Influence of Hormones and Selection of Stable Cell Lines of Plumbago rosea for Accumulation of Plumbagin

  • Callus and suspension cultures derived from leaf explants of Plumbago rosea were established on Murashige and Skoog's medium containing 1 mg/L IAA, 0.5 mg/L NAA and 0.3 mg/L BAP. Callus cultures were tested for their growth and accumulation of plumbagin, a naphthoquinone and was identified by $^1H$ NMR and electron ionization mass spectroscopy. While auxins (not 2,4-D) influenced growth and plumbagin accumulation, cytokinins did not influence them much. Increasing concentrations of IAA in presence of NAA and BAP increased plumbagin in suspensions only up to 1 mg/L. Growth of callus was optimum (8.3 g DCW/I) at a hormonal combination of 1.5 mg/L IAA, 0.5 mg/L NAA and 0.3 mg/L BAP, but high plumbagin accumulation (4.9 mg/g DCW) was recorded at 1.0 mg/L IAA plus 0.3 mg/L BAP. Since instability in growth and secondary metabolite accumulation was noticed, several cell lines/clumps of callus were screened for plumbagin accumulation by visual and analytical methods. Biomass and accumulation of plumbagin showed a negative correlation in several cell lines. But one cell line showed stability both in terms of biomass and plumbagin accumulation over a period of 6 months.

Reduction of Fat Accumulation in Broiler Chickens by Sauropus Androgynus (Katuk) Leaf Meal Supplementation

  • The present study was designed to evaluate the usefulness of Sauropus. androgynus leaf (SAL) meal on reducing fat accumulation in broiler chickens. Eighty unsexed broiler chickens were allocated to four treatment groups with five replicates of four chickens each. SAL meal supplementation had no effect on body, leg, back, breast, wing, liver and heart weights, carcass protein, moisture and ash contents (p>0.05). Broilers fed diets supplemented with 30 g of SAL meal had lower feed intake with better feed conversion ratio (p<0.05) than did the control chickens. SAL supplementation at all levels significantly reduced fat accumulation in abdomen region, and liver (p<0.01), and in carcass (p<0.05). Higher SAL supplementation resulted in lower fat accumulation in the carcass ($r^{2}=0.94$; p<0.01), abdomen ($r^{2}=0.99$; p<0.01) and liver ($r^{2}=0.98$; p<0.01). The current study showed that a 30 g supplementation of SAL meal to the broiler diet was effective to improve feed conversion ratio without reducing body weight. SAL meal supplementation to the diet reduce fat accumulation in broiler chickens.

Elicitors which Induce the Accumulation of p-Coumaroylamino Acids in Ephedra distachya Cultures

  • Some ammonium oxalate soluble pectic fragments prepared from cultured cell wall of Ephycla distrahya elicited the accumulation of p-coumarocylamino acids (p-CAA) in E. distachya cultures while water soluble and alkali soluble fractions had no activity. Partial purification of the pectic fragments fraction using DEAE-cellulose chromatography afforded two active fractions (PS-I and PS-II) which were composed of mainly uronic acids (98-99 w/w %). They elicited the accumulation of p-CAA in an amount of 52-60 nmol per gram fresh weight of cultures. The acidic sugar compositions of PS-I and PS-II were found to be galacturonic acid and glucuronic acid by TLC analysis. They were supposed to act as endogenous elicitors of p-CAA accumulation. In order to investigate the effect of ethylene on p-CAA accumulation, Ethrel, which is known as ethylene generator, and ACC(1-aminocyclopropane-1-carboxylic acid), a direct precusor of ethylene biosynthesis, were added to the culture. However, they did not glycopeptide elicitor [(Con A-II)], either. Consequently, no relationships between ethylene and p-CAA accumulation were recognized. Several tentative elicitors were teted for their activity. Commercial yeast glucan, $CuCl_2$, laminarin and laminariheptaose had slight activity whereas ${\alpha}$-methylmannopyranoside and commercial yeast mannan had no elicitor activity. ${\alpha}$-methylmannopyranoside which has been known as a tentative inhibitor of glucan elicitor in Glycine max did not affect on the elicitor activity of Con A-II.

