Luteolin Inhibits Extracellular Signal-Regulated Kinase Pathway Through Protease-Activated Receptors (-2 and -4) and Their Agonist Activity

  • Luteolin is a major flavonoid of Lonicera japonica and has anti-inflammatory effect. The activation of proteinase-activated receptor (PAR)-2 and -4 by trypsin appears to play a role in inflammation, In the present study, we examined the inhibitory effects of luteolin on activation of trypsin-induced human leukemic mast cells (HMC-1). HMC-1 cells were stimulated with trypsin, PAR-2 and PAR-4 agonist, in the presence or absence of luteolin. The level of TNF-${\alpha}$ secretion was measured by enzyme-linked immunosorbent assay (ELISA). The expression of tryptase and phosphorylated-extracellular signal-regulated kinase (ERK) were assessed by Westem blot analysis. Moreover, trypsin activity was measured by the substrate Bz-DL-Arg-p-nitroanilide (BAPNA). TNF-${\alpha}$ secretion and Tryptase expression in trypsin-stimulated HMC-1 cells were markedly inhibited by pretreatment of luteolin. Furthermore, the pretreatment of luteolin resulted in the reduction of ERK phosphorylation and trypsin activity. These results suggest that luteolin might has the inhibitory effects on the PAR-2 and -4-dependent inflammation.

Analysis of Polycyclic Aromatic Hydrocarbons in Agricultural Soils by Gas Chromatography-Ion Trap Tandem Mass Spectrometry

  • An investigation has been carried out on collision-induced dissociation (CID) in the development of an analytical protocol for the determination of polycyclic aromatic hydrocarbons (PARs) by ion trap tandem mass spectrometry. Two different considerations were used to choose the optimal CID conditions for complex matrix environmental samples, namely, to determine the highest signal-to-noise (SIN) ratio and the other to eliminate the background interferences originated from complex matrix samples. The PAR content of agricultural soil was measured to estimate overall distribution of PAR in throughout the country, we collected and analyzed 226 soil samples from paddy and upland soil. The average content of total PAR in all samples was 236 ${\mu}g$ $kg^{-1}$, and the range was from 23.3 to 2, 834 ${\mu}g$ $kg^{-1}$. The overall distribution of PAR was found to be closely related to the pollution sources, the size of city and the type of industry.

Anti-inflammatory Effect of Baenong-san in Proteinase-activated Receptor-2-mediated Paw Edema (배농산이 프로테이나아제 활성수용체-2에 의한 흰쥐 발바닥 부종에 미치는 항염효과)

  • The Baenong-san has long been used for treatment of inflammatory in Korea. In this study, the anti-inflammatory effects of Baenong-san water extract (BWX) were investigated in proteinase-activated receptor-2 (PAR2)-mediated rat paw edema. Paw edema was induced by injection of trypsin or trans-cinnamoyl-LIGRLO-NH₂ (to-NH₂) into hindpaw of rats. BWX (10, 50, 100 and 200mg/kg) was orally administered 1 h before induction of inflammation. At doses of 50, 100 and 200 mg/kg, BWX showed significant inhibition of both change in paw volume and vascular permeability. BWX(100mg/kg) significantly also inhibited PAR2 agonist-induced myeloperoxidase (MPO) activity in paw tissue. This study demonstrated that BWX has an anti-inflammatory action for PAR2-mediated paw edema.

Skin Barrier Recovery by Protease-Activated Receptor-2 Antagonist Lobaric Acid

  • Atopic dermatitis (AD) results from gene and environment interactions that lead to a range of immunological abnormalities and breakdown of the skin barrier. Protease-activated receptor 2 (PAR2) belongs to a family of G-protein coupled receptors and is expressed in suprabasal layers of the epidermis. PAR2 is activated by both trypsin and a specific agonist peptide, SLIGKV-$NH_2$ and is involved in both epidermal permeability barrier homeostasis and epithelial inflammation. In this study, we investigated the effect of lobaric acid on inflammation, keratinocyte differentiation, and recovery of the skin barrier in hairless mice. Lobaric acid blocked trypsin-induced and SLIGKV-$NH_2$-induced PAR2 activation resulting in decreased mobilization of intracellular $Ca^{2+}$ in HaCaT keratinocytes. Lobaric acid reduced expression of interleukin-8 induced by SLIGKV-$NH_2$ and thymus and activation regulated chemokine (TARC) induced by tumor necrosis factor-a (TNF-${\alpha}$) and IFN-${\gamma}$ in HaCaT keratinocytes. Lobaric acid also blocked SLIGKV-$NH_2$-induced activation of ERK, which is a downstream signal of PAR2 in normal human keratinocytes (NHEKs). Treatment with SLIGKV-$NH_2$ downregulated expression of involucrin, a differentiation marker protein in HaCaT keratinocytes, and upregulated expression of involucrin, transglutamase1 and filaggrin in NHEKs. However, lobaric acid antagonized the effect of SLIGKV-$NH_2$ in HaCaT keratinocytes and NHEKs. Topical application of lobaric acid accelerated barrier recovery kinetics in a SKH-1 hairless mouse model. These results suggested that lobaric acid is a PAR2 antagonist and could be a possible therapeutic agent for atopic dermatitis.

