Effect of Sources and Levels of Carbohydrates on Fermentation Characteristics and Hydrogenation of Linoleic Acid by Rumen Bacteria In Vitro

  • An in vitro study was conducted to examine the effect of sources and the addition levels of carbohydrates on fermentation characteristics, bacterial growth, and hydrogenation of linoleic acid ($C_{18:2}$) by mixed ruminal bacteria. Starch and cellobiose were added to the 200 ml non-selective basal media at the levels of 0.20 and 0.35% (w/v), respectively. Linoleic acid (66.8~79.6 mg) in the absorbed form into the pieces of nylon cloth was also added to the media of 5 treatments including control which was not added with carbohydrate. Three mls of rumen fluid strained through 12 layers of cheese cloth were added to each medium, and were incubated anaerobically in the shaking incubator of $39^{\circ}C$ for 24 hours. During 24 h incubation the pH in incubation media of all treatments was maintained above 6.6 by the addition of sodium bicarbonate. The pH and ammonia concentration of incubation media were not clearly influenced by the sources and addition levels of carbohydrates while additions of carbohydrates increased (p<0.0001) VFA concentration at the 24 h incubation. Molar proportions of acetate were reduced (p<0.0004) while those of propionate were increased (p<0.0006) by the addition of carbohydrates. But the differences in concentration and molar proportions of the VFA were small between the sources or the addition levels. Bacterial growth was faster (p<0.0004) in the starch added treatments than in the cellobiose added ones and control, but no differences were found between addition levels. Increased (p<0.0487) hydrogenation was observed from the starch added treatments compared to the cellobiose added ones, but there was no difference between addition levels.

Effects of the Addition of Ecklonia cava Powder on the Selected Physicochemical and Sensory Quality of White Pan Bread

  • Physicochemical properties and consumer perception of white pan bread as influenced by the addition of Ecklonia cava powder (ECP) were investigated. Freeze-dried Ecklonia cava were ground, sieved through a laboratory sieve and a fraction with particles less than $250{\mu}m$ was used. Amount of ECP added (0~3%) to the bread was found to affect the bread quality significantly (P<0.05). pH, bread height, and volume of the control was significantly higher than others (P<0.05) and decreased significantly (P<0.05) with the addition of ECP. Moisture content showed no significant differences (P>0.05). There were distinctive color changes with the addition of the powder: $L^*$- and $a^*$-values decreased but $b^*$-value increased significantly (P<0.05). The hardness of bread was found to increase but both cohesiveness and springiness showed a reverse trend with the addition of the powder. Consumer acceptance test indicated that ECP content 1% on wheat flour could be the recommended supplementation level for the consumers without sacrificing sensory quality.

  • The kinetics of the addition of 1-benzylindole-3-(p-substituted) acetophenone derivatives was investigated by ultraviolet spectrophotometery in 30% dioxane -$H_2O\;at\;25^{\circ}C$. A rate equation which can be applied over wide pH range was obtained. The Substituent effects on 1-benzylindole-3-(p-substituted) acetophenone derivatives were studied, and addition were facilitated by electron attracting groups. On the base of the rate equation, substituent effect, and general base effect the plausible addition mechanism was proposed : Below pH 3.0, only neutral thiourea molecule was added to the carbon-carbon double bond, and in the range of pH 0.0${\sim}$14.0, netural thiourea molecule and thiourea anion competitively attacted the double bond. By contrast, above pH 10.0, the reaction was dependent upon only the addition of thiourea anion.

  • 견의 저온염색용 조제(K-1)를 사용하여 염료 농도, 염욕의 pH, 염색시간에 따른 염색속도를 구하여 이 저온염색용가 염색에 미치는 영향을 분석한 결과를 다음과 같이 요약한다. 1. pH 7에서 K-1의 농도가 증가할수록 무첨가구에 비해 흡진율은 증가 하였으며, K-1의 농도가 3% o.w.f. 이상에서는 증가의 경향이 둔화되었다. 2. pH 5에서도 무첨가보다 K-1의 첨가구에서 높은 흡진율을 보였으며, pH 7의 5$0^{\circ}C$ 첨가구가 pH 5, 5$0^{\circ}C$의 무첨가구보다 확산성은 높게 나타났다. 3. 염료 농도별로 볼 때 pH 7과 pH 5에서 모두 K-l 첨가구에서는 무첨가구보다 흡진율이 클 뿐만 아니라 염료 농도에 따른 흡진율 감소가 적게 일어났다.

