  • Isoimperatorin and Oxypeucedanin isolated from root of Angelica Koreana Max. (Umbelliferae) that is used as "Kang-Whal" a botanical drug, in Korea, Show following general pharmacologic activities. The treatment with Oxypeucedanin on the exercised duodenum of the rabbit increased the motility of it while isoimperatorin shows little effects. But on the exercised heart of the frogs, both components depressed its contractility. Whereas Oxypeucedanin depressed both blood pressure of the carotied artery and respiration of the rabbit, Isoimperatorin tends to increase blood pressure, respiration and the tonus of the muscle of the earth worm. Oxypeucedanin did not affect the tonus of the muscles of the earth worm, but depressed the uterus contraction of the rabbit, which was, on the contrary, increased by Isoimperatorin.

Effects of oxypeucedanin hydrate isolated from Angelica dahurica on myoblast differentiation in association with mitochondrial function (백지에서 추출한 oxypeucedanin hydrate의 미토콘드리아 기능 관련 근생성 효과)

  • Purpose: Mitochondria play a crucial role in preserving skeletal muscle mass, and damage to mitochondria leads to muscle mass loss. This study investigated the effects of oxypeucedanin hydrate, a furanocoumarin isolated from Angelica dahurica radix, on myogenesis and mitochondrial function in vitro and in zebrafish models. Methods: C2C12 myotubes cultured in media containing 0.1, 1, 10, or 100 ng/mL oxypeucedanin hydrate were immunostained with myosin heavy chain (MHC), and then multinucleated MHC-positive cells were counted. The expressions of markers related to muscle differentiation, muscle protein degradation, and mitochondrial function were determined by quantitative reverse transcription polymerase chain reaction. To investigate the effects of oxypeucedanin hydrate on mitochondrial dysfunction, Tg(Xla.Eef1a1:mito-EGFP) zebrafish embryos were treated with 5-fluorouracil, leucovorin, and irinotecan (FOLFIRI) with or without oxypeucedanin hydrate and analyzed for mito-EGFP intensity and mitochondrial length. Results: Oxypeucedanin hydrate significantly increased MHC-positive multinucleated myotubes (≥ 3 nuclei) and increased the expression of the myogenic marker myosin heavy chain 4. However, it decreased the expressions of muscle-specific RING finger protein 1 and muscle atrophy f-box (markers of muscle protein degradation). Furthermore, oxypeucedanin hydrate enhanced the expressions of markers of mitochondrial biogenesis (peroxisome proliferator-activated receptor-gamma coactivator 1 alpha, transcription factor a mitochondrial, succinate dehydrogenase complex flavoprotein subunit A, and cytochrome c oxidase subunit 1) and mitochondrial fusion (optic atrophy 1). However, it reduced the expression of dynamin-related protein 1 (a mitochondrial fission regulator). Consistently, oxypeucedanin hydrate reduced FOLFIRI-induced mitochondrial dysfunction in the skeletal muscles of zebrafish embryos. Conclusion: The study indicates that oxypeucedanin hydrate promotes myogenesis by improving mitochondrial function, and thus, suggests oxypeucedanin hydrate has potential use as a nutritional supplement that improves muscle mass and function.

  • Four furocoumarins and two dihydrofuranocoumarin glycosides have been isolated from the benzene-soluble and butanol-soluble portions of the roots of Ostericum koreanum Kitagawa, respectively. Four furocoumarins have been determined as imperatorin, isoimperatorin, oxypeucedanin and oxypeucedanin hydrate, and two dihydrofuranocoumarin glycosides determined as marmesinin and $4'-O-{\beta}-D-glucopyranosyl-3'-hydroxymarmesin$ on the basis of spectral data and physico-chemical evidence.

  • Five furanocoumarins including a new one were isolated from the root of Angelica dahurica by repeated silica gel column chromatography. Their chemical structures were determined to be isoimperatorin (1), oxypeucedanin hydrate-3"-butyl ether (2), imperatorin (3), knidilin (4), and oxypeucedanin hydrate (5). This represents the first study in which the compound 2 has been isolated and identified, The long-range coupling ($^{5}J$) in the $^1H$-NMR spectrum observed in the linear furanocoumarin skeleton was also investigated in detail.

