THE EFFECT OF HYDROGEN AND OXYGEN CONTENTS ON HYDRIDE REORIENTATIONS OF ZIRCONIUM ALLOY CLADDING TUBES

  • To investigate the effect of hydrogen and oxygen contents on hydride reorientations during cool-down processes, zirconium-niobium cladding tube specimens were hydrogen-charged before some specimens were oxidized, resulting in 250 ppm and 500 ppm hydrogen-charged specimens containing no oxide and an oxide thickness of $0.38{\mu}m$ at each surface. The nonoxidized and oxidized hydrogen-charged specimens were heated up to $400^{\circ}C$ and then cooled down to room temperature at cooling rates of $0.3^{\circ}C/min$ and $8.0^{\circ}C/min$ under a tensile hoop stress of 150 MPa. The lower hydrogen contents and the slower cooling rate generated a larger fraction of radial hydrides, a longer radial hydride length, and a lower ultimate tensile strength and plastic elongation. In addition, the oxidized specimens generated a smaller fraction of radial hydrides and a lower ultimate tensile strength and plastic elongation than the nonoxidized specimens. This may be due to: a solubility difference between room temperature and $400^{\circ}C$; an oxygen-induced increase in hydrogen solubility and radial hydride nucleation energy; high temperature residence time during the cool-down; or undissolved circumferential hydrides at $400^{\circ}C$.

Fenbendazole의 항암활성에서 활성산소종의 관련성 (Involvement of reactive oxygen species in the anti-cancer activity of fenbendazole, a benzimidazole anthelmintic)

  • Fenbendazole (FBZ) is a benzimidazole anthelmintic that has been widely used in treatments for gastrointestinal parasites including pinworms and roundworms in animals. Recently, some studies demonstrated that FBZ has anti-cancer effects related to disruption of microtubule polymerization. In this study, we investigated whether FBZ has anti-cancer activity in HL-60 cells, a human leukemia cell line, and assessed its relationship with the production of reactive oxygen species (ROS). FBZ treatment at 0.25-1 μM significantly decreased the metabolic activity of HL-60 cells. The mitochondrial membrane potential of FBZ-treated HL-60 cells decreased in a concentration-dependent manner. Apoptosis analysis using annexin V-FITC/propidium iodide staining demonstrated that 1 μM FBZ increased the percentages of cells in apoptosis and necrosis. In addition, Hoechst 33342 staining showed the presence of broken nuclei in HL-60 cells treated with 0.5 and 1 μM FBZ. To investigate the anti-cancer mechanism of FBZ, HL-60 cells were treated with FBZ in the absence or presence of N-acetyl cysteine (NAC), an inhibitor of ROS production. NAC significantly recovered the decreased metabolic activity of HL-60 induced by 0.5 and 1 μM FBZ treatments. This study provides evidence that FBZ has anti-cancer activity in HL-60 cells provided, in part, via ROS production.

Experimental Verification of a Kinetic Model of Zr-Oxidation

  • It has long been known that the oxidation kinetics of Zr-based alloys undergoes a crossover from parabolic to cubic in the pretransition period (before breakaway of the oxide scale). This kinetic crossover, however, is not fully understood yet. We have earlier proposed a model for the Zr-oxidation kinetics, in a closed form for the first time, by taking into account a compressive strain energy gradient as a diffusional driving force in addition to a chemical potential gradient of component oxygen across the ZrO$_2$ scale upon Zr [J. Nucl. Mater., 299 (2001) 235]. In this paper, we experimentally reconfirm the validity of the proposed model by using the thermogravimetric data on mass gain of Zr in a plate and wire form, respectively, in air atmosphere at different temperatures in the range of 500$^{\circ}$ to 800$^{\circ}C$, and subsequently report on the numerical values for oxygen chemical diffusivity and strain energy gradient across the oxide scale.

직접에탄올 연료전지의 운전조건에 관한 연구 (A Study on an Operating Conditions for the Direct Ethanol Fuel Cell)

  • The goal of this paper is to find an operating conditions of the single direct ethanol fuel cell such as the cell temperature, and flow rates of ethanol and oxygen. To investigate the output characteristics, the electrical current increased from 0[A] with interval of 0.001[A] every 2[s], and the cell voltage was increased until the voltage became 0.05[V]. Related to the effect of the cell temperature, the output characteristics both voltage and power were increased upto 80[$^{\circ}C$] according to the increase of the current density, but those were decreased over that temperature. In addition, the optimal flow rate of ethanol in anode was identified as of 2[mL/min] due to the dependence of generation rate such as the hydrogen ion and electron. And the flow rate of oxygen in cathode was desirable to about 300[sccm/min], it might be affected by the chemical reaction rate of the water formation among hydrogen ion, electron, and oxygen. Consequently, the fundamental conditions were identified in this work, and it will be carried out to find the best conditions of membrane by the effect of the plasma surface treatment, and the effect of other catalysts except for a platinum.

충격파관을 이용한 메탄/산소 혼합기의 점화지연시간 측정 (Measurement of Ignition Delay Time of Methane/Oxygen Mixtures by Using a Shock Tube)

  • Ignition delay time of methane/oxygen mixture is measured experimentally with the shock tube in order to obtain the data for high pressure conditions where gas turbines and internal combustion engines are operating. The shock tube experiment is validated first over the temperature range of 1400-2000 K at 10 bar and with the various equivalence ratios of 0.5, 1 and 2. The measured ignition delays are compared with the data from the literatures. And then, experiments are conducted for non-explored conditions, i.e., at 40 bar and with the equivalence ratio of 1.5. The present experimental data show a good agreement with the available ones from the literatures and reasonable dependence on pressure and equivalence ratio. In addition, the effects of the temperature and equivalence ratio on ignition delay time are analyzed.

