Genome-wide Survey of Copy Number Variants Associated with Blood Pressure and Body Mass Index in a Korean Population

  • Hypertension is the major factor of most death and high blood pressure (BP) can lead to stroke, myocardial infarction and cardiac failure. Moreover, hypertension is strongly correlated with body mass index (BMI). Although the exact causes of hypertension are still unclear, some of genetic loci were discovered from genome-wide association study (GWAS). Therefore, it is essential to study genetic variation for finding more genetic factor affecting hypertension. The purpose of our study is to conduct a CNV association study for hypertension-related traits, BP and BMI, in Korean individuals. We identified 2,206 CNV regions from 3,274 community-based Korean participants using the Affymetrix Genome-Wide Human SNP Array 6.0 platform and performed a logistic regression analysis of CNVs with two hypertension-related traits, BP and BMI. Moreover, the 4,692 participants in an independent cohort were selected for respective replication analyses. GWAS of CNV identified two loci encompassing previously known hypertension-related genes: LPA (lipoprotein) on 6q26, and JAK2 (Janus kinase 2) on 9p24, with suggestive p-values (0.0334 for LPA and 0.0305 for JAK2 ). These two positive findings, however, were not evaluated in the replication stage. Our result confirmed the conclusion of CNV study from the WTCCC suggesting weak association with common diseases. This is the first study of CNV association study with BP and BMI in Korean population and it provides a state of CNV association study with common human diseases using SNP array.

Evaluation of Temporomandibular Disorders with Tension-Type Headache by Gender (성별에 따른 측두하악장애 환자의 긴장성 두통 양상)

  • This study was designed to evaluate the Temporomandibular Disorders(TMD) with Tension-Type Headache(TTH) by gender. Patients with TMD and/or TTH visited the Department of Oral Medicine, Kyung Hee University Dental Hospital were recruited to this study. Experimental group(n=60) is composed of TMD with TTH and control group(n=111) is composed of TMD without TTH. Evaluation list was pain quality, pain intensity, pain laterality, pain increase by routine physical activity and then it was analyzed statistically. The results were as follows ; 1. In the control group, pain quality was significantly different by gender(p=0.04). But, in the experimental group, pain quality was not significantly different by gender. 2. In the control group, pain intensity was not significantly different by gender. And, in the experimental group, pain intensity was not significantly different by gender. 3. In the control group, pain laterality was not significantly different by gender. And, in the experimental group, pain laterality was not significantly different by gender. 4. In the control group, pain increase by routine physical activity was not significantly different by gender. And, in the experimental group, pain increase by routine physical activity was not significantly different by gender. Therefore, it is considered that not temporomandibular disorder patients with tension-type headache but temporomandibular disorder patients without tension-type headache was influenced by gender in the pain quality.

Evaluation of the Effect of Mine Drainage on the Aquatic Environment by Quantitative Real-time PCR (실시간 정량 중합효소연쇄반응을 이용한 광산 배수의 수계 영향 평가)

  • Metals and sulfate can be considerably dissolved at low pH condition in the acid mine drainage(AMD) and it would make an environmental problems. There are only few of acid mine drainage treatment systems in Korea which are operating, but these still have an effect on the surrounding stream. In this study, quantification of indicator microorganisms was conducted to judge the environmental impact of AMD on microflora by quantitative real-time PCR in the drainage samples of four mines and the water samples of each surrounding stream. Two species of iron reducing bacteria(Rhodoferax ferrireducens T118 and Acidiphilium cryptum JF-5) were selected for indicator bacteria based on 16S rRNA cloning analysis, and sulfate reducing bacteria(Desulfosporosinus orientus), iron and sulfur oxidizing bacteria(Acidothiobacillus ferrooxidans) and iron oxidizing bacteria(Leptosprillum ferrooxidans) were included into indicator since these were found in the previous studies on the mining area. Thereafter, the comparative analysis of four mines were established by the microbiological variation index and it was determined that the biological environment effect of AMD is highest in Samtan mine which doesn t contain treatment system by the value.

C/N/O/S stable isotopic and chemometric analyses for determining the geographical origin of Panax ginseng cultivated in Korea

