The Annual Reproductive Cycle, Proximate Composition, Fatty Acid and Amino Acid Content of Pacific Oyster, Crassostrea gigas (Magallana gigas), in Gadeok-do, Korea

  • Environmental factors impact oyster growth, condition, and gonadal development, which is linked to gamete characteristics observed through histology. The reproductive cycle of bivalves is related to energy storage and utilization. Therefore, in this study, the year-round growth change and gonadal development of oysters were observed using histological analysis, and the biochemical composition changes were confirmed. The oysters used in this study are being nurtured in Gadeok-do, and 40 oysters were randomly sampled monthly from March 2021 to February 2022. Result of histological analysis of gonads, oysters were showed early development from December to February, late development from March and April, mature and ripe from May to July, spawned from August to October, and spent from November to December. Condition index values of oysters decreased in summer and autumn and increased again when entered the spent after spawning. The protein content of oysters was high in May, the maturity period, and the lipid content decreased during the spawning period. In addition, EPA and DHA, the major fatty acids of oysters, were low during the spawning period and high during the maturation period. As a result, this study suggested a close relationship between changes in oyster growth, biochemical composition, and the reproductive cycle.

열처리에 의한 굴(Crassostrea gigas)의 영양성분 변화 (Effect of Heat Treatment on Biochemical Characteristics of Pacific Oyster Crassostrea gigas)

  • The aim of this study was to investigate changes in the biochemical characteristics of the Pacific oyster Crassostrea gigas in response to boiling water bath ($85^{\circ}C$, $100^{\circ}C$) and autoclaving ($100^{\circ}C$) heat treatments for 5, 10, 15, and 20 min. The weight loss (7.6-9.5%) of oysters exposed to a $100^{\circ}C$ boiling water bath for 15 min was lower compared to those receiving no treatment. Oysters exposed to the $100^{\circ}C$ boiling water bath for 20 min lost 27.8% of their total weight. The $100^{\circ}C$ autoclaving treatment resulted in weight loss of 13.0-26.1%, with the highest weight loss occurring after 10 min of treatment (26.1%). Heat-treated oysters had significantly higher levels of crude lipid and crude protein relative to untreated oysters (P<0.05). In the heat-treated oysters, total amino acids (including proline and glutamic acid) were increased, but they were decreased by the 20-min $100^{\circ}C$ boiling water bath and autoclaving treatments. The fatty acid composition of oysters was not significantly affected by the water bath heat treatment, but oysters treated with $100^{\circ}C$ autoclaving for 20 min had decreased polyunsaturated fatty acids. Oysters exposed to the water bath heat treatment had significantly increased vitamin A content, but $100^{\circ}C$ autoclaving for 20 min was associated with reduced vitamin A content, as compared to all of the other heat treatments. Therefore, oysters were affected by heat treatment, undergoing metamorphosis and weight loss, but nutrient quality did not change significantly compared to controls, except with the 20-min $100^{\circ}C$ boiling water bath and autoclaving treatments.

Development of a Virus Elution and Concentration Procedure for Detecting Norovirus in Oysters

  • Low levels of virus contamination and naturally occurring reverse transcription-polymerase chain reaction (RT-PCR) inhibitors restrain virus detection in oysters. A rapid and efficient oyster-processing procedure that can be used for sensitive virus detection in oysters was developed. Poliovirus type 1 Sabin strain was used to evaluate the efficacy of virus recovery. The procedure included (a) acid-adsorption and elution with buffers (0.25M glycine-0.14 M NaCl, pH 7.5; 0.25M threonine-0.14M NaCl, pH 7.5); (b) polyethylene glycol (PEG) precipitation; (c) resuspension in Tween 80/Tris solution and chloroform extraction; (d) the second PEG precipitation; (e) viral RNA extraction with TRIzol and isopropanol precipitation; and (f) RT-PCR combined with semi-nested PCR. The overall recovery of elution/concentration was 19.5% with poliovirus. The whole procedure usually takes 19 hr. The overall detection sensitivity was 4 RT-PCR units of genogroup I norovirus (NoV) and 6.4 RT-PCR units of genogroup II Nov/25 g of oysters initially seeded. The virus-detecting method developed in this study should facilitate the detection of low levels of NoV in oysters.

Distribution of Polycyclic Aromatic Hydrocarbons in Farmed Oysters (Crassostrea gigas) around Tongyeong, Korea

  • To evaluate the culture conditions in oyster-farming waters, chemical and biological measurements were made in seawater and oysters from six bays around Tongyeong in November and December 2003. Nutrient levels in the seawater were higher in the western area than in the eastern area, in contrast to particulate organic matter and dissolved oxygen levels. The mean total polycyclic aromatic hydrocarbon ({\sum}PAH) content of the oysters was 194.5-375.9 ng/g dry weight, with four-ring compounds constituting 34.1%-79.6% of PAH. Despite wide temporal variations, a "western > eastern" spatial distribution of PAH was apparent. These low concentrations of PAHs indicate that Tongyeong waters are pristine in terms of PAH contamination. Among the hemocytic biomarkers, only lysosomal activity was significantly reduced in Hansan-Goje Bay, but did not correlate closely with PAH content. This finding indicates that the impact of PAH on cultured oysters is negligible around Tongyeong waters.