High Temperature Characterization of Accumulation-mode Pi-gate pMOSFETs (고온에서 accumulation-mode Pi-gate p-MOSFET 특성)

  • The device performances of accumulation-mode Pi-gate pMOSFETs with different fin widths have been characterized at high operating temperatures. The device fin height is 10nm and fin widths are 30nm, 40nm, and 50nm. The variation of the drain current, threshold voltage, subthreshold swing, effective mobility, and leakage current have been investigated as a function of operating temperatures. The drain current at high temperature is slightly larger than at room temperature. The variation of the threshold voltage as a function of the operating temperature is smaller than that of the inversion-mode MOSFETs. The effective mobility is decreased with the increase of operating temperature. It is observed that the effective mobility is enhanced as the fin width decreases.

Effect of External Factors on Heavy Metal Accumulation in the Cell of Heavy Metal-Tolerant Microorganisms (중금속내성균의 중금속 축적에 미치는 외부요인의 영향)

  • This study was performed to develop the biological treatment technology of wastewater polluted with heavy metals. Heavy metal-tolerant microorganisms, such as Pseudomonas putida, P. aeruginosa, P. chlororaphis and P. stutzeri possessing the ability to accumulate cadmium, lead, zinc and copper, respectively, were isolated from industrial wastewater and mine wastewater polluted with various heavy metals. The effect of several external factors, such as temperature, pH and heavy metal compounds on heavy metal accumulation in the cells was investigated. The amount of heavy metal accumulation into cells according to the kind of heavy metal compound was slightly increased in the case of the heavy metal compound with -nitrate group, but generally, there is little change according to the kind of compound in the amount of heavy metal accumulation. The amount of heavy metal accumulation according to the precultured time was increased in the case of the cell precultured for 24 hours, but generally the precultured time did not affect to the amount of heavy metal accumulation. Heavy metal accumulation into cells was affected by several external factors, such as temperature and pH. The optimum temperature and optimum pH of the accumulation of heavy metal into cells were $20{\sim}37^{\circ}C$ and pH $6{\sim}8$, respectively. By increasing the concentration of each heavy metal-tolerant microorganism in the solution, the total amount of heavy metal accumulated was increased, whereas the amount of heavy metal accumulated per cell(mg, heavy metal/g, dry cells) was decreased. These results indicated that the amount of heavy metal accumulated was not proportional to the concentration of microorganisms.

Effect of Triton X-100 on Intracellular Accumulation of Cadmium in Hansenula anomala (카드뮴의 Hansenula anomala 세포내 축적에 미치는 Triton X-100의 효과)

  • As a pary of investgation on effective accumulation of cadmium in yeast cells, effect of surfactant was studied on intracellular accumulation of cadmium in extremely cadmium-tolerant yeast, Hansenula anomala B-7. Cadmium accumulation was enhanced up to approximately 40% by the addition of non-ionic surfactant, Triton X-100 and its optimal concentration was found to be 0.1-0.2%. Furthermore, optimum conditions for intracellular accumulation of cadmium were at $40^{\circ}C$ and initial pH 6.0 for 48 hours under shaking culture.

Tu-Chung Leaf Meal Supplementation Reduced an Increase in Lipid Accumulation of Chickens Stimulated by Dietary Cholesterol

  • The effect of tu-chung (Eucommia ulmoides, Oliver) leaf meal on reducing lipid accumulation in chickens fed 1% cholesterol containing diet was studied. Forty male White Leghorn chickens aged 56 days were weighed and divided into four groups of ten chickens, and fed diets with or without 1% dietary cholesterol which were supplemented with 0 and 5% tu-chung. Tu-chung supplementation to the diet without cholesterol increased acetyl-CoA carboxylase (p<0.01) but decreased 3-hydroxy-3-methylglutaryl-CoA reductase activities (p<0.01) with no effect on fatty acid synthetase activities. However, its supplementation to the diet with cholesterol had no effect on these three enzyme activities as compared with the cholesterol containing diet without tu-chung. Tu-chung supplementation to the diet without cholesterol increased hepatic triglyceride (p<0.01), whereas its supplementation to the diet with cholesterol decreased it (p<0.01). Tu-chung supplementation to the diet with cholesterol decreased plasma cholesterol ester, free cholesterol, phospholipids (p<0.05) and triglyceride (p<0.01) as compared with the cholesterol containing diet without tu-chung. Supplementation of tu-chung to the diet without cholesterol decreased plasma free cholesterol (p<0.05). It is concluded that tu-chung leaf meal reduced an increase in lipid accumulation in chickens stimulated by 1% cholesterol feeding.

