Characterization of Cysteine Residues in Cabbage Phospholipase D by Sulfhydryl Group Modifying Chemicals (설프히드릴 변형 화합물질들에 의한 양배추 포스포리파제 D의 시스테인 잔기의 특성)

  • SH group modifying chemicals were used to characterize the eight cysteine residues of cabbage PLD. 5,5-dithiobis(2-nitrobenzoate)(DTNB) was used to titrate the SH group of cysteine residues . Based on the optical density at 412nm due to the reduced DTNB, 4 SH groups are found to be present in a native PLD while 8 SH groups in the denatured PLD whose tertiary structure was perturbed by 8M urea. The results imply that among the 8 cysteine residues of PLD, the half(4) are exposed on the surface whereas the other half are present at the interior of the enzyme tertiary structure. The PLD was inactivated by SH modifying reagents such as p-chloromercuribenzoate(PCMB), iodoacetate, iodoacetamide, and N-ethylmaleimide. At the addition of dithiothreitol(DTT) only the PCMB inhibited PLD activity was recovered reversibly. The micro-environment of the exposed SH group of cysteine residues was examined with various disulfide compounds with different functional groups and we found that anionic or neutral disulfides appear to be more effective than the positively charged cystamine for inactivating the PLD activity. The effect of redox state of cysteine residues on the PLD activity was further explored with H2O2. The oxidation of SH groups by H2O2 inhibited the PLD activity more than 70%, which was mostly recovered by DTT. From these results, we could confirm chemically that all the cysteine residues of PLD are present as in their reduced SH forms and the 4 SH groups exposed on the surface of the enzyme may play important roles in the regulation of PLD activity.

  • Because of the important role LD converters play in the production of high quality steel, various dynamic models have been attempted in the past by many researchers not only to understand the complex chemical reactions that take place in the converter process but also to assist the converter operation itself using computers. And yet no single dynamic model was found to be completely satisfactory because of the complexity involved with the process. The process indeed involves dynamic energy and mass balances at high temperatures accompanied by complex chemical reactions and transport phenomena in the molten state. In the present study, a mathematical model describing the dynamic behavior of LD converter process has been developed. The dynamic model describes the time behavior of the temperature and the concentrations of chemical species in the hot metal bath and slag. The analysis was greatly facilitated by dividing the entire process into three zones according to the physical boundaries and reaction mechanisms. These three zones were hot metal (zone 1), slag (zone 2) and emulsion (zone 3) zones. The removal rate of Si, C, Mn and P and the rate of Fe oxidation in the hot metal bath, and the change of composition in the slag were obtained as functions of time, operating conditions and kinetic parameters. The temperature behavior in the metal bath and the slag was also obtained by considering the heat transfer between the mixing and the slag zones and the heat generated from chemical reactions involving oxygen blowing. To identify the unknown parameters in the equations and simulate the dynamic model, Hooke and Jeeves parttern search and Runge-Kutta integration algorithm were used. By testing and fitting the model with the data obtained from the operation of POSCO #2 steelmaking plant, the dynamic model was able to predict the characteristics of the main components in the LD converter. It was possible to predict the optimum CO gas recovery by computer simulation

Effect of Ce Addition on Catalytic Activity of Cu/Mn Catalysts for Water Gas Shift Reaction (수성가스전이반응(Water Gas Shift Reaction)을 위한 Ce 첨가에 따른 Cu/Mn 촉매의 활성 연구)

