This study was carried out to characterize virus-like particles (VLPs) in Pleurotus ostreatus. The white and the dark gray mutants frequently observed in mushroom farms of Pleurotus ostreatus (Wonhyeong-neutari). A 5.8kb segments of dsRNA was detected only in the white mutants but not in the dark gray mutants. The VLPs were purified from the fruit bodies by Polyethylene Glycol (PEG) and ultracentrifugation. Electron microscopy analysis showed that VLPs were isometric about 14, 20~45nm in diameter. Further study is needed to reveal the morphological and yield variations of mushroom strains including VLPs observed in the mushroom farms. Also it is needed to maintain fundamental research for taxonomy, diagnosis, and physiology of VLPs in the mushroom strains.

Pyrrosia lingua which belongs to Polypodiaceae has been used as a traditional medicine for the treatment of urinary system inflammation, urination disorder, and bronchitis. However, there are not enough phytochemical and pharmacological studies of P. lingua up to now. Here in this study, the protective effect of MeOH extract of whole plant of Pyrrosia lingua (MPL) against 2% glucose-induced toxicity was investigated using Caenorhabditis elegans (C. elegans) model system. We found that MPL significantly extended the lifespan of wild-type nematode under normal culture condition. MPL also effectively recovered the decreased lifespan caused by 2% glucose-toxicity. In addition, MPL efficiently attenuated the increased glucose concentration inside of nematode. Further studies evaluating diabetes-related factors revealed that MPL reduced both intracellular ROS and lipid accumulation which were up-regulated under 2% glucose supplement condition. Our data also showed that MPL improved the 2% glucose-induced shortened body movement of nematode. Lastly, we carried out genetic studies using several single gene knockout mutants to establish the possible target of MPL. Our results demonstrated that genes such as daf-2 and daf-16 were responsible for the protective activity of MPL against 2% glucose-induced toxicity. These results indicate that MPL exerts protective action against 2% glucose via regulation of insulin/IGF-1 sinaling pathway and FOXO activation.

