Effects of herbal-aupuncture solution from vitis labrusca root on LPO, RNS and cytochrome P450 (포도근 약침액이 LPO, RNS 및 Cytochrome P450에 미치는 효과)

  • Objectives : In this study, we determinated protective effects of Vitis labrusca Root herbal-acupuncture solution (VLHA) against atherosclerosis and cardiovascular disease induced by oxidative damage and cytochrome P450 (CYP) induction. Methods : Antioxidant capacity of VLHA were evaluated by the ${\beta}-carotene/linoleic$ acid emulsion system and FeCl2-ascorbic acid stimulted lipid peroxidation in rat liver homogenate, respectively. Scavenging effects of VLHA on ONOO- and NO were measured by DHR 123, DAF-2 assay. Furthermore, we measured inhibitory activity of VLHA on CYP 2E1 in rat liver microsome using by Aniline assay. Results : VLHA exhibited potent inhibition rate(74%) on FeCl2-ascorbic acid induced lipid peroxidation, and revealed a strong scavenging effects on ONOO- and NO. In addition, VLHA showed inhibitory effect on CYP 2E1. Conclusions : The present study concludes that our results suggest that VLHA could be used as a good source in the prevention of atherosclerosis and alcoholic liver injury.

Removal of Contaminants Deposited on Surfaces of Matrices by Using Low-Temperature Argon Plasma Treatment (저온 아르곤 플라즈마처리를 이용한 모재 표면의 오염물 제거)

  • The possibility of a low-temperature argon plasma treatment as a mean of restoration technology for contaminated invaluable archive materials and artefacts, and evidencing documents was investigated along with an oxygen plasma treatment for comparison. For this purpose, the degree of color changes, ${\Delta}E^*ab$, and surface morphological changes due to plasma treatments as an evaluation of removal performance of artificial contaminants such as brilliant green dye and carbon deposit on cellulose acetate and plain paper as matrices, respectively, were measured and analyzed using a spectrophotometer and a field emission scanning electron microscope. Compared to the argon plasma treatment with sputtering characteristic, that of the oxygen plasma with characteristic of an oxidation reaction has shown superior results in removing the contaminants; the oxygen plasma has proven to damage the matrices significantly due to its oxidative characteristic, and post-plasma reactions has anticipated to be also detrimental to the surfaces, whereas, the problems caused by the counterpart has resulted in being negligible and rather has thought to be an appropriate mean for delicate restoration and/or removal operations of contaminants.

Protective Effects of the Nuclear Factor Kappa B Inhibitor Pyrrolidine Dithiocarbamate on Experimental Testicular Torsion and Detorsion Injury

  • Testicular torsion results with the damage of the testis and it is a surgical emergency. Pyrrolidine dithiocarbamate (PDTC) is a low-molecular-weight antioxidant and potent inhibitor of nuclear factor kappa B (NF-${\kappa}B$) activation. In this study, we aimed to investigate the effects of PDTC to testicular torsion-detorsion (T/D) injury. Forty adult male Sprague-Dawley rats were separated into four groups. A sham operation was performed in group I. In group II, torsion is performed 2 hours by 720 degree extravaginally testis. In group III, 4 h reperfusion of the testis was performed after 2 h of testicular torsion. In group IV, after performing the same surgical procedures as in group III, PDTC (100 mg/kg, intravenous's) was administered before 30 min of detorsion. The testes tissue malondialdehyde (MDA), superoxide dismutase (SOD) catalase (CAT) level was evaluated. Histological evaluations were performed after hematoxylin and eosin staining. Testicular tissue MDA levels were the highest in the T/D groups compared with treatment group. Administration of PDTC prevented a further increase in MDA levels. Significant decrease occurred in CAT and SOD levels in treatment group compared with the control group. The rats in the treatment group had normal testicular architecture. The results suggest that PDTC can be a potential protective agent for preventing the biochemical and histological changes related to oxidative stress in testicular injury caused by testis torsion.

Scavenging Effects of Ginkgo biloba Extract on Paraquat Induced Toxicity (Paraquat 유도독성에 대한 Ginkgo biloba Extract의 독성경감효과(I))

