BIOLOGICAL RESPONSES OF OSTEOBLAST-LIKE CELLS TO DIFFERENT TITANIUM SURFACE BY ANODIZING MODIFICATION

  • Statement of problem. To improve a direct implant fixation to the bone, various strategies have been developed focusing on the surface of materials. The surface quality of the implant depends on the chemical, physical, mechanical and topographical properties of the surface. The different properties will interact with each other and a change in thickness of the oxide layer may also result in a change in surface energy, the surface topography and surface, chemical composition. However, there is limited the comprehensive study with regard to changed surface and biologic behavior of osteoblast by anodization. Purpose of study. The aim of this study was to analyze the characteristics of an oxide layer formed and to evaluate the cellular biologic behaviors on titanium by anodic oxidation (anodization) by cellular proliferation, differentiation, ECM formation and gene expression. And the phospholipase activity was measured on the anodized surface as preliminary study to understand how surface properties of Ti implant are transduced into downstream cellular events. Methods and Materials. The surface of a commercially pure titanium(Grade 2) was modified by anodic oxidation. The group 1 samples had a machined surface and other three experimental specimens were anodized under a constant voltage of 270 V(Group 2), 350 V(Group 3), and 450 V(Group 4). The specimen characteristics were inspected using the following five categories; the surface morphology, the surface roughness, the thickness of oxide layer, the crystallinity, and the chemical composition of the oxide layer. Cell numbers were taken as a marker for cell proliferation. While the expression of alkaline phosphatase and Runx2 (Cbfa1) was used as early differentiation marker for osteoblast. The type I collagen production was determined, which constitutes the main structural protein of the extracellular matrix. Phospholipase $A_2$ and D activity were detected. Results. (1) The anodized titanium had a porous oxide layer, and there was increase in both the size and number of pores with increasing anodizing voltage. (2) With increasing voltage, the surface roughness and thickness of the oxide film increased significantly (p<0.01), the $TiO_2$phase changed from anatase to rutile. During the anodic oxidization, Ca and P ions were more incorporated into the oxide layer. (3) The in vitro cell responses of the specimen were also dependant on the oxidation conditions. With increasing voltage, the ALP activity, type I collagen production, and Cbfa 1 gene expression increased significantly (p<0.01), while the cell proliferation decreased. (4) In preliminary study on the relation of surface property and phospholipase, PLD activity was increased but $PLA_2$ activity did not changed according to applied voltage. Conclusion. The anodized titanium shows improved surface characteristics than the machined titanium. The surface properties acquired by anodization appear to give rise more mature osteoblast characteristics and might result in increased bone growth, and contribute to the achievement of a tight fixation. The precise mechanism of surface property signaling is not known, may be related to phospholipase D.

Stability of Partial Nitrification and Microbial Population Dynamics in a Bioaugmented Membrane Bioreactor

  • Bioaugmentation of bioreactors focuses on the removal of numerous organics, with little attention typically paid to the maintenance of high and stable nitrite accumulation in partial nitrification. In this study, a bioaugmented membrane bioreactor (MBR) inoculated with enriched ammonia-oxidizing bacteria (AOB) was developed, and the effects of dissolved oxygen (DO) and temperature on the stability of partial nitrification and microbial community structure, in particular on the nitrifying community, were evaluated. The results showed that DO and temperature played the most important roles in the stability of partial nitrification in the bioaugmented MBR. The optimal operation conditions were found at 2-3 mgDO/l and $30^{\circ}C$, achieving 95% ammonia oxidization efficiency and nitrite ratio ($NO_2^-/{NO_x}^-$) of 0.95. High DO (5-6 mg/l) and low temperature ($20^{\circ}C$) had negative impacts on nitrite accumulation, leading to nitrite ratio drop to 0.6. However, the nitrite ratio achieved in the bioaugmented MBR was higher than that in most previous literatures. Denaturing gradient gel electrophoresis (DGGE) and fluorescence in situ hybridization (FISH) were used to provide an insight into the microbial community. It showed that Nitrosomonas-like species as the only detected AOB remained predominant in the bioaugmented MBR all the time, and coexisted with numerous heterotrophic bacteria. The heterotrophic bacteria responsible for mineralizing soluble microbial products (SMP) produced by nitrifiers belonged to the Cytophaga-Flavobacterium-Bacteroides (CFB) group, and $\alpha$-, $\beta$-, and $\gamma$- Proteobacteria. The fraction of AOB ranging from 77% to 54% was much higher than that of nitrite-oxidizing bacteria (0.4-0.9%), which might be the primary cause for the high and stable nitrite accumulation in the bioaugmented MBR.

