Optimization of the Conditions of Flavonoid Extraction From Tartary Buckwheat Sprout Using Response Surface Methodology (반응표면분석법을 이용한 타타리메밀싹에서 플라보노이드 추출 최적화)

  • Tartary buckwheat is a grain with many flavonoids, such as rutin, quercetin, kaempferol, and myricetin. This study aimed to optimize extraction conditions to maximize the rutin, quercetin, and myricetin contents of tartary buckwheat sprout extracts using response surface methodology. A BoxBehnken design containing 15 experiments was employed to evaluate the effects of extraction conditions, such as temperature (X1, 50~70℃), extraction time (X2, 5~9 hr), and ethanol concentration (X3, 60~90%). The coefficients of determination (R2) for all the dependent variables (extraction temperature, extraction time, and extraction ethanol concentration) were determined to be over 0.95, indicating significance. The p-value of the model in lack of fit was over 0.1 than means, indicating that the model was well predicted. The optimal extraction conditions for rutin, quercetin, and myricetin contents were obtained at X1 = 51.03, X2 = 6.62, and X3 = 69.16, respectively. Under these optimal conditions, the predicted rutin, quercetin, and myricetin contents were 808.467 ㎍/ml, 193.296 ㎍/ml, and 37.360 ㎍/ml, respectively. For the validation of the model, ten experiments were performed and the experimental rutin and quercetin contents were measured at 802.84±8.49 ㎍/ml, 193.76±2.80 ㎍/ml, and 34.84±0.43 ㎍/ml, respectively. The experimental rutin and quercetin contents were similar to the predicted contents, but the experimental myricetin content was lower than predicted.

  • Bio-conversion manufacturing technology has been developed to produce ginsenoside Rd which is increasingly in demand as a cosmetic material due to various possibilities related to improving skin function. In order to convert ginsenoside Rb1 which is contained in red ginseng saponin (RGS) into Rd, several commercial enzymes were tested. Viscoflow MG was found to be the most efficient. In order to optimize the conversion of RGS to ginsenoside Rd by enzymatic transition was carried out using response surface methodology (RSM) based on Box-Behnken design (BBD). The main independent variables were RGS concentration, enzyme concentration, and reaction time. Conversion of ginsenoside Rd was performed under 17 conditions selected according to BBD model and optimization conditions were analyzed. The concentration of the converted ginsenoside Rd ranged from 0.3113 g/L to 0.5277 g/L, and the highest production volume was obtained under condition of reacting 2% RGS and 1.25% enzyme for 13.5 hours. Consequently, RGS concentration, enzyme concentration which is 0.05 less than p-value and among the interactions between the independent variables, the interaction between enzyme concentration and reaction time was confirmed to be the most influential.

Dermal Papilla Cells Proliferation Constituent of Schisandra chinensis Fruits and Optimization Using Response Surface Methodology (오미자의 모유두세포 증식 활성성분과 반응표면분석을 이용한 추출조건의 최적화)

  • In the present study, we have refined gomisin N, which represents activity in the proliferation of dermal papilla cells (HFDPCs) from the fruit of Schisandra chinensis (S. chinensis), and have identified optimal extraction conditions for obtaining extracts with high content of gomisin N. The activity of the extracts and fractions was evaluated, and the results indicated approximately 29% proliferation activity in the group treated with 1 ㎍/mL of n-hexane fraction. Column chromatography was used to assess the active ingredient in the n-hexane fraction, and two compounds, namely gomisin N(1) and schisandrin(2), were isolated and identified. When the HFDPCs proliferation activity was tested for the isolated compounds, gomisin N exhibited ≥ 20% proliferation activity. Thus, via response surface methodology (RSM), the optimum extraction conditions to obtain the maximum level of gomisin N from the fruit of S. chinensis were determined, where ethanol proportion, extraction time, and extraction temperature were used as the independent variables. The results revealed coefficient of determination ≥ 0.95 and p-value ≤ 0.05, which confirmed the fit of the model. The optimum extraction conditions to achieve the maximum content of gomisin N were as follows: ethanol proportion 83.8%, extraction temperature 80 ℃, and extraction time 8.7 h. The content of gomisin N using these conditions was predicted as 378,300 ppm, and a mean value close to the predicted value (376,884 ppm) was obtained while validating the aforementioned conditions.