Fatty Acid Compositions of Cultured Oyster (Crassostrea gigas) from Korean and Japanese Spats

  • Changes in lipid content, lipid class and fatty acid compositions of the cultured oysters in shallow-water, Bukman bay (Tongyeong, Korea), using both Korean and Japanese spats were investigated. The content of non-polar lipid (NL) comprised approximately $60- 80\%$ of total lipid (TL) in the cultured oysters. There was a positive correlation between NL content and meat weight, y=0.287lx-15.309 (r=0.834l, p<0.001). The prominent fatty acids of the oysters were 16:0, 20:5n-3 (eicosapentaenoic acid, EPA), 22:6n-3 (docosahexaenoic acid, DHA), 18:0, l8:1n-7, l8:1n-9, l6:1n-7, 14:0 and l6:4n-3. During the growth of the oysters, l6:4n-3 showed the highest coefficient of variation, accounting for $41.8\%$ for the Korean oyster and $32.3\%$ for the Japanese one, respectively. Both oysters showed low level of n-3 fatty acids such as DHA and EPA and high level of n-6 fatty acid, 20:4n-6, in the spawning period (August). During growth of the oysters, both EPA and DHA were the richest fatty acids in the harvest period (December, 314 mg/100g sample) and in the pre-spawning period (July, 237-247 mg/100g sample), respectively. Consequently, the cultured oyster with Japanese spat contained approximately two times more n-3 fatty acids per oyster individual than those with Korean one in the harvest season.

Endogenous Rhythm in Oxygen Consumption by the Pacific Oyster Crassostrea gigas (Thunberg)

  • Pacific oysters Crassostrea gigas (Thunberg) were collected on April, 1999 and March­September, 2000 from Goseung Bay along the southern coast of Korea. The oysters tested cp;;ected from a depth of 0.5-2 m in which they cultured by a long line hanging method. The oxygen consumption rates (OCR) of oysters held under constant temperature and darkness (CC), were determined using an automatic intermittent-flow-respirometer (AIFR). Depending on holding periods after oyster collection, the experiments were divided into two groups: Group 7-d (held to ambient temperature for ca. 7 days) and Group 2l-d (held to ambient temperature for ca. 21 days). The OCR for Group 7-d single oyster displayed two peaks every day under CC, while Group 2l-d single oyster showed one peak every day. It is likely that the rhythmic patterns 02.6-12.8 hours) of the OCR in the Group 7-d single oyster may have been influenced by tidal currents at the sampling site. The rhythmic patterns (24.3-24.7 hours) in the Group 2l-d single oyster may have been shifted from two peaks to one peak each day under CC. The present study concludes that the OCR rhythm of wild oysters in nature is governed by two lunar-day clocks (24.8 hours); one driving one peak and the other driving the second peak. When oysters are subjected to the long-term CC conditions, one of the two-clock systems is depressed or only intermittently becomes active. Jpwever. the OCR rhythms by two to three oysters occurred arrhythmic patterns during the experiments and exhibited some evidence of weak rhythmicity of compared to those of a single oyster. It could be partly due to differences group effects.

전국 여러 지역산 굴의 참굴큰입흡충 피낭유충 감염 상황 (A nationwide survey of naturally produced oysters for infection with Gymnophalloides seoi metacercariae)

  • 참굴큰입흡충(Gymnophalloides seoi)의 국내 분포상황을 알기 위하여 동해안 남해안 및 서해안 등 여러 해안 지역에서 참굴을 채집한 후 참굴큰입흡충 피낭유충 감염상황을 조사하였다. 전라남도 신안군의 13개 면과 신안군 외 14개 군(경기도, 전라북도, 전라남도, 경상남도, 경상북도, 강원도)의 씨개 면을 대상지역으로 하였고. 굴의 피낭유충 감염 여부 및 감염량을 조사하였다. 신안군 13개 면 중 압해면 및 안좌면을 포함한 10개 면의 굴에서 피낭유충 감염이 확인되었고 지역별 감염률은 1.7-100%이었다. 그 중 압해면산 굴이 개체당 평균 785.9개의 피낭유충을 보유하고 있어 감염량이 가장 높았다 신안군 이외의 24개 면 중에서는 전라북도 부안군 변산면만 제외하고 모두 음성의 결과를 나타내었으며, 부안군 변산면에서는 굴 50개 중 1개에서 피낭유충 15마리가 검출되었다. 이번 연구 결과로 보아 참굴큰입흡충의 농후한 유행은 전라남도 신안군에 국한된 것으로 보이며, 전라북도 부안군 등 인근 지역에 약간의 유행이 있는 것으로 판단되었다.

