Octane Biodegradability by Crude Oil4 tilizing Bacteria Carrying OCT Plasmid (OCT 플라스미드를 갖는 원유 분해세균에 의한 Octane 분해능)

  • Xanthomonns curnpestris M12, Xunthornonas sp. M28, Acinetuhucter Iwofz GI, and Klebsiella pneumoniae L25, Pseudomonas rnaltophiliu N246 were screened to increase the ability for crude oil utilization. All of these could utilize hexadecane and octane with the exception of N246 strain for only octane biodegradation. Thus N246, M12, and M28, strains were specially examined for octane oxidation. Octane biodegradation by three strains showed the optimal conditions at $30^{\circ}C$, pH 7.0~9.0, and 0.2~0.3% octane concentration as a substrate. It was found that P. multofihila N246 and X. curnpestns M12 had plasmid and the cured plasmid from N246 strain lost octane uitilization. Therefore, it was confirmed that certain genes for octane utilization were Iocated on OCT plasmid in N246 strain. The size of OCT plasmid in N246 strain was 118 kb. The N246 strain was resistant to ampicillin.

Improvement of uniformity in chemical vapor deposition of silicon carbide using CFD (탄화규소 화학기상증착 공정에서 CFD를 이용한 균일도 향상 연구)

  • In order to increase the thickness uniformity in chemical vapor depositon of silicon carbide, we have carried out CFD studies for a CVD apparatus having a horizontally-rotated 3-stage susceptor. We deposited silicon carbide films of 3C-SiC phase showing quite uniform thickness between stages but not uniform one in the stage. The cause of this nonuniformity is thought to be originated from the high rotational speed. And the uniformity between stages can be further increased with the $120^{\circ}$ split type nozzles from CFD results. Through the formation of silicon carbide film on graphite substrates we can make oxidation-resistant and dust-free graphite components with high hardness for the semiconductor applications.

Formation of DNA-protein Cross-links Mediated by C1'-oxidized Abasic Lesion in Mouse Embryonic Fibroblast Cell-free Extracts

  • Oxidized abasic residues arise as a major class of DNA damage by a variety of agents involving free radical attack and oxidation of deoxyribose sugar components. 2-deoxyribonolactone (dL) is a C1'-oxidized abasic lesion implicated in DNA strand scission, mutagenesis, and covalent DNA-protein cross-link (DPC). We show here that mammalian cell-free extract give rise to stable DPC formation that is specifically mediated by dL residue. When a duplex DNA containing dL at the site-specific position was incubated with cell-free extracts of Po ${\beta}-proficient$ and -deficient mouse embryonic fibroblast cells, the formation of major dL-mediated DPC was dependent on the presence of DNA polymerase (Pol) ${\beta}$. Formation of dL-specific DPC was also observed with histones and FEN1 nuclease, although the reactivity in forming dL-mediated DPC was significantly higher with Pol ${\beta}$ than with histones or FEN1. DNA repair assay with a defined DPC revealed that the dL lesion once cross-linked with Pol ${\beta}$ was resistant to nucleotide excision repair activity of cell-free extract. Analysis of nucleotide excision repair utilizing a model DNA substrate containing a (6-4) photoproduct suggested that excision process for DPC was inhibited because of DNA single-strand incision at 5' of the lesion. Consequently DPC mediated by dL lesion may not be readily repaired by DNA excision repair pathway but instead function as unusual DNA damage causing a prolonged DNA strand break and trapping of the major base excision repair enzyme.

  • The Ni-base super-heat-resistant alloy, GTD-111, is employed in gas turbines because of its high temperature strength and oxidation resistance. It is important to predict the fatigue life of this superalloy in order to improve the efficiency of gas turbines. In this study, low-cycle fatigue tests are performed as variables of total strain range and temperature. The relationship between the strain energy density and number of cycles to failure is examined in order to predict the low-cycle fatigue life of the GTD-111 superalloy. The fatigue life predicted by using the strain-energy methods is found to coincide with that obtained from the experimental data and from the Coffin-Manson method.

