To evaluate the ewe reproductive performances of three Iranian local sheep breeds, namely Moghani, Chal and Zel, 27 rams and 473 ewes were used as the foundation flock in a crossbreeding program. The three breeds were crossed amongst themselves reciprocally. Ram breed and ewe breed showed a two-way interaction on apparent fertility. The Zel ewes showed higher fertility than the Chal and Moghani ewes (94.4 vs 77.3 and 84.6%). Prolificacy among main and individual mating groups were not significantly different (p>0.05). Zel ewes were 22.3 and 14.8% more reproductive (p<0.05) than Chal and Moghani ewes, respectively. Zel and Moghani ewes showed higher (p<0.05) ewe productivity than Chal ewes. Ram breed had a significant (p<0.05) effect on ewe efficiency. The Zel rams were not completely successful to mate with Chal ewes, this caused a to way-interaction between ram and ewe breeds. The Zel ewes mated with Chal and Moghani rams showed 26.1 and 28.5% more efficiency than those mated with Zel rams. This observation strongly supports the use of crossbreeding to improve the efficiency of the Zel ewes. With decreasing lamb mortality, through supplemented feeding of lambs in pre-weaning period, and increasing litter weight, through crossbreeding, it should be possible to improve the efficiency of Zel ewes even better than Chal and Moghani ewes. Small body size of Zel ewes that needs the lower maintenance requirements would be a well advantageous to using Zel ewes as the dam herd to produce commercial lambs in north area of Iran.
A rapid, sensitive and selective hydrophilic interaction liquid chromatography-tandem mass spectrometric(HILIC-MS/MS) method for the determination of tiapride in human plasma was developed. Tiapride and internal standard, metoclopramide were extracted from human plasma with dichloromethane at basic pH and analyzed on an Atlantis HILIC silica column with the mobile phase of acetonitrile-ammonium formate (190 mM, pH 3.0) (94:6, v/v). The ana-Iytes were detected using an electrospray ionization tandem mass spectrometry in the multi-ple-reaction-monitoring mode. The standard curve was linear (r=0.999) over the concentration range of 1.00-200 ng/mL. The coefficient of variation and relative error for intra- and inter-assay at three QC levels were 6.4∼8.8% and -2.0∼3.6%, respectively. The recoveries of tiapride ranged from 96.3 to 97.4%, with that of metoclopramide (internal standard) being 94.2%. The lower limit of quantification for tiapride was 1.00 ng/mL using 1 00 $\mu$L of plasma sample.
The muscarinic antagonist 1-[benzilic 4, 4'-$[^3H]$ QUINUCLIDINYL BENZILATE $([^3H]$ QNB) bound to a single class of muscarinic receptors with high affinity in rabbit ileal membranes. The $K_D\;and\;B_{ max}$ values for $([^3H]$ QNB calculated from analysis of saturation isotherms were 52.5 pM AND 154 fmol/mg, respectively. Chlopheniramine (CHP), histamine $H_1$ blocker, increased $K_D$ vlue for $([^3H]$QNB without affecting the binding site concentrations and Hill coefficient. The $K_i$ value of CHP for inhibition of $([^3H]$QNB binding in ileal membranes was 1.44\mu{M}$ and the pseudo-Hill coefficient for CHP was close to unit. In the functional assay carbachol, muscarinic agonist, increased the contractile force of ileum with $ED_{50}$ value of $0.11\mu{M}$. CHP caused the rightward shift of the dose-response curve to carbachol. The $pA_2$ value of CHP determined from Schild analysis of carbacholinduced contraction was 5.77 and the slope was unity indicating competitive antagonism with carbachol. The dissociation constant $(K_i)$ of CHP obtained in competitive experiments with $([^3H]$ QNB was similar to the $K_A$ value (1.69 \mu{M)}$ of CHP as inhibitor of carbachol induced contraction in rabbit ileum. This result suggest that the binding of $H_i$ blocker. CHP, vs $([^3H]$QNB to muscarinic receptors in ileal membranes represents an interaction with a receptor of physiological relevance.