Juniperus chinensis extract induces apoptosis via reaction oxygen species (ROS) generation in human pancreatic cancer cell lines

  • Pancreatic cancer is among the most difficult-to-treat tumors. More than half of patients with this cancer have very few symptoms at the early stages, allowing the development of distant metastases and resistance to cancer treatment. In this study, we found that Juniperus chinensis extract (JCX) decreased the cell viability and migration activity of PANC-1 and SNU-213 pancreatic cancer cells in a dose-dependent manner. JCX increased caspase-3 activation and generation of reactive oxygen species (ROS). N-acetylcysteine treatment blocked JCX-induced ROS generation and the negative effects on pancreatic cancer cell viability. In addition, JCX down-regulated the levels of phospho-focal adhesion kinase (p-FAK) and phospho-extracellular signal-regulated kinase (p-ERK). Together, these results indicate that JCX induces apoptosis in human pancreatic cancer cell lines through ROS production, downregulating FAK/ERK signaling and activating caspase-3. We propose that JCX-derived compounds represent candidates for the development of alternative medicines for the treatment of pancreatic cancer.

일상적인 음식 섭취 후 발생한 급성 메트헤모글로빈혈증 (Acute methemoglobinemia after a normal diet)

  • Methemoglobin is generated by the oxidation of ferrous iron to ferric iron within a hemoglobin molecule. Methemoglobin is unable to bind and transport oxygen, resulting in methemoglobinemia, which can lead to fatal tissue hypoxia. The most common cause of methemoglobinemia is poisoning by oxidizing agents such as dapsone, benzocaine, and primaquine. However, methemoglobinemia can also be caused by normal dietary sources. We present two cases of methemoglobinemia that developed after a normal diet in two male patients. In this case report, the patients suddenly developed dyspnea and cyanosis after eating the same meal. They had no history of suspected poisoning, such as the use of drugs, exposure to chemicals, or gas inhalation. Their symptoms did not improve even after a high dose of oxygen was administered; further, an abnormal 'oxygen saturation gap' was observed. Because of CO-oximetry, the methemoglobin levels of the patients were 50.0% and 46.6%, respectively. We administered methylene blue (1 mg/kg), and the patients recovered completely without any complications. Emergency physicians should, therefore, be aware that methemoglobinemia can also be caused by normal dietary sources. In addition, if the source and route of contamination are unclear, an epidemiological investigation should be conducted.

Supplementation of cryoprotective extender with resveratrol decreases apoptosis index and reactive oxygen species levels in post-thaw dog sperm

  • Resveratrol (RSV, 3,5,4'-trihydroxytrans-stilbene) protects sperm from cryo-induced damage in various animal and human species. In this study, we aimed to assess the effect of dog sperm cryoprotective extender containing RSV on the quality of post-thaw dog sperm. Sperm were collected from 4 Beagles and supplemented with different concentrations of RSV (0, 100, 200, and 400 µM). After thawing, apoptosis index, and reactive oxygen species (ROS) levels were assessed to determine post-thaw sperm quality. Dog sperm cryopreserved with 400 µM RSV showed significant improvement in post-thaw sperm quality with lower apoptosis index and ROS levels (p < 0.05). Our results showed that the supplementation of dog sperm cryoprotective extender with RSV at a concentration of 400 µM improved the post-thaw dog sperm quality in the term of sperm ROS production and apoptosis. In addition, we emphasize the necessity of testing the ROS levels and apoptosis index using flow cytometry to determine the quality of post-thaw semen.

Tropical red alga Compsopogon caeruleus: an indicator of thermally polluted waters of Europe in the context of temperature and oxygen requirements

  • The red alga Compsopogon caeruleus can generally be found in tropical and subtropical waters worldwide. In addition to its natural habitats, this species may be found in waters that receive abnormally hot water, e.g., from powerhouses. To date, the presence of C. caeruleus has not been observed in thermally polluted lacustrine ecosystems in Poland, which has a moderate climate. The thalli of this red alga were found growing on Vallisneria spiralis in Lichenskie Lake. Importantly, this paper presents a previously unknown relationship between the temperature (20, 25, 30, 35, and 40℃) and oxygen requirements of C. caeruleus (based on ex situ measurements of O2 consumption by thalli). Surprisingly, 35℃ can be the optimum temperature for C. caeruleus, and this temperature is higher than the values reported by some previous thermal analyses by approximately 10℃. Additionally, we reviewed and mapped the distribution of this nonnative and mesophilic red alga in natural / seminatural water ecosystems in Europe. Finally, we propose that the occurrence of C. caeruleus mature thalli can be a novel, simple and easy-to-recognize bioindicator of artificially and permanently heated waters in moderate climate zones by a regular discharge of postindustrial water.

NDRG3-mediated lactate signaling in hypoxia

  • Hypoxia is associated with many pathological conditions as well as the normal physiology of metazoans. We identified a lactate-dependent signaling pathway in hypoxia, mediated by the oxygen- and lactate-regulated protein NDRG family member 3 (NDRG3). Oxygen negatively regulates NDRG3 expression at the protein level via the PHD2/VHL system, whereas lactate, produced in excess under prolonged hypoxia, blocks its proteasomal degradation by binding to NDRG3. We also found that the stabilized NDRG3 protein promotes angiogenesis and cell growth under hypoxia by activating the Raf-ERK pathway. Inhibiting cellular lactate production abolishes NDRG3-mediated hypoxia responses. The NDRG3-Raf-ERK axis therefore provides the genetic basis for lactate-induced hypoxia signaling, which can be exploited for the development of therapies targeting hypoxia-induced diseases in addition to advancing our understanding of the normal physiology of hypoxia responses.