Mesenchymal stem cells (MSCs) are multipotent stem cells that can be differentiated into a variety of cell types, including adipocytes, osteoblasts, chondrocytes, β-pancreatic islet cells, and neuronal cells. MSCs have been reported to exhibit immunomodulatory effects in many diseases. Many studies have reported that MSCs have distinct roles in modulating inflammatory and immune responses by releasing bioactive molecules. Exosomes are cell-derived vesicles present in biological fluids, including the blood, urine, and cultured medium of cell cultures. In this study, we investigated the immunomodulatory effects of mouse adipose tissue-derived MSCs (mAD-MSCs), cultured medium (MSC-CM) of mAD-MSCs, and mAD-MSC-derived exosomes (MSC-Exo) on lipopolysaccharide (LPS)-induced RAW 264.7 cells. We observed that the expression levels of IL-1β, TNF-α, and IL-10 were significantly increased in LPS-stimulated RAW 264.7 cells compared to those in LPS-unstimulated RAW 264.7 cells. Additionally, these values were significantly (p < 0.05) decreased in mAD-MSCs-RAW 264.7 cell co-culture groups, MSC-CM-treated groups, and MSC-Exo-treated groups. MSCs can modulate the immune system in part by secreting cytokines and growth factors. We observed that immunomodulatory factors such as IL-1β, TNF-α, and IL-10 were secreted by mAD-MSCs under co-culturing conditions of mAD-MSCs with activated RAW 264.7 cells. In addition, mAD-MSC-derived exosomes exhibited similar immunomodulatory effects in activated RAW 264.7 cells. Therefore, our results suggest that mAD-MSCs have an immunomodulatory function through indirect contact.
Beta-catenin is very important in early development including involvement in cell adhesion, cell signaling, and developmental fate specification. Cell-cell interaction is an important process during mammalian embryonic development. In preimplantation embryos, embryonic compaction is the process of increased cellular flattening and adhesion of junctional complexes and results in a polarized distribution. Beta-catenin is associated with embryonic compaction in mammals. Here, we examined the relationship between beta-catenin expression and compaction in porcine embryos derived from in vitro fertilization. First of all, we investigated beta-catenin expression in each embryonic developmental stage and also focused on expression pattern according to full, partial and non-compaction at morula stage. We used the immunocyto-chemical method in this research. To confirm compaction affects on the embryonic development, we compared between compaction and developmental rates to the blastocyst. The result showed that compaction and non-compaction rates were 14.6% and 63.8% at 4 days after IVF, respectively The developmental rates to the blastocyst and their total cell number were 50.9% vs 36.4% and 41.4$\pm$11.5 vs 26.8$\pm$12.7 in compaction and non-compaction groups. Although no difference was detected in the ratio of ICM to total cells between two groups, total cell number of the blastocysts in compaction group was superior to that of the blastocysts in non-compaction group (P<0.05). Expression of beta-catenin appeared in the boundary of membrane surface between blastomeres in 2- and 4-cell stage, and observed irregular pattern from 8-cell to blastocyst stage. We also investigated beta-catenin expression pattern according to the degree of compaction in the 3 groups; full, partial (>50%) and non-compaction. The expression signal in fully compacted embryos was stronger than those of partial and non-compacted embryos. Especially, beta-catenin expression appeared various patterns in morula stage suggesting the aberrant distribution of beta-catenin is affected by compaction patterns. Our results suggest that abnormal beta-catenin expression was affected by embryo quality and further development in porcine embryos in vitro.
The study was performed to investigate the effect of incorporation of green manures (GM) into a sandy loam soil for organic vegetable production in the condition of plastic film house, relating to growth and yield of tomato crop. Three species of GM as perennial ryegrass, sudangrass and soybean are cultivated during the rest time of summer season and incorporated into soil just after the harvest. Thereafter tomato crop was transplanted as the following crop to soil incorporated GM. Among GM, soybean was proper as GM crop for organic farming, due to the effect of yield increase by continuous supply of nitrogen on following the tomato crop. Yield of tomato crop after soybean incorporation into soil was $4.2Mg\;ha^{-1}$ similar to $4.4Mg\;ha^{-1}$ of N-P-K standard fertilization (conventional) treatment. But perennial ryegrass and sudangrass were improper, because the biomass yield of perennial ryegrass was very low due to growth retardation by high temperature during summer season and soil incorporation of sudangrass as GM results in yield decrease of following the tomato crop caused by high C/N ratio of sudangrass itself. In conclusioa soybean incorporation into soil had advantage of producing conventional level on following the tomato yield and therefore it could recommend as GM for organic vegetable production.
Cyclic adenosine 3'5'-monophosphate (cyclic AMP) has been frequently accepted as an intracellular messenger for receptor-mediated action of opioids. In this experiment, it was designed to determine the interaction of dopaminergic and opioidergic system in the mouse striatum in normal and chronic haloperidol treated groups. Haloperidol 750ug/kg I.P. for 10 days was performed for dopamine denervation. The morphine, DAGO, DPDPE, and U5O,488H inhibited the increase of haloperidol-induced cyclic AMP content in chronic haloperidol treated mouse striatum. The inhibition of DAGO and DPDPE showed significant increase compared to normal mouse striatum. Naloxone showed antagonistic effect on the morphine and U5O,488H in chronic haloperidol treated group, and showed antagonistic effect on morphine, DAGO, DPDPE, and U5O, 488H in normal mouse striatum. These findings support that there is a functional interrelationship of dopaminergic and opioidergic pathway in the striatum. This result provides an evidence that following destruction of striatal dopaminergic neuron, there are some changes of cAMP content on the ${\mu},\;{\gamma},\;and\;{\kappa}$ opioid receptor, but the ${\kappa}$ opioid receptor still has its function.
Purpose : Glomerular filtration rate (GFR) is a fundamental parameter in assessing renal function and predicting the progression of chronic renal disease. Because the use of serum creatinine has several disadvantages, many studies have investigated the use of cystatin C for estimating GFR. We compared creatinine clearance and GFR with formulas using serum creatinine and cystatin C. Methods : We retrospectively analyzed 211 patients with various renal diseases and classified them into two groups according to creatinine clearance (Group 1: CrCl >$90mL/min/1.73m^2$, Group 2: CrCl <$90mL/min/1.73m^2$). We measured serum creatinine, cystatin C, and creatinine clearance. We calculated GFR using the Schwartz, Counahan, Filler and Lepage, Bokencamp et al, and Grubb et al formulas. Results : GFR determined by the Schwartz formula had the highest correlation to creatinine clearance (r=0.415, P=0.00). GFR determined by various formulas using cystatin C had lower correlation to creatinine clearance (r=0.187, r=0.187, r=0.291). The Schwartz and Counahan formulas showed greater diagnostic accuracy in detecting decreased GFR than cystatin C in group 2 (areas under the curve: Schwartz, 0.596; Counahan, 0.572; Filler, 0.512; Bokencamp, 0.508; and Grubb, 0.514). Conclusion : GFR determined by the Schwartz and Counahan formulas using serum creatinine showed higher correlation coefficient than that determined by formulas using cystatin C. The formulas using cystatin C were not superior to those using serum creatinine in detecting decreased GFR. Cystatin C measurement was not satisfactory for assessing GFR in patients whose renal function was not severely decreased.

