Fisheries Resources in Garolim Bay (가로림만 어업자원에 관하여)

  • Garolim Bay is not only important fishing ground but also expected area for the tidal power plant. The construction and operation of tidal power plant will make change the ecosystem of this bay. Therefore, the actual fisheries stocks should be precisely understood for the effect estimation and overall utilization of the bay after the construction of the tidal power plant. During the study period from January through December in 1981, forty-six adult fishes species, 3 species of fish egg and 25 fishes larvae species have occurred in the bay. Considering the result on monthly distribution of eggs and larvae, the inner area of the bay seems to be important as nursing ground of larvae spawned at the outside bay in winter, e. g., Ammodytes personatus, and Enedrias sp. This inner bay is also major spawning ground for many species spawning in spring and summer, e. g., Gobiidae, Konosirus punctatus, Engraulis japonica, etc. Taking into consideration the annual mean production for three years($1978{\sim}1980$), there are two major fishing seasons. The one is in May-June for Enedrias larvae stock, and the another in October-November for big eyed herring stock. For the mariculture stocks, short necked clam, oyster and laver are important species. After construction of the tidal power plant, the migratory species, i. e., larvae of Enedrias and Ammodytes personatus, Mugil cephalus, Konosirus punctatus, etc. will be directly damaged by the interuption of migration route. On the otter hand, the change of physico-chemical factors of seawater will also affect the ecosystem of the bay. Consequently, for the overall utilization of the bay after construction, the actual ecosystem including the fisheries stocks, must bs precisely revealed, and the mechanical designs, e. g., sluice position and its demension, should be also considered with these biological characters of the bay.

Quality Characteristics and Antioxidant Activities of Fermented Deodeok Tea with Pleurotus eryngii Mycelium (Pleurotus eryngii Mycelium으로 발효한 더덕 차의 품질 및 항산화 특성)

  • This study was conducted to obtain the baseline data for fermented tea with King oyster (Pleurotus eryngii) mushroom mycelium as well as to investigate the possibility of development for deodeok processing products. For this research, the physicochemical properties and antioxidant activity for fermented tea were confirmed by different roasting temperatures and roasting times. The results of the physicochemical properties, browning, turbidity, reducing sugar, total polyphenol and total flavonoid contents, were increased with increasing roasting temperature and roasting time, whereas the pH was decreased. The results of the antioxidant activity test of fermented tea, DPPH radical-scavenging activity, FRAP and reducing power, were increased with increasing roasting temperature and roasting time. The preference of fermented tea was confirmed as being the highest when roasted at $180^{\circ}C$ for 30 min. Although fermented tea did not have a unique flavor of mushroom and burnt flavor, it had strong color, flavor and taste. For this reason, fermented tea production for high content of functional materials and high preference is possible by roasting at $180^{\circ}C$ for 30 min. As for the results of this study, the physical properties of fermented deodeok were relatively stable on high temperature exposure and furthermore, the development of a variety of foods will be possible because fermented deodeok has an antioxidant capacity.

Analysis of Environment Factors in Pleurotus eryngii Cultivation House (새송이버섯 재배사의 환경요인 분석)

  • Pleurotus eryngii(King oyster) is one of the most promising mushrooms produced on the domestic farms. The quality as well as quantity of Eryngii is sensitively affected by micro climate factors such as temperature, relative humidity, $CO_2$ concentration, and light intensity. To safely produce high-quality Eryngii all the year round, it is required that the environmental factors be carefully controlled by well designed structures equipped with various facilities and control systems. At the commercial mushroom cultivation house(A,B), this study was carried out to find out reasonable range of each environmental factor and yield together with economic and safe structures influencing on the optimal productivity of Eryngii. this experiment was conducted for about two-month from Nov. 11, 2002 to Dec. 30, 2002 in Eryngii. cultivation house-A, B. Ambient temperature during the experiment period was not predominantly different from that of a normal year. The capacity of the hot water boiler and the piping systems were not enough. Maximum air temperature difference between the upper and lower growth stage during a heating time zone was about 2~3$^{\circ}C$. The max. and min. relative humidity were ranged approximately 60~100%, and average relative humidity was ranged approximately 80~100%. And $CO_2$concentration increased until maximum 1,600~1,800 ppm with the passing growing period. The illuminance in cultivation house was widely distributed from 20lx to 160 lx in accordance with position, and it was maintained lower than the recommended illuminance range 100~200 lx. The average yield per bottle was about 67~85g. But the optimal productivity will be evaluated by considering the quality and quantity of mushroom production, energy requirements, facility construction and management cost, etc.

Production and utilization of organic compost from spent mushroom (Pleurotus eryngii) substrate (큰느타리버섯 수확 후 배지를 이용한 유기퇴비 제조 및 활용)

  • We investigated the possibility of utilizing spent mushroom culture-medium as raw material to produce organic compost. Mushroom culture-medium contained 2.69% nitrogen, 41.07% carbon, and 0.99% phosphoric acid. Nitrogen and carbon content in rice bran were 3.08% and 47.34%, respectively. Nitrogen, carbon, and phosphoric acid content in mushroom Pleurotus eryngii were 3.41%, 34.63%, and 1.70%, respectively. Our study aimed to evaluate the degree of decay of compost produced from the substrate, used to culture King Oyster mushrooms by analyzing seed germination indexes in cucumber, radish, lettuce, and Chinese cabbage. Our results showed that compost from spent mushroom culture-medium completed the stabilization process in 4 to 6 weeks, as evaluated by the appropriate organic compost maturity point for the plantation.