  • Cu/Mn/Ce catalysts for water gas shift (WGS) reaction were synthesized by urea-nitrate combustion method with the fixed molar ratio of Cu/Mn as 1:4 and 1:1 with the doping concentration of Ce from 0.3 to 0.8 mol%. The prepared catalysts were characterized with SEM, BET, XRD, XPS, $H_2$-TPR, $CO_2$ TPD, $N_2O$ chemisorption analysis. The catalytic activity tests were carried out at a GHSV of $28,000h^{-1}$ and a temperature range of 200 to $400^{\circ}C$. The Cu/Mn(CM) catalysts formed Cu-Mn mixed oxide of spinel structure ($Cu_{1.5}Mn_{1.5}O_4$) and manganese oxides ($MnO_x$). However, when a small amount of Ce was doped, the growth of $Cu_{1.5}Mn_{1.5}O_4$ was inhibited and the degree of Cu dispersion were increased. Also, the doping of Ce on the CM catalyst reduced the reduction temperature and the base site to induce the active site of the catalyst to be exposed on the catalyst surface. From the XPS analysis, it was confirmed that maintaining the oxidation state of Cu appropriately was a main factor in the WGS reaction. Consequently, Ce as support and dopant in the water gas shift reaction catalysts exhibited the enhanced catalytic activities on CM catalysts. We found that proper amount of Ce by preparing catalysts with different Cu/Mn ratios.

Hygienic Consideration on the Quality Change of Perilla Oil (들기름의 품질변화에 대한 위생적 고찰)

  • Perilla oil was measured on hygiene state and quality change for oil press condition. All sample .was commercially salted perilla oil, and was tested standard items. The result showed violative rate of $23.1\%$, and violative items were acid value and iodine value. Relationship between D.B.I. and iodine value was 0.78, so that unsuitability of iodine value should be caused of oxidation factor. But acid value was not relationship comparatively. The quality change appeared very small by roasting conditions, but quality of perilla seed gaye many influence on quality of oil. Therefore use of fresh perilla seed is a matter of great importance to quality of perilla oil. Perilla oil was demanded many attention on Quality management for stage and sold period because of high possibility of quality change.

Safety Evaluation of Water-soluble Chromium in Makeup Products (메이크업 화장품에서 수용성 크롬의 안전성 평가)

  • There is now a growing concern about the possible toxicity of heavy metals in cosmetics. Heavy metals can be used as cosmetic ingredients or may be present as low level impurities in some of the raw materials. Chromium derivatives are used as pigments in cosmetics. Chromium is essential and toxic trace elements. Chromium may cause skin allergy. However, the regulations related to cosmetic products give no limit values for Chromium. Hexavalent chromium is significantly more toxic than trivalent chromium. Hexavalent chromium may present a carcinogenic risk at high concentrations. Therefore, it is important to consider oxidation state of chromium when analyze chromium. The purpose of this study is to determine the concentrations of water-soluble trivalent and hexavalent chromium in samples of makeup products, and to assess the safety of cosmetics on the basis of animal sensitization tests using guinea pig. The present study of chromium in 48 makeup products of 12 manufacturers provides a basis for assessing safety of makeup products. Water-soluble hexavalent chromium was not detected in any product. Water-soluble trivalent chromium was detected in only 9 eye shadows out of 48 makeup products, and could not be quantified 3 out of 9 eye shadows. The highest level of water-soluble trivalent chromium was about 10 mg/kg in spite of 90,000 mg/kg of total chromium. The results of animal sensitization tests show that 200 mg/kg of trivalent chromium and 5 mg/kg of hexavalent chromium have no harmful effect. No cross-reaction among these metals was found. Accordingly, the concentrations of water-soluble chromium in makeup products seemed to be safe. The overall results indicate that chromium in cosmetics probably have no significant toxicological effects. However, It is necessary to set guidelines on the maximum permissible concentration of water-soluble chromium in cosmetics.

  • The thermal behavior of NiFe$_2$O$_4$ prepared by a solid-state reaction was investigated for H$_2$ generation by the thermochemical cycle. The reduction of NiFe$_2$O$_4$ started from 800 $^{\circ}C$, and the weight loss was 0.2-0.3 wt% up to 1000 $^{\circ}C$. At this reaction, NiFe$_2$O$_4$ was reduced by release of oxygen bonded with the Fe$^3$ion in the B site of NiFe$_2$O$_4$. In the $H_2O$ decomposition reaction, H$_2$ was generated by oxidation of reduced NiFe$_2$O$_4$. The crystal structure of NiFe$_2$O$_4$ for redox reaction maintained spinel structure. Then, NiFe$_2$O$_4$ is excellent material in the thermochemical cyclic reaction due to release oxygen at low temperature for the reduction reaction and produce H$_2$ maintaining crystal structure for redox reaction.