Objective: Our aim was to investigation the roles of MHC class I chain-related gene A(MICA) and natural killer cell group 2D(NKG2D) in human renal cancer cells. Materials and Methods: The expression of membrane MICA (mMICA) on renal cells and NKG2D on NK cells were detected by flow cytometry (FCM); the content of sMICA were detected by enzyme linked immunosorbent assay (ELISA) and the distribution of mMICA on renal tumor tissues by immunohistochemistry; the interaction between MICA and NKG2D was observed by antibody closed method. Results: Our results showed that the expression of mMICA in renal cancer tissues was significantly higher than in controls, where the soluble MICA was not expressed. Cytotoxic activity of NK cells was significantly reduced after exposure to NKG2D and MICA antibodies (P<0.05), and serum containing sMICA can obviously lower the function of NKG2D (P<0.05). Conclusions: The interaction of mMICA and NKG2D play important roles in mediation of cytotoxicity of NK cells in RCC. On the other hand, sMICA may mediate tumor immune escape through down- regulated NKG2D expression.
A carbonaceous sorbent was prepared from rice husk via sulphuric acid treatment. After preparation and washing, the wet carbon with moisture content 85% was used in its wet status in this study due to its higher reactivity towards Cr(VI) than the dry carbon. The interaction of Cr(VI) and the carbon was studied and two processes were investigated in terms of kinetics and equilibrium namely Cr(VI) removal and chromium sorption. Cr(VI) removal and chromium sorption were studied at various initial pH (1.6-7), for initial Cr(VI) concentration (100 mg/l). At equilibrium, maximum Cr(VI) removal occurred at low initial pH (1.6-2) where, Cr(III) was the only available chromium species in solution. Cr(VI) removal, at such low pH, was related to the reduction to Cr(III). Maximum chromium sorption (60.5 mg/g) occurred at initial pH 2.8 and a rise in the final pH was recorded for all initial pH studied. For the kinetic experiments, approximate equilibrium was reached in 60-100 hr. Cr(VI) removal data, at initial pH 1.6-2.4, fit well pseudo first order model but did not fit pseudo second order model. At initial pH 2.6-7, Cr(VI) removal data did not fit, anymore, pseudo first order model, but fit well pseudo second order model instead. The change in the order of Cr(VI) removal process takes place in the pH range 2.4-2.6 under the experimental conditions. Other two models were tested for the kinetics of chromium sorption with the data fitting well pseudo second order model in the whole range of pH. An increase in cation exchange capacity, sorbent acidity and base neutralization capacity was recorded for the carbon sorbent after the interaction with acidified Cr(VI) indicating the oxidation processes on the carbon surface accompanying Cr(VI) reduction.
Objectives: The aim of this study was to compare the knowledge and attitudes about AIDS between an activity-oriented and a lecture-oriented AIDS prevention education in middle school boys. Methods: Each of 60 middle school boys in M city, Gyeongsangnam-do, was assigned to the experimental or control group. The activity-oriented AIDS prevention education in experimental group and the lecture-oriented education in control group were provided for a 45 minutes class per week for 4 weeks, respectively. Date were collected with questionnaires and analyzed through $x^2$-test, t-test, and repeated measures ANOVA. Results: The experimental group and control group were homogeneous in knowledge and attitudes about AIDS prevention before the study. In AIDS prevention knowledge, there was no significant interaction of times by groups (F=0.033, p=.930); but the main effect of times was significant (F=63.950, p<.001). In attitudes toward AIDS, there was a significant interaction of times by groups (F=8.892, p<.001). Conclusion: The activity-oriented AIDS prevention education was an effective teaching strategy to foster positive attitudes toward AIDS prevention in middle school boys. Therefore, systematic and sustained development of AIDS prevention education reflecting learners' characteristics is required.