  • Many coating technologies have been developed so far to improve the corrosion resistance, strength, abrasion resistance and other surface properties of materials and equipment. Among them, the formation of CCO (CaCoO, then CCO) thin films has been studied and used in the electronic material field. One of the characteristics of CCO thin films is that it is resistant to high temperature heat. Particularly, the method of forming the CCO thin film is relatively simple, and it was judged that it could be introduced into the existing equipment. Therefore, in this study, an experiment and analysis were carried out to determine whether the coating of CCO thin films can be applied to hot dip galvanizing facilities. A CCO thin film was formed on the surface of STS304 base material and oxidized in a Zn fume atmosphere in a $650^{\circ}C$ furnace with an air atmosphere. Oxidation was carried out for 30 days, after which the shape of the CCO thin film was confirmed by SEM and its corrosivity was analyzed through a potentiodynamic polarization experiment.

Micro Structure and Surface Characteristics of NiCr Thin films Prepared by DC Magnetron Sputter according to Annealing Conditions (DC 마그네트론 스퍼터링 NiCr 박막의 열처리 조건에 따른 미세구조 및 표면특성)

  • Ni/Cr thin film is very interesting material as thin film resistors, filaments, and humidity sensors because their relatively large resistivity, more resistant to oxidation and a low temperature coefficient of resistance (TCR). These interesting properties of Ni/Cr thin films are dependent upon the preparation conditions including the deposition environment and subsequent annealing treatments. Ni/Cr thin films of 250 nm were deposited by DC magnetron sputtering on $Al_2O_3/Si$ substrate with 2-inch Ni/Cr (80/20) alloy target at room temperature for 45 minutes. Annealing treatments were performed at $400^{\circ}C,\;500^{\circ}C,\;and\;600^{\circ}C$ for 6 hours in air or $H_2$ ambient, respectively. The clear crystal boundaries without crystal growth and the densification were accomplished when the pores were disappeared in air ambient. Most of surface was oxidic including NiO, $Ni_2O_3$ and $Cr_xO_y$(x=1,2, y=2,3) after annealing in air ambient. The crystal growth in $H_2$ ambient was formed and stabilized by combination with each other due to the suppression of oxidized substance on film surface. Most oxidic Ni was restored when the oxidic Cr was present due to its stability in high-temperature $H_2$ ambient.

Comparison of biological and chemical assays for measuring the concentration of residual antibiotics after treatment with gamma irradiation

  • Antibiotic pollution is one of the factors contributing to the spread of antibiotic-resistant bacteria in the environment. Advanced oxidation and irradiation processes have been introduced to eliminate antibiotics from water and wastewater. However, few studies have reported the toxic effects of residual antibiotics and their byproducts induced by a treatment system. In this study, we compared the efficacies of chemical (high-performance liquid chromatography (HPLC)) and biological (antimicrobial susceptibility test) assays for measuring the concentrations of residual antibiotics after gamma irradiation for degrading amoxicillin, cephradine, lincomycin, and tetracycline. The concentrations of residual antibiotics estimated using the two assay methods were almost identical, except cephradine. In the case of cephradine, inhibited bacterial growth was observed that was equivalent to twice the concentration measured by HPLC in the samples subjected to gamma irradiation. The observed inhibition of bacterial growth suggested the generation of potentially toxic intermediates following antibiotic degradation. These results indicate that biological and chemical assays should be used in concert for monitoring antibiotic contamination and the toxic derivatives of antibiotic degradation. The results demonstrate that these four antibiotics can be decomposed by 2.0 kGy gamma-irradiation without toxic effects of their byproducts.