Neuronal injury in AIDS dementia: Potential treatment with NMDA open-channel blockers and nitric oxide-related species

  • The neurological manifestations of AIDS include dementia, encountered even in the absence of opportunistic superinfection or malignancy. The AIDS Dementia Complex appears to be associated with several neuropathological abnormalities, including astrogliosis and neuronal injury or loss. How can HIV-1 result in neuronal damage if neurons themselves are only rarely, if ever, infected by the vitus\ulcorner In vitro experiments from several different laboratiories have lent support to the existence of HIV- and immune-related toxins. In one recently defined pathway to neuronal injury, HIV-infected macrophages/microglia as well as macrophages activated by HIV-1 envelope protein gp120 appear to secrete excitants/neurotoxins. These substances may include arachidonic acid, platelet-activating factor, free radicals (NO - and O$_2$), glutamate, quinolinate, cysteine, cytokines (TNF-${\alpha}$, IL1-B, IL-6), and as yet unidentified factors emanating from stimulated macrophages and possibly reactive astrocytes. A final common pathway for newonal suscepubility appears to be operative, similar to that observed in stroke, trauma, epilepsy, and several neurodegenerative diseases, including Huntington's disease, Parkinson's disease, and amyotrophic lateral sclerosis. This mechanism involves excessive activation of N-methyl-D-aspartate (NMDA) receptor-operated channels, with resultant excessive influx of Ca$\^$2+/ leading to neuronal damage, and thus offers hope for future pharmacological intervention. This chapter reviews two clinically-tolerated NMDA antagonists, memantine and nitroglycerin; (ⅰ) Memantine is an open-channel blocker of the NMDA-associated ion channel and a close congener of the anti-viral and anti-parkinsonian drug amantadine. Memantine blocks the effects of escalating levels of excitotoxins to a greater degree than lower (piysiological) levels of these excitatory amino acids, thus sparing to some extent normal neuronal function. (ⅱ) Niuoglycerin acts at a redox modulatory site of the NMDA receptor/complex to downregulate its activity. The neuroprotective action of nitroglycerin at this site is mediated by n chemical species related to nitric oxide, but in a higher oxidation state, resulting in transfer of an NO group to a critical cysteine on the NMDA receptor. Because of the clinical safety of these drugs, they have the potential for trials in humans. As the structural basis for redox modulation is further elucidated, it may become possible to design even better redox reactive reagents of chinical value. To this end, redox modulatory sites of NMDA receptors have begun to be characterized at a molecular level using site-directed mutagenesis of recombinant subunits (NMDAR1, NMDAR2A-D). Two types of redox modulation can be distinguished. The first type gives rise to a persistent change in the functional activity of the receptor, and we have identified two cysteine residues on the NMDARI subunit (#744 and #798) that are responsible for this action. A second site, presumably also a cysteine(s) because <1 mM N-ethylmaleimide can block its effect in native neurons, underlies the other, more transient redox action. It appears to be at this, as yet unidentified, site on the NMDA receptor that the NO group acts, at least in recombinant receptors.

Reduction of Hexavalent Chromium on PVC Filters (혜산산업보건학술상 수상논문 - PVC 여과지에 채취된 6가 크롬의 환원)

  • Chromium exists at various valences, including elemental, trivalent, and hexavalent chromium, and undergoes reduction-oxidation reactions in the environment. Since hexavalent chromium is known as a human carcinogen, it is most important to evaluate the oxidationreduction characteristics of the hexavalent chromium species. Although hexavalent chromium can be reduced to trivalent state, the detailed information on this in workplace environments is limited. The purpose of this study was to investigate hexavalent chromium reduction in time in various conditions. A pilot chrome plating operation was prepared and operated in a laboratory for this study. There was evidence that the hexavalent chromium was reduced by time after mist generation. The percentage ratio (with 95% confidence intervals in parentheses) of hexavalent chromium to total chromium was almost 100% (99.1 ; 102.3) immediately after mist generation, and was reduced to 87.4% (84.8 ; 89.9) at 1 hour and 81.0% (78.3 ; 83.5) at 2 hours, respectively. Another test indicated that hexavalent chromium collected on PVC filters was also reduced by time after sampling. Hexavalent chromium was reduced to 90.8% (88.2 ; 93.3) at 2 hours after sampling. It also was found that hexavalent chromium was reduced during storage in air. It is recommended that air samples of hexavalent chromium be protected against reduction during storage.

