Effects of Aralia elata Water Extracts on Activities of Hepatic Oxygen Free Radical Generating and Scavenging Enzymes in Streptozotocin-Induced Diabetic Rats (두릅열수추출물이 당뇨유발 흰쥐의 간조직 중 유해 활성산소 대사효소계 활성에 미치는 영향)

  • Oxidative stress is currently suggested as a mechanism underyling diabetes. Accordingly, the present study was designed to evaluate the effect of Aralia elate water extracts (AEW) on activities of hepatic oxygen free radical generating and scavenging enzymes in streptozotocin (STZ)-induced diabetic rats. Male Wistar rats divided into nondiabetic group, diabetic group, and diabetic-AEW supplemented group. The extract was supplemented in 1.14% of raw Aralia elata/kg diet for 7 weeks. Diabetes was induced by injecting STZ (55 mg/kg BW, ip) once 2 weeks before sacrifying. The hepatic cytochrome P-450 content, xanthine oxidase and aminopyrine N-demethylase activities were significantly lowered in the diabetic group compared to the nondiabetic group. Whereas, the activities of aniline hydroxylase and oxygen free radical scavenging enzymes, superoxide dismutase, catalase, glutathione peroxidase, glutathione reductase, glucose-6-phosphate dehydrogenase and glutathione S-transferase, were significantly higher in the diabetic group than in the nondiabetic group. However, the supplementation of AEW normalized these enzyme activities in STZ-induced diabetic rats. When the AEW was supplemented with the diabetic rats, hepatic glutathione content was markedly elevated as well as lipid peroxide level was significantly lowered compared to those of the diabetic group. Thus, these results suggested that AEW supplement enhanced the activities of oxygen species metabolizing enzymes in STZ-induced diabetic rats.

The Simultaneous Nitrification and Organics Oxidation of Wastewater in Airlift Biofilm Reactors (공기리프트 생물막 반응기에서의 폐수 질화 및 유기물 동시산화)

  • The effects of organic supplement (acetate) and dissolved oxygen concentration on the nitrification rate of wastewater were investigated in the 27.7 L pilot-scale airlift biofilm reactor with the granular activated carbon media of 0.613 mm diameter. The ammonium oxidation rate increased stepwise up to 5 kg N/㎥$.$d at the riser air velocity of 0.063 m/s, when the air velocity and the ammonium loading rate were raised alternately. The nitrite build-up was observed during the early stage of the biofilm formation, which disappeared after the reactor operation of 128 days. As increasing the organic loading rate, the organic oxidation rate increased up to 25.0 kg COD/㎥$.$d with the removal efficiency of 94% but the oxidation rates of ammonium and nitrite decreased. The oxidation rates of ammonium and nitrite increased with the dissolved oxygen concentrations. When the pure oxygen was sparged, the ammonium oxidation rate was almost five times higher than that with air at the same velocity.

  • This research evaluated how the composition of Yak-sun(oriental diet therapy), taken as a nutritional supplement with daily meals, can effect the medical condition of those suffering from modern diseases like obesity and hyperlipideia. We produced Yak-sun tea, determined its nutrient composition and physical characteristics and evaluated how this tea effects affects the active oxygen concentration inside the body by clinical practices. The study results indicated that this tea significantly decrease active oxygen concentration. We think that scientific and objective evaluation was done on the components of Yak-sun tea prescription. We concluded that we could apply the components not only in the form of tea, but also in other food forms. The information we received from this conclusion will provide basic information for the application of oriental medicinal resources into other foods and will also be a steppingstone in the field of functional food research for the development of medicinal herbs which is already an important research field world-wide.

Protectins Effects of Vitamin E against Immobilization Stress-Induced Oxidative Damage in Rat Brain (스트레스로 인한 뇌조직의 산화적 손상에서 Vitamin E의 방어 효과)

  • The remarkable change of phenomenon induced by stress increase energy metabolism that can induce many reactive oxygen species (ROS) production. ROS can peroxidize cellular macromolecules including lipid and protein. The object of this study was to investigate whether stress may induce cellular damage by producing ROS and whether vitamin E, as a strong lipid-soluble antioxidant, can protect cells against reactive oxygen species produced by noise and immobilization stress in SD rats. The stress group increased 5-hydroxyindole aceti acid (5-HIAA) , one of the stress hormone, in brain tissue and free fatty acid in plasma. Vitamin I treatment had no effect on 5-HIAA but free fatty acid contents decreased with a fortified vitamin I diet. Furthermore, the body weight of vitamin I-treated rats increased more than that of the stress group. Lipid peroxidation and protein degradation as an index of oxidative damage in brain tissue decreased with the use of the fortified vitamin I diet supplement. The results suggest that vitamin I supplements have a protective effect against noise and immobilization stress-induced oxidative damage in brain tissue.

