Precipitation-Filtering Method for Reuse of Uranium Electrokinetic Leachate (우라늄 오염 동전기 침출액의 재이용을 위한 침전-여과 방법)

  • A large volume of uranium electrokinetic leachate has been generated during the electrokinetic decontamination to remove uranium from contaminated soil. The treatment technology for the reuse of the uranium leachate was developed. The concentration of uranium in the generated uranium leachate was 180 ppm and concentrations of Mg(II), K(I), Fe(II), and Al(III) ions ranged from 20 ppm to 1,210 ppm. The treatment process for uranium leachate consisted mainly of mixing and cohesion, precipitation, concentration, and filtration. In order to obtain the pH=11 of a precipitate solution, the calcium hydroxide needs to be 3.0g/100ml and the sodium hydroxide needed to be 2.7g/100ml. The results of several precipitation experiments showed that a mixture of NaOH+0.2g alum+0.15g magnetite was an optimal precipitant for filtration. The average particle size of precipitate with NaOH+alum+0.15g magnetite was $600\;{\mu}m$. Because the total value of metal concentrations in supernatant at pH=9 was the smallest, sodium hydroxide should be added with 0.2g alum and 0.15g magnetite for pH=9 of leachate.

Preparation of Whelk Internal Organ Jeotgal with the Addition of Commercial Proteolytic Enzymes (상업용 단백질 가수분해 효소를 첨가한 골뱅이 내장 젓갈의 제조)

  • For the utilization of the by-products of whelk processing, whelk internal organ with the addition of commercial proteolytic enzymes - Flavourzyme, Neutrase, Protease NP, Prozyme - were used to make jeotgal, Korean traditional salted and fermented seafood sauce. The products were prepared at salt concentration of 25% with enzyme contents 0.05 and 0.1%. The samples were stored at $10^{\circ}C$ and the chemical properties were evaluated for 6 months. The pH in all samples were decreased from near 6.8 in the beginning stage to 6.1-6.4 in the final stage of incubation. Amino nitrogen of jeotgal increased with enzyme concentration and showed maximum value, 646 mg%, at 0.1% of Flavourzyme. Total nitrogen content was increased till four months, but rapidly decreased after that. Protein degradations of whelk internal organ during maturation of jeotgals were investigated by SDS-PAGE. The patterns of degradation were different with added enzymes.

EFFECT OF ORCHARDGRASS GROWTH STAGE ON POOL SIZE AND KINETICS OF DIGESTA PARTICLES IN THE RUMEN OF SHEEP

  • The differences in rumen particle pool size, passage rate and rumen degradability in sheep receiving three varieties of orchardgrass hay harvested at pre-heading (H1), early-bloom (H2) and late-bloom (H3) were investigated using four ruminal-cannulated wethers (68 kg) fed 1,300 g of the hay once a day. Representative samples of whole rumen contents were collected at different times after feeding and the quantities of rumen particle pools [large particle pool (LPP), retained on a $1,180{\mu}m$ sieve; small particle pool (SPP), retained on a 47 but passed a $1,180{\mu}m$ sieve; and soluble fraction (SOL), passed a $47{\mu}m$ sieve (SOL)] were determined by a wet-sieving technique. The fullowing results were obtained: 1) The dry weight of whole rumen contents were significantly lower (p < 0.05) for HI than for H2 or H3. The reduction rate of whole rumen contents was slightly but significantly greater for HI that, the other hay varieties. 2) The LPP disappearance rates were 26.2, 25.3 and 21.7 g DM/h for H1, H2 and H3, respectively, and no statistical differences were found among the hay varieties. Appreciable changes were not observed with SPP and SOL throughout measurements for all hay varieties; however the SPP was markedly greater (p <0.05) for H2 and ill than for HI, while SOL did not differ among hay varieties. 3) The SPP passage rate (g DM/h) and effective rumen degradability (%) for HI, H2 and ill were, respectively, 9.7, 56.6; 16.9, 42.3; and 18.0, 28.9. The ruminal tum-over rate for SPP appeared to be higher for HI than for the other hay varieties.

Assessments of nutrient intake and metabolic profiles in Korean adolescents according to exercise regularity using data from the 2008 Korean National Health and Nutrition Examination Survey