Optimization of an Industrial Medium and Culture Conditions for Probiotic Weissella cibaria JW15 Biomass Using the Plackett-Burman Design and Response Surface Methodology

  • The objective of this study was to optimize industrial-grade media for improving the biomass production of Weissella cibaria JW15 (JW15) using a statistical approach. Eleven variables comprising three carbon sources (glucose, fructose, and sucrose), three nitrogen sources (protease peptone, yeast extract, and soy peptone), and five mineral sources (K2HPO4, potassium citrate, ⳑ-cysteine phosphate, MgSO4, and MnSO4) were screened by using the Plackett-Burman design. Consequently, glucose, sucrose, and soy peptone were used as significant variables in response surface methodology (RSM). The composition of the optimal medium (OM) was 22.35 g/l glucose, 15.57 g/l sucrose, and 10.05 g/l soy peptone, 2.0 g/l K2HPO4, 5.0 g/l sodium acetate, 0.1 g/l MgSO4·7H2O, 0.05 g/l MnSO4·H2O, and 1.0 g/l Tween 80. The OM significantly improved the biomass production of JW15 over an established commercial medium (MRS). After fermenting OM, the dry cell weight of JW15 was 4.89 g/l, which was comparable to the predicted value (4.77 g/l), and 1.67 times higher than that of the MRS medium (3.02 g/l). Correspondingly, JW15 showed a rapid and increased production of lactic and acetic acid in the OM. To perform a scale-up validation, batch fermentation was executed in a 5-l bioreactor at 37℃ with or without a pH control at 6.0 ± 0.1. The biomass production of JW15 significantly improved (1.98 times higher) under the pH control, and the cost of OM was reduced by two-thirds compared to that in the MRS medium. In conclusion, OM may be utilized for mass producing JW15 for industrial use.

Optimization of finely ground meat ball analogue formulations using proteins and polysaccharides from plant origin (세절형 식물성 미트볼의 저작 특성 모방을 위한 식물성 단백질과 다당류 소재의 배합비 최적화)

  • This study optimized the finely ground meat ball analogue formulation by a series of experiments. Replacing isolated soy protein (ISP) to isolated mung bean protein (IMP) in total 27% protein caused an increase in hardness while adhesiveness, springiness, and chewiness were decreased (p<0.05), and the best ISP to IMP ratio was 10:17. When protein content was changed from 19% (w/w) to 31% (w/w) with the best ISP:IMP ratio, adhesiveness was increased with decreasing protein content, but all texture profiles were greatly decreased comparing to control (p<0.05). To modify texture of plant meat ball, potato starch (PS), κ-carrageenan (KC), methyl cellulose (MC), konjac (KJ), and potato protein (PP) were applied in formulation. Finally, KJ and MC were combined with and without PP in formulation. As a result, 0.5% KJ+MC+PP and 1% KJ+MC were best formulation for plant meat ball, nevertheless, highly harder texture than control must be regulated which warranted further exploration.

Formulation Optimization Study of Carvedilol and Ivabradine Fixed-dose Combination Tablet Using Full-factorial Design (완전요인배치법을 이용한 carvedilol 및 ivabradine 이층정 복합제 내 carvedilol 속방층 제형 최적화 연구)

  • This study was conducted to optimize the formulation conditions of the immediate-release layer of carvedilol in the development of a two-layer tablet formulation for carvedilol and ivabradine. Using a 24+3 full-factorial design of experiments, excipients (microcrystalline cellulose, citric acid, and crospovidone) of the carvedilol immediate-release layer (wet granulation part) and process parameters for the tablet compression process (main compression) were optimized, and seven types of each dependent variable (assay, content uniformity, hardness, friability, disintegration, and dissolution [pH 1.2 and 6.8]) were evaluated using design expert software. The analysis of variance results confirmed that the main compression has a significant effect on hardness, friability, and disintegration time and that microcrystalline cellulose has a major effect on friability and dissolution. In addition, it was confirmed that citric acid has a significant effect on friability. Crospovidone affects friability and dissolution. According to the design space from the design of the experiment results, the optimized range is microcrystalline cellulose (~18.0-32.0 mg), citric acid (~0.5-12 mg), and main compression (~615-837 kgf). Consequently, this study confirmed the availability of manufacturing the carvedilol immediate-release layer in which all risk factors evaluated in the initial risk assessment are removed.