Wear Characteristics According of Heat Treatment of Si3N4 with Different Amounts of SiO2 Nano-Colloid (SiO2 나노 콜로이드 량이 다른 Si3N4의 열처리에 따른 마모 특성)

  • This study sintered $Si_3N_4$ with different amounts of $SiO_2$ nano-colloid. The surface of a mirror-polished specimen was coated with $SiO_2$ nano-colloid, and cracks were healed when the specimen was treated at a temperature of 1273 K for 1 h in air. Wear specimen experiments were conducted after heat treatments for 10 min at 1073, 1273, and 1573 K. The heat-treated surface that was coated with the $SiO_2$ nano-colloid was slightly rougher than the noncoated surface. The oxidation state of the surface according to the heat treatment temperature showed no correlation with the surface roughness. Moreover, the friction coefficient, wear loss, and bending strength were not related to the surface roughness. $Si_3N_4$ exhibited an abrasive wear behavior when SKD11 was used as an opponent material. The friction coefficient was proportional to the wear loss, and the bending strength was inversely proportional to the friction coefficient and wear loss. The friction coefficient and wear loss increased with increasing amounts of the $SiO_2$ nanocolloid. In addition, the friction coefficient was slightly increased by increasing the heat treatment temperature.

Effect of Panax Ginseng Saponin on Metabolism and Ion Transport in Human Erythrocytes (인삼이 적혈구세포의 해당과정 및 막 투과도에 미치는 영향)

  • Red cell glycolytic intermediates, metabolites and metabolic ratios were studied. Glycolytic intermediates were measured in neutralized perchloric acid extracts of red cell suspensions after 3 hr incubation at $37^{\circ}C$ in the presence and absence of saponin. Adenosine triphosphate(ATP), adenosine diphosphate(ADP), pyruvate and lactate were measured by enzymatic procedures involving stoichiometric oxidation or reduction of a pyridine nucleotide. Glucose was determined using glucose oxidase after zinc hydroxide extraction. The redox state was calculated from the lactate dehydrogenase equilibrium. Adenosine triphosphatase activity(ATPase) was measured by determining the amount of phosphate released from ATP by washed erythrocyte membranes(ghost) during 20 min. incubation. Both total hydrolysis and the amount of hydrolysis that occured in the presence of ouabain were measured. The second measurement yields Mg-ATPase and represents nonspecific ATPase activity of the membranes. The difference between total and Mg-ATPase activity can be attributed to Na-K-ATPase. For the measurement of sodium fluxes, human erythrocytes were preincubated in $^{22}Na$ for 3 hr at $37^{\circ}C$, washed and suspended in a tracer-free medium. The amount of $^{22}Na$ transported out of cells at any time was determined by analysis of supernatant samples taken at various time after addition of the labeled cells to isotope-free medium. The cells and medium were separated and the radioactivity appearing in the medium was measured. From the total radioactivity in the suspension and the radioactivity appearing in the medium at known time, the rate constant for sodium release was computed. The results are summarized as follows: 1) ATP and ATP/ADP were found to increase at every concentration of saponin tested whereas ADP declined at every cone. of saponin. The increase in pyruvate and lactate were observed at every cone, of saponin and thus $NAD^+/NADH$ computed from pyruvate/lactate also increased. Glucose utilization was stimulated by saponin. 2) $Na^+-K^+-ATPase$ activities showed a biphasic response to saponin, first increasing in lower concentration and then decreasing in higher concentration of saponin. 3) The efflux of sodium was significantly increased by saponin in the range of 5 to 10 mg%. The stimulatory effect of saponin on the rate constants for active(ouabain-sensitive) sodium efflux was inhibited by addition of ouabain.
