) as observed in patients with Parkinson's disease. The effects of SKF38393 (a $D_1$ receptor agonist) and Quinpirole (a $D_2$ receptor agonist) were compared in parkinsonian rat models with 6- hydroxydopamine (6-OHDA) after STN lesion. Methods : SKF38393 and Quinpirole were consecutively injected intrastriatally. SNpr was microrecorded to ascertain the activity of the basal ganglia output structure. The effect of SKF38393 or Quinpirole injection on the firing rate and firing patterns of SNpr was investigated in medial forebrain bundle (MFB) lesioned rats and in MFB+STN lesioned rats. Results : The administration of SKF38393 decreased SNpr neuronal firing rates and the percentage of burst neurons in the MFB lesioned rats, but did not alter them in MFB+STN lesioned rats. The administration of Quinpirole significantly decreased the spontaneous firing rate in the MFB lesioned rats. However, after an additional STN lesion, it increased the percentage of burst neurons. Conclusion : This study demonstrated that dopamine agonists and STN lesion decreased the hyperactive firing rate and the percentage of burst neurons of SNpr neurons in 6-OHDA lesioned rats, respectively. Quinpirole with STN lesion increased a percentage of burst neurons. To clear the exact interactive mechanism of $D_1$ and $D_2$ agonist and the corresponding location, it should be followed a study using a nonselective dopamine agonist and $D_1$, $D_2$ selective antagonist.

Geotechnical characteristics and empirical geo-engineering relations of the South Pars Zone marls, Iran

  • This paper evaluates the geotechnical and geo-engineering properties of the South Pars Zone (SPZ) marls in Assalouyeh, Iran. These marly beds mostly belong to the Aghajari and Mishan formations which entail the gray, cream, black, green, dark red and pink types. Marls can be observed as rock (soft rock) or soil. Marlstone outcrops show a relatively rapid change to soils in the presence of weathering. To geotechnically characterise the marls, field and laboratory experiments such as particle-size distribution, hydrometer, Atterberg limits, uniaxial compression, laboratory direct-shear, durability and carbonate content tests have been performed on soil and rock samples to investigate the physico-mechanical properties and behaviour of the SPZ marls in order to establish empirical relations between the geo-engineering features of the marls. Based on the experiments conducted on marly soils, the USCS classes of the marls is CL to CH which has a LL ranging from 32 to 57% and PL ranging from 18 to 27%. Mineralogical analyses of the samples revealed that the major clay minerals of the marls belong to the smectite or illite groups with low to moderate swelling activities. The geomechanical investigations revealed that the SPZ marls are classified as argillaceous lime, calcareous marl and marlstone (based on the carbonate content) which show variations in the geomechanical properties (i.e., with a cohesion ranging from 97 to 320 kPa and a friction angle ranging from 16 to 35 degrees). The results of the durability tests revealed that the degradation potential showed a wide variation from none to fully disintegrated. According to the results of the experiments, the studied marls have been classified as calcareous marl, marlstone and argillaceous lime due to the variations in the carbonate and clay contents. The results have shown that an increase in the carbonate content leads to a decrease in the degradation potential and an increase in the density and strength parameters such as durability and compressive strength. A comparison of the empirical relationships obtained from the regression analyses with similar studies revealed that the results obtained herein are reasonably reliable.

Distribution and Ultrastructure of Dopaminergic Neurons in the Substantia Nigra of Mongolian Gerbil (Meriones unguiculates) (모래쥐 흑색질의 도파민성 신경세포의 분포와 미세구조)

  • The substantia nigra of the Mongolian gerbil was studies by tyrosine hydroxylase immunohistochemistry and immunoelectron microscopy with preembedding method. The purpose was to obtain information on the distribution and ultrastructure of the Tyrosine hydroxylase immunoreactive and dopaminergic neurons in the substantia nigra, in order to provide the necessary background for the gerbil. Large number of tyrosine hydroxylase immunoreactive neurons were located in the compact part of substantia nigra. Findings in the gerbil, compared to observations in the other species, included the presence of prominent bundles of tyrosine hydroxylase immunoreactive cytoplasmic processes passing in the dorsoventral direction from pars compacta into pars reticulata at middle and caudal levels of the substantia nigra, and the presence of a distinct tyrosine hydroxylase immunoreactive substantia nigra pars lateralis. Tyrosine hydroxylase immunoreactive neurons had well-developed cell organelles, especially rough endoplasmic reticulum, free ribosome and poly-ribosome, and showed the infoldings of the nuclear envelope. We anticipate that the present description of the cellular organization of the tyrosine hydroxylase immunoreactive dopaminergic area in the substantia nigra of gerbil will be useful for the animal experimental model of Parkinson's disease.