We examined the effects of ocean acidification (OA) and eutrophication on the physiology of a red alga, Gracilariopsis chorda, using specimens collected at Wando Island, Korea, in July of 2015. The samples were transported to a laboratory and placed on growth media for treatments involving low or high levels of ammonium ($4{\mu}M$ or $60{\mu}M\;NH_4{^+}$) and low or high pH(7.5 or 8.2). The control treatment used filtered seawater (pH 8.2 and $4{\mu}M\;NH_4{^+}$). All experiments were conducted at $20^{\circ}C$ and under a lighting intensity of $80{\mu}mol\;photons\;m^{-2}\;s^{-1}$, with or without an injection of $CO_2$ (pH 7.5). In addition, we calculated rates of respiration under darkness, at a pH of 7.5 and $60{\mu}M\;NH_4{^+}$. Fluctuations in pH as well as the evolution of photosynthetic oxygen and $NH_4{^+}$ uptake rates were monitored for 6 h. The greatest increase in pH levels, from 7.50 to 8.65, occurred in response to $60{\mu}M\;NH_4{^+}$, whereas the largest decrease, from 7.50 to 7.42, was associated with elevated respiration rates. At a pH of 7.5, rates of oxygen evolution were higher (236% saturation) for samples treated with $60{\mu}M\;NH_4{^+}$ than for the control (121% saturation). Ammonium uptake was highest at pH 7.5 and $60{\mu}M\;NH_4{^+}$, with a rate of $0.526{\pm}0.002{\mu}mol\;g^{-1}\;FW\;h^{-1}$, followed in order by the treatments of $pH\;8.2/60{\mu}\;NH_4{^+}$, $pH\;7.5/4{\mu}M\;NH_4{^+}$, and the control ($pH\;8.2/4{\mu}M\;NH_4{^+}$). We speculated that the rates of photosynthesis and $NH_4{^+}$ uptake could be enhanced at a higher ammonium concentration and lower pH because $CO_2$ concentrations were increased through greater photosynthetic activity. Therefore, these findings suggest that the physiology of G. chorda populations can be improved by the interaction of optimized $CO_2$ concentrations and an adequate supply of essential nutrients such as ammonium.
DNase I footprinting과 gel retardation 실험 결과, RNA polymerase와 SSB 단백질은 229-nt ssDNA 단편에 stable interaction을 하며, 시발체 RNA를 합성하였다.
NHERF ($Na^+/H^+$ exchanger regulatory factor) and E3KARP (NHE3 kinase A regulatory protein) play important roles in membrane targeting, trafficking and sorting of ion channels, transmembrane receptors and signaling proteins in many tissues. Each of these proteins contains two PDZ (PSD-95/Dlg-1/ZO-1) domains, which mediate the assembly of transmembrane and cytosolic proteins into functional signal transduction complexes. The interaction between NHERF and E3KARP was investigated by surface plasmon resonance spectroscopy (BIAcore), fluorescence measurement, His-tagged pull-down experiment, and size-exclusion column (SEC) chromatography. BIAcore experiments revealed that NHERF bound to E3KARP with an apparent $K_D$ of 7 nM. Fluorescence emission spectra of the NHERF-E3KARP complex suggested that the tight interaction between these proteins was accompanied by significant conformational changes in one or both. The CD spectra of NHERF and E3KARP show that the conformational changes of these proteins were dependent on pH and temperature. These results implicate that the NHERF-E3KARP complex allows intracellular signaling complexes to form through PDZ-PDZ interactions.
Due to neutron radiation, atomic displacement has a significant effect on material in nuclear reactors. A range of secondary displacement models, including the Kinchin-Pease (K-P), Lindhard, Norgett-Robinson-Torrens (NRT), and athermal recombination-corrected displacement per atom (arc-dpa) have been suggested to calculate the number of displacement per atom (dpa). As neutron elastic interaction is the main cause of displacement damage, the focus of the current study is to calculate the atomic displacement caused by the neutron elastic interaction in order to estimate the exact amount of yield strength in a WWER-1000 reactor pressure vessel. To achieve this purpose, the reactor core is simulated by MCNPX code. In addition, a program is developed to calculate the elastic radiation damage induced by the incident neutron flux (RADIX) based on different models using Fortran programming language. Also, due to non-elastic interaction, the displacement damage is calculated by the HEATR module of the NJOY code. ASME E-693-01 standard, SPECTER, NJOY codes, and other pervious findings have been used to validate RADIX results. The results showed that the RADIX(arc-dpa)/HEATR outputs have appropriate accuracy. The relative error of the calculated dpa resulting from RADIX(arc-dpa)/HEATR is about 8% and 46% less than NJOY code, respectively in the ¼ and ¾ vessel wall.