Characteristics and breeding of a new cultivar Pleurotus eryngii, Seolsong (큰느타리버섯 신품종 '설송'의 육성 및 그 특성)

  • To develop a new cultivar of king oyster mushroom(Pleurotus eryngii), G09-21 as parental strain was selected by the method of Di-mon crossing between monokaryotic strain ASI 2824-21 derived from ASI 2824(Keunneutari No. 2) and dikaryotic strain ASI 2887(Aeryni 3). The Pe21-53(G09-21-10 x ASI 2844-9) was shown the best cultural characteristics, selected to be a new cultivar and designated as 'Seolsong'. The 'Seolsong' was distinctly formed incompatibility line in the confrontation growth of parental strains Keunneutari No. 2, Aeryni 3 and ASI 2844. Analysis of the genetic characteristics of the new cultivar 'Seolsong' showed a different DNA profile as that of the control strains, Keunneutari No. 2, Aeryni 3 and ASI 2844, when RAPD(Random Amplified Polymorphic DNA) primer URP4 was used. The optimum temperature and pH arrange for mycelial growth were $25{\sim}30^{\circ}C$ and pH 5~8, respectively. This new cultivar 'Seolsong' of fruiting body production per bottle was about $131.4{{\pm}}43.1$ g which is about 102% quantity compared to that of other cultivar Keunneutari No. 2. And also the stipe is thick and long, but the number of available stipe is few. Particularly, it was tolerant of high moisture above 90% during the growth period after primodia formation. We therefore expect that this new strain will save of labor and cost of cultivation by without culling work.

Evaluation of the Influence of Inland Pollution Sources on Shellfish Growing Areas after Rainfall Events in Geoje Bay, Korea (강우에 따른 거제만해역 육상오염원의 영향평가)

  • The influences of inland pollution sources because of rainfall events on the bacteriological water quality in Geoje Bay, a major shellfish production area in Korea, were investigated. The sanitary status of sea water and shellfish after rainfall events was also evaluated. The flow rates of 13 streams around Geoje Bay showed 6 to 7-fold increases after 15 to 21 mm of rainfall. Peak pollution was observed in the Naegan Stream, the Gandeok Stream and the Seojeong Stream. The calculated impact area of inland pollution sources was 3.1 $km^2$ immediately after 15 mm of rainfall and expanded to 3.5 $km^2$ after 24 hours. These calculations of impacted area matched results from fecal coliform analyses with sea water. The distance between the major pollution source in the bay (the Gandeok Stream) and the station with the worst bacteriological water quality immediately after 15 mm of rainfall, which was below the Korean standard, was 0.8 km in a straight line; this distance increased to 2.0 km after a period of 24 hours. The area impacted by inland pollution sources after a 15 mm rainfall event was wider than after a 21 mm rainfall. Although the flow rate from inland pollution sources was higher, the concentration of fecal coliform in the discharged water was lower after higher rainfall events. These observations corresponded with the results of fecal coliform analyses with sea water samples. According to the evaluation of the influences of inland pollution sources and fecal coliform analyses on sea water and shellfish samples in Geoje Bay, pollutants from inland sources did not reach the boundary line of the shellfish growing area after rainfall events of 15 or 22 mm. The bacteriological water quality of the shellfish growing area in Geoje Bay met the Korean standard and US NSSP requirements for approved shellfish growing areas.

Effects of Different Natural Mordants and Mordanting Methods on the Dyeing Degree of Silk Using Extracts from Coreopsis drummondii (금계국 추출물에 의한 천연염색시 천연매염제 종류 및 매염방법에 따른 견직물의 염색성)

  • This study was conducted to clarify the effects of different natural mordants and mordanting methods on the dyeing degree of silk using extracts from Coreopsis drummondii. Surface colors were shown as an order of descent Y in control, and an order of descent Y and YR treated by mordants. Extracts from Coreopsis drummondii was sufficient for the natural dyestuffs. $10\%,\;20\%\;and\;30\%$ mordants made from lime-juice of Camellia japonica, oyster shells. and Symplocos chinensis (Lour) Druce for. pilosa (Nakai) Ohwi were used for the dyeing of silk. Surface color of silk was not affected by the concentration of mordants and mordanting methods, however, $20\%$ mordant was suitable for the dye coloring. It was shown that dye coloring concentrations were highly in the order of mordanting later > mordanting ahead > co-mordanting, and so on. Silks were naturally dyed by the extracts from Coreopsis drummondii, and twenty-nine kinds of natural mordants were treated and screened. Surface color of silk was shown as an order of descent YR treated by the mordants made from the lime-juice of Camellia japonica and Eurya japonica, and as an order of descent Y by the other mordants. By the treatment of natural mordants, concentrations of dye coloring were significantly increased compared with control (none-mordanting), and the potentials as a natural mordant for the effective dye coloring was sufficiently ascertained.