  • Reactive oxygen species (ROS) are highly reactive molecules due to their unpaired electron. They have been suspected as one of the major tissue damage inducers in biological metabolic systems. Antioxidant enzymes, such as catalase and superoxide dismutase, could not repair all the oxidative damages resulting from those excessive toxic ROS. It is, therefore, urgent to develop effective antioxidants to relieve from the oxidatire damages. In this study antioxidative effects were investigated by using two flavonoids such as quercetin and naringenin and a flavonoid-rich extract, Ginkgo biloba extract in combination with paraquat that is known as a strong generator of oxygen radicals. The results are summeringed as follows: 1. To assess radical scavenging ability reduction concentrations (IC$_{50}$) of 1,1-diphenyl-2-picrylhydrazine (DPPH) within 15 minutes were measured. The values of the IC$_{50}$ of quercetin and Ginkgo biloba extract were 15.4 $\mu$M and 13.2$\mu$g/ml, respectively. Their radical removing activities showed concentration-dependent manners. 2. In the hydrogen peroxide assay by using PMS-NADH system, quercetin, naringenin and Ginkgo biloba extract led to removing hydrogen peroxide in concentrationdependent manner whose removing abilities at 100$\mu$M or 100 $\mu$g/ml were 75.6, 25.8 and 26.0%, respectively. 3. In the hydrogen peroxide-induced rat blood hemolysis assay all three compounds led to similar effects whose hemolysis inhibition ratios at 100$\mu$M or 100$\mu$g/ml were 68.0, 5.14 and 55.8%, respectively. 4. In the xanthinee oxidase assay by measuring degree of NADH oxidation in the presence of hypoxanthine and xanthinee oxidase, both quercetin and Ginkgo biloba extract showed excellent activities showing 42.8 and 24.2% inhibiting xanthine oxidase activity at 100$\mu$M or 100$\mu$g/ml concentrations, respectively.

Causes of age-related sarcopenia and frailty: the role of exercise and nutrition for prevention (노화 관련 근감소증과 노쇠함의 원인과 예방을 위한 운동과 영양의 역할)

  • The purpose of this study is not only to define and cause of sarcopenia and frailty due to aging, but also to explore prevention and delay through regular exercise and right nutrition. Sarcopenia and frailty are known to cause frailty, cachexia, osteoporosis, metabolic syndrome and early death due to decreased muscle mass and muscle function caused by aging. The two disease were related to nervous system damage and lack of protein synthesis within the muscles due to the increase in the amount of oxidative stress, and inadequate nutrition and lack of physical activity. It also emphasizes the role and importance of right nutrition and physical activity (such as aerobic exercise, resistance movement, etc) to prevent and improve such syndromes. Conclusions, it is in proposing the supply of high-quality protein, including regular exercise, as the most beneficial way to prevent and overcome aging-related sarcopenia and frailty.

Effect of Black Ginseng on Memory Improvement in the Amnesic Mice Induced by Scopolamine

  • This study compared the effects of black, white, and red ginseng extracts (WGE, RGE, BGE, 200 mg/kg, p.o.) on learning and memory deficits associated with scopolamine treatment (SCOP, 2 mg/kg, i.p.). Tacrine (THA, 10 mg/kg, p.o.) was used as a positive control. Ginseng significantly reversed SCOP-induced memory impairment in the passiveavoidance test and also reduced escape latency in training trials of the Morris water maze test. The increased acetylcholinesterase (AChE) activity produced by SCOP was significantly inhibited by WGE and RGE (p<0.001). SCOP administration had no effect on choline acetyltransferase (ChAT) activity, but RGE and BGE significantly increased ChAT activity (p<0.05). SCOP administration increased oxidative damage in the brain. Treatment of amnesic mice with ginseng extracts decreased malondialdehyde (MDA) levels and restored superoxide dismutase (SOD) and catalase (CAT) activity to control levels. These results suggest that black ginseng enhances cognitive activity by regulation of cholinergic enzymes and antioxidant systems.

Korean red ginseng extract alleviates advanced glycation end product-mediated renal injury

  • The effect of Korean red ginseng (KRG) on diabetic renal damage was investigated using streptozotocin (STZ)-induced diabetic rats. The diabetic rats showed loss of body weight gain, and increases in kidney weight and urine volume, whereas the oral administration of KRG at a dose of 100 or 250 mg/kg of body weight per day for 28 d prevented these diabetes-induced physiological abnormalities. Among the kidney function parameters, elevated plasma levels of urea nitrogen and creatinine in diabetic control rats tended to be lowered in KRG-treated rats. In addition, administration of KRG at a dose of 100 mg/kg body weight in the diabetic rats showed significant decreases in serum glucose and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), implying that KRG might prevent the pathogenesis of diabetic complications caused by impaired glucose metabolism and oxidative stress. KRG also significantly reduced advanced glycation end product (AGE) formation and secretion from kidney of diabetic rats. Furthermore, KRG decreased the levels of N-(carboxymethyl) lysine and expression of AGE receptor. KRG also reduced the overexpression of cyclooxygenase-2 and inducible nitric oxide synthase in the kidney via deactivation of nuclear factor-kappa B. We also found that KRG prevented STZ-induced destruction of glomerular structure and significantly suppressed high glucose-induced fibronectin production. Taken together, KRG ameliorates abnormalities associated with diabetic nephropathy through suppression of inflammatory pathways activated by TNF-${\alpha}$ and AGEs. These findings indicate that KRG has a beneficial effect on pathological conditions associated with diabetic nephropathy.