The Effect of Carbon Monoxide Inhalation on the Uterine Motility of the Nonpregnant Rabbit (일산화탄소흡입(一酸化炭素吸入)이 비임신(非妊娠) 토끼자궁운동(子宮運動)에 미치는 영향(影響))

  • Adult nonpregnant female rabbits were subjected to the study of the effects of carbon monoxide inhalation on the uterine motility. Animals were anesthetized with intravenous injection of nembutal, 35 mg/kg, and the uteri were exposed. Polyethylene tubing which had a small hole near the blind tip was inserted in the loop and normal saline was infused at a constant rate of 1.5 ml/min. On the other end of the loop, an outlet of fluid was made. When a peristaltic wave proceeded to the hole, a rise of the pressure was ensued and it was transmitted to the pressure transducer, making an upward deflection of the recording pen on the physiograph. Carbon monoxide, 1,000 ppm in the concentration, was inhaled through a tracheal cannula for 30 minutes, following fresh air for 30 minutes. In some cases, pure oxygen was also supplemented for another 30 minutes. Uterine motility was expressed in terms of the impulse that was the time integral of the pressure and of the frequency of the peristaltic waves. The results obtained were as follows. 1. When 1,000 ppm carbon monoxide was inhaled for 30 minutes, the impulse dropped to $72{\pm}16.5%$ and the frequency to $75{\pm}22.7%$ of the values obtained before the gas administration. 2. By fresh air for 30 minutes, the impulse and the frequency restored to $77{\pm}25.7%$ and $92{\pm}21.1%$, respectively. 3. By the supplement of pure oxygen for 30 minutes, no remarkable improvement were revealed, showing $89{\pm}35.2%$ in the impulse and $91{\pm}10.8%$ in the frequency, respectively. 4. There was an appreciable discrepancy in the recovery courses of the impulse and the frequency, suggesting different mechanisms attributable to the alteration by carbon monoxide inhalation.

Enhanced (R)-2-(4-Hydroxyphenoxy)Propionic Acid Production by Beauveria bassiana: Optimization of Culture Medium and H2O2 Supplement under Static Cultivation

  • (R)-2-(4-hydroxyphenoxy)propionic acid (HPOPA) is a key intermediate for the preparation of aryloxyphenoxypropionic acid herbicides (R-isomer). In order to improve the HPOPA production from the substrate (R)-2-phenoxypropionic acid (POPA) with Beauveria bassiana CCN-A7, static cultivation and H2O2 addition were attempted and found to be conducive to the task at hand. This is the first report on HPOPA production under static cultivation and reactive oxygen species (ROS) induction. On this premise, the cultivation conditions and fermentation medium compositions were optimized. As a result, the optimal carbon source, organic nitrogen source, and inorganic nitrogen source were determined to be glucose, peptone, and ammonium sulfate, respectively. The optimal inoculum size and fermentation temperature were 13.3% and 28℃, respectively. The significant factors including glucose, peptone, and H2O2, identified based on Plackett-Burman design, were further optimized through Central Composite Design (CCD). The optimal concentrations were as follows: glucose 38.81 g/l, peptone 7.28 g/l, and H2O2 1.08 g/l/100 ml. Under the optimized conditions, HPOPA titer was improved from 9.60 g/l to 19.53 g/l, representing an increase of 2.03-fold. The results obtained in this work will provide novel strategies for improving the biosynthesis of hydroxy aromatics.

Effects of Alpha-G Rutin Supplementation in Sperm Freezing Extender on Dog Sperm Cryopreservation

  • This study was designed to investigate the effects of alpha-glucosyl rutin (G-rutin) and its comparative effects with other antioxidants (glutathione: GSH, catalase: CATA and beta-mercaptoethanol : ${\beta}ME$) on dog sperm freezing. In the first experiment (E1), the spermatozoa were diluted in freezing extender supplemented with 0 (control), 0.001, 0.01, or 0.1% G-rutin and frozen using liquid nitrogen ($LN_2$). The progressive motility, reactive oxygen species (ROS) level and apoptosis of spermatozoa were assessed after sperm thawing at $37^{\circ}C$ for 25 sec. In the second experiment (E2), 0.1% G-rutin group was compared with 10 mM ${\beta}-ME$, $5{\mu}M$ GSH and $50{\mu}M$ CATA groups by assaying progressive motility, viability and gene expression of Bcl-2 and SMCP after sperm freezing and thawing. In E1, 0.1% G-rutin group showed higher (P < 0.05) post-thaw progressive motility and lower (P < 0.05) ROS levels. In E2, the expressions of SMCP in G-rutin group were higher (P < 0.05) than in CATA group while Bcl-2 expression of G-rutin group was higher (P < 0.05) than ${\beta}-ME$ and CATA groups. However, there were no significant differences in progressive motility and viability. Therefore, we suggest that G-rutin can be used as a potentially antioxidative supplement in dog sperm freezing extender on the basis of gene expression related to motility and apoptosis as well as ROS level.

