Whitening Effects of Angelica dahurica Radix Ethanol Extract (백지 에탄올추출물의 미백효능 연구)

  • To investigate whitening Effects of Angelica dahurica Radix Ethanol Extract (ADEE), we used melan-a cell line, brown guinea pig, and HMB-45. We treated with ADEE of 6.25, 12.5, 25, and 50 ${\mu}g/m{\ell}$ concentration in order to evaluate the effect of ADEE on cell viability and on morphological observation of melan-a cells. Also we were induced the artificial tanning spots by 1,500 mJ/$cm^2$ of ultraviolet B radiation on the backs of brown guinea pigs (approximately 450~500g) and then the test agent of $30{\mu}{\ell}$ was applied on the spots twice a day, five days a week, for five weeks respectively. The visible whitening effect was evaluated once a week. At the end of the experiment, the animals were sacrificed under anesthetization. The artificial tanning spots were obtained by biopsy punch and stained with HMB-45 to observe the gp100 proteins which were melanosomes. Our results show that cell viability was not reduce at ADEE concentrations between 6.25 and 50 ${\mu}g/m{\ell}$, melanin synthesis and melanocyte dendricity were decreased in ADEE treated melan-a cells increasing ADEE concentration. In the gross observation, ADEE treated groups had lower pigmentation than the vehicle control groups. And in the histological observation, ADEE treated groups had lower melanocytes than the vehicle control groups. Also in the quantitative analysis of the gp100 proteins using image analysis software, ADEE treated groups had a significantly lower value (p<0.001) than the vehicle control group and this resultsagreed with the results of observation under microscope. From these results, weconcluded that ADEE had positive whitening effect.

  • The embryos of Pinus taeda, P. rigida, and P. taeda ${\times}$ rigida were cultured for adventitious shoot regeneration in vitro. Culture media were modified from Gresshoff and Doy (MGD), Murashige and Skoog (MMS), Lloyd and McCown (MLM), and Schenk and Hildebrandt (MSH). NAA was added to initiation media at a concentration of 0.1 or 0.01 mg/l. BAP was used at the concentrations of 0.1. 0.5, 1, 2, or 5mg/l. Each explant was induced for 3-4 weeks on solid medium. All explants were cultured up to 16 weeks. Illumination was about $1506{\pm}540lux$ at the level of the tissues in the growth room with a temperature of $25{\pm}2^{\circ}C$. A 16-hour photoperiod per 24 hours was used. Half-strength medium was used for all the subcultures. For shoot production by loblolly pine, MMS, MLM, or MSH is preferred with 5 mg/l BAP with either 0.1 or 0.01 mg/l NAA. For shoot production by pitch pine, MMS, MLM, or MSH is recommended with 2 or 5 mg/l BAP with 0.1 mg/l NAA. For shoot production by the hybrid pine, MMS or MLM is more effective with 1, 2 or 5 mg/l BAP with 0.1 mg/l NAA. There were no differences recognized among the species tried in the patterns of bud formation and shoot development. Different composition of media, in major and minor salts or possibly in vitamins, should be tested for the two developmental stages of adventitious shoots ; the induction of shoot buds and the elongation of them into shoots.

Anti-oxidant and Anti-inflammatory Effects of Ethanol Extracts from Leonurus japonicus Houtt. on LPS-induced RAW 264.7 Cells (익모초 에탄올 추출물의 항산화 및 항염증 활성)

  • Leonurus japonicus (L. japonicus) Houtt., a biennial plant in the Lamiaceae family is broadly distributed in Asia such as Korea, China, Japan. The aerial part of L. japonicus is used as a traditional medicine to treat uterine disease including dysmenorrhea, amenorrhea, sterility. In this study, we examined the antioxidant and anti-inflammatory effects of L. japonicus ethanol extracts. The antioxidant activity of L. japonicus was measured by total polyphenol and flavonoid content, and DPPH, ABTS scavenging, reducing power activity, and intracellular ROS expression assay. The anti-inflammatory effects were measured by nitric oxide (NO), cytokines (TNF-α and IL-1β) production and inflammatory protein expression in LPS-induced RAW 264.7 cells. Total polyphenol and flavonoid content of L. japonicus were 51.40 ± 0.47 mg of gallic acid equivalents/g and 73.28 ± 0.10 mg of rutin equivalents/g respectively. DPPH, ABTS radical scavenging activity and reducing power activity tended to increase concentration-dependent and treatment L. japonicus with 400 ㎍/mL reduced ROS production by 69.5%. Furthermore, L. japonicus inhibited NO, TNF-α and IL-1β production in a concentration-dependant manner and reduced the expression levels of inflammatory proteins via regulating NF-κB, MAPK pathway. Therefore, we suggest that L. japonicus could be a natural antioxidants and medicinal source to treat oxidative stress and inflammation-related disease.