  • The purpose of this study was to assess the metabolic profile and nutrient intake data in Korean adolescents according to exercise regularity using the forth Korean National Health and Nutrition Examination Survey from 2008 data. A total of 834 Korean adolescents (440 boys, 394 girls) aged 12 to 18 years were classified into exercising male (EM), non-exercising male (NM), exercising female (EF), and non-exercising female (NF) groups. EM exhibited significantly higher weight (P<0.001), waist circumference (WC) (P<0.01) and body mass index (EMI) (P<0.001) than NM. EF showed significantly greater height (P<0.01), weight (P<0.001), we (P<0.001), and BMI (P<0.001) than NF. Although ratios of macronutrient intake were within the appropriate range in all groups, energy, riboflavin, vitamin e and calcium were assessed as in adequate by the nutritional adequacy ratio (NAR) in all groups. Moreover, EF and NF had inadequate intake of vitamin A and iron according to NAR, respectively. There were significant correlations between height and NAR protein (r=0.249, P<0.001), and mean adequacy ratio (MAR) (r=0.177, P<0.01), and between weight and NAR protein (r=0.180, P<0.01), and MAR(r=0.136, P<0.05) in EM. On the other hand, there were significant correlations between weight and NAR protein (r=0.270, P<0.01), we and NAR protein (r=0.271, P<0.01), and BMI and NAR protein (r=0.326, P<0.01) in NM. There were significant correlations height and NAR Fe (r=0.153, P<0.05) in EF. However, there were no significant correlations between metabolic factors and nutritional adequacy in NF. Although we noted no significant differences in MAR between the groups, the exercising groups showed higher MAR values than the non-exercising groups. Therefore, practicing of ideal dietary behaviors appears to be induced through physical activity and regular exercise in Korean adolescents.

Systematic and Evolutionary Study on the Genus Parus(Passeriformes : Paridae) in Korea (한국산 박새속(참새목, 박새과) 조류의 계통진화)

  • Electrophoretic methods were used to examine the degree of genic variation and genetic relatedness in 11 populations representing 6 species and subspecies of the genus Parus in Korea. The average degree of genic variation of 3 subspecies of P. major was =1.2, =24.4% , D=0.042, and G=0.058, whereas the rest of the species showed slightly lower degree of genic variation than P. major. Genetic relatedness between subspecies and species in the genus Parus showed similar to those reported at comparable taxnonomic levels in other birds. But it appears to be considerably less than that of non avian taxa. Genetic relatedness between 3 subspecies of P.major and P. varius varius was closely related(=0.80), whereas between P.palustrius hellmayri and P.ater amurensis was relatively remote (=0.67). The presumed divergent times of P.palustrius hellmayri, P, ater amurensis , and P. varius varius were about 1.8, 1.6, and 1.0million years before present respectively, and 3 subspecies of P. major were recently differentiated about 100 thousands years before present.

Virtual Screening and Testing of GSK-3 Inhibitors Using Human SH-SY5Y Cells Expressing Tau Folding Reporter and Mouse Hippocampal Primary Culture under Tau Cytotoxicity

  • Glycogen synthase kinase-3β (GSK-3β) is an important serine/threonine kinase that implicates in multiple cellular processes and links with the neurodegenerative diseases including Alzheimer's disease (AD). In this study, structure-based virtual screening was performed to search database for compounds targeting GSK-3β from Enamine's screening collection. Of the top-ranked compounds, 7 primary hits underwent a luminescent kinase assay and a cell assay using human neuroblastoma SH-SY5Y cells expressing Tau repeat domain (TauRD) with pro-aggregant mutation ΔK280. In the kinase assay for these 7 compounds, residual GSK-3β activities ranged from 36.1% to 90.0% were detected at the IC50 of SB-216763. In the cell assay, only compounds VB-030 and VB-037 reduced Tau aggregation in SH-SY5Y cells expressing ΔK280 TauRD-DsRed folding reporter. In SH-SY5Y cells expressing ΔK280 TauRD, neither VB-030 nor VB-037 increased expression of GSK-3α Ser21 or GSK-3β Ser9. Among extracellular signal-regulated kinase (ERK), AKT serine/threonine kinase 1 (AKT), mitogen-activated protein kinase 14 (P38) and mitogenactivated protein kinase 8 (JNK) which modulate Tau phosphorylation, VB-037 attenuated active phosphorylation of P38 Thr180/ Tyr182, whereas VB-030 had no effect on the phosphorylation status of ERK, AKT, P38 or JNK. However, both VB-030 and VB-037 reduced endogenous Tau phosphorylation at Ser202, Thr231, Ser396 and Ser404 in neuronally differentiated SH-SY5Y expressing ΔK280 TauRD. In addition, VB-030 and VB-037 further improved neuronal survival and/or neurite length and branch in mouse hippocampal primary culture under Tau cytotoxicity. Overall, through inhibiting GSK-3β kinase activity and/or p-P38 (Thr180/Tyr182), both compounds may serve as promising candidates to reduce Tau aggregation/cytotoxicity for AD treatment.