Leg Fracture Recovery Monitoring Simulation using Dual T-type Defective Microstrip Patch Antenna (쌍 T-형 결함 마이크로스트립 패치 안테나를 활용한 다리 골절 회복 모니터링 모의실험)

  • In this paper, we present the design and optimization process of an on-body microstrip patch antenna with a paired T-type defect for monitoring fracture recovery of human legs. This antenna is designed to be light, thin and compact despite the improvement of return loss and bandwidth performance by adjusting the size of the T-type defect. The structure around the applied human leg is structured as a 5-layer dielectric plane, and the complex dielectric constant of each layer is calculated using the 4-pole Cole-Cole model parameters. In a normal case without bone fracture, the return loss of the on-body antenna is -66.71dB at 4.0196GHz, and the return loss difference ΔS11 is 37.95dB when the gallus layer have a length of 10.0mm, width of 1.0mme, and height of 2.0mm. A 3'rd degree polynomial is presented to predict the height of the gallus layer for the change in return loss, and the polynomial has a very high prediction suitability as RSS = 1.4751, R2 = 0.9988246, P-value = 0.0001841.

  • The result of respiration amount on the growth step of P. ostreatus shows as follows. On the early and middle mycelial culture step, respiration amount per minute in the 16 bottles(850cc) maintained 184.75ppm, 185.8ppm respectively. Therefore we are able to calculate how much fresh air replace in the growth chamber for optimization optimal CO2 amount. This means early respiration amount was increased than middle a little on the proceeding mycelial culture. On the early, middle, last and post harvest growth step, the respiration amount per minute in the 16 bottles (850cc) maintained 154.5ppm, 148.1ppm, 157.1ppm and 101.6ppm respectively.

  • The ocean accounts for over 70% of the Earth's surface and is a space of largely unexplored unknowns and opportunities. Korea is a peninsula surrounded by the sea on three sides, emphasizing the importance of marine research. The ocean has an extremely complex environment with immense biological diversity. In terms of microbiology, the marine environment has varying factors like extreme temperature, pressure, solar radiation, salt concentration, and pH, providing ecologically unique habitats. Due to this variety, marine organisms have very different phylogenetic classifications compared with terrestrial organisms. Although various microorganisms inhabit the ocean, studies on the diversity, isolation, and cultivation of marine microorganisms and the secondary metabolites they produce are still insufficient. Research on bioactive substances from marine microorganisms, which were rarely studied until the 1990s, has accelerated in terms of natural products from marine Actinomycetes since the 2000s. Since then, industries for bioplastic and biofuel production, carbon dioxide capture, probiotics, and pharmaceutical discovery and development of antibacterial, anticancer, antioxidant, and anti-inflammatory drugs using bacteria, archaea, and algae have significantly grown. In this review, we introduce current research findings and the latest trends in life science and biotechnology using marine microorganisms. Through this article, we hope to create consumer awareness of the importance of basic and applied research in various natural product-related discovery fields other than conventional pharmaceutical drug discovery. The article aims to suggest pathways that may boost research on the optimization and application of future marine-derived materials.

The Effect of Engineering Design Based Ocean Clean Up Lesson on STEAM Attitude and Creative Engineering Problem Solving Propensity (공학설계기반 오션클린업(Ocean Clean-up) 수업이 STEAM태도와 창의공학적 문제해결성향에 미치는 효과)

  • The purpose of this study was to investigate the effects of engineering design-based ocean cleanup classes on STEAM attitudes and creative engineering problem-solving dispositions. Furthermore, during this process, we tried to determine interesting points that students encountered in engineering design-based classes. For this study, a science class with six lessons based on engineering design was developed and reviewed by a professor who majored in engineering design, along with five engineering design experts with a master's degree or higher. The subject of the class was selected as the design and implementation of scientific and engineering measures to reduce marine pollution based on the method implemented in an actual Ocean Clean-up Project. The engineering design process utilized the engineering design model presented by NGSS (2013), and was configured to experience redesign through the optimization process. To verify effectiveness, the STEAM attitude questionnaire developed by Park et al. (2019) and the creative engineering problemsolving propensity test tool developed by Kang and Nam (2016) were used. A pre and post t-test was used for statistical analysis for the effectiveness test. In addition, the contents of interesting points experienced by the learners were transcribed after receiving descriptive responses, and were analyzed and visualized through degree centrality analysis. Results confirmed that engineering design in science classes had a positive effect on both STEAM attitude and creative engineering problem-solving disposition (p< .05). In addition, as a result of unstructured data analysis, science and engineering knowledge, engineering experience, and cooperation and collaboration appeared as factors in which learners were interested in learning, confirming that engineering experience was the main factor.