Amylase, Pretease, Peroxidase and Ascorbic Acid Oxidase Activity of Kimchi Ingredients (김치원료의 amylase, protease, peroxidase, ascorbic acid oxidase 활성)

  • Several enzymes of kimchi ingredients were assayed to improve the product quality using these quality related enzyme information. Among various hydrolases, amylase and protease were selected with respect to lactic acid fermentation. Peroxidase and ascorbic acid oxidase were studied for off flavor production and ascorbic acid destruction. The amount of protein in kimchi ingredients, specific and total enzyme activity of sample were compared. Regarding total enzyme activity of sample, ${\alpha}-amylase$ activity of salted and fermented anchovy, dried red pepper and salted and fermented shrimp were higher than other ingredients. Activity of salted and fermented anchovy was 2,790.0 units/g sample. Salted and fermented anchovy, oyster and Chinese radish showed the highest ${\beta}-amylase$ activity (4.4, 2.1, 1.0 units/g sample, respectively). Salted and fermented anchovy showed the highest protease activity of 13.4 PU/g sample, followed by salted and fermented shrimp and dried red pepper. For peroxidase, Chinese radish, cucumber, green onion showed the highest activity of 7.2, 6.8 and 5.6 units/g sample, respectively. In case of ascorbic acid oxidase, salted and fermented anchovy showed the strongest enzyme activity (331.4 units/g sample), followed by dried red pepper and salted and fermented shrimp.

Some Properties and Microbial Community Changes of Gul (Oyster) Jeotgal during Fermentation

  • Gul jeotgals (GJs) were prepared using solar salt aged for 3 years. One sample was fermented using starters, such as Bacillus subtilis JS2 and Tetragenococcus halophilus BS2-36 (each $10^6CFU/g$), and another sample was fermented without starters for 49 days at $10^{\circ}C$. Initial counts of bacilli and lactic acid bacteria (LAB) in non-starter GJ were found to be $3.20{\times}10^2$ and $7.67{\times}10^1CFU/g$ on day 0, and increased to $1.37{\times}10^3$ and $1.64{\times}10^6CFU/g$ on day 49. Those of starter GJ were found to be $2.10{\times}10^5$ and $3.30{\times}10^7CFU/g$ on day 49, indicating the growth of starters. The pH values of GJ were $5.93{\pm}0.01$ (non-starter) and $5.92{\pm}0.01$ (starter) on day 0 and decreased to $5.78{\pm}0.01$ (non-starter) and $5.75{\pm}0.01$ (starter) on day 49. Amino-type nitrogen (ANN) production increased continuously during fermentation, and $407.19{\pm}15.85$ (non-starter) and $398.04{\pm}13.73$ (starter) mg% on day 49. Clone libraries of 16S rRNA genes were constructed from total DNA extracted from non-starter GJ on days 7, 21, and 42. Nucleotide sequences of Escherichia coli transformants harboring recombinant pGEM-T easy plasmid containing 16S rRNA gene inserts from different bacterial species were analyzed using BLAST. Uncultured bacterium was the most dominant group and Gram - bacteria such as Acidovorax sp., Afipia sp., and Variovorax sp. were the second dominant group. Bacillus amyloliquefaciens (day 7), Bacillus velezensis (day 21 and 42), and Bacillus subtilis (day 42) were observed, but no lactic acid bacteria were detected. Acidovorax and Variovorax species might play some role in GJ fermentation. Further studies on these bacteria are necessary.

Effect of Postmortem Phases on Lamb Meat Quality: A Physicochemical, Microstructural and Water Mobility Approach

  • To investigate the effect of postmortem phases on lamb meat quality, the physicochemical quality, microstructure and water mobility of oyster cut, short loin, knuckle and silverside muscles from Small-Tail Han sheep were evaluated in the pre-rigor, rigor mortis and post-rigor phases. Pre-rigor lamb meat had higher pH and water holding capacity (WHC), whereas lower CIE L*, b*, hue angle values than rigor mortis and post-rigor meat (p<0.05). The Warner-Bratzler shear force (WBSF) values were higher in rigor mortis short loin and silverside than their pre-rigor and post-rigor counterparts, pre-rigor short loin had lower WBSF value than its post-rigor counterpart (p<0.05). Muscle fibers shrank laterally and longitudinally during the onset of rigor mortis. Rigor mortis and postrigor lamb meat exhibited wide I-bands, dark A-bands, short sarcomeres and large inter-myofibrillar spaces. The shift of immobilized water to free water and repulsion from the intra-myofibrillar space to the extracellular space result in the increase of water loss in rigor mortis and post-rigor lamb meat. The results of the principal component analysis (PCA) indicated that rigor mortis and post-rigor lamb meat had similar quality properties but different from pre-rigor lamb meat. In conclusion, the lamb meat in the pre-rigor phase had good tenderness, color and WHC. The results of this research could provide some theoretical references for lamb meat production and processing.