Postembryonic Development of Leucokinin I-Producing Neurons in the Brain of Insect Spldoptera litura

  • Antisera against the myotropic neuropeptide leucokinin I, originally isolated from head extracts of the cockroach Leucophaea maderae, have been used to investigate the distribution of the leucokinin I-immunoreactive (LK I-IR) neurons in the brain of the common cutworm, Spodoptera Iitura, during postembryonic development. The LK I-IR neurons are found at the larval stages (excluding first instar larval stage), pupal stages, and adult stage, of which the brains have been examined in this experiment. The number of the LK I-IR neurons in the brain increases from the second instar larva to the fifth instar larva which has about 32, the largest number in all postembryonic stages. Thereafter, the LK I-IR neurons begin to decrease in number. During the pupal stages, smaller number of LK I-IR neurons persist in the brains; 6 or 4. At adult stage the brain contains 8 LK I-IR neurons. The LK I-IR cell bodies are distributed in each dorsal cortex of both cerebral hemispheres in the second instar larva and through all the neuromeres of the brain during later larval stages, despite of being a large number of the LK I-IR cell bodies in dorsolateral neuromeres. At pupal stages, most of the LK I-IR cell bodies are found in the pars intercerebralis. Extremely small number of the LK I-IR cell bodies are localized in the pars lateral is. Adult brain contains the LK I-IR cell bodies in the pars intercerebralis and the middle cortex of the posterior brain. The LK I-IR nerve processes can be easily found in the neuropils of almost all the neuromeres in the brains of third, fourth, fifth and sixth instar larvae. Most of the LK I-IR nerve fibers in those brains are originated from the LK I-IR cell bodies located in the brains. The LK I-IR cell bodies which have very weak reactivities to the antisera do not show projection of the LK I-IR nerve processes in the brains.

Electrical properties and ATP-sensitive K+ channel density of the rat substantia nigra pars compacta neurons (랫드 흑질 신경세포의 전기적 특성과 ATP-sensitive K+채널의 전류밀도)

  • Substantia nigra is known to highly express glibenclamide binding site, a protein associated to ATP-sensitive $K^{+}$ ($K_{ATP}$) channel in the brain. However, the functional expression of $K_{ATP}$ channels in the area is not yet known. In this work, we attempted to estimate the functional expression of $K_{ATP}$ channels in neurons of the substantia nigra pars compacta (SNC) in young rats using slice patch clamp technique. Membrane properties and whole cell currents attributable to $K_{ATP}$ channel were examined by the current and voltage clamp method, respectively. In SNC, two sub-populations of neurons were identified. Type I (rhythmic) neurons had low frequency rebound action potentials ($4.5{\pm}0.25Hz$, n=75) with rhythmic pattern. Type II (phasic) neurons were characterized by faster firing ($22.7{\pm}3.16Hz$, n=12). Both time constants and membrane capacitance in rhythmic neurons ($34.0{\pm}1.27$ ms, $270.0{\pm}11.83$ pF) and phasic neurons ($23.7{\pm}4.16$ ms, $184{\pm}35.2$ pF) were also significantly different. The current density of $K_{ATP}$ channels was $6.1{\pm}1.47$ pA/pF (2.44~15.43 pA/pF, n=8) at rhythmic neurons of young rats. Our data show that in SNC there are two types of neurons with different electrical properties and the density of $K_{ATP}$, channel of rhythmic neuron is about 600 channels per neuron.

Anatomical studies on pattern of branches of portal veins in Korean native cattle (한우문맥(韓牛門脈)의 분지(分枝)에 관한 해부학적(解剖學的) 연구(硏究))