원유 유출 지역에서 자연암반과 양식 기질에 서식하는 참굴(Crassostrea gigas)의 초기 가입 특성 및 성장 비교 (Comparison of Recruitment and Growth Patterns of Pacific Oysters (Crassostrea gigas) between a Natural Rocky Shore and Farming Substrate Within an Oil Spill Contaminated Area of Korea)

  • Macrobenthic biodiversity in the rocky intertidal areas of the Tae-an region, Republic of Korea, has decreased since the Hebei Spirit oil spill in December 2007. We aimed to investigate ecological roles of Pacific oyster (Crassostrea gigas) because recruitment and growth of oysters are critical to the recovery of damaged rocky shore ecosystem. We surveyed two sites monthly: natural rocky substrate and farming substrate, from July 2012 to January 2013 to identify and compare the changes in macrobenthic fauna. The abundance of young oysters was higher at the natural site. On the other hand, the mean height of oyster on the farming substrate was more than twice as great. The abundance of oyster at the natural site increased until October and then continuously decreased until end of study period. However, the abundance of oyster at the farming site constantly decreased from the beginning of study period. These different growth patterns might be attributable to spatial competition between oyster and a barnacle species (Balanus albicostatus) and environmental factors. At the natural site, physical stress factors including dramatic temperature changes and desiccation a few of the major factors limiting growth during aerial exposure. In addition, motile macrobenthos could be detrimental to oysters because they interrupt filter-feeding activities and hence hamper the growth of oysters. We show the higher recruitment of oysters at the natural site and healthy growth in the farming substrate are due to complicated differences in physical and biological stress factors.

훈건 굴을 이용한 분말조미소재의 가공 및 품질안전성 (Processing and Shelf-life Stabilities of Flavoring Substances of the Smoke-Dried Oysters)

  • This study was conducted to evaluate the optimal processing conditions of smoke-dried powdered oysters and to determine their shelf-life during storage for development of a natural oyster flavoring substance. The optimal conditions for processing of smoke-dried oyster powder with freshy oyster were as follows. Raw shelled oysters were rinsed with 3% saline solution, drained, boiled for 10 minutes at $98^{\circ}C$, and then smoked for 1 hour at $50^{\circ}C$, followed by drying for 4 hours at $80^{\circ}C$ Smoke-dried oyster powder with oyster scraps were prepared as flavoring material. The smoked oyster scraps were submerged in oyster sauce far 10 minutes at room temperature and then dried with hot air for 5 hours at $50^{\circ}C$. The smoke-dried oysters and smoke-dried oyster scraps were then pulverized to 50 mesh and packed in tea bags or vacuum-packed in laminated plastic film bags (PE/PVDC/CPP, $12{\mu}m/15{\mu}m/50{\mu}m$). Compared to non smoke-dried powdered oysters, the smoking and dipping in oyster sauce enhanced the flavor and prevented lipid oxidation of the smoke-dried powdered oyster product. Shelf-life tests indicated that the vacuum-packaging method preserved the quality of smoke-dried powdered oysters stored for 150 days at room temperature.

양식산 굴(Crassostrea gigas)의 생굴 및 가공소재용으로서 화학적 및 생물학적 위생 특성 (Chemical and Biological Properties on Sanitary of Cultured Oyster Crassostrea gigas Intended for Raw Consumption or Use in Seafood Products)

  • Oysters Crassostrea gigas are a globally popular shellfish for human consumption. As filter-feeding bivalve mollusks, oysters may harbor many microorganisms and chemicals that could pose potential human health risks. The objective of this study was to investigate the suitability of cultured oysters for raw consumption or use in seafood products by measuring concentrations of harmful microorganisms and chemicals in their flesh. Microbial concentrations in cultured oysters were found to be: $1.0{\times}10^2-6.0{\times}10^4CFU/g$ (viable cell counts), not detected $(ND)-5.4{\times}10^3CFU/g$ (coliform bacteria), $ND-1.3{\times}10^2CFU/g$ (E. coli), and $ND-4.6{\times}10^3CFU/g$ (Vibrio parahaemolyticus). Other pathogenic bacteria, including Enterohemorrhagic E. coli (EHEC), Listeria monocytogenes, Staphylococcus aureus, and Salmonella spp., were not detected in any samples. Heavy metal concentrations of cultured oysters were ND-0.239 mg/kg (total mercury), ND-1.091 mg/kg (lead), ND-0.968 mg/kg (cadmium). The concentrations of benzo(a)pyrene ranged from $0.280-0.880{\mu}g/kg$. Paralytic shellfish poison ranged from ND-0.58 mg/kg, while diarrhetic shellfish poison was not detected. No radioactivity was detected. These results suggest that oysters intended for raw consumption or use in seafood products should be subjected to chemical and biological controls.