Heavy Metal Interactions during Accumulation and Elimination of Cadmium and Copper in the Liver of Juvenile Flounder, Paralichthys olivaceus

  • Experiments were carried out to investigate the effects of metal interaction on the accumulation and elimination of Cd and Cu in the liver of flounder, Paralichthys olivaceus, exposed to sub-chronic Cd (0, 5, 10, 50, 100 ${\mu}g/L$)/Cu $(10 {\mu}g/L)$ mixture. Cd exposure resulted in an increased Cd accumulation in the liver of flounder for exposure periods and concentration, and Cd accumulation increased linearly with exposure time. Cu accumulation profiles were similar to those of Cd. Cd concentration in the liver significantly decreased at the 10th depuration period and elimination rate was $66.20\%,\;86.22\%$ in 50 and $100 {\mu}g/L$at the end of depuration periods, respectively. Although, Cu elimination was similar to Cd elimination phase, Cd elimination rate was higher than that of Cu. Co-relationship of Cd and Cu have a positive correlation coefficient r=0.8620 (P<0.001) and support the strong relationship between Cd and Cu accumulation. As increase with the Cd exposure concentration, there were significant (P<0.001) differences between Cd and Cu accumulation.

  • The objective of this study was to assess parameters affecting nitrite accumulation, which offers advantages in terms of less aeration energy and carbon consumption for denitrification. The influence of the alkalinity to $NH_4{^+}-N$ concentration ratio, pH, FA(free ammonia) concentration and temperature on nitrite accumulation was investigated. The experiment was performed with supernatant from dewatering process of anaerobic digested sludge using a submerged biofilm reactor. The influent contains high strength of ammonium nitrogen and the alkalinity was insufficient for complete nitrification. An increased nitrite accumulation was observed with increase in alkalinity to $NH_4{^+}-N$ concentration ratio. The increase in alkalinity to $NH_4{^+}-N$ concentration ratio has been a maior reason for the high pH value and FA concentration in the reactor. It can be considered that selective inhibition of Nitrobacter can be causes of nitrite accumulation. The nitrite accumulation increased with increment of temperature at fixed alkalinity to $NH_4{^+}-N$ concentration ratio.

P(3HB) Accumulation in Alcaligenes eutrophus H16(ATCC 17699) under Nutrient-Rich Condition and Its Induced Production from Saccharides and Their Derivatives

  • Poly(3-hydroxybutyrate)(P(3HB)) accumulation under nutrient-rich condition with various amounts of $(NH_4)_2 SO_4$ was systematically investigated. The results of the electron-microscopy and the solvent extraction showed that the P(3HB) accumulation is unavoidable even under nutrient-rich condition. This indicates that in a two-step culture of Alcaligenes eutrophus H16, the researches should be careful in interpreting the data of polyhydroxyalkanoates(PHAs) accumulation in terms of the carbon-source fed in the second step because the two-step culture product contains the P(3HB) produced under nutrient-rich condition. The polyester production capability in a two-step batch culture of A. eutrophus H16(ATCC 17699) was also investigated using various saccharides and their derivatives such as glucose, fructose, gluconic acid, glucaric acid, sorbitol, lactose, galactose, and mannose. The polyesters synthesized were characterized by 500 MHz$^{1}H-NMR$ spectroscopy, intrinsic viscosity$[\eta]$ measurement in chloroform and differential scanning calorimetry(DSC). 500 MHz $^{1}H-NMR$ analysis showed that all polyesters synthesized generally contained 1~2 mol% of 3HV. Another finding is that the glucose utilization can be increased by changing the autoclaving procedure of the substrate to enhance the P(3HB) production yield up to 46 wt% of P(3HB) in dry cells.