Effects of Rearing Density Stress on Malformation and Stress and Immune Related Gene Expression of Juvenile Olive Flounder Paralichthys olivaceus (자어기 넙치(Paralichthys olivaceus)의 사육 밀도에 따른 기형 발생과 스트레스 및 면역 유전자 발현 분석)

  • Stocking density is associated with Paralichthys olivaceus growth; thus, fish should be rapidly reared at high densities for commercial reasons. Studies have reported that high stocking density retards growth; however, few have investigated the malformations caused by stocking density stress. This study compared the growth and malformation rates of P. olivaceus at different densities and stress- and immune-related gene expression between malformed and normal fish. Forty days post-hatching, fish (total length, 1.49±0.02 cm) were reared at 800 (low density; LD), 1500 (medium density; MD), and 4000 (high density; HD) fish/m2, and the growth rate was measured weekly. On day 30, RNA was extracted from the kidneys, and the expression of stress-, immune-, and malformation-related genes was analyzed using qRT-PCR. The malformation rate in the HD groups was approximately three times higher (62%) than that in the LD and MD groups (approximately 20%), and growth was lower regarding length and weight. The stress-related (HSP70 and GPX) and immune-related (PIR and IgM) genes showed higher mRNA expression in the HD group and malformed fish than in the LD group and normal fish. However, TLR3 showed the opposite results. In summary, high stocking density suppressed growth and increased malformation risk in P. olivaceus.

Side Shear Resistance of Drilled Shafts in Weathered Rock (풍화된 암반에 근입된 현장타설말뚝의 주면지지력)

  • In this research, the effect of rock mass weathering on the side shear resistance of drilled shaft socketed into igneous-metamorphic rock was investigated. For that, 23 cast-in-place concrete piles with diameters varying from 400mm to 1,500mm were constructed at four different sites, and the static axial load tests were performed to examine the resistant behavior of the piles. A comprehensive field/laboratory testing program at the field test site was also performed to describe the in situ rock mass conditions quantitatively. The side shear resistance of rock socketed piles was found to have no intimate correlation with the compressive strength of the intact rock. However, the global rock mass strength, which was calculated by the Hoek and Brown criteria, was found to closely correlate to the side shear resistance. The ground investigation data regarding the rock mass conditions (e.g. $E_m$, $E_{ur}$, $p_{lm}$, RMR, RQD, j) were also found to be highly correlated with the side shear resistance, showing the coefficients of correlation greater than 0.75 in most cases. Additionally, the applicability of existing methods for the side shear resistance of weathered granite-gneiss was verified by comparison with the field test data. The existing methods which consider the effect of rock mass condition were modified and/or extended for weathered rock mass where mass factor j is lower than 0.15, and RQD is below 50%.

  • A feeding experiment was conducted using juvenile Korean rockfish (Sebastes schlegeli) to evaluate supplemental vitamin premix in a test diet containing fish meal as protein source for nutritional study. Four vitamin premixes were prepared by adjusting different quantity and combination of vitamins. Triplicate groups of the 50 fish averaging 4.25 g were fed one of four isoproteic ($47\%$) and isolipidic ($9\%$) experimental diets to satiation twice a day for 10 weeks. Weight gain, feed efficiency and protein retention in fish fed the diet with vitamin premix-1 were significantly higher than those in fish fed the other diets (P<0.05). Moisture, protein and lipid contents of muscle and whole body were not affected by different dietary vitamin premix (P>0,05). These results indicate that premix-1 can be used to adequate supplemental vitamin premix in test diet containing fish meal as protein source for juvenile Korean rockfish.