  • Background: The geographical origin of Panax ginseng Meyer, a valuable medicinal plant, is important to both ginseng producers and consumers in the context of economic profit and human health benefits. We, therefore, aimed to discriminate between the cultivation regions of ginseng using the stable isotope ratios of C, N, O, and S, which are abundant bioelements in living organisms. Methods: Six Korean ginseng cultivars (3-yr-old roots) were collected from five different regions in Korea. The C, N, O, and S stable isotope ratios in ginseng roots were measured by isotope ratio mass spectrometry, and then these isotope ratio profiles were statistically analyzed using chemometrics. Results: The various isotope ratios found in P. ginseng roots were significantly influenced by region, cultivar, and the interactions between these two factors ($p{\leq}0.001$). The variation in ${\delta}^{15}N$ and ${\delta}^{13}C$ in ginseng roots was significant for discriminating between different ginseng cultivation regions, and ${\delta}^{18}O$ and ${\delta}^{34}S$ were also affected by both altitude and proximity to coastal areas. Chemometric model results tested in this study provided discrimination between the majority of different cultivation regions. Based on the external validation, this chemometric model also showed good model performance ($R^2=0.853$ and $Q^2=0.738$). Conclusion: Our case study elucidates the variation of C, N, O, and S stable isotope ratios in ginseng root depending on cultivation region. Hence, the analysis of stable isotope ratios is a suitable tool for discrimination between the regional origins of ginseng samples from Korea, with potential application to other countries.

  • When routine behavior control is impossible, midazolam is often used for sedation, because it has wide margin of safety, relatively few side-effects and multiple route of administration. Although intramuscular administration of all administration route is frequently used, it is a major source of anxiety, discomfort, and trauma in children. To the contrary, oral administration of midazolam is easily administered and accepted by children. But, it's therapeutic drug concentration has not been established. The purpose of this study was to compare sedation effect and physiologic parameter of oral midazolam which palliate demerits of intramuscular administration in sedating young pediatric patients with intramuscular midazolam Twelve negative children, mean age 62.5 months, who needed at least two separate restorative visits, requiring local anesthesia participated in this study On every visit, one of the following 2 different sedative regimen was given : (1) 0.30mg/kg midazolam by intramuscular administration (2) 0.75mg/kg by oral administration. Physiologic parameter(oxygen saturation, heart rate) was recorded by ten procedure and behavior was videotaped and rated using Ohio State University Behavior Rating Scale and Automated Counting System by one investigator, blind to administration route The analyzed sedative effect of oral midazolam resulted in good sedative effect, comparing to intramuscular route, And there is no statistically difference between oral and intramuscular administration of midazolam (p>0.05).

Growth Responses of 4 Species to NaCl Concentration in Artificial Soil (NaCl 농도별 토양 처리에 대한 4수종의 생장 반응)

  • In order to inspect growth responses of Fraxinus rhynchophylla Hance, Koelreuteria paniculata Laxmann, Quercus acutissima Carruther and Ulmus parvifolia Jacquin to NaCl treatment, NaCl solution was treated for four months with 0, 25, 50, 100 and 200 mM concentrations, then survival rate, change of relative growth rate, weight, dry weight and pigment content of leaf were investigated. According to NaCl treatment, pH and EC (electrical conductivity) of soil increased, and growth rates of four tree species fell apparently as treatment time became longer. U. parvifolia had the highest survival rate with 15% in the 200mM treatment, and the other three species withered in the treatment. Relative growth rate, weight and dry weight decreased when NaCl treatment time grew longer. The total chlorophyll declined after it rose to 60 days, and the total chlorophyll and carotenoid of the all species according to NaCl treatment did not change very much. With the result from anlayzing growth responses of four tree species to oxidative stress which occurs during NaCl treatment, U. parvifolia has the highest tolerance, followed by K. paniculata, F. rhynchophylla and Q. acutissima.

Structural and Piezoelectric Properties of MnO2-Doped PZT-PSN Ceramics for Ultrasonic Vibrator (초음파 진동자용 MnO2가 Doping된 PZT-PSN 세라믹스의 구조 및 압전 특성)

  • For use in ultrasonic actuators, we investigated the structural and piezoelectric properties of $(1\;-\;x)Pb(Zr_{0.515}Ti_{0.485})O_3$ - $xPb(Sb_{1/2}Nb_{1/2})O_3$ + 0.5 wt% $MnO_2$ [(1 - x)PZT - xPSN + $MnO_2$] ceramics with a variation of x (x = 0.02, 0.04, 0.06, 0.08). All the ceramics, which were sintered at $1250^{\circ}C$ for 2 h, showed a typical perovskite structure, implying that they were well synthesized. A homogeneous micro structure was also developed for the specimens, and their average grain size was slightly decreased to $1.3{\mu}m$ by increasing x to 0.8. Moreover, a second phase with a pyrochlore structure appeared when x was above 0.06, which resulted in the deterioration of their piezoelectric properties. However, the 0.96PZT-0.04PSN+$MnO_2$ ceramics, which corresponds with a morphotropic phase boundary (MPB) composition in the (1 - x)PZT - xPSN + $MnO_2$ system, exhibited good piezoelectric properties: a piezoelectric constant ($d_{33}$) of 325 pC/N, an electromechanical coupling factor ($k_p$) of 70.8%, and a mechanical quality factor ($Q_m$) of 1779. The specimens with a relatively high curie temperature ($T_c$) of $305^{\circ}C$ also showed a significantly high dielectric constant (${\varepsilon}_r$) value of 1109. Therefore, the 0.96PZT - 0.04PSN + $MnO_2$ ceramics are suitable for use in ultrasonic vibrators.

