Keratinase Production by Recalcitrant Feather Degrading Pseudomonas Geniculata and Its Plant Growth Promoting Activity (난분해성 우모분해 Pseudomonas geniculata에 의한 케라틴 분해효소 생산 및 식물성장 촉진 활성)

  • We investigated the optimal conditions for keratinase production by feather-degrading Pseudomonas geniculata H10 using one variable at a time (OVT) method. The optimal medium composition and cultural condition for keratinase production were determined to be glucose 0.15% (w/v), beef extract 0.08% (w/v), $KH_2PO_4$ 0.12% (w/v), $K_2HPO_4$ 0.02% (w/v), NaCl 0.07% (w/v), $MgSO_4{\cdot}7H_2O$ 0.03%, $MgCl_2{\cdot}6H_2O$ 0.04% along with initial pH 10 at 200 rpm and $25^{\circ}C$, respectively. The production yield of keratinase was 31.6 U/ml in an optimal condition, showing 4.6-fold higher than that in basal medium. The strain H10 also showed plant growth promoting activities. This strain had ammonification activity and produced indoleacetic acid (IAA), siderophore and a variety of hydrolytic enzymes such as protease, lipase and chitinase. Therefore, this study showed that P. geniculata H10 could be not only used to upgrade the nutritional value of feather wastes but also useful in situ biodegradation of feather wastes. Moreover, it is also a potential candidate for the development of biofertilizing agent applicable to crop plant soil.

THE INFLUENCE OF CURING WITH LIGHT TRANSMITTING WEDGE INSERTED INTO THE COMPOSITE RESIN BODY ON THE DEGREE OF MICROLEAKAGE AND MICROHARDNESS (광 전달 쐐기를 이용한 중합 방법이 레진 충전물의 미세 누출과 미세 경도에 미치는 효과)

  • The purpose of this study was to test the hypothesis that the more thorough cure of lightactivated composite resin could be obtained if a new way of delivering the light source deep through the resin body is developed when compared to the conventional bulk cure or incremental cure. Using cylindrical resin blocks and natural teeth, various curing conditions were tested for their effects on the degree of microleakage and microhardness. Data were analyzed statistically using One-Way ANOVA and Scheff's Multiple Range test. The results of the present study were as follows: 1. The increment of reduction in microhardness with depth was shown to be higher in group I and II than in group II and V. 2. At the level of lower inner surface and the bottom surface, significantly lower microhardness values were obtained in group I and II when compared to group III, IV and V. Group IV showed the lowest value among three groups(III, IV, V) representing the only statistically significant difference from group V(p<05). 3. Differences between groups in the degree of microleakage detected were shown to be statistically significant (p<05) with the exception of group IV and V at both occlusal and gingival margins. 4. Based upon the above-mentioned results, commonly cited recommendation of incremental curing is strongly supported and the light-transmitting wedge insertion method can be considered as a effective and feasible clinical procedure for the better curing of composite resin. However, more studies under a variety of conditions should be completed before this method is applied actually to the clinical setting.

  • Bond strength between alloy and ceramic plays an important role in deciding the quaring the processing of metalceramic restorations. Now the author had an experiment on the measurement of bond strength differences accoding to the contents of recast alloy used during the processing of metal-ceramic restorations. In the experiment, Anusavice's Planar Interface Shear Bond Test was employed to determine the bond strength. Total 25 specimens were divided as 5 groups, that is, 5 specimens in a group respections : Group I (new alloy 100%) Group II (new 75%$\cdot$recast 25%), Group III (new 50%$\cdot$recast 50%), Group IV (new 25%$\cdot$recast 75%), Group V(recast 100%). All specimens examined micropically and respective strength values of the group specimens were checked simultaneously. The results were as follows, 1. In the analtsis of variance the result showed the significant differences of 1%(P<0.01) among the each group classifid according to the recast contents. 2. When the interfaces of specimens were examined with an electron microscope, the air bubble were evenly occurred in all the specimens, and the occurrence frequencies and the sizes of air bubble were different between Group I and Group III, especially marked different Group I and Croup V. 3. In respective verification of each group through T-test, between Group I and Group II, between Group I and Group III did not show significant differences. 4. There was significant difference between Group I and Group IV, between Group I and Group V as 1%(P<0.01). 5. In the Shear Bond Test of all the groups, Croup V showed the lowest value.   • The possibility of $ZrO_2$ thin film as insulator for Metal-Ferroelectric-Insulator-Semiconductor(MFIS) structure was investgated. $SrBi_2Ta_2O_9$ and $SrBi_2Ta_2O_9$(SBT) thin films were deposited on P-type Si(111) wafer by R. F. magnetron sputtering method. The electrical properties of MFIS gate were relatively improved by inserting the $ZrO_2$ buffer layer. The window memory increased from 0.5 to 2.2V in the applied gate voltage range of 3-9V when the thickness of SBT film increased from 160 to 220nm with 20nm thick $ZrO_2$. The maximum value of window memory is 2.2V in Pt/SBT(160nm)/$ZrO_2$(20nm)/Si structure with the optimum thickness of $ZrO_2$. These memory windows are sufficient for practical application of NDRO-FRAM operating at low voltage.