Effect of heat stress on growth performance and physiological changes of pigs in commercial farm (고온스트레스가 일반 양돈농가의 돼지 생산성 및 생리 변화에 미치는 영향)

  • This study investigated the effect of heat stress on the performance and blood characteristics in commercial pig farms. A total of 180 growing pigs and 180 finishing pigs were assigned to two treatments consisting of thermal-neutral period(TNP) and high-temperature period(HTP) with three replications in floor pen, respectively. Feeding trials in the TNP and HTP were individually performed in autumn and summer seasons, respectively. Temperature-humidity index(THI) was calculated by temperature and humidity. Performance and physiological responses were identified per growth stages and feeding trial. Average temperature and THI were $16.8^{\circ}C$ and 61.4 at the TNP, and $25^{\circ}C$ and 74.3 at the HTP, respectively. Growing pigs in HTP exhibited lower BW, ADG and ADFI than in TNP(p<0.01). Similarly, finishing pigs showed lower growth parameters in HTP than in TNP(p<0.01). Lymphocytes and neutrophils of growing pigs were lower in HTP than in TNP(p<0.05). The serum T-PRO and NEFA in finishing pigs were higher in HTP than in TNP(p<0.05). In HTP, finishing pigs had higher cortisol levels than in TNP. Therefore, HTP can negatively influence growth performance and nutritional metabolism in pigs. Our results may provide useful information for developing feeding programs and diets to control heat stress for swine farms.

Distribution of Organochlorines and PCB Congeners In Korean Human Tissues

  • In order to investigate the residual amounts of organochlorines and polychlorinated biphenyls (PCBs) in Korean human tissues (blood, adipose tissue, liver, kidney cortex, and lung), the samples were collected from the autopsied cadavers of 40 men and 40 women (from teens to seventies of age). ${\alpha}-BHC,\;{\beta}-BHC,\;{\gamma}-BHC,\;{\delta}-BHC$, p,p'-DDT, p,p'-DDD, p,p'-DDE, endrin, dieldein, aldrin, and 7 marker PCBs (28, 52, 101, 118, 138, 153, and 180) were determined in human tissues. The levels of organochlorines and PCB congeners indicated that they have been widely distributed in Korean human body. Positive correlations in terms of age were observed for the following cases: p,p'-DDE, p,p'-DDT, ${\Sigma}-DDT$, PCB 118, PCB 138, PCB 153, and ${\Sigma}-PCB$ in the adipose tissue, and p,p'-DDE in the lung. Concentration of these compounds showed a significant age-related increase. Accumulation of these compounds in aged people revealed that these compounds were more slowly eliminated in our environment and risk assessment was necessary for further proper action. Significant differences in the levels of PCBs between genders were found for PCB 118 in the adipose tissue and PCB 138 in the liver. Positive correlation coefficients between tissues were detected with p,p'-DDE and ${\beta}-BHC$.

Degradation of Phenol in Water Using Circulation Dielectric Barrier Plasma Reactors (순환식 유전체 장벽 플라즈마 반응기를 이용한 수중 페놀 처리)

  • Objectives: The purpose of this study was evaluating the applicability of the circulation dielectric barrier plasma process (DBD) for efficiently treating non-biodegradable wastewater, such as phenol. Methods: The DBD plasma reactor system in this study consisted of a plasma reactor (discharge, ground electrode and quartz dielectric tube, external tube), high voltage source, air supply and reservoir. Effects of the operating parameters on the degradation of phenol and $UV_{254}$ absorbance such as first voltage (60-180 V), oxygen supply rate (0.5-3 l/min), liquid circulation rate (1.5-7 l/min), pH (3.02-11.06) and initial phenol concentration (12.5-100 mg/l) were investigated. Results: Experimental results showed that optimum first voltage, oxygen supply rate, and liquid circulation rate on phenol degradation were 160 V, 1 l/min, and 4.5 l/min, respectively. The removal efficiency of phenol increased with the increase in the initial pH of the phenol solution. To obtain a removal efficiency of phenol and COD of phenol of over 97% (initial phenol concentration, 50.0 mg/l), 15 min and 180 minutes was needed, respectively. Conclusions: It was considered that the absorbance of $UV_{254}$ for phenol degradation can be used as an indirect indicator of change in non-biodegradable organic compounds. Mineralization of the phenol solution may take a relatively longer time than that required for phenol degradation.

Changes in Radical Scavenging Activity and α-Glucosidase Inhibitory Activity of Dried Daraesoon (Shoot of Hardy Kiwi, Actinidia arguta) during Cooking (건조 다래순의 조리 중 라디칼 소거 활성과 알파글루코시데이스 억제 활성의 변화)

  • This study evaluated the in vitro radical scavenging and ${\alpha}$-glucosidase inhibitory activities of dried daraesoon (shoot of hardy kiwi) during cooking involving rehydration and subsequent heating at $180^{\circ}C$ with or without perilla oil. Pigments and antioxidants were quantified by HPLC and spectrophotometry. Unlike the tocopherol content, the polyphenol, flavonoid, chlorophyll, and carotenoid contents as well as the DPPH radical scavenging and ${\alpha}$-glucosidase inhibitory activities of daraesoon extract were significantly decreased by rehydration (p<0.05). Heating the rehydrated daraesoon for 10 or 20 min increased its radical scavenging activity irrespective of perilla oil addition, whereas the ${\alpha}$-glucosidase inhibitory activity increased significantly only after heating with perilla oil (p<0.05). During cooking, changes in both activities showed a similar pattern to that showed by polyphenol content changes. These results suggest that the health functionality of daraesoon can be enhanced by an appropriate cooking process that retains polyphenols.