Pollutants Classification based on Trend Analysis and Assessment of Water Pollutants Achievement in Subbasins of Han River Basin (한강수계 중권역별 오염물질 추세분석 및 달성도 평가를 통한 우선관리물질 선정)

  • The objectives of this study were to analyze trends of water pollutants and to evaluate the achievement of water quality standards by subbasins in the Han River. The trends of 40 water pollutants at 232 water quality measurement points were analyzed. Chemical oxygen demand (COD), Total organic carbon (TOC), Total coliforms (TC), et cetera were found to be worsening trend. For evaluation of achievement, we evaluated water quality arithmetic mean with river environment standards and human health standards at representative points of the subbasin. Biochemical oxygen demand (BOD), TOC, Total phosphorus (T-P), Fecal coliforms (FC), TC exceeded water quality standards, and water quality of human health standards was all satisfied. So, we prioritized pollutants. If pollutants exceed water quality standards or were worse, they were classified first pollutants. Although BOD and T-P are first pollutants because of water quality standards excess, they are continuously improved. Also, it is better to maintain current status because water quality management system of BOD and T-P is well prepared. Meanwhile, TOC, TC, and FC exceed water quality standards. Furthermore, they were worse gradually, but there is a lack of management systems such as water quality standards of the effluence facilities. Therefore, it is necessary to supplement the system. The results of this study can be used as primary data for the establishment of water quality standards and selection of management pollutants.

Overlap Syndrome:Obstructive Sleep Apnea-Hypopnea Syndrome in Patients with Chronic Obstructive Pulmonary Disease (중첩증후군:만성 폐쇄성 폐질환을 가 진 폐쇄성 수면무호흡-저호흡 증후군)

  • Overlap syndrome can be defined as a coexistence of chronic obstructive pulmonary disease (COPD) and sleep apnea-hypopnea syndrome (SAHS). The association of COPD and SAHS has been suspected because of the frequency of both diseases. Prevalence of COPD and SAHS is respectively 10 and 5% of the adult population over 40 years of age. However, a recent study has shown that the prevalence of SAHS is not higher in COPD than in the general population. The coexistence of the two diseases is only due to chance. SAHS does not affect the pathophysiology of COPD and vice versa. Prevalence of overlap syndrome is expected to occur in about 0.5% of the adult population over 40 years of age. Patients with overlap syndrome have a more profound hypoxemia, hypercapnia, and pulmonary hypertension when compared with patients with SAHS alone or usual COPD patients without SAHS. To treat the overlap syndrome, nocturnal noninvasive ventilation (NIV) or nasal continuous positive airway pressure (nCPAP) can be applied with or without nocturnal oxygen supplement.

Study on prestressed concrete beams and poles with cement replaced by steel dust

  • Cement acts as the most important component of concrete as it binds and holds the concrete together. But it is one of the major $CO_2$ emitters all over the world, during manufacturing (900 kg of $CO_2$ per 1000 kg). Some of the modern construction methods aim at reducing the amount of usage of cement and came out with numerous solutions for replacement of the same. One such supplement in current trend is the Steel dust or the Electric Arc Furnace Dust (EAFD), which is a waste product from the electric arc furnace when the scrap metal is melted. When the concrete containing steel dust is exposed to atmosphere, the environmental oxygen and moisture play role to form rust and ultimately the member becomes harder. As Cement is the binder of conventional concrete, only certain percentage of the same could be replaced by the new material, steel dust. Tests were conducted for the 28 days cube strength of M45 grade (suitable for prestressing) concrete which has 0%, 10%, 20%, 30%, 40% and 50% steel dust instead cement. From the test, the optimum percentage replacement of steel dust was obtained, for which the beams and overhead poles were cast, prestressed and tested for the failure load and deflections. A conventional concrete beam and overhead pole were also cast, prestressed and tested to compare the results with those of the beam and pole that contained steel dust. The load vs. deflection plot and other results from the test is also discussed.