  • The distribution of portal veins within the liver in 30 Korean native cattle were observed. Vinylite solution was injected into portal veins of eighteen specimens for cast preparation. The angiography was prepared in twelve specimens by injecting 30% barium sulfate solution into portal veins, and then radiographed on a X-ray apparatus(Shimadzu 800MA 120Kvp). The results were summarized as follow: 1. The Vena portae was divided immediately upon entering the liver into a very short Truncus dexter venae portae($14.75{\pm}4.86$ : 6.9~23.1mm) and a long Truncus sinister venae portae($94.16{\pm}9.62$ : 110~150mm). 2. The Truncus sinister venae portae runs of first in the long axis of the liver from the Porta hepatis toward the left lobe. At the boundary between the quadrate and left lobes it bends sharply 50 to 80 degrees toward the Incisura ligamentum teretis, and after a course of 36. 5 to 54.mm between the quadrate and left lobes, ends abruptly. The Truncus sinister venae portae is divided for description into the Pars transversa, from the Porta hepatis to the flexure, and the Pars umbilicalis, from the flexure to the end. 3. The branches of Venae portae were Ramus ventralis lobi sinistri, Ramus intermedius lobi sinistri, Ramus dorsalis lobi sinistri, Ramus lobi quadratii, Ramus ventralis lobi dextri, Ramus intermedius lobi dextri, Ramus dorsalis lobi dextri, Rami processus caudatorum and Rami processus papillarum. 4. The Ramus intermedius lobi sinistri was arised from the left surface of the Pars umbilicalis, and was origined on the common trunk with Ramus dorsalis lobi sinistri(3 cases, 10%) or Ramus ventralis lobi sinistri(3 cases, 10%). 5. The Rami lobi quadratii consisted of the vein(15 cases, 50%) or two veins(15 cases, 50%), and was observed on the arched-shaped at 2 cases (6.6%) of the liver. 6. The Rami processus caudatorum consisted of one vein(28 cases, 93.3%) or two veins(2 cases, 6.6%). The former were formed common trunk with R, dorsalis lobi dextri(7 cases, 23.3%) or R. ventralis lobi dextri (2 cases, 6.6%). 7. The Rami processus papillarum were arised from the dorsal border of Pars transversa, and also gave off many small branches supplied papillary process of the caudate lobe. 8. The anastomosis on the branches of Vena portae was observed in the intralobar and interlobar areas. 9. The Truncus dexter venae portae and Truncus sinister venae were ramified many secondary branches that were radiated within the liver. 10. On the diaphragmatic surface, small vessels of the portal veins were observed, while there were big ones on the visceral surface. 11. The ramified angles at Ramus dorsalis lobi dextri, Rami processus papillarum, Ramus dorsalis lobi sinistri, Ramus intermedius lobi sinistri, Ramus ventralis lobi sinistri, Rami lobi quadratii, Rami processus caudatorum, Ramus ventralis lobi dextri and Ramus intermedius were 10~50, 70~110, 100~150, 140~170, 185~220, 270~330, 240~300, 270~320 and 340~10 degrees, respectively.

Anatomical studies on pattern of branches of hepatic arteries in Korean native cattle (한우간동맥(韓牛肝動脈)의 분지(分枝)에 관한 해부학적(解剖學的) 연구(硏究))

  • The distribution of the hepatic arteries within the liver in 30 Korean native cattle were observed. Vinylite solution was injected into the hepatic arteries of 10 livers for cast preparation. The angiography was prepared in 20 livers by injected 30% barium sulfate solution into the hepatic arteries and then radiographed on X-ray apparatus. The results were summarized as follow ; 1. The hepatic arteries were divided into the Ramus (R.) sinister, R. dexter and R. intermedius. The intrahepatic arteries were derived from the R. sinister and R. dexter. The R. intermedius divided into the Arteria(A.) gastrica dextra, A. gastroduodenalis and A. cystica. 2. The right branch (ramus dexter) of the hepatic artery supplies the right lobe and caudate process. It consisted of the A. dorsalis lobi dextri, A. ventralis lobi dextri and A. lobi caudati. They formed one common trunk with three arteries(6 cases, 20%), and two arteries formed common trunk with the A. ventralis lobi dextri and A. lobi caudati (16 cases, 52.8%), or with A. dorsalis lobi dextri and A. lobi caudati (5 cases, 16.5%). The three arteries arose from the A. hepatia independently(3 cases, 10%). 3. The left branch (ramus sinister), larger than the right and often double (6 cases, 20%), supplies the caudate, quadrate and left lobes, and often gives origin to the A. gastrica dextra. 4. The branches of hepatic artery in the left lobe were derived from A. dorsalis lobi sinistri, A. ventralis lobi sinistri and A. intermedius lobi sinistri. The A. intermedius lobi sinistri were absent in some cases (3 cases, 10%). They almostly originated a single branch and often two branches. 5. The A. cystica arose from the R. intermedius(27 cases, 90%) and A. hepaica(3 cases, 10%). 6. The Rami processus papillarum arose from dorsal border of the Pars transversa of the left branch(19 cases, 62.7%), the left branch and right branch (9 cases, 30%) or A. hepatica(2 cases, 6.6%). 7. The A. lobi quadrata arose from either the Pars transversa of left branch (12 cases, 40%) or A. hepatica(4 cases, 13.2%). It also arose from both Pars transversa of left branch and A. ventralis lobi sinistri (3 cases, 10%). 8. The anastomosis on the branches of A. hepatica was observed in intrahepatic and cystic arteries. The left and right branches ramified several secondary radiating branches within the liver. 9. The hepatic arteries formed a very short common trunk of the right branch ($18.75{\pm}11.89$ : 2.20~44.8mm) and a long Pars transversa of the left branch($72.85{\pm}20.04$ : 28.2~104.1mm). 10. The ramified angles at the A. dorsalis lobi dextri, A. ventralis lobi dextri, A. lobi sinistri, A. ventralis lobi sinistri, A. lobi quadrata, A. cystica and Rami processus papillarum were 50-80, 270-300, 340-20, 120-160, 160-290, 200-230, 250~290, 230~280 and 80~120 degrees, respectively.

