Comparative Study on Anti-thrombotic Effects of Gamihyunbooleekyungtang with Decoction Method (탕전기의 추출방법(抽出方法)에 따른 가미현부이경탕(加味玄附理經湯)의 항혈전(抗血栓) 효능(效能) 비교(比較) 연구(硏究))

  • Purpose: In this study, we investigated anti-thrombotic effects of Gamihyunbooleekyungtang currently used clinical treatment of dysmenorrhea and compared effects by decoction methods. Methods: Gamihyunbooleekyungtang was extracted by pressure extractor (GHYT) and nonpressure extractor(GHYT-1). Inhibitory effect of platelet cohesion, suppression effect of GPIIb/IIIa activity, inhibitory effect of $TXB_2$ and $PEG_2$ biosynthesis, and oxidative damage suppression effect were tested in vitro. Also suppression of pulmonary embolism was studied in vivo. Results: In this study, both GHYT and GHYT-1 extract showed a safety in cytotoxicity of hFCs. Both showed dose-dependent inhibitory effect on platelet coagulation. GHYT-1 extract is usually higher than GHYT, especially for $E_{max}$. GHYT and GHYT-1 extract showed dose-dependent inhibitory effect on GPIIb/IIIa activities. GHYT extract suppressed significantly $TXB_2$ biosynthesis at 1000, $500{\mu}g/m{\ell}$ and also suppressed significantly $PEG_2$ biosynthesis at $1000{\mu}g/m{\ell}$ as compared with the control group. GHYT-1 suppressed significantly $TXB_2$ biosynthesis at $1000{\mu}g/m{\ell}$ as compared with the control group. Also it suppressed pulmonary embolism triggered by collagen and epinephrine by respectively 50%, 75% as compared with the control group. Both GHYT and GHYT-1 extract showed dose-dependent decrease of oxidative damages caused by DPPH, whereas dose-dependent increase of superoxide dismutase like activity was observed. Conclusion: These results suggest that Gamihyunbooleekyungtang can be used for treating dysmenorrhea caused by thrombosis. Nonpressure decoction method is a little more effective, but more detailed studies would be needed.

Hepatoprotective Effects of GongJin-dan, on Ethanol-mediated Experimental Liver Damage in Rats

  • Background : A traditional Oriental medicine, GongJjn-dan (GJD), is one of the most well-known tonic agents in Korea. Among 6 types of GJD components, antler, red ginseng, and Cornus fructus have shown antioxidant effects, while EtOH-induced tissue damage may be a consequence of oxidative stress. Objectives & Methods : The hepatoprotective effects of GJD were evaluated on EtOH-mediated experimental liver damaged rats at 50, 100, 250 and 500mg/kg comparing with 100mg/kg of silymarin as a reference drug in the present study. Test substances were dosed once a day for 60 days with oral administration of 20% ethanol 2.5ml/100g body weight twice a day (equivalent to 7.9g ethanol/kg/day). Each of 8 rats per group was selected using body weight at 10 days after acclimatization. Experimental animals were sacrificed after 60 days of continuous oral treatment of test substances with 20% ethanol treatment, and changes on the body weight, liver weight, and serum AST and ALT were observed. Results : There were dramatic decreases of body weight and increases of liver weight and serum AST and ALT. Similar inhibition effects on the EtOH-induced hepatic damages were detected between equal dosages of GJD and silymarin. Conclusion : Based on these results. it is concluded that GJD showed clear hepatoprotective effects on EtOH-induced hepatic damage.

The Effects of Jihwangyeumja and GamiJihwangyeumja water extract on The Cultured Primary Hippocampal Cell Damaged by XO/HX (지황음자와 가미지황음자 유출액이 XO/HX로 손상된 배양 해마신경세포에 미치는 효과)

  • The purpose of this study is to examine the toxic effects caused by xanthine oxidase/hypoxanthine(XO/HX) and the effects of herbal extracts such as JHYJ and GJHYJ on the treatment of the toxic effects. For this purpose, experiments with the cultured hippocampal cells from new born mice were done. The results of these experiments were as follows. 1. XO/HX, a oxygen radical-generating system, decreased the survival rates of the cultured cells on XTT assay and INT assay, the amount of DNA syntheses, and the amount of neurofilaments, and increased the lipid peroxidation. 2. JHYJ and GJHYJ have the efficacy of increasing the survival rates of the cultured cells. 3. JHYJ and GJHYJ have the efficacy of increasing the amount of neurofilaments and of decreasing the lipid peroxidation. 4. JHYJ and GJHYJ have the efficacy of increasing the amount of DNA syntheses. From the above results, it is suggested that Jihwangyeumja and Gamijihwangyeumja have marked efficacy as a treatment for the damages caused by the XO/HX-mediated oxidative stress. And Jihwangyeumja and Gamijihwangyeumja are thought to have certain pharmacological effects. Further dinical study of this pharmacological effects of Jihwangyeumja and Gamijihwangyeumja should be complemented.