Potential Feeding Value of Deoiled Rice Bran by Japanese Quails. 1. The Metabolisable Energy Content

  • The apparent metabolisable energy (AME) and N-corrected ME (AMEn) of deoiled rice bran (DORB) were determined with adult quails at 6 and 10- week of age. The DORB obtained from two types of extraction process, Batch (DORB-B) and the Continuous (DORB-C), was each included in a practical type of the reference diet at 20 or 40% level. The analysed crude protein, ether extract, total ash, calcium, phosphorus, glucose and starch content of DORB-B and DORB-C were found at 19.0, 0.79, 17.05, 0.11, 1.92, 2.3, 11.22, and 15.02, 1.56, 13.0, 0.40, 2.76, 2.16, 19.0, respectively. The level of inclusion of DORB in diet appeared to exert a significant effect on the AME and AMEn values. When bioassayed at 20% inclusion level the DORB was found to have a significantly (p<0.01) lower value than that obtained at 40% inclusion level. However, no significant effect of age of quails on the AME values of DORB was evident. The ME bioassays with quails gave significantly (p<0.01) higher AME values for DORB-C than DORB-B thereby indicating that the continuous system of solvent extraction of rice bran is superior to the batch system from this point of view. The AME value of DORB predicted from its chemical composition also revealed that the DORB-C contained approximately 15% more energy than that in DORB-B.

An Electrophysiologic Study on the Ulnar Digital Nerves (척골 지단 신경의 전기생리학적 연구)

  • The ulnar nerve extends down the arm, across the elbow, and into the hand. It provides sensation to the little and ring fingers and activates many of the small muscles in the hand. The determination of peripheral nerve conduction velocity is an important part of ulnar nerve evaluation. The electrodiagnostic value as neurophysiologic investigative procedure has been known for many years but normal value of digital nerve was not reported in Korea. The purpose of this investigation was to measure the digital nerve conduction velocity of ulnar nerve for obtain clinically useful reference value and compare difference in each fingers and then compare with the other countries. 71 normal Korean volunteers (age, 19-65 years; 142 hands) examined who has no history of peripheral neuropathy, diabetic mellitus, chronic renal failure, endocrine disorders, anti-cancer medicine, anti-tubercle medicine, alcoholism, trauma, radiculopathy. Nicolet Viking II (EMG machine) was use for detected conduction velocity and amplitude of digital nerves in ulnar nerve. Data analysis was performed using SPSS. Descriptive analysis was used for obtain mean and standard deviation and independent t-test was used to compare with ring and little finger. Conduction velocity of the right ring finger was 57.44m/sec and little finger was 55.32msec. The left ring finger was 55.55msec and little finger was 54.11msec. Amplitude of the right ring finger was $30.28{\mu}V$ and little finger was $48.36{\mu}V$. The left ring finger was $30.67{\mu}V$ and little finger was $52.76{\mu}V$. There were significantly difference between ring and little in amplitude (p<.05) but there were no statistically difference between conduction velocity of ring and little finger (p>.05). The amplitude of little finger are greater than ring finger. The present results revealed that electodiagnosis can easily perform in little finger for digital nerve of ulnar nerve study.

Feasibility of Mixed-Energy Partial Arc VMAT Plan with Avoidance Sector for Prostate Cancer (전립선암 방사선치료 시 회피 영역을 적용한 혼합 에너지 VMAT 치료 계획의 평가)