Comparison of the Sonodegradation of Naphthalene and Phenol by the Change of Frequencies and Addition of Oxidants or Catalysts (주파수 변화 및 보조제 첨가에 따른 나프탈렌 및 페놀의 초음파 분해효율 비교)

  • The research seeks to find the optimal conditions for sonodegradation of naphthalene and phenol as exemplary organic pollutants to be subjected to ultrasound in varying frequencies (28 kHz, 580 kHz, and 1,000 kHz) and in the presence of different kinds of additive (T$TiO_2$, $H_2O_2$, $FeSO_4$, Zeolite, and Cu). In cases of both naphthalene and phenol, 580 kHz of ultrasound has proven to be the most effective among others at sonodegradation. Based on the observation that OH radicals are also produced in maximum under exposure of 580 kHz of ultrasound, we concluded that this frequency of ultrasound creates hospitable condition for the combined process of degradation by pyrolysis and oxidization. $FeSO_4's$ degradation rate and k1 value have increased by approximately 1.8 times compared with the results of the solutions without any additives. This seems to be the result of ultrasound reaction which, accompanied by Fenton's reaction, increased the oxidative degradation and the production of OH radicals. However, application of ultrasound and Fenton's reaction is limited to the batch type conditions, as its use in continuous system can cause loss of iron or decay of the cistern, thereby creating additional pollutants. When the additive is replaced with $TiO_2$, on the contrary, the rate of sonodegradation has increased up to 20% compared to when there was no additive. We therefore conclude that $TiO_2$ could prove to be an effective additive for ultrasound degradation in continuous treatment system.

Study of Corrosion Characteristics of Corroded Iron Objects from Underwater by Sulfides (해저 철제유물의 황화물에 의한 부식특성 연구)

  • This study is on the corrosion of iron objects caused by sulfides in undersea environment. The corrosion state of objects in seawater and their damage state after underwater and left in highly humid air were studied. The samples of this study were four iron objects which had been taken out from undersea mud layer located in Taean Mado, Chungcheongnamdo. SEM-EDS and XRD analyse on the objects to check whether they have sulfides or not. The result of analysis suggested that the major component of corrosion product generated in undersea deposit soil is sulfur(S) and iron sulfide(FeS) is formed as sulfide. However, there was no clear corrosion on the surface of objects which was exposed to sea water because of the impact of concretion which covered the surface. In order to check the damage status of iron objects after they had been taken out of sea water, exposure tests in high humidity environment and dehumidified environment were done on the corrosion products. The result of the test suggested that the oxidization of iron sulfide corrosion product makes iron sulfate ($FeSO_4$) and sulfuric acid ($H_2SO_4$) and they can cause secondary corrosion of iron objects. Therefore, it is believed that the iron sulfide corrosion product of iron objects taken out from underwater environment should be removed by all means and the keeping environment of the iron objects should also maintain dehumidified state.

The Study of Ag Thin Film of Suitable Anode for T-OLED: Focused on Nanotribology Methode (UV 처리에 의한 T-OLED용 산화전극에 적합한 Ag 박막연구: Nano-Mechanics 특성 분석을 중심으로)

  • The work function of Ag (silver) is too low (~4.3 eV) to be used as an electrode of T-OLED (Top Emission Organic Light Emitting Diode). To solve this weakness, researches used plasma-, UV-, or thermal treatment on Ag films in order to increase the work function (~5.0 eV). So, most of studies have focused only on the work function of various treated Ag films, but studies focusing on nanomechanical properties were very important to investigate the efficiency and life time of T-OLED etc. In this paper, we focused on the mechanical properties of the Ag and $AgO_x$ film. The Ag was deposited on a glass substrate with the thickness of 150 nm by using rf-magnetron sputter with the power was fixed at 100 W and working pressure was 3 mTorr. The deposited Ag film was UV treated by UV lamp for several minutes (0~9 min). We measured the sheet resistance and mechanical property of the deposited film. From the experimental result, there were some differences of the sheet resistance and surface hardness of Ag thin film between short time (0~3 min) and long time UV treatment. These result presumed that the induced stress was taken place by the surface oxidation after UV treatment.

Effect of oral administration of egg yolk on oxidation and esterification of hepatic fatty acid in rats (랫드에서 난황의 경구투여가 간 지방산의 산화, 에스터화에 미치는 영향)

  • The purpose of the present study was to investigate the mechanism of metabolic partitioning between oxidization and esterification of liver fatty acids synthesized and secreted by the liver from egg yolk-fed laboratory rats. Animals were divided into four groups and orally administered egg yolk daily for 30 days: CON (control group, 1.0 g of saline solution), T1 (1.0 g of pork belly oil), T2 (1.0 g of egg yolk), and T3 (1.0 g of pork belly oil and 1.0 g of egg yolk, alternatively each week). The accumulation rate of [14C]-labelled lipid in liver was lowest in T2 among all treatments (P<0.05). Phospholipid secretion was higher in T2 than other treatments (P<0.05). The triglycerol secretion was highest in T1 and higher in the order of CON, T3, and T2 (P<0.05). Metabolic partitioning rate of phospholipid from total glycerolipid was highest in T2, followed by T3, CON and T1 (P<0.05). The 14CO2 production from total glycerolipid was the highest in T2 and showed a high oxidation rate compared to CON, T1, and T3 (P<0.05). Metabolic partitioning of glycerolipid from the liver decreased in triglycerol of T2 compared to CON, T1, and T3, whereas phospholipids of T2 increased (P<0.05).