  • This paper describes an efficient coding method for Ko-rean characters (alphabet) using a three-state transition graph. Par-allel hangul Dynamic Coding Method (PHDCM) compresses about 3.5 bits per Korean character compared with other coding techinques. When we ran the method on a MasPar machine it achieved a 49.314-fold speedup with 64 processors having 10 million orean characters

Performance Comparison of Two Parallel LU Decomposition Algorithms on MasPar Machines

  • This paper presents a performance study of two LU decomposition algorithms on two massively parallel SIMD machines: the 16K processor MasPar MP-1 and the 4K processor MasPar MP-2. The paper presents experimental results and an analysis of the algorithms to explain the results. While the blocked and the nonblocked algorithms for LU decomposition have been studied individually by others, we compare the two algorithms and identify the tradeoffs between them. Our analysis of the blocked algorithm shows how the block size affects the interprocessor communication cost and the memory read/write overhead. The analysis in this paper is useful to determine an optimum block size for the blocked algorithm.

  • This study was conducted to investigate the influence of shading treatment on the photosynthetic rate, transpiration rate, stomatal conductance and its any correlations in Liriope platyphylla $W_{ANG}\;et\;T_{ANG}$. Followings were achieved as a conclusion. The net photosynthetic rate was increased as the PAR was increased and reached maximum at the $700-1000{\mu}mol/m^2/s$ of PAR in all of leaves, also this treatment caused a higher net photosynthetic rate in comparison with control. It shows the tendency of increasing stomatal conductance caused by the increment of PAR. The diurnal changes of photosynthesis, transpiration rate and stomatal conductance were increased as the PAR was increased in the morning, but they indicated a decreased tendency in broad day. The relationship between net photosynthetic rate and stomatal conductance is well fit by the first regression linear equation. However, the values obtained from the linear equation have the different, respectively, and have highly significance. From the above results, net photosynthetic rate of shading treatment is higher than control in the same stomatal conductance. Different first regression linear equation were obtained between the transpiration rate and stomatal conductance, photosynthesis and stomatal conductance in during the control and shading treatment, too.

Plasma Catalytic Methane Conversion over Sol-gel Derived Pt/TiO2 Catalyst in a Dielectric-barrier Discharge Reactor (DBD 반응기에서 솔-젤 법으로 제조된 Pt/TiO2 촉매를 이용한 메탄의 플라즈마 전환반응)

  • Plasma catalytic methane conversion was carried out in the presence of sol-gel derived $Pt/TiO_2$ catalysts within a dielectric-barrier discharge (DBD) reactor. Plasma-assisted reduction (PAR) was applied to reduce the prepared $Pt/TiO_2$ catalysts in DBD reactor, and prepared catalysts were successively reduced by PAR within 20 min irrespective of the Pt loading and the calcination temperature. The highest methane conversion was 40% when 3 wt% $Pt/TiO_2$ and 5 wt% $Pt/TiO_2$ catalysts were used after calcination at $600^{\circ}C$. The selectivities of light alkanes ($C_2H_6$, $C_3H_8$, $C_4H_{10}$) were highly increased when $Pt/TiO_2$ catalysts were used in DBD reactor.

  • If a hydrogen explosion occurs in a containment building, its multiplex defense wall may be destroyed and a large amount of radioactive material may be released. The hydrogen occurred interacting with melting fuel rods must be effectively controlled and removed. however, the countermeasures for reducing explosion risk are difficult to carry out, due owing to the various variety of accident scenarios causes and the irregularity of hydrogen distribution and behavior. In this study, We examine the guide structures while considering the ambient flows, in order to improve the efficiency of PAR the widely used Passive Autocatalytic Recombiner(PAR). We simulate the fluid behavior and the hydrogen reduction rate were simulated when a guide is attached to the two-step catalyst PAR. For an upward flow, the consisting of a height of 150mm, a gap of 0mm, and a performs $60^{\circ}$ showed the best. In contrast, for a sideways flow, a consisting of the height of 150mm, a gap of 100mm, and a performs $60^{\circ}$ showed the best in the case of side ward flow. for a downward flow, a consisting of the height of 50mm and a directly attached guide produce the best in the case of down ward flow results.