  • Purpose: The purpose of this work was to investigate the dosimetric impact of mixed energy partial arc technique on prostate cancer VMAT. Materials and Methods: This study involved prostate only patients planned with 70Gy in 30 fractions to the planning target volume (PTV). Femoral heads, Bladder and Rectum were considered as oragan at risk (OARs). For this study, mixed energy partial arcs (MEPA) were generated with gantry angle set to 180°~230°, 310°~50° for 6MV arc and 130°~50°, 310°~230° for 15MV arc. Each arc set the avoidance sector which is gantry angle 230°~310°, 50°~130° at first arc and 50°~310° at second arc. After that, two plans were summed and were analyzed the dosimetry parameter of each structure such as Maximum dose, Mean dose, D2%, Homogeneity index (HI) and Conformity Index (CI) for PTV and Maximum dose, Mean dose, V70Gy, V50Gy, V30Gy, and V20Gy for OARs and Monitor Unit (MU) with 6MV 1 ARC, 6MV, 10MV, 15MV 2 ARC plan. Results: In MEPA, the maximum dose, mean dose and D2% were lower than 6MV 1 ARC plan(p<0.0005). However, the average difference of maximum dose was 0.24%, 0.39%, 0.60% (p<0.450, 0.321, 0.139) higher than 6MV, 10MV, 15MV 2 ARC plan, respectively and D2% was 0.42%, 0.49%, 0.59% (p<0.073, 0.087, 0.033) higher than compared plans. The average difference of mean dose was 0.09% lower than 10MV 2 ARC plan, but it is 0.27%, 0.12% (p<0.184, 0.521) higher than 6MV 2 ARC, 15MV 2 ARC plan, respectively. HI was 0.064±0.006 which is the lowest value (p<0.005, 0.357, 0.273, 0.801) among the all plans. For CI, there was no significant differences which were 1.12±0.038 in MEPA, 1.12±0.036, 1.11±0.024, 1.11±0.030, 1.12±0.027 in 6MV 1 ARC, 6MV, 10MV, 15MV 2 ARC, respectively. MEPA produced significantly lower rectum dose. Especially, V70Gy, V50Gy, V30Gy, V20Gy were 3.40, 16.79, 37.86, 48.09 that were lower than other plans. For bladder dose, V30Gy, V20Gy were lower than other plans. However, the mean dose of both femoral head were 9.69±2.93, 9.88±2.5 which were 2.8Gy~3.28Gy higher than other plans. The mean MU of MEPA were 19.53% lower than 6MV 1 ARC, 5.7% lower than 10MV 2 ARC respectively. Conclusion: This study for prostate radiotherapy demonstrated that a choice of MEPA VMAT has the potential to minimize doses to OARs and improve homogeneity to PTV at the expense of a moderate increase in maximum and mean dose to the femoral heads.

Effect of rare earth dopants on the radiation shielding properties of barium tellurite glasses

  • Rare earth doped barium tellurite glasses were synthesised and explored for their radiation shielding applications. All the samples showed good thermal stability with values varying between 101 ℃ and 135 ℃ based on dopants. Structural properties showed the dominance of matrix elements compared to rare earth dopants in forming the bridging and non-bridging atoms in the network. Bandgap values varied between 3.30 and 4.05 eV which was found to be monotonic with respective rare earth dopants indicating their modification effect in the network. Various radiation shielding parameters like linear attenuation coefficient, mean free path and half value layer were calculated and each showed the effect of doping. For all samples, LAC values decreased with increase in energy and is attributed to photoelectric mechanism. Thulium doped glasses showed the highest value of 1.18 cm-1 at 0.245 MeV for 2 mol.% doping, which decreased in the order of erbium, holmium and the base barium tellurite glass, while half value layer and mean free paths showed an opposite trend with least value for 2 mol.% thulium indicating that thulium doped samples are better attenuators compared to undoped and other rare earth doped samples. Studies indicate an increased level of thulium doping in barium tellurite glasses can lead to efficient shielding materials for high energy radiation.

Study on Production and Properties of Galactooligosaccharide from Soybean Arabinogalactan (대두 Arabinogalactan으로부터 갈락토올리고당의 생산과 그 특성에 관한 연구)

  • For the purpose of enzymatic production of galactooligosaccharides from soybean arabinogalactan (SAG) hydrolysis, the $\beta$-1, 4-D-arabinogalactanase($\beta$-1, 4-galactanase) from Bacillus sp. HJ-12 was used. The soybean galactooligosaccharides(SOS) were optimally produced in SAG 1%(w/v), pH 8.0, 5$0^{\circ}C$, $\beta$-1, 4-galactanase 20unit/g SAG and 24-40 hour reaction conditions. The produced galactooligosaccharides had visocity of 11,000 cp at 75%(w/v), $25^{\circ}C$. The viscosity of galactooligosaccharides was 80 fold increasing value than that of sucrose solution. Temperature dependence of viscosity of SOS was 4.6 fold higher value than surose solution below than 5$0^{\circ}C$. Less than 50 Brix, the viscosity of SOS was similar with sucrose solution(20-40 cp), but increasing of concentration, the difference of viscosity between SOS and sucrose solution was increased. And, SOS was very stable at pH and temperature.

  • While the enhanced biological phosphorus removal(EBPR), in anaerobic/aerobic condition, was known to remove phosphorus by means of metabolism of poly-P microorganisms, the phosphorus removed could be released in the form of ortho-P in the aerobic fixed biofilm reactor. This study was initiated to investigate the cause of ortho-P release in the aerobic fixed biofilm reactor. The resutls indicated that the phosphorus release was caused by autooxidation. The synthesis and release of phosphrous were related to the ORP and the boundary value for the phase change was about 170mV. In the synthesis phase, the phosphorus removal rate per COD removed was $0.023mgP_{syn}/mgCOD_{rem}$. The phosprous contents of the microorganism were 4.3 ~ 6.0% on a dry weight basis.