  • ${\alpha}$-(n-Butyl)-N-Phenylnitrone 유도체의 첨가 반응속도상수를 25^{\circ}C$의 수용액에서 자외선 분광광도법으로 측정하여 넓은 pH범위에서 잘 맞는 반응속도식을 유도하였다. 첨가반응생성물은 ${\alpha}$-phenylthiobutylidene-aniline이었으며, 첨가반응속도상수 측정과 반응속도식의 유도, 일반기염 효과, 치환기 효과 및 최종생성물의 결과로부터 반응 메카니즘을 제안하였다. 즉, pH 3.0 이하에서는 티오페놀의 첨가로 반응이 시작되며, pH 3.0∼10.0에서는 티오페놀과 티오페녹시드 음이온이 경쟁적으로 첨가되어 진행되며, pH10.0이상에서는 티오페녹시드 음이온의 첨가에 의해서 반응이 진행된다.

  • 3,4-methylenedioxyphenylmethylene malononitrile에 대한 cysteine의 친핵성 첨가반응 속도를 측정하여 염기성 및 산성용액에서의 반응메카니즘을 제안하였고 넓은 pH범위에서 잘 맞는 반응속도식을 구했다. 이 반응속도식으로 부터, pH 9.0 이상에서는 hydroxide ion 농도에만 비례하고, pH 6.0 이하에서는 cysteine의 중성분자만이 첨가하며, pH 7~9.0에서는 cysteine 중성분자와 그의 음이온이 경쟁적으로 첨가됨을 알 수 있었고 첨가되는 cysteine 중성분자와 음이온의 비도 계산할 수 있었다.

  • The present experiment was performed in order to know the therapeutic effect of laserpuncture and aquapuncture with methionine on recovery in artificially induced hepatic damaged rats by carbon tetrachloride. The animals were divided into control, laserpuncture and aquapuncture groups. The changes of serum enzyme activities(ALT and AST), serum total protein contents, protein fractions(ALB and GLB) and A/G ratio were examined before and after application of laserpuncture and aquapuncture. In change of serum ALT activity, recovery with treatment in laser and aquapuncture groups was more rapid than that of control. The significant decreased value was shown on 3rd(P < 0.05) and 7th day(P < 0.01) after treatment in laserpuncture group and on 7th day(P < 0.05) after treatment in aquapuncture group. In addition, significance was detected on 2nd day(P < 0.05) between laserpuncture and aquapuncture groups. In change of serum AST activity, recovery with treatment in laserpuncture and aquapuncture groups was more rapid than that of control. The significant lower values on 7th day(P<0.01) in aquapuncture groups. In addition, significant low value was detected on 7th day(P < 0.05) in aquapuncture group comparing with that of laserpuncture group. The change of serum TP contents showed similar pattern in control, laser and aquapuncture groups. The significances were detected on 7th day(P < 0.05) in both experimental groups. In addition, significant high value was detected on 2nd day(P < 0.05) in laserpuncture group comparing with that of aquapuncture group. The change of serum ALB content with treatment in laser and aquapuncture groups was more rapid than that of control. The significant high value was shown on 2nd(P < 0.01), 5th(P < 0.05) and on 7th day(P < 0.01) in laserpuncture group. In addition, significant high value was detected on 3rd day(P<0.01) only in aquapuncture group comparing with laserpuncture group. The change of serum GLB content showed similar pattern among groups. The significant low values were detected on 2nd day(P < 0.05) in aquapuncture group. In addition significant high value was detected on 5th day(P < 0.01) only in laserpuncture group comparing with that of aquapuncture group. In change of A/G ratio laser and aquapuncture groups showed higher values than that of control. The significant high values were detected on 1st day(P < 0.Of) and 2nd day(P < 0.05) in laserpuncture group, and on 1st day(P < 0.05) and 2nd day(P < 0.01) in aquapuncture group. In addition, significant high value was detected on 5th day(P < 0.05) in aquapuncture group comparing with that of laserpuncture. Considering above findings collectively, it was considered that both laserpuncture and aquapuncture were effective; aquapuncture was more effective than laserpuncture for recovery of hepatic damage.

  • ${\beta},\;{\beta}$-Dichlorostyrene 유도체(p-H, p-Cl, $p-CH_3,\;및\;p-OCH_3$)에 대한 thioglycolic acid 및 cysteine의 친핵성 첨가반응속도를 자외선 분광분석법으로 측정하여 pH에 따르는 반응속도상수의 변화, 일반염기(general base) 및 치환기 효과등으로 부터 넓은 pH범위에서 실험치와 잘 일치하는 반응속도식을 구하였고, 실험사실에 잘 맞는 반응 메카니즘을 제안하였다. 즉 pH 9.0 이상에서의 반응속도는 sulfide anion이 pH $9.0{\sim}7.0$에서는 thioglycolic acid 및 cysteine의 중성분자와 그의 anion들이 경쟁적으로 첨가되며 pH 7.0 이하에서는 thioglycolic acid 및 cysteine의 중성분자만이 첨가됨을 알았다.