Biological Properties of Different Types and Parts of the Dandelions: Comparisons of Anti-Oxidative, Immune Cell Proliferative and Tumor Cell Growth Inhibitory Activities

  • Dandelions have been reported to have medicinal properties and bioactive components that impact human health. However, the precise biological properties of dandelions and the parts of the plants possessing bioactive components remain uncertain. In this study, we evaluated 3 different types of dandelions based on their cultivation origin (Songpa, Uiryung, and native Uiryung types) as well as their 4 different plant parts (leaf, flower, root, skin). Each sample was extracted with $80\%$ methanol and then compared for the biological activities (anti-oxidative, immune cell proliferative and tumor cell growth inhibitory activities). All 3 types of dandelions possessed a degree of biological functions including the hydroxyl radical scavenger activity, immune cell proliferative activity and tumor cell growth inhibitory activity. However, there was no significant difference in these activities between the 3 dandelion types. Leaves of all three dandelion types showed the highest levels of all biological activities. To a lesser degree, the flower and root parts displayed biological activities. In the skin parts, anti-oxidative activity was also detected only at higher doses of dandelion extracts. Heating the dandelion leaf extract did not affect the biological activity, suggesting a heat-stable nature of the biological compounds. Taken together, these collective data suggest that dandelions, in particular their leaves, possess a high concentration of heat-resistant biological compounds, which are responsible for anti-oxidative, immune cell proliferative and tumor cell growth-inhibitory activities.

EFFECTS OF SURFACTANTS ON THE FENTON DEGRADATION OF PHENANTHRENE IN CONTAMINATED SEDIMENTS

  • Laboratory batch experiments were conducted to evaluate the Fenton degradation rates of phenanthrene. Fenton reactions for the degradation of phenanthrene were carried out with aqueous and slurry phase, to investigate the effects of sorption of phenanthrene onto solid phase. Various types of surfactants and electrolyte solutions were used to evaluate the effects on the phenanthrene degradation rates by Fenton's reaction. A maximum 90% removal of phenanthrene was achieved in aqueous phase with 0.9% of $H_2O_2$ and 300 mg/L of $Fe^{2+}$ at pH 3. In aqueous phase reaction, inhibitory effects of synthetic surfactants on the removal of phenanthrene were observed, implying that surfactant molecules acted as strong scavenger of hydroxyl radicals. However, use of $carboxymethyl-{\beta}-cyclodextrin$ (CMCD), natural surfactant, showed a slight enhancement in the degradation of phenanthrene. It was considered that reactive radicals formed at ternary complex were located in close proximity to phenanthrene partitioned into CMCD cavities. It was also show that Fenton degradation of phenanthrene were greatly enhanced by addition of NaCl, indicating that potent radical ion ($OCI^-$) played an important role in the phenanthrene degradation, although chloride ion might be acted as scavenger of radicals at low concentrations. Phenanthrene in slurry phase was resistant to Fenton degradation. It might be due to the fact that free radicals were mostly reacting with dissolved species rather than with sorbed phenanthrene. Even though synthetic surfactants were added to increase the phenanthrene concentration in dissolved phase, low degradation efficiency was obtained because of the scavenging of radicals by surfactants molecules. However, use of CMCD in slurry phase, showed a slight enhancement in the phenanthrene degradation. As an alternative, use of Fenton reaction with CMCD could be considered to increase the degradation rates of phenanthrene desorbed from solid phase.

Optimal Aluminizing Coating on Incoloy 909 (Incoloy 909 합금의 최적 알루미나이징 확산 코팅)

  • An Fe-Ni-Co based superalloy Incoloy 909 (Incoloy 909) has been used for gas turbine engine component material. This alloy is susceptible to high temperature oxidation and corrosion because of the absence of corrosion resistant Cr. For the improvement of durability of the component of Incoloy 909 aluminizing-chromate coating by pack cementation process has been investigated at relatively low temperature of about $550^{\circ}C$ to protect the surface microstructure and properties of Incoloy 909 substrate. As a previous study to aluminizing-chromate coating by pack cementation of Incoloy 909, the optimal aluminizing process has been investigated. The size effects of source Al powder and inert filler $Al_O_3$ powder and activator selection have been studied. And the dependence of coating growth rate on aluminizing temperature and time has also been studied. The optimal aluminizing process for the coating growth rate is that the mixing ratio of source Al powder, activator $NH_4Cl$ and filler $Al_O_3$ are 80%, 1% and 19% respectively at aluminizing temperature $552^{\circ}C$ and time 20 hours.