Temporal changes of Chl-α, physical and chemical factors were investigated by diurnal observation at 2-hour interval at three fixed stations in the western Chinhae Bay from 12 Aug. to 13 Aug. 1999. Difference of dissolved oxygen between surface and bottom layer was maximum when the thermocline were strong. Organic distribution such as COD was affected by the growth of phytoplankton. Limitting factor was nitrogen, that is, inorganic nitrogen plays a significant role on regulating the algal growth. Surface distribution of dissolved inorganic nitrogen was very low compared to bottom layer by uptake of organisms. Maximum value of Chl-α at station C2 and C11 were observed from subsurface layer, ranges of which exceeded possibility concentration of red tide outbreak, 10 mg/㎥. On the other hand, that of C15 exist at surface layer. In this area, DIN and DIP concentrations increased by input sources such as rainfall and benthic flux before the bloom of phytoplankton. Accumulation of phytoplankton occurred at subsurface layer by the rapid uptake of DIN, especially nitrate ion, when strong thermocline existed as approach to the afternoon, which led to the increase of organics in water column and oxygen deficiency water mass at bottom layer until late at evening. Since then, DIN increases gradually as water temperature decrease to minimum. The quantitative understanding of nitrogen of fluxed to and from the various sources is necessary for environmental management.

This study was conducted to compare the biological activities of 7 melania snails from the family Pleuroceridae (Semisulcospira coreana, Koreanomelania nodifila, Semisulcospira forticosta, Koreoleptoxis globus ovalis, Semisulcospira libertina, Semisulcospira tegulata and Semisulcospira gottschei) in Korea. Among the 7 species, S. coreana, Korean. nodifila, S. forticosta and S. gottschei showed over 80% cytotoxicities on three cancer cell lines (SNU-1, A549 and Hep 3B) compared to the non-treatment, whereas S. libertina and S. tegulata showed almost no growth inhibition activities on the same cancer cell lines. In relation to ACE inhibition activity, only S. coreana, Korean. nodifila, and S. forticosta showed over 60% ACE inhibition activities, whereas other melania snails exhibited inhibition activities of lower than 25%. DPPH radical scavenging activities were also determined, and used to categories melania snails into three groups based on Duncan's multiple range test at P<0.05. The amount of TNF-${\alpha}$ produced by in vitro mouse peritoneal macrophage was determined according to bioactivity on L-929 cells. Three melania snails, S. coreana, Korean. nodifila and S. gottschei, exhibited 95.2%, 89.7% and 93.7% cell death(%) on L-929 cells, respectively. Glucose-6-phosphate dehydrogenase inhibitory activity was also obtained in the extract of S. coreana (31.9%) and Korean. nodifila (28.1%), showing that these extracts can be used as supplemental dietary health foods. In conclusion, we believe that the extracts of melania snails should be given due consideration in functional health food development.

The purpose of this study was to investigate muscle activation of lower extremity such as rectus femoris, tibialis anterior and soleus according to 0cm(bare foot), 4cm and 7cm heel height of shoe on the rocking surface in older women. 20 older women who did not have any lower musculoskeletal and neurological disorders in the past were participated in this study. Each subject was standing for 15 seconds on the level 8 of Biodex Stability System (BSS) while wearing 4cm and 7cm heel height shoes including bare foot. Electromyography was used to measure muscle activation of lower extremity, and the muscle activation was expressed as a percentage of maximal voluntary isometric contraction (% MVIC). We measured % MVIC of three muscles during 5 seconds except for the first 5 seconds and last 5 seconds. SPSS 12.0 program was used for this study. Repeated one-way analysis of variance(ANOVA) was performed to compare the significant difference among the muscles of lower extremities according to heel heights of shoe on the rocking plate. % MVIC of each muscle such as rectus femoris, tibialis anterior and soleus regarding heel heights of shoe had statistically significant differences (p<0.05). The results of contrast test were as follows; 1) % MVIC of rectus femoris had significant differences between barefoot and 4cm, and barefoot and 7cm. 2) % MVIC of tibialis anterior had significant differences between barefoot and 4cm, barefoot and 7cm, and 4cm and 7cm. 3) % MVIC of soleus had significant differences between barefoot and 7cm, 4cm and 7cm. The results indicate that all commonly responsive muscle on the conditions of barefoot, 4cm, and 7cm shoe height on the rcoking surface is tibialis anterior muscle. We found out that the more heels of shoe high, the more muscle activation increases. High-heeled shoes above 7cm remarkably increase the muscle activation of lower extremity and may result in muscle fatigue. Thus, these shoes may summate risk factors of falls in older women. We can acknowledge that the heels above 4 cm affect each muscle activation in lower extremity on the rocking surface.