Mechanical Properties of The CO2 Free Vacuum Carburized in SCM415H (CO2 무 배출 침탄 열처리된 SCM415H 소재의 기계적 성질)

  • Vacuum carburizing is supposed to be the superior process to the gas carburizing. However, the vacuum carburizing has the stage in which hydrocarbon gas is supplied into the furnace to be pyrolysis, and consequently the stable heat treatment is hard to achieve due to the soot from the hydrocarbon pyrolysis. Recently, many studies have been made which utilize acetylene gas to overcome this defects. In this paper, the carburizing and the diffusion periods have been selected based on the Harris experimental formula, and the mechanical properties of the vacuum carburized specimen have been compared with those of the gas carburized SCM415H specimen to identify the feasibility of the $CO_2$ free vacuum carburizing process. The result showed that the vacuum carburized materials used have no oxidization of the grain boundaries, and show the 29.8% higher effective hardness depth and the acceptable tensile strength.

Characterization of CH4-oxidizing and N2O-reducing Bacterial Consortia Enriched from the Rhizospheres of Maize and Tall Fescue (옥수수와 톨페스큐 근권 유래의 메탄 산화 및 아산화질소 환원 세균 컨소시움 특성)

  • CH4-oxidizing and N2O-reducing bacterial consortia were enriched from the rhizosphere soils of maize (Zea mays) and tall fescue (Festuca arundinacea). Illumina MiSeq sequencing analysis was performed to comparatively analyze the bacterial communities of the consortia with those of the rhizosphere soils. Additionally, the effect of root exudate on CH4 oxidation and N2O reduction activities of the microbes was evaluated. Although the inoculum sources varied, the CH4-oxidizing and N2O-reducing consortia derived from maize and tall fescue were similar. The predominant methanotrophs in the CH4-oxidizing consortia were Methylosarcina, Methylococcus, and Methylocystis. Among the N2O-reducing consortia, the representative N2O-reducing bacteria were Cloacibacterium, Azonexus, and Klebsiella. The N2O reduction rate of the N2O-reducing consortium from maize rhizosphere and tall fescue rhizosphere increased by 1.6 and 2.7 times with the addition of maize and tall fescue root exudates, respectively. The CH4 oxidization activity of the CH4-oxidizing consortia did not increase with the addition of root exudates. The CH4-oxidizing and N2O-reducing consortia can be used as promising bioresources to mitigate non-CO2 greenhouse gas emissions during remediation of oil-contaminated soils.

Design and operating parameters of multi-functional floating island determined by basic experiments of unit processes (단위공정별 기초실험을 통한 다기능 융복합부도의 설계·운전인자 도출)

  • Water quality improvement processes for stagnant area consist mainly of technologies applying vegetation and artificial water circulation, and these existing technologies have some limits to handle pollution loads effectively. To improve the purification efficiency, eco-friendly technologies should be developed that can reinforce self-purification functions. In this study, a multi-functional floating island combined with physical chemical biological functions ((1) flotation and oxidization using microbubbles, (2) vegetation purification and (3) bio-filtration with improved adsorption capacity) has been developed and basic experiments were performed to determine the optimal combination conditions for each unit process. It has been shown that it is desirable to operate the microbubble unit process under conditions greater than $3.5kgf/cm^2$. In vegetation purification unit process, Yellow Iris (Iris pseudacorus) was suggested to be suitable considering water quality, landscape improvement and maintenance. When granular red-mud was applied to the bio-filtration unit process, it was found that T-P removal efficiency was good and its value was also stable for various linear velocity conditions. The appropriate thickness of filter media was suggested between 30 and 45 cm. In this study, the optimal design and operating parameters of the multi-functional floating island have been presented based on the results of the basic experiments of each unit process.

Complete genome sequence of Comamonas sp. NLF-7-7 isolated from biofilter of wastewater treatment plant (폐수처리장의 바이오 필터로부터 분리된 Comamonas sp. NLF-7-7 균주의 유전체 염기서열 해독)

  • Comamonas sp. NLF-7-7 was isolated from biofilter of wastewater treatment plant. The whole-genome sequence of Comamonas sp. NLF-7-7 was analyzed using the PacBio RS II and Illumina HiSeqXten platform. The genome comprises a 3,333,437 bp chromosome with a G + C content of 68.04%, 3,197 total genes, 9 rRNA genes, and 49 tRNA genes. This genome contained pollutants degradation and floc forming genes such as sulfur oxidization pathway (SoxY, SoxZ, SoxA, and SoxB) and floc forming pathway (EpsG, EpsE, EpsF, EpsG, EpsL, and glycosyltransferase), respectively. The Comamonas sp. NLF-7-7 can be used to the purification of wastewater.
