Antifungal and Antioxidative Activities of Yucca smallina Fern

  • The antifungal activity of crude methanolic extract and fractions from Yucca smalliana Fern. leaves, roots and flowers were investigated in vitro against a panel of plant pathogenic fungi. The minimal inhibitory concentration(MIC) was determined by an agar dilution method. Preliminary liquid culture and agar plate assays showed that the growth of Fu sarium oxysporum, Phytophthora capsici, Rhizoctonia solani and Botrytis cinerea were inhibited by Y. smalliana extracts. The extracts from flowers and leaves showed antifungal activity of 64.0% and 34.0% against F. oxysporum, 66.0% and 62.0% against P. capsici, and 27.0% and 41.0% against B. cinerea, respectively. The methanolic extract from Y. smallina leaves in distilled water was fractionated using solvents of increasing polarity: hexane, ethyl acetate and butanol. These fractions had a broad spectrum of antifungal activity, found to reside entirely in the butanol and aqueous fraction. The aqueous fraction showed inhibition rate of 60.0, 67.8, 84.6 and 58.3% against F. oxysporum, R. solani, C. gloeosporioides, and B. cinerea, respectively, and the butganol fracgtion showed 36.0, 46.0, 66.1 and 58.3%, respectively. Phenolics(e.g. flavonoids, steroids and terpenoids) were observed in the thin layer profile of the different fractions. Leave extract showed a prominent antioxidant activity totally scavenging the free radical of DPPH at a concentration of 1 mg/ml.

Analysis of Partial Discharge Characteristics in SF6 Gas Insulation (SF6 가스절연에서 부분방전의 특성분석)

  • This paper deals with the characteristics of partial discharge (PD) for the purpose of a condition based maintenance (CBM) of gas insulated switchgears (GIS) in power equipment. Four types of electrode systems such as a protrusion on enclosure (POE), a particle on spacer (POS), a free particle (FP) and a Floating were designed and fabricated. PD pulses were measured using UHF sensor with a frequency range of 300 MHz~1.4 GHz and a DAQ with a sampling rate of 250 MS/s. Discharge inception voltage (DIV), discharge extinction voltage (DEV), and phase resolved partial discharge (PRPD) were analyzed depending on electrode systems. The average DIV in the POS was 28.8 kV. It was about 1.7 times higher than that in the FP, which was the lowest value of 17.2 kV. The FP shuffled and jumped at the applied voltage of 23.5 kV. Over 95% of PD pulses in the POE were generated in the negative polarity ($181^{\circ}{\sim}360^{\circ}$) of applied voltage. The results showed the phase (${\Phi}$)-magnitude (dBm) of PD pulses by UHF sensor, a cluster was formed separately depending on electrode systems.

Induction of Quinone Reductase, an Anticarcinogenic Marker Enzyme, by Extract from Chrysanthemum zawadskii var. latilobum K.

  • Induction of NAD(P)H:(quinone-acceptor) oxidoreductase (QR) which promotes obligatory two electron reduction of quinones and prevents their participation in oxidative cycling and thereby the depletion of intracellular glutathione, has been used as a marker for chemopreventive agents. Induction of phase II enzyme is considered to be an important mechanism of cancer prevention. In our previous study, we assessed the quinone reductase QR-inducing activities of 216 kinds of medicinal herb extracts in cultured murine hepatoma cells, BPRc1 and hepalc1c7 cells. Among the 216 herbal extracts tested in that study, extracts from Chrysanthemum zawadskii showed significant induction of QR. In this study, we examined QR-inducing activity of solvent fractions of the herbal extract. The dichloromethane fraction of the herb showed the highest QR induction among the samples fractionated with four kinds of solvents with different polarity. The fraction also significantly induced the activity of glutathione S-transferase (GST), one of the major detoxifying enzymes, at $4{\mu}g/mL\;and\;2{\mu}g/mL$ in hepalc1c7 and BPRc1 cells, respectively. In conclusion, dichloromethane-soluble fraction of Chrysanthemum zawadskii which showed relatively strong induction of detoxifying enzymes merits further study to identify active components and evaluate their potential as cancer preventive agents.