The Scavenging Effect on Nitric Oxide in Ursi Fel ${\cdot}$ Bovis Calculus Herbal-acupuncture Solution (웅담합우황(熊膽合牛黃) 약침액(藥鍼液)의 Nitric Oxide에 대한 억제효과(抑制效果))

  • Oxidative damage to DNA, proteins, lipids and other molecules may contribute to the development of neurodegenerative disease. This study was designed to find out whether Ursi Fel ${\cdot}$ Bovis Calculus Herbal-Acupuncture Solution(UBHAS) can scavenge nitric oxide(NO) or not. NO concentration was estimated after treatment with Vit. C or 1, 10, $100{\mu}g/m{\ell}$ of UBHAS. There was no significant scavenging effect of UBHAS on NO in 1, $100{\mu}g/m{\ell}$ group. while significant scavenging in $l0{\mu}g/m{\ell}$ group after 2hrs. UBHAS showed significant scavenging effects on NO in 1, 10, $100{\mu}g/m{\ell}$ group after 6hrs. There was a significant scavenging effect in 10, $100{\mu}g/m{\ell}$ group. even though no significant in $1{\mu}g/m{\ell}$ group after 12, 24 hrs. This study showed that UBHAS might be used for aging related disease. These results suggest that UBHAS has scavenging effect on NO. however, further studies are required to investigate the antioxidative effects of it.

Formation of DNA-Protein Crosslink at Oxidized Abasic Site Mediated by Human DNA Polymerase Iota and Mitochondrial DNA Polymerase Gamma

  • Human genomic DNA is continuously attacked by oxygen radicals originated from cellular metabolic processes and numerous environmental carcinogens. 2-deoxyribonolactone (dL) is a major type of oxidized abasic (AP) lesion implicated in DNA strand scission, mutagenesis, and formation of covalent DNA-protein crosslink (DPC) with DNA polymerase (Pol) ${\beta}$. We show here that human DNA polymerase (Pol)${\iota}$ and mitochondrial $Pol{\gamma}$ give rise to stable DNA-protein crosslink (DPC) formation that is specifically mediated by dL lesion. $Pol{\gamma}$ mediates DPC formation at the incised dL residue by its 5'-deoxyribose-5-phosphate (dRP) lyase activity, while $Pol{\gamma}$ cross links with dL thorough its intrinsic dRP lyase and AP lyase activities. Reactivity in forming dL-mediated DPC was significantly higher with $Pol{\gamma}$ than with $Pol{\iota}$. DPC formation by $Pol{\gamma}$, however, can be reduced by an accessory factor of $Pol{\gamma}$ holoenzyme that may attenuate deleterious effects of crosslink adducts on mitochondrial DNA. Comparative kinetic analysis of DPC formation showed that the rate of DPC formation with either $Pol{\iota}$ or $Pol{\gamma}$ was lower than that with $Pol{\beta}$. These results revealed that the activity of catalytic lyase in DNA polymerases determine the efficiency of DPC formation with dL damages. Irreversible crosslink formation of such DNA polymerases by dL lesions may result in a prolonged strand scission and a suicide of DNA repair proteins, both of which could pose a threat to the genetic and structural integrity of DNA.

Effects of the Antioxidative Components to Ginsenoside in the Liver of 40-week-old Mice (40주령의 생쥐 간에 미치는 ginsenoside의 항산화효과)

  • To study on antioxidant effects of saponin fractions, we investigated effects in the liver of 40-week-old mice to which were pretreated with 5 mg/kg per body weight of saponins for 5 days. The ability of saponins to protect against oxidative damage to the mouse liver was examined by determining the level malondialdehyde (MDA),hydrogen peroxide and the activity of superoxide dismutase (SOD) and catalase (CAT). The only panaxadiol (PD)among the ginseng saponin fractions significantly increased the hepatic SOD activities (p<0.01), Whereas PD, panaxatriol (PT), ginsenoside Rd (G-Rd) (p<0.01) and ginsenoside Re (G-Re) (p<0.05) significantly decreased the contents of hydrogen peroxide. It was only G-Rd that significantly increased CAT activities (p<0.05). The level of MDA was significantly decreased by G-Rd and PD. In conclusion, PD and G-Rd among the saponin fractions were especially increased in the activity of hepatic antioxidative enzyme and decreased the lipid peroxidation that was expressed in term of MDA formation.