Error Performance Analysis of DS/CDMA-Trellis Coded QPSK Signal with MRC Diversity Reception in Wireless Data Communication (무선 데이터 통신에서 MRC 다이버시티 수신시의 DS/CDMA-Trellis Coded QPSK 신호의 오율 해석)

  • In this paper, we have analyzed the packet error probability of DS/CDMA-Trellis Coded QPSK modulation signal with MRC(Maximum Ratio Combining) diversity reception in Rician fading, multi-user interferences and multipath channel. And then we have evaluated the performance and capacity of DS/CDMA-Trellis Coded QPSK system using the MRC diversity reception as a function of direct power to indirect power ratio ($K_R$), the number of diversity branch(M), the number of multi-user(U), PN chip rate(PN), the number of multipath channel($L_P$), and packet length(PL). From the results, we know that the coding gain of DS/CDMA-Trellis Coded QPSK system with 2 branch MRC diversity is about 6 dB against uncoded DS/CDMA BPSK system with 2 branch MRC diversity in Rician fading ($K_R=6dB), 5 multi-user interferences, and 3 multipath channel. And, we know that coded QPSK signal designed for the AWGN channel also perform well on a Rician fading channel with MRC diversity reception. Consequently, we expected that proposed system structure is reliable to the wireless data communication system in Rician fading, multi-user interferences, and multipath channel.

Detection of Novel Mutations in the FABP3 Promoter Region and Association Analysis with Intramuscular Fat Content in Pigs (돼지 FABP3 Promoter 부위 내 신규 돌연변이 탐색과 근내지방도와의 연관성 분석)

  • Intramuscular fat content(lMF) is considered as one of major economic traits in the pig breeding and industry. In general, high IMF results in better meat quality. Several approaches to detect quantitative trait 10ci( QTL) for IMF indicated a strong possibility of the existence of a QTL related to IMF between the microsatellite marker SW71 and SW1881 on SSC6q. Porcine FABP3 has been considered as a candidate gene affecting IMF due to its physiological roles and position on the pig genome. Two novel mutations, g.-114T> C and g.-158T>G were detected by duplicate sequencing of the porcine FABP3 promoter region. These two mutations were identified as absolute linkage disequilibrium. The g.-158T> G mutation was used for investigating relationships with growth and fat deposition traits. The GG genotype of the g.-158T> G polymorphism showed highly negative effects(P< 0.01) on body weights at 3 and 12 weeks of age, and a positive effect(P< 0.05) on IMF. However, backfat thickness(BF) and carcass fat(CF) content were not significantly associated with the genotype. The result indicates that the novel mutations, identified in this study, could be utilized as possible genetic markers to improve IMF, independent with BF.

Stage specific transcriptome analysis of liver tissue from a crossbred Korean Native Pig (KNP × Yorkshire)

  • Korean Native Pig (KNP) has a uniform black coat color, excellent meat quality, white colored fat, solid fat structure and good marbling. However, its growth performance is low, while the western origin Yorkshire pig has high growth performance. To take advantage of the unique performance of the two pig breeds, we raised crossbreeds (KNP ${\times}$ Yorkshire to make use of the heterotic effect. We then analyzed the liver transcriptome as it plays an important role in fat metabolism. We sampled at two stages: 10 weeks and at 26 weeks. The stages were chosen to correspond to the change in feeding system. A total of 16 pigs (8 from each stage) were sampled and RNA sequencing was performed. The reads were mapped to the reference genome and differential expression analysis was performed with edgeR package. A total of 324 genes were found to be significantly differentially expressed (${\left|log2FC\right|}$ > 1 & q < 0.01), out of which 180 genes were up-regulated and 144 genes were down-regulated. Principal Component Analysis (PCA) showed that the samples clustered according to stages. Functional annotation of significant DEGs (differentially expressed genes) showed that GO terms such as DNA replication, cell division, protein phosphorylation, regulation of signal transduction by p53 class mediator, ribosome, focal adhesion, DNA helicase activity, protein kinase activity etc. were enriched. KEGG pathway analysis showed that the DEGs functioned in cell cycle, Ras signaling pathway, p53 signaling pathway, MAPK signaling pathway etc. Twenty-nine transcripts were also part of the DEGs, these were predominantly Cys2His2-like fold group (C2H2) family of zinc fingers. A protein-protein interaction (PPI) network analysis showed that there were three highly interconnected clusters, suggesting an enrichment of genes with similar biological function. This study presents the first report of liver tissue specific gene regulation in a cross-bred Korean pig.