Anti-glycation Activities from Various Agricultural Products (단백질 glycation 저해효과가 있는 식품소재)

  • The ethanolic extracts of 83 kinds of agricultural products, including cereals, vegetables, and Chinese herbs, were tested for their inhibitory activities on protein cross-linking using the $[^{14}C]$-N-formyl-lysine incorporation method. Most of the extracts inhibited, but some extracts accelerated, the cross-linking of protein. Of those items with relatively high activities, we selected 20 samples to test for activity against AGE fonnation using the fluorophotometric method. The ethanol extract of buckwheat that was genninated for 1 day (GB-01) was detennined to have the highest activity with both methods. The ethanol extract of GB-01 was further fractionated by organic solvents, including chloroform, ethyl acetate, butanol, and water, in order of increasing polarity. The fraction that was extracted with ethyl acetate presented the highest protein glycation inhibitory activity (95.2% inhibition at the 100 ug/mL addition level). Polyphenol content analysis by HPLC showed that the amounts of rutin and quercetin were increased with the separation procedures. Finally, there was a significant relationship between activity and polyphenol content in the partially purified samples (p<0.05).

Electrotransport of Donepezil Hydrochloride from Poly(ethylene oxide) Hydrogel (폴리에틸렌옥사이드 하이드로겔을 이용한 도네페질염산염의 이온토포레시스 피부투과)

  • The objective of this work is to study transdermal delivery of donepezil hydrochloride (DH) using iontophoresis and to evaluate various factors which affect the transdermal transport. After the flux study using 4 kinds of hydrogel, hydrogel containing 8% poly(ethylene oxide) (PEO) was chosen as the hydrogel for further studies. Under experimental condition, DH was stable. We have studied the effect of polarity, current density, drug concentration and current profile on transdermal flux and compared the results. In vitro flux study was performed at $33^{\circ}C$, using side-by-side diffusion cell and full thickness hairless mouse skin. DH is positively charged at pH 7.4, and anodal delivery was much larger than cathodal and passive delivery at all current densities studied (0.2, 0.4 and 0.6 mA/$cm^2$). Cathodal delivery showed higher flux than passive flux. Flux increased as the concentration of DH in hydrogel increased. Pulsatile application of current showed smaller flux value than the application of continuous current. Based on these results, we have evaluated the possibility of delivering enough amount of DH to reach the therapeutic level. The maximum cumulative amount of DH transported for 12 hours was 455 ${\mu}g/cm^2{\cdot}hr$ when the amount of DH in the hydrogel was 3 mg/mL and the current density was 0.4 mA/$cm^2$. If the patch size is 10 $cm^2$, then we can deliver 4.6 mg for 12 hours. Because the daily dosage of DH is 5 mg, it seems possible to deliver clinically effective amount of DH using iontophoresis. This study also provides some information about the role of electrorepulsion and electroosmosis during the transport through skin.

Enhanced Iontophoretic Delivery of Risedronate Sodium Across Hairless Mice Skin (무모생쥐 피부에 리제드로네이트 소디움의 이온토포레시스 경피전달)

  • Osteoporosis was traditionally defined by the occurrence of nontraumatic fractures, especially of the spine, in the setting of low bone mass. Bisphosphonates are an important group of therapeutic agents for the management of osteoporosis, as they inhibit bone resorption and increase bone density, thereby potentially decreasing fracture risk. Risedronate sodium is a bisphosphonate class used by oral formulation. In this study, risedronate was transdermally delivered by iontophoresis. Effects of polarity, pH, current density, and drug concentration were studied using a side-by-side diffusion cell including the hairless mice skin. In addition we studied effect of enhancers. The flux was evaluated by HPLC/UV system. The amount of transported drug under iontophoretic delivery was approximately 186 fold higher than that under passive delivery. Flux was proportional to the increase of drug concentration and current density. The flux was observed about 0.68mg/$cm^2$ when the amout of Propyleneglycol monolaurate (PGML) used 1% as enhancer. Results indicated that iontophoresis is an effective method for transdermal administration of risedronate sodium

Paleomagnetic Study of the Yeonil Group in Pohang Basin (포항분지(浦港盆地)에 분포(分布)하는 연일층군(延日層群)의 고지자기(古地磁氣) 연구(硏究))