Effects of Fatty Acids and Vitamin E Supplementation on Antioxidant Systems in the Liver and Serum of the Second Generation Rat (지방산 및 비타민 E 보충 식이가 제2세대 흰쥐 간조직과 혈청의 항산화체계에 미치는 영향)

  • Effects of dietary fatty acids and vitamin E on antioxidant system were studied in rat liver and serum. Sources of dietary fat (10 wt%) were safflower oil (SO) poor in $\omega$3 fatty acid and mixed oil (MO) with computer-adjusted fatty acid ratios (AA/DHA=1.4, $\omega$6/$\omega$3=6.3, P/M/S=1.0/l.5/1) with (ME) and without (MO) vitamin E (500 mg/kg diet). Rats were fed the three kinds of diet from 3~4 wks prior to the conception. At the age of 3 and 9 wks of the second generation rat, antioxidant vitamins and glutathione peroxidase (GSH-Px), superoxide dismutase (SOD) activities were measured in the liver and serum. The concentrations of $\beta$-carotene were lower in ME than in MO and SO in the liver at the age of 3 wks. It seemed that vitamin E has an inhibitory action on the uptake of $\beta$-carotene or acts as a preferred antioxidant to $\beta$-carotene. The concentrations of lycopene were lower in SO than in MO in the liver at the age of 3 wks. The concentrations of cryptoxanthin showed no significant changes within groups. The activities of GSH-Px tended to increase in ME compared to MO and the ratios of SOD/GSH-Px tended to decrease in ME compared to MO in the liver at the age of 3 weeks. The activities of antioxidant enzyme at the age of 3 weeks and 9 weeks were similar. This suggested that the activity level of antioxidant enzymes reached to the adult level at the age of 3 weeks which is the end point of lactation period.

  • In order to determine the optimum operational paramsters in cow manure composting, 4 laboratory scale composters were established. The cow manure was mixed with certain amount of saw dust to adjust the initial C/N ratio to 24, initial pH to 6.9 and composting was performed with varying operational conditions. It was found that the optimum aeration rate was 1000 ml/min kg. VS, the optimum moisture content 50% and no significant difference was found with different initial pH condition. Microorganisms were counted under the optimum conditions determined in this study. At the end of the experimental period, the number of bacteria, actinomycetes and fungi was $1.5{\times}10^9$ cells, $1.1{\times}10^8$ cells and $3.0{\times}10^8$ cells/g dry compost, respectively. At day 0, the number of coliforms, fecal coliforms and fecal streptococci was $3.1{\times}10^3$ cells, $7.5{\times}10^2$ cells and $5.6{\times}103$ cells/g dry composting material, respectively. Their population was decreased with time lapse, However, their survival time was longer than those reported by other researchers. Microorganisms were identified at the end of the experiment. Genus Bacillus was the most dominant comprising 89.3% of the total population. Among the Genus Bacillus, B. circulans compoex was the most abundant, followed by B. Stearothermophilus, B. Sphericus, B. licheniformis and B, brevis.

Studies on Raw Silk Cohesion for Promotion (생사의 포합향상에 관한 연구)