  • The purpose of this study is to examine the toxic effects caused by glucose oxidase (GO) and the effects of Hyungbangjihwangtang (HJT) water extracts on the treatment of the toxic effects. For this purpose, experiments with the cultured hippocampal cells from new born mice were done. The results of these experiments were as follows. 1. GO decreased the survival rate of the cultured cells on MTT assay and NR assay. 2. HJT has the efficacy of decreasing the lipid peroxidation. 3. HJT has the efficacy of increasing the amount of neurofilaments. 4. HJT has the efficacy of decreasing the activation of protein kinase C(PKC). From the above results, it is concluded that HJT has marked efficacy as a treatment for the damages caused by the GO-mediated oxidative stress. Further clinical study of this pharmacological effects of H]T should be completed.

For this purpose, experiments with the cultured nerve cells from the spinal motor neurons of new born mice were done. The results of these experiments were as follows. XO/HX, a oxygen radical-generating system, decreased the survival rate of the cultured cells on NR assay. MTT assay, the amount of neurofilaments and increased the amount of total proteinand increased the lipid peroxidation and the amount of LDH JST has the efficacy of increasing the amount of neurofilaments and total protein, and decreasing the lipid peroxidation and the amount of LDH, GJST has efficacy of increasing the amount of neurofilaments and total protein, and decreasing lipid peroxidation and the amount of LDH. From the above results, it is concluded that JST and GJST have marked efficacy as a treatment for the damages caused in the XO/HX mediated oxidative stress. And JST and GJST are thought to have certain pharmacologicall effects. Further clinical study of this pharmacological effects of JST and GJST should be complemented.

  • As the average life span have been lengthened and the rate of senile population have been raised, chronic degenerative diseases incident to aging has been increased rapidly and become a social problem. With this social background, recently, the facts that oxygen radicals(OR) have toxic effects on Central Nervous System and Peripheral Nervous System and cause neuropathy such as Parkinson's Disease, Alzheimer Disease have been turned out, and accordingly lots of studies on the mechanism of the toxic effects of OR on nerves, the diseases caused by OR and the approaches to curing the diseases have been made. The purpose of this study is to examine the toxic effects caused by Glucose Oxidase(GO) and the effects of herbal extracts such as Chilbokyeum(CBY), Chilbokyeumga Acori Rhizoma(CAR), Acori Rhizoma(AR) on the treatment of the toxic effects. For this purpose, experiments with the cultured cell from the cerebrums of new born mice were done. The results of these experiments were as follows. 1. GO, a oxygen radical, decreased the survival rate of the cultured cells on NR assay, MTT assay and amount of neurofilaments and increased the amount of total protein, lipid peroxidation and the amount of LDH. 2. CBY have efficacy of increasing the amount of neurofilaments and total protein and decreasing lipid peroxidation and the amount of LDH. 3. CAR have efficacy of increasing the amount of neurofilaments and total protein and decreasing lipid peroxidation and the amount of LDH. 4. AR have efficacy of increasing the amount of neurofilaments and total protein. From the above results, It is concluded that Chilbokyeumgamibang has marked efficacy as a treatment for the damages caused in the GO-mediated oxidative process. And Chilbokyeumgamibang is thought to have certain pharmacological effects on controlling over aging and treating Dementia. Further clinical study of this pharmacological effects of Chilbokyeumgamibang should be complemented.

