Surface Characteristics of Type II Anodized Ti-6Al-4V Alloy for Biomedical Applications

  • Titanium and its alloys offer attractive properties in a variety of applications. These are widely used for the field of biomedical implants because of its good biocompatibility and high corrosion resistance. Titanium anodizing is often used in the metal finishing of products, especially those can be used in the medical devices with dense oxide surface. Based on SAE/AMS (Society of Automotive Engineers/Aerospace Material Specification) 2488D, it has the specification for industrial titanium anodizing that have three different types of titanium anodization as following: Type I is used as a coating for elevated temperature forming; Type II is used as an anti-galling coating without additional lubrication or as a pre-treatment for improving adherence of film lubricants; Type III is used as a treatment to produce a spectrum of surface colours on titanium. In this study, we have focused on Type II anodization for the medical (dental and orthopedic) application, the anodized surface was modified with gray color under alkaline electrolyte. The surface characteristics were analyzed with Focused Ion Beam (FIB), Scanning Electron Microscopy (SEM), surface roughness, Vickers hardness, three point bending test, biocompatibility, and corrosion (potentiodynamic) test. The Ti-6Al-4V alloy was used for specimen, the anodizing procedure was conducted in alkaline solution (NaOH based, pH>13). Applied voltage was range between 20 V to 40 V until the ampere to be zero. As results, the surface characteristics of anodic oxide layer were analyzed with SEM, the dissecting layer was fabricated with FIB method prior to analyze surface. The surface roughness was measured by arithmetic mean deviation of the roughness profile (Ra). The Vickers hardness was obtained with Vickers hardness tester, indentation was repeated for 5 times on each sample, and the three point bending property was verified by yield load values. In order to determine the corrosion resistance for the corrosion rate, the potentiodynamic test was performed for each specimen. The biological safety assessment was analyzed by cytotoxic and pyrogen test. Through FIB feature of anodic surfaces, the thickness of oxide layer was 1.1 um. The surface roughness, Vickers hardness, bending yield, and corrosion resistance of the anodized specimen were shown higher value than those of non-treated specimen. Also we could verify that there was no significant issues from cytotoxicity and pyrogen test.

Production and Separation of Angiotension Converting Enzyme Inhibitor during Natto Fermentation (납두 발효과정 중 Angiotensin Converting Enzyme 저해물질의 생성 및 분리)

  • As functionality investigation of a soybean fermentation food, a angiotensin converting enzyme inhibitory peptide was separated during natto fermentation by Bacillus natto and inhibitory effect was investigated. After incubation at each 2$0^{\circ}C$, 3$0^{\circ}C$, 4$0^{\circ}C$, 5$0^{\circ}C$, 6$0^{\circ}C$ for the 0~72 hr, protein content, protease activity and angiotensin converting enzyme inhibition were determined. The protein content and protease activity were increased and reached maximum at 60 hr fermentation with 4$0^{\circ}C$ and decreased after the 60 hr fermentation during natto fermentation. The optimum condition for angiotensin converting enzyme inhibitors was appeared at fermentation for 60 hr at 4$0^{\circ}C$. Crude extract of natto was partially purified by Amicon membrane YM-3 and Sephadex G-10, G-25 gel filtration, stepwise. The inhibitory rate was increased in a concentration dependent manner, espcially the most potent activity about 74.74% at 1.0 mg peptide content. The most prominent amino acid of the peptide from natto was alanine, followed by phenylalnine, histidine.

Lumbar Herniated Disc Resorption of 78 Patients after Korean Medicine Treatment (요추 추간판 탈출증 환자 78명의 한방치료 후 디스크 재흡수 증례 보고)

  • Objectives To analyze the resorption of herniated lumbar intervertebral disc on MRI in patients who were treated with Korean Medicine. Methods 78 patients with lumbar disc herniation were included, 47 males and 32 females. Patients' diagnosis was based on magnetic resonance imaging (MRI). All of the patients were only treated with Korean Medicine for 6 months treatment, and underwent MRI examination twice, pre-treatment and post-treatment. MR images were assessed by reduction in diameter of T2-weighted image between pre-treatment and post-treatment, and analyzed by differences in sex, age and disc type. Results 91.2% (71 people) of total patients had extrusion type. Korean medicine treatment reduced lumbar disc herniation diameter of all the patients from 7.11 mm to 3.67 mm on average (p<0.001). The reduction of diameter in the male group was 3.52 mm on average. The reduction of diameter in the female group was 3.32 mm on average. The reduction of diameter was 3.28 mm in the group below age 29, 3.88 mm in the 30~39 age group, 3.39 mm in the 40~49 age group, and 2.71 mm in the above 50 age group on average. Conclusions The MRI results suggest that lumbar herniated disc can resorb with Korean Medicine treatment. The likelihood of lumbar disc resorption was higher at extrusion type. but there was not a significant difference between the sexes and between ages.