  • 본 연구는 외기에 노출된 경골목조벽체의 수분축적을 감소시킬 수 있는 벽체 구성을 찾기 위해 수행되었다. 이를 위해 다음 다섯 가지 형태의 벽체 구성을 대상으로 벽체 내 수분이동과 수분축적을 평가하였다; 일반적인 벽체구성(기본벽체 T), 실내 측 방습지 추가(P1), 실내 측과 단열재 구간의 방습지 추가(P2), 환기를 위한 사이딩 뒷면의 공기층 추가(P3), P3벽체에서 실외 측 방습지의 실내 측으로 이동(P4). 옥외 실험가옥에 기본벽체와 제안벽체를 설치하여 외기에 노출된 벽체의 수분이동과 수분축적을 평가하였다. 실험결과 계절에 따라 추가 방습지와 공기층의 개선 정도가 다르게 나타났으나 연간 벽체 내 수분축적 감소에 유리한 벽체 구성은 1) 공기층 추가 및 실내 측으로 방습지 이동(P4), 2) 실내 측 방습지 추가(P1), 3) 기본벽체(T), 4) 공기층 추가(P3), 5) 실내 측 및 단열재 구간 방습지 추가(P2) 순으로 나타났다.

Effects of Addition Level and Chemical Type of Propionate Precursors in Dicarboxylic Acid Pathway on Fermentation Characteristics and Methane Production by Rumen Microbes In vitro

  • Two in vitro experiments were conducted to examine the effects of propionate precursors in the dicarboxylic acid pathway on ruminal fermentatation characteristics, $CH_4$ production and degradation of feed by rumen microbes. Fumarate or malate as sodium salts (Exp. 1) or acid type (Exp. 2) were added to the culture solution (150 ml, 50% strained rumen fluid and 50% artificial saliva) to achieve final concentrations of 0, 8, 16 and 24 mM, and incubated anaerobically for 0, 1, 3, 6, 9 and 12 h at $39^{\circ}C$. For both experiments, two grams of feed consisting of 70% concentrate and 30% ground alfalfa (DM basis) were prepared in a nylon bag, and were placed in a bottle containing the culture solution. Addition of fumarate or malate in both sodium salt and acid form increased (p<0.0001) pH of culture solution at 3, 6, 9 and 12 h incubations. The pH (p<0.0001) and total volatile fatty acids (VFA, p<0.05) were enhanced by these precursors as sodium salt at 3, 6 and 9 h incubations, and pH (p<0.001) and total VFA (p<0.01) from fumarate or malate in acid form were enhanced at a late stage of fermentation (9 h and 12 h) as the addition level increased. pH was higher (p<0.001) for fumarate than for malate as sodium salt at 3 h and 6 h incubations. Propionate ($C_3$) proportion was increased (p<0.0001) but those of $C_2$ (p<0.05) and $C_4$ (p<0.01 - p<0.001) were reduced by the addition of sodium salt precursors from 3 h to 12 incubation times while both precursors in acid form enhanced (p<0.011 - p<0.0001) proportion of $C_3$ from 6h but reduced (p<0.018 - p<0.0005) $C_4$ proportion at incubation times of 1, 3, 9 and 12 h. Proportion of $C_3$ was increased (p<0.05 - p<0.0001) at all incubation times by both precursors as sodium salt while that of $C_3$ was increased (p<0.001) from 6h but $C_4$ proportion was decreased by both precursors in acid form as the addition level increased. Proportion of $C_3$ was higher (p<0.01 - p<0.001) for fumarate than malate as sodium salt from 6 h incubation but was higher for malate than fumarate in acid form at 9 h (p<0.05) and 12 h (p<0.01) incubation times. Increased levels (16 and 24 mM) of fumarate or malate as sodium salt (p<0.017) and both precursors in acid form (p<0.028) increased the total gas production, but no differences were found between precursors in both chemical types. Propionate precursors in both chemical types clearly reduced (p<0.0001 - p<0.0002) $CH_4$ production, and the reduction (p<0.001 - p<0.0001) was dose dependent as the addition level of precursors increased. The $CH_4$ generated was smaller (p<0.01 - p<0.0001) for fumarate than for malate in both chemical types. Addition of fumarate or malate as sodium type reduced (p<0.004) dry matter degradation while both precursors in both chemical types slightly increased neutral detergent fiber degradability of feed in the nylon bag.