  • Paleomagnetic investigations have been carried out on the Tertiary sedimentary formations (Yeonil Group) in Pohang basin, southeastern Korea. A total of 215 samples were collected from 26 sites. Progressive thermal demagnetization indicates that many samples have unstable magnetization and do not reveal a characteristic direction. However, some samples from the lower and upper Duho Formation show a characteristic direction $D/I=7.8^{\circ}/48.3^{\circ}$ (${\alpha}_{95}=3.7^{\circ}$, k=174.1). Stepwise thermal demagnetization data show that some samples from the Hagjeon and middle Duho Formations reveal great-circle distributions moving from the present to a reversed direction of geomagnetic field. The mean of intersection points of the great-circles is nearly antipodal to the characteristic normal direction of the lower and upper Duho Formation. We infer that the Hagjeon Formation was formed during the reversed polarity chron C5B (16.2~14.7 Ma) and the Duho Formation 14.7~11.6 Ma based on our results and previous paleontologic and age dating data. Paleomagnetic direction for the Middle Miocene of Korea, analysed from the combined results of stable endpoints and great circles, is $D/I=8.7^{\circ}/53.9^{\circ}$ (${\alpha}_{95}=4.2^{\circ}$, k=74.8), and the corresponding paleopole is Lat./Long.=$82.7^{\circ}/230.2^{\circ}$ (${\delta}p=2.8^{\circ}$, ${\delta}m=5.9^{\circ}$). On the basis of this, we interpret that the opening of the East sea (Japan sea) or the synchronous clockwise rotation of the Southwest Japan exerted no structural influence on the Yeonil Group in the Middle Miocene.

Efficient One-Pot Three-Component Synthesis of Monomethine Cyanine Dyes with Quinoline Nucleus and Their Spectral Properties

  • An improved method for the preparation of monomethine cyanine dyes with quinoline nucleus by one-pot three-component using 1-methyl-2-quinolinethione, quaternized 2- or 4-methylheterocyclic compounds and methyl p-toluenesulfonate as starting materials was described. Compared with the traditional methods, the new synthetic method reduced the reaction steps, shortened the reaction time, avoided the separation and purification of the intermediate and reduced cost. The dyes absorbed in the region 478.0-563.0 nm and had molar extinction coefficients of $1.3{\times}10^4-9.4 {\times}10^4L\;mol^{-1}\;cm^{-1}$. Their fluorescence maxima and Stokes shifts were in the range of 525.2-594.4 nm and 16.2-80.6 nm in different solvents, respectively. From the spectral properties of the dyes in different solvents, it could be found that the ${\lambda}_{max}$ of the dyes were shorter in protonic solvents, and showed hypsochromic shifts with the increase of polarity of the solvents.

Study on the Change of Physical Characteristics by Polarity and Additives of SiC DPF Binder for Diesel Engine Application (디젤엔진에 적용하기 위한 SiC DPF용 접합제의 극성 및 첨가물에 따른 물리적 특성 변화에 관한 연구)

  • Fine dust has become a significant social problem. Diesel engines are used as the main propulsion power source in ships. This study introduces a diesel particulate filter (DPF) that is used as an exhaust after-treatment system for diesel engines to reduce particulate matter known as diesel fine dust. Two materials are used for the DPF: Cordierite and silicon carbide (SiC). In this study, to improve the physical properties of the binder used in the SiC DPF, cordialite was used instead of the SiC-based materials used as the conventional binder to evaluate the thermal durability against high-temperature deformation through the change of the coefficient of thermal expansion. In addition, the physical properties of the silica sol, as a main component of the base coating solution for determining the bond between the binder and the segment, were confirmed. Based on this, the change effect of the binder physical properties was confirmed through experiments by either adding a silane coupling agent or SiC to increase the reactivity of the silica sol.

A Role of YlBud8 in the Regulation of Cell Separation in the Yeast Yarrowia lipolytica

  • The spatial landmark protein Bud8 plays a crucial role in bipolar budding in the budding yeast Saccharomyces cerevisiae. The unconventional yeast Yarrowia lipolytica can also bud in a bipolar pattern, but is evolutionarily distant from S. cerevisiae. It encodes the protein YALI0F12738p, which shares the highest amino acid sequence homology with S. cerevisiae Bud8, sharing a conserved transmembrane domain at the C-terminus. Therefore, we named it YlBud8. Deletion of YlBud8 in Y. lipolytica causes cellular separation defects, resulting in budded cells remaining linked with one another as cell chains or multiple buds from a single cell, which suggests that YlBud8 may play an important role in cell separation, which is distinct from the function of Bud8 in S. cerevisiae. We also show that the YlBud8-GFP fusion protein is located at the cell membrane and enriched in the bud cortex, which would be consistent with a role in the regulation of cell separation. The coiled-coil domain at the N-terminus of YlBud8 is important to the correct localization and function of YlBud8, as truncated proteins that do not contain the coiled-coil domain cannot rescue the defects observed in $Ylbud8{\Delta}$. This finding suggests that a new signaling pathway controlled by YlBud8 via regulation of cell separation may exist in Y. lipolytica.