  • Ten coumarins were isolated from the root of Angelica dahurica by repeated silica gel column chromatography. Their chemical structures were elucidated on the basic of physicochemical and spectroscopic data. Among them, oxypeucedanin hydrate acetonide (7) was isolated for the first time from this plant. Cytotoxicity of coumarins isolated were determined in vitro against L1210, HL-60, K562, and B16F10 tumor cell lines by MTT method. Pangelin (5) and oxypeucedanin hydrate acetonide (7) showed a potent cytotoxic activity with the $IC_{50}$ values of 8.6 to 14.6 ${\mu}g$ /mL against four kinds of tumor cell lines. Other compounds showed the moderate cytotoxic activity or no activity against the tumor cell lines.

The Comparison of Constituents Distributed in the Root of Angelica dahurica (구릿대 뿌리의 부위별 성분 비교)

  • In this study, we analyzed and quantified the amounts of bioactive phenolic constituents, xanthotoxin (1), oxypeucedanin (2), and imperatorin (3) in each part of the root of Angelica dahurica by HPLC, which validated by ICH guide lines comparing the linearity, intra day precision, inter-day precision. As a result, the amount of imperatorin 2.96% in the rootlet was two fold higher than that of the main root 1.32%. On the other hand, the amounts of xanthotoxin and oxypeucedanin in the rootlet showed similar to those of main root. In addition, the cortex of root is more plentiful of three consitituents (0.66%, 0.53%, and 1.85%) than those of xylem (0.29%, 0.05%, and 0.07%). These results show that the rootlet and cortex contain a large amount of bioactive phenolic constituent including xanthotoxin, oxypeucedanin, and imperatorin than other parts of the root.

Effect of Imperatorin on Adenosine Triphosphate-stimulated Mucin Secretion from Airway Epithelial Cells

  • Angelicae Koreanae Radix has been used for controlling inflammatory respiratory diseases in folk medicine and their components, imperatorin, marmesinin and oxypeucedanin were reported to have diverse biological effects. In this study, we investigated whether imperatorin, marmesinin and oxypeucedanin affect adenosine triphosphate(ATP)-induced mucin secretion from cultured airway epithelial cells. Confluent primary hamster tracheal surface epithelial(HTSE) cells were metabolically radiolabeled using $^3H$-glucosamine for 24 h and chased for 30 min in the presence of varying concentrations of each agent to assess the effects on $^3H$-mucin secretion. The results were as follows: 1) imperatorin significantly inhibited ATP-induced mucin secretion; 2) However, marmesinin and oxypeucedanin did not affect ATP-induced mucin secretion, significantly. We conclude that imperatorin might inhibit ATP-induced mucin secretion by directly acting on airway mucin-secreting cells. Therefore, imperatorin should further be investigated for the possible use as mucoregulators in the treatment of inflammatory airway diseases.

Comparative Studies on Medicinal Constituents of Korean and Chinese Angelicae Dahuricae Radix

  • The medicinal constituents contained in Korean and Chinese Angelicae dahuricae Radix were compared by confirming their qualities. From the extracts of Angelicae dahuricae Radix, oxypeucedanin, imperatorin, and alloisoimperatorin etc. furanocoumarin derivatives were identified by GC/MS analysis. Through GC/FID analysis, the furanocoumarin derivatives content of Baizi cultivated in Korea was more than that cultivated in China, except for one cultivated at Ankuk province, and so confirmed to possess, on the whole, good quality medicinal constituents by content, as compared with the ones cultivated in China.

  • To investigate biological active constituents from natural products, we have studied the roots of Angelica koreana Max. (Umbelliferae). Fifteen compounds were isolated from the MeOH extract by column chromatography on a silica gel. The compounds were identified as isoimperatorin, oxypeucedanin, oxypeucedanin hydrate, osthol, nodakenin, 2-hydroxy-4-methylacetophenone, cimifugin, falcarindiol, heraclenin, pabulenol, umbelliferone, demethylsuberosin, hamaudol, sec-O-glucosylhamaudol, and prim-O-glucosylcimifugin, respectivelv, by spectroscopic means. Among these, the latter eight compounds were isolated for the first time form this plant.