  • The study on the nerve cells in the pars intercerebralis(IP) of 5-day-old cabbage butterfly Pieris rapae L. was performed to observe their ultrastructures and classify them on the basis. of the differences in size, shape and relative distribution cf cell organelles. The brain-subesophageal ganglion complex was fixed in 1% paraformaldehyde-1% gluaraldehyde mixture and embedded in araldite mixture. The transverse thin sections of IP were stained with uranyl acetate and lead citrate and examined by Hitachi 500 and ]EM 100B electron microscope. Five distinct types. of nerve cells are recognized and are arbitrarily designated as Type I, Type II Type III, Type IV and Type V. Type I neurone: These neurones are neurosecretory cells. Several neurosecretory cells are. recognized in the pars intercerebralis. They are roughly round or peach-shaped cells measuring $13{\sim}25{\mu}m$ in diameter. The rounded nucleus shows about $5{\sim}10{\mu}m$ in diameter. The chromatin is predominantly diffused with only occasional dense patches. The perikaryon contains numerous. mitochondria, free polyribosomes and neurosecretory granules. The neurosecretory granules are relatively uniform in electron density, and each one is about $100{\sim}400{\mu}m$ in diameter and surrounded by a single membrane. The granules are also observed mostly as in groups. In one group of neurones the cisternae of endoplasmic reticulum are distended or in other group of neurones are not distended. Golgi saccules are slightly dilated at their lateral extremities and contains. frequenty dense rounded materials. Type II neurone: Thes have the largest soma in the pars intercerebralis about $30{\sim}35{\mu}m$ in diameter. They also show roughly polygonal in shape. The nucleus is elongated or sickle-shaped. The chromatin is mainly in the euchromatin form. The perikarya in these cells are well populated with populated with free ribosomes and contain numerous mitochondria and Golgi bodies. The cisternae of granular endoplasmic reticulum are also well distributed. Type III neurone: They are oval or spindle-shaped and also medium-sized. neurones approximately $15{\sim}17{\mu}m$ in length. The nucleus is oval or slightly elongated in shape and $8{\sim}9{\mu}m$ in length. The chromatin occurs in diffused form. The cytoplasm contains many filamentous or oval mitochondria. The perikaryon has also numerous free polyribosomes and cisternae of granular endoplasmic reticulum. Type VI neurone: They are roughly polygonal in shape probably due to the close approximation of the adjacent cells. The soma is about $7{\sim}8{\mu}m$ in diameter. The nucleus is round or oval in shape and $5.0{\sim}5.8{\mu}m$ in diameter. The necleus also occupies a large proprion of the cell body. The perikaryon is well populated with free ribosomes and contains several mitochondria and cistenae of granular endoplasmic reticulum. Type V neurone: These neurones are similar to Type VI neurones in various respects such as cell size and cell inclusion, but they differ from Type IV neurones in shape. The soma is oval or slightly elongated. The cell body contains several filamentous and oval mitochondria.

  • 국내의 첫액금로가 될 KALIMER의 안전등급 잔열제거 계통인 PSDRS의 설계 및 설계/해석방법론 설정을 위하여 기존의 PSDRS 해석 코드인 기존의 PARS를 개선시킨 PARS2-iB를 개발하고 이를 이용하여 PSDRS내부 전열모드의 정성적 및 정량적 특성, 그리고 PSDRS 설계에서의 우선 고려 대상 설계인자 및 PSDRS 설계 해석 방법론 설정을 위한 PSDRS 작동 mechanism의 열유체적 연결 특성을 파악하였다.