Protective Effects of Dohongsamul-tang on Zinc-mediated Cytotoxicity in H9c2 Cardiomyoblast Cells (산화적 손상에 의해 유발된 심근세포 독성에 대한 도홍사물탕의 방어효과)

  • The water extract of Dohongsamul-tang(DHSMT)has been traditionally used for treatment of ischemic heart in oriental medicine. However, little is known about the mechanism by which the water extract of DHSMT rescues cells from these damages. Therefore, this study was designed to evaluate the protective effects of DHSMT on zinc-mediated cytotoxicity in H9c2 cardiomyoblast cells. This study demonstrates that treatment of H9c2 cells with zinc caused a decrease in cell viability in a dose dependent manner and a chromatin condensation. Zinc induced the cleavage of poly(ADP-ribose) polymerase (PARP). In addition, zinc induced the decrease of Bcl-2, as well as increase of Bak expression and mitochondrial dysfunction. Zinc-induced H9c2 cell death was remarkably prevented by the pretreatment of DHSMT with consistent suppression of the cleavage of poly(ADP-ribose) polymerase (PARP), mitochondrial dysfunction and the expression of Bak and Bcl-2. Taken together, the results suggest that zinc induced severe cell death in H9c2 cardiomyoblast cells via intracellular GSH(reduced glutathione) depletion and the protective effects of DHSMT against oxidative injuries may be achieved through modulation of mitochondrial dysfunction and scavenging of ROS(reactive oxygen species).

  • Cells express several antioxidant enzymes to scavenge reactive oxygen species (ROS) responsible for oxidative damages and various human diseases. Therefore, antioxidant enzymes are considered biomedicine candidates. Among them, extracellular superoxide dismutase (SOD3) had showed prominent efficacy against asthma and inflammation. Despite its advantages as a biomedicine, the difficulty in obtaining large quantity of active recombinant human SOD3 (rhSOD3) has limited its clinical applications. We found that a significant fraction of over-expressed rhSOD3 was composed of the inactive apo-enzyme and its potency against inflammation depended on the rate of metal incorporation. Also, purified rhSOD3 was unstable and lost its activity very quickly. Here, we suggest an ideal preparative method to express, purify, and store highly active rhSOD3. The enzymatic activity of rhSOD3 was maximized by incorporating metal ions into rhSOD3 after purification. Also, albumin or polyethylene glycol prevented rapid inactivation or degradation of rhSOD3 during preparative procedures and long-term storage.

Antioxidant Activity and Contents of Bioactive Components in Polar Microalgae (극지미세조류의 유용성분 함량 및 항산화 활성)

  • In this study, bioactive components such as polyohenols, flavonoids and tocopherols were determined in cultured polar microalgae (Fragilariopsis pseudonana, Chaetoceros neogracile, Stellarima microtrias, Porosiara pseudodenticular). Antioxidant activity of methanol extracts of polar microalgae was also investigated. ${\alpha}-Tocopherol$ contents in Fragilariopsis pseudonana were almost two times higher than those of Chaetoceros neogracile in. The antioxidant activity of methanol extracts of Fragilariopsis pseudonana methanol extracts determined by ABTS assay was higher than other algae. Total polyphenol contents of methanol extracts also showed a similar trend as antioxidant activity. The protective activity against oxidative damages induced by glutamate in PC 12 cells was shown in only Chaetoceros neogracile.

Various Fatty Acids Induce Cell Damages Differently in CYP2E1-transduced HepG2 Cells, E47 Cells, Compared to C34 Cells

  • The differential effects of various fatty acids such as n-3 and n-6 types or degrees of unsaturation on the CYP2E1 induction and the production of lipid peroxidation (LPO) were investigated. The CYP2E1-transduced human hepatoma HepG2 cells (E47) were cultured in RPMI 1640 media containing different concentrations of various fatty acids up to 48 h incubation compared to 04 cells and CYP2E1-null cells. Treated fatty acids were linoleic acid (LA:n-6, C18:2), arachidonic acid (AA:n-6, C20:4) and docosahexaenoic acid (DHA:n-3, C22:6). The cell survival rate was decreased corresponding to the degree of unsaturation (LA>AA $\cong$DHA) and to LPO production in E47 and 04 cells. The four or five unsaturation degree of fatty acids, AA and DHA, caused time- and dose-dependent cell death in E47 cells but not as much as in C34 (without CYP2E1), suggesting an important role of CYP2E1 in the DHA mediated damage. In the levels of lipid peroxides (LPO), AA also elevated LPO by 3- and 5- fold compared to DHA or LA treated E47 cells. However, AA did not increase LPO until 48 h incubation in C34 cells. In conclusion, the polyunsaturated fatty acids induced CYP2E1 induction might be changed by the elevated levels of lipid peroxide (LPO) and oxidative stress through the connection of CYP2E1 and degrees of unsaturated fatty acids.
