3차원 강뼈대구조의 비선형 해석 (Nonlinear Analysis of 3-D Steel Frames)

  • 본 논문에서는 3차원 강뼈대구조물의 비선형 해석 기법을 개발하였다. 본 해석은 재료적 비선형과 기하학적 비선형을 고려하였다. 재료적 비선형으로 휨에 의한 점진적인 소성화를 고려하였다. 기하학적 비선형으로 $P-{\delta}$$P-{\Delta}$ 효과를 고려하였다. 절점에서의 재료적 비선형성은 여러개의 화이버로 구성되어 있는 P-M 힌지 개념을 사용함으로써 고려하였다. 기하학적 비선형성은 안정함수 (Stability function)를 사용하여 고려하였다. 단 전단과 비틀림에 의해 발생하는 비선형형은 고려하지 않았다. 수치해석법으로는 수정변위증가법을 사용하였다. 본 연구에서 제안된 해석방법으로 예측된 하중-변위가 다른 해석기법의 결과들과 잘 일치하였다.

섬유소 분해 효소의 고생산을 위한 복합균주 개발 (Development of a Microbial Consortium with High Cellulolytic Enzyme Production)

  • 본 연구의 목적은 기존의 단일 균주보다 두 종의 서로 다른 균주를 복합시켰을 때 두 균주간의 상호 보완적인 면을 이용하여 효소생산을 극대화하기 위함이며, 다음과 같은 결론을 얻었다. 실험에서 사용된 T viride와 FB01는 최적 PH와 온도가 비슷하여 혼합배양이 가능하였으며, 이들의 복합균주를 개발하는데 성공하였다. 즉 두 균주는 회분배양 하였을 때, 서로의성장속도에 차이가 있었기 때문에 두 균주의 생장균형을 유지하기 위해 접종시기를 조절할 필요가 있었다. 접종시기를 변화시킴으로서 두 균주간의 상호작용으로 인해 단일 균주일때보다 CMCase, $\beta$-glucosidase 및 avicelase의 활성이 우수했으며 또한 계대배양을 통한 두 균주의 활성유지에 관한 실험에서는 pH 조절을 통해 $\beta$-glucosidase 활성이 최고 3.2배까지증가함을 알 수 있었다. 따라서 배양액내에서 초기 pH의 영향은 생산된 cellulase complex의 다양한 구성요소의 상대적인 농도를 결정하는 실질적인 요인으로 판명된다. pH 4.5에서의 enzyme activity는 12.5 U/mL CMCase, 2.46 U/mL If-glucosidase, 0.93 U/mL avicelase였으며 개발당시의 복합균주활성인, 2.04 U/ml CMCase, 0.78U/ml $\beta$-glucosidase, 0.2 U/mL avicelase과 비교할 때 각각 6, 3.2, 4.7배의 높은 활성을 보였다. 복합균주의 탄소원으로 볏짚을 이용했을 때 복합균주에 의한 효소생산이 단일 균주인 T. virile나 FB01보다 훨씬 높게 나타났으며, 이때의 볏짚의 최적 농도는 5%(w/v)였다. 이러한 배양조건에서 복합균주를 장기적으로 계대배양하면서 효소의 생산성 향상과 복합균주의 안정성을 관찰할 필요가 있다고 사료된다.

  • A comprehensive study was conducted using PPSMV resistant (BSMR 736) and susceptible (ICP 8863) genotypes to develop a segregating population and understand the inheritance of PPSMV resistance. The observed segregation was comparable to 13 (susceptible): 3 (resistant). Hence, the inheritance was controlled by two genes, SV1 and SV2, with inhibitory gene interaction.

  • A xylanase from the Bacillus sp. strain DSNC 101, isolated from soil, was purified to homogeneity by anion-exchange and hydrophobic interaction chromatography followed by gel filtration chromatography. The enzyme cleaved xylan, but not carboxymethyl cellulose, Avicel, soluble starch, and pNPX. The main product of oat spelts xylan hydrolysates was xylobiose. The xylanase had a molecular weight of 25 kDa determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Optimum temperature and pH for the xylanase activity were $50^{\circ}C$ and 6.0, respectively. $K_{m}\;and\;V_{max}$ of the enzyme for oat spelts xylan were 12.5 mg of xylan/ml and 869.5 unit/mg of protein, respectively. Xylanase was completely inhibited by Hg, Cu, and N-bromosuccinimide, but was stimulated by Ca, Co, and Mg.

Effects of Endocrine Disruptors (NP, DBP and BPA) on Sperm Characteristics and Development of IVF Embryos in Pig