  • The purpose of this studies is to improve the cohesion of raw silk through various analyses on cocoon drying, cooking, reeling, re-reeling, and on the properties of water. Also we investigated the correlation between silk testing items which we have reached to the following results. 1. Drying of cocoon When cocoons were slowly dried with 100$^{\circ}C$, the results of cohesion became much better. On the other hand, the results were considerably decreased in case the temperature with 115$^{\circ}C$. 2. Cooking of cocoon In case of the cooking of cocoon, we found that the result of cohesion was best with incomplete cooking, that of the control was next, while in over cooking, the results were very low. Also the results of cohesion were much better when using the method of over cooking with sericin arrestive agent than that of incomplete cooking with sericin agglutinating agent. 3. Reeling of cocoon A) When the temperature of reeling bath was 25-45$^{\circ}C$, the results of cohesion test were much better, but at the temperature below 25$^{\circ}C$ or above 45$^{\circ}C$, the results became worse. B) With out the process of croissieur, the results of cohesion were too bad, but in case of croissieur more than 1cm, cohesion became better rapidly. Further more, we understood that the results of cohesion were improving slightly with longer length of croissieur. C) When the velocity of reeling was increased, the results of cohesion also improved. The best results were shown when reeling velocity was 180-220 r.p.m. But when the velocity was increased more than 220 r.p.m., the results of cohesion got worse more or less. D) When the temperature of the drying pipe in reeling machine was raised, the results of cohesion also showed a tendency to improve. 4. Re-reeling A) We could net reach a conclusion as to have correlation between the number of dipping repeat in vacuum tank and the results of cohesion before re-reeling process. B) When we used Seracol 500 as an agglutination protective agent with l/1000 to l/2000 of water, the results of cohesion test were better. C) When we used Pearl-lite as an agglutination protective agent with 1/1000 to 1/2000 of water, the results of cohesion were considerably better. D) We gained tile best results when used Cohesion Improving Chemical, A-80, with 500-1500 times diluted. 1) Results of cohesion was improved when humidity was low or temperature was high in the rereeling machine. 5. Filature water A) The water pH near the isoelectric point of protein showed the best cohesion, but the farther water pH, the worser results were obtained. B) With the increasing of M-alkalinity in filature water, the results of cohesion were worse. Above all, we understood the tendency of the results of cohesion get worse when the M-alkalinity is increased above 200 ppm. C) By increasing the total hardness of the filature water, it improved the results of cohesion. Especially, when the total hardness was above 300ppm, the results were extremely high. 6. Effects combination of each results A) The result of effects combination in filature processes with the obtained best conditions was distinctively improved. But the results could not reach in mathematicaly double effect. When reelied under worse conditions, the results of cohesion test were too bad. There was "effect limit" for the promotion. B) Generally the results of cohesion were bad when the filature conditions(the temperature, pressure and the properties of water, etc) are processed as sericin loss to be high. On the other hand, the results were very good when lower sericin loss was controlled in filature conditions. C) When filature conditions such as reeling velocity and croissieur length provide pysical cohesion ability and when raw silk dry fast during reeling and re-reeling, we found the result of cohesion was better. 7. Correlation of silk testing items. A) A negative correlation exists between the results of cohesion test and cleanness defect. Another word, the result of cohesion test was found to be worse as cleanness defect increased. B) In cleanness, cohesion has negative correlation against the number of slugs, but we could not find any correlation against long loops, loose ends. C) Cohesion has negative correlation against average neatness and low neatness defect. The better the results of neatness respectively, the better the results of cohesion found. D) There is no correlation between tenacity and the results of cohesion test, but there was high positive correlation between the results of elongation and those of cohesion test. The more elongation, the better the results of cohesion was found.

Salinity Monitoring of Soups of The Institutions Enrolled at Center for Children's Foodservice Management (어린이급식관리지원센터 등록 시설 국물음식의 염도 모니터링)

  • The salinity of soups of institutions enrolled at the Center for Children's Foodservice Management was monitored to reduce sodium intake of children by continuously managing salinity of soups provided by children food service. Monitoring of salinity was conducted by a salinity meter (SB-1500H, HM digital) three times in 2016 by patrolling children's institutions for the instruction of public health. The average salinity of soups provided by total children's institution showed a significant decrease in salinity - $1^{st}$ 0.45%, $2^{nd}$ 0.41% and $3^{rd}$ 0.39% - as monitoring was proceeded (p<0.01). When salinity of soups was analyzed according to the type of children's institution, the salinity of national and public institutions showed no significant decrease. In contrast, the salinity of private institutions decreased significantly (p<0.05). Depending on the enrollment of the food service, there was no significant difference in average salinity of soups between large food services, which have over 50 children, and small food services, which have less than 50 children. However, all food services showed a salinity decrease from the $1^{st}$ monitoring to the $3^{rd}$ monitoring. Salinity according to the type of soup showed that salinity of clear soup, miso soup, and stew except beef soup decreased from the $1^{st}$ monitoring to $3^{rd}$ monitoring. However, there was no significant difference in salinity according to the type of soup which can be attributed to a large difference in salinity among the same type of soup. The individual soup showing the biggest difference between maximum and minimum salinity of soup was egg soup (0.62%), potato soup (0.54%), and mushroom soup (0.64%). The present average salinity of soup provided by children's institutions was less than 0.5% which is the recommended standard of the Ministry of Food and Drug Safety. However, the salinity of soups should be managed within a certain range including the minimum salinity regarding the taste education of children.