Voltammetric Determination of Cu(II) Ion at a Chemically Modified Carbon-Paste Electrode Containing 1-(2-pyridylazo)-2-naphthol (1-(2-Pyridylazo)-2-naphthol 수식전극을 사용한 Cu(II) 이온의 전압전류법적 정량)

  • Cu(II) ion-responsive chemically modifed electrodes (CMEs) were constructed by incorporating 1-(2-pyridylazo)-2-naphthol (PAN) into a conventional carbon-paste mixture of graphite powder and Nujol oil. Cu(II) ion was chemically deposited on the surface of the PAN-chemically modified electrode in the absence of an applied potential by immersion of the electrode in a buffer solution (pH 3.2) containing Cu(II) ion, and then reduced at a constant potential in 0.1 M KNO$_3$. And a well-defined voltammetric peak could be obtained by scanning the potential to the positive direction. The electrode surface could be regenerated with exposure to acid solution and reused for the determination of Cu(II) ion. In 5 deposition / measurement / regeneration cycles, the response could be reproduced with 6.1${\%}$ relative standard deviation. In case of using the differential pulse voltammetry, the calibration curve for Cu(II) was linear over the range of 2.0 ${times}$ 10$^{-7}$ ∼ 1.0 ${times}$ 10$^{-6}$ M. And the detection limit was 6.0 ${times}$ 10$^{-8}$ M. Studies of the effect of diverse ions showed that Co, Ni, Zn, Pb, Mg and Ag ions added 10 times more than Cu(II) ion did not influence on the determination of Cu(II) ion, except EDTA and oxalate ions.

Estimation and Comparison of Stem Volume for Larix kaempferi in South Korea using the Stem Volume Model (수간재적모델에 따른 일본잎갈나무의 수간재적 추정 및 비교)

  • This study aimed to develop an equation for estimating stem volume for Larix kaempferiin South Korea using independent variables, diameter at breast height (DBH), and height as being closely associated with stem volume. Analysis was conducted on the growth performance of 2,840 Larix kaempferi samples across South Korea after felling them and gleaning diameter data according to both stem height and stem analyses. In order to test the fitness of six different stem taper equations, empirical assessment was conducted for fitness index (FI), bias, mean, and absolute deviation (MAD), and coefficient variation (%CV). The two selectedmodels found to be optimal were the following: model one (V=a+bDBH2), established by employing DBH only; and model four (V=a+bDBH2H), established by utilizing DBH and height, respectively. The findings of non-linear regression indicated statistical significance (p < 0.05) in a and b, which were the coefficients for the intercepts and slopes of the models. The FI of the models ranged between 94% and 99%, and the bias was close to zero, while MAD ranged from 0.01 to 0.05, and %CV from 5.97 to 14.43, indicating a high level of fitness. Thus, using the suggested models, the basic information necessary for forest management was obtained, and an estimation of the stem volume was effected without delay soon after effecting DBH and height measurements.

Effects of Acanthopanacis Cortex Roots 50% Ethyl Alcohol Extracts on the Cerebral Hemodynamics and Cytokine Production in Cerebral Ischemic Rats (오갈피나무 뿌리 50% 에탄올 추출물이 항허혈에 미치는 실험적 효과)

  • This experimental Study was designed to investigate the mechanism of Acanthopanacis Cortex Roots(ACR) 50% ethyl alcohol extract on the improvement of regional cerebral blood flow and cytokines production in cerebral ischemic rats. And was designed to investigate whether ACR inhibits lactate dehydrogenase(LDH) activity in neuronal cells The results were as follows; ACR significantly inhibited LDH activity in neuronal cells. These results suggest that ACR prevents the neuronal death. rCBF was significantly and stably increased by ACR(10 mg/kg, i.p.) during the period of cerebral reperfusion, which contrasted with the findings of rapid and marked increase in control group. In cytokine production of serum by drawing from femoral arterial blood at 1 hr after middle cerebral arterial occlusion, experimental group was significantly decreased $IL-1{\beta}$ and $TNF-{\alpha}$ production, and significantly increased IL-10 production compared with control group. In cytokine production of serum by drawing from femoral arterial blood at 1 hr after reperfusion, experimental group was significantly decreased $IL-1{\beta}$ and $TNF-{\alpha}$ production, and significantly increased IL-10 production compared with control group. According to above results, the author suggest that ACR had an anti-ischemic effect through the improvement of cerebral hemodynamics, and inhibitive effect on the brain damage by inhibited $IL-1{\beta}$ and $TNF-{\alpha}$ production, and accelerated IL-10 production.