  • This study was to examine single or combined in vitro effects of environmental endocrine disruptors on boar sperm characteristics, oxidative stress damage in sperm and development of porcine IVF embryos. Addition of various concentration of NP (10, 20, $30{\mu}M$), DBP (10, 50, $100{\mu}M$) and BPA (1, 5 or $10{\mu}g/ml$) on boar sperm characteristics such as percentages of sperm motility, viability, membrane integrity and mitochondrial activity were dose-dependently decreased within 3, 6 or 9 hr incubation period (p<0.05). The overall detrimental effects increased with incubation time increasement. NP, DBP and BPA showed the detrimental effects on sperm membrane and mitochondria of energy production organelles affecting cell viability with the dependancy of dose and incubation time. In combination effects, NP ($10{\mu}M$) + DBP ($10{\mu}M$) significantly decreased boar general sperm characteristics for 3 or 6 hr incubation period compared with control (p<0.05). When both of NP and DBP concentrations (NP; $30{\mu}M$, DBP; $100{\mu}M$) increase, the detrimental effects on sperm characteristics were larger than those of low concentration combination (p<0.05). The inhibitory effects of NP ($30{\mu}M$) + BPA ($10{\mu}g/ml$) on sperm characteristics were larger than those of NP ($10{\mu}M$) + BPA ($1{\mu}g/ml$) (p<0.05). DBP ($100{\mu}M$) + BPA ($10{\mu}g/ml$) decreased sperm characteristics compared with the low concentration combination (DBP $10{\mu}M$ + BPA $1{\mu}g/ml$, p<0.05). This result indicates the detrimental effects of both chemicals on sperm characteristics were dose dependent. Addition of NP ($30{\mu}M$) + DBP ($100{\mu}M$), NP ($30{\mu}M$) + BPA ($10{\mu}g/ml$), DBP ($10{\mu}M$) + BPA ($1{\mu}g/ml$) or DBP ($100{\mu}M$) + BPA ($10{\mu}g/ml$) significantly increased lipid peroxidation for 3 or 6 hr incubation period (p<0.05) compared with no addition control. NP (${\geq}20{\mu}M$) decreased the percentages of IVF embryo development from morulae and blastocyst stages (p<0.05) and its detrimental effects were dose-dependant. BPA 0, 1, 5 or $10{\mu}g/ml$ decreased significantly and dose-dependently the percentage of morulae plus and blastocysts (p<0.05). Combinations of DBP ($100{\mu}M$) plus NP ($30{\mu}M$) and DBP ($100{\mu}M$) plus BPA ($10{\mu}g/ml$) did not affect on morulae and blastocyst development, but NP ($30{\mu}M$) plus BPA ($10{\mu}g/ml$) has significant detrimental effect on embryo development at these stages (p<0.05). These overall results indicate that the partial detrimental effects on boar sperm characteristics and embryo development by NP, DBP, BPA or the combination of these chemicals might be due to the increasement of lipid peroxidation and free radical formation in the cell and there were no specific interaction effects on boar sperm and embryo degeneration among the combined treatments.

  • Binding parameters of some edible dyes with bovine serum albumin were determined at pH 7.4 buffer solution. The edible dyes used were fast green, brilliant blue FCF, indigo carmine and allura red AC, and the final concentrations of each dyes were $3{\sim}9{\times}10^{-6}M$, $6{\times}10^{-6}M$, $3{\times}10^{-5}M$ and $3{\times}10^{-5}M$, respectively. The final concentrations of bovine serum albumin were $1{\sim}7{\times}10^{-5}M$. The values of binding free energy between edible dye and protein were ranged from -6,024 to -6,800 cal/mole.

Design and simulation of resonance based DC current sensor

  • A novel resonance based proximity DC current sensor is proposed. The sensor consists of a piezo sensed and actuated cantilever beam with a permanent magnet mounted at its free end. When the sensor is placed in proximity to a wire carrying DC current, resonant frequency of the beam changes with change in current. This change in resonant frequency is used to determine the current through the wire. The structure is simulated in micro and meso scale using COMSOL Multi physics software and the sensor is found to be linear with good sensitivity.

  • The interaction between adherent molecules and gas molecules was modeled in the molecular scale and simulated by the molecular dynamics method in order to understand evaporation and removal processes of adherent molecules on metallic surface using high temperature gas flow. Methanol molecules were chosen as adherent molecules to investigate effects of adhesion quantity and gas molecular collisions because the industrial oil has too complex structures of fatty acid. Effects of adherent quantity, gas temperature, surface temperature and adhesion strength for the evaporation rate of adherent molecules and the molecular removal mechanism were investigated and discussed in the present study. Evaporation and removal rates of adherent molecules from metallic surface calculated by the molecular dynamics method showed the similar dependence on the surface temperature shown in the experimental results.

  • To realize magnetic sails, momentum of the solar wind should be efficiently transferred to a spacecraft via magnetic field, which is produced around a spacecraft. In this paper, two important physical processes are addressed: 1) diffusive processes caused by plasma turbulence at the magnetospheric boundary around the spacecraft; and 2) field aligned current loops that will electrically connect the magnetospheric boundary and the spacecraft. The idea of the magnetic sails will be demonstrated by an experimental simulator, in which a fast plasma beam will penetrate into a dipole magnetic field. For that purpose, the two important physical processes should be scaled down to a small laboratory experiment in a space chamber. From the scaling considerations, the interaction can be scaled down if high-speed and high-density $(10^{19}m^{-3})$ plasma jet is used with 1-T-class magnetic field.

  • A partially purified preparation of L-galactonolactone oxidase which catalyzes the last step of L-ascorbic acid biosynthesis was obtained from Saccharomyces cerevisiae ATCc 26787. The purification procedures included Triton X-100 treatment, protamine sulfate precipitation, ammonium sulfate precipitation, DEAE-Sepharose CL-6B ion exchange chromatography, Sephadex G-150 gel filtration chromatography, and Phenyl-Sepharose CL-4B hydrophobic interaction chromatography. The optimum temperature for the enzyme activity was about $34^{\circ}C$ and the optimum pH was 6.8-7.0. The substrate specificity was confined to L-aldonolactones, L-galactono-1,4-lactone and L-gulono-1,4-lactone. An apparent Km value of 0.294mM with L-galactono-1,4-lactone as a substrate was found. By comparing the substrate specificities of this enzyme with those of isofunctional enzymes of higher plants and animals, it becomes evident that the enzyme of S. cerevisiae ATCC 26787 is rather similar to the L-gulonolactone oxidase of animals than the galactonolactone dehydrogenase of higher plants.