Clinical Antagonistic Effect of Atipamezole in Cats Anesthetized with Tiletamine-Zolazepam and Medetomidine (고양이에서 tiletamine-zolazepam과 medetomidine 마취에 대한 atipamezole의 임상적 길항 효과)

  • The purpose of the study is to evaluate the clinical antagonistic effect of atipamezole(0.25 mg/kg, IM) in cats anesthetized with tiletamine-zolazepam ($Zoletil^{(R)}$, 10 mg/kg, IM) and medetomidine (0.05 mg/kg, IM). Twelve healthy 1 year old Korean mixed breed cats were used for this study. They were 4 males and 8 females. These cats were randomly assigned to two groups. One was control group ($Zoletil^{(R)}$ + medetomidine, ZM), and the other was treatment group ($Zoletil^{(R)}$ + medetomidine and antagonism by atipamezole, ZMA). All cats were examined 15 minutes before, 5, 25, 65 and 105 minutes after administration of tiletamine-zolazepam and medetomidine. Atipamezole was injected intramuscularly 20 minutes after ZM administation. Recovery time, heart rate, respiratory rate, total plasma protein and blood glucose were significantly different between ZM group and ZMA group (P<0.05). However, rectal temperature was not significantly different between ZM group and ZMA group. Two groups were able to induce sternal recumbency within 2 minutes and lateral recumbency within 4 minutes after the anesthetics injection. Mean sternal position time ($mean{\pm}SD$) was $174.0{\pm}44.6\;and\;116.2{\pm}27.3$ minutes, and mean standing position time was $210.8{\pm}45.6\;and\;154.2{\pm}21.1$ minutes in ZM and ZMA group, respectively. In these two groups, adverse effects during recovery time from anesthesia were not seen. As a result, the ZMA group had a faster recovery than the ZM group. Thus it was concluded that atipamezole could exert a useful reversal effect in cats anesthetized with medetomidine-tiletamine/zolazepam combination.

Effect of Soil Organic Matter on Arsenic Adsorption in the Hematite-Water Interface: Chemical Speciation Modeling and Adsorption Mechanism (비소의 적철석 표면 흡착에 토양유기물이 미치는 영향: 화학종 모델링과 흡착 기작)

  • This study was performed to investigate the effect of humic acid on the adsorption of arsenic onto hematite and its binding mechanism through the chemical speciation modeling in the binary system and the adsorption modeling in the ternary system. The complexation modeling of arsenic and humic acid was suitable for the binding model with the basis of the electrostatic repulsion and the effect of bridging metal. In comparison with the experimental adsorption data in the ternary system, the competitive adsorption model from the binary intrinsic equilibrium constants was consistent with the amount of arsenic adsorption. However, the additive rule showed the deviation of model in the opposite way of cationic heavy metals, because the reduced organic complexation of arsenic and the enhanced oxyanionic competition diminished the adsorption of arsenic. In terms of the reaction mechanism, the organic complex of arsenic, neutral As(III) and oxyanionic As(V) species were transported and adsorbed competitively to the hematite surface forming the inner-sphere complex in the presence of humic acid.

Reliability and Validity of Korean version of GRIT (한국판 GRIT 척도 : 신뢰도, 타당도 및 요인구조 연구)

  • Objective : GRIT is a non-cognitive trait which is defined as perseverance and passion for long-term goals. It predicts success, performance and thedifference from other traits. The purpose of this study was to examine the reliability and validity of the Korean version of the GRIT scale. Methods : A total of 92 patients were enrolled in the study. All patients received psychiatric assessment including Clinical Useful Depression Outcome Scale (CUDOS), Clinical Useful Anxiety Outcome Scale (CUXOS), Patient Health Questionnaire (PHQ-15), Connor-Davidson Resilience Scale (CDRS), Brief Resilience Scale (BRS), and GRIT as well as demographic assessment. Cronbach's alpha coefficient of total GRIT score and the split-half reliability of each item was calculated to assess test reliability. Exploratory and confirmatory factor analyses were performed to select the best fitting model and assess construct validity. Finally, a correlation analysis was performed to check convergent and discriminant validity. Results : Cronbach's alpha coefficient for GRIT was found to be 0.85 and all Cronbach's alpha were more than 0.8 even in cases where all items were deleted. We found 3 appropriate factor models in exploratory factor analysis, compared them with 3 models and chose the 2-factor model as the most suitable based on the best fit test. Finally, correlation of the GRIT with CUDOS, CUXOS, PHQ-15, CDRS and BRS were statistically significant (all p<0.01), with relatively low correlation coefficient. Conclusion : This study indicates that the Korean version of GRIT is a reliable and valid instrument for investigating individual power of passion and perseverance.

Antimicrobial Activity of Aqueous Ethanol Extracts of Perilla frutescens var. acuta Leaf (차조기의 에탄올과 물 혼합 추출물의 항균활성)

  • This study was conducted to examine the antimicrobial activity of Perilla frutescens var. acuta leaf fractions extracted with a mixture of ethanol and water. The Ca and Mg contents of the leaf were 595.75 mg% and 467.0 mg%, respectively, and they were the highest among all of the test minerals. The extract yield increased w e content of water in e extraction solvent. Antimicrobial activity against Staphylococcus aureus, Bacillus subtilis and Pseudomonas aeruginosa was found in the 50,70 and 95% ethanol extracts. Of the various fractions extracted from the mixture of ethanol and water, the ethyl acetate fraction showed antibacterial activity against all microorganisms tested in this experiment, and the antibacterial activity of ethyl acetate fraction from the water extract was the strongest. The phenol and flavonoid content in the ethyl acetate fraction showed no correlation with the concentration of ethanol in the extract solvent; however, their contents were higher in the 30% ethanol and water extraction which the antimicrobial activity of the extract was the strongest.
