Helicobacter pylori Infection and Vitamin C: Past, Present and Future Perspectives (Helicobacter pylori 감염과 비타민 C: 과거, 현재, 미래)

  • Helicobacter pylori is the causative agent of chronic gastritis and has a role in the pathogenesis of peptic ulcer diseases, and gastric cancer. There have been reports suggesting a close link between these gastroduodenal disorders and a state of vitamin C deficiency. In this paper, the past, present and future perspectives on H. pylori infection and vitamin C will be discussed under the following view points. Since the ecological niche of H. pylori is the mucus layer and intercellular junctions of the gastric epithelium, the various kinds of host inflammatory cells motivated by the local and systemic immune responses cannot eliminate the microorganisms. When the invading foreign body is not removed, despite full activation of defense mechanisms, adverse consequences of the immune responses develop on the host gastric mucosa. The reasons for the body vitamin C depletion could be explained as follows; 1) the increased vitamin C consumption by increased oxygen free radical production through the prolonged hypersensitivity reactions in the gastric mucosa, 2) the increased vitamin C oxidation by the nitrite which is formed from nitrate reduction by the intragastric bacteria proliferated in the hypochlorhydric gastric cavity, 3) the strong ${\gamma}$-glutamyltranspeptidase activity of H. pylori which depletes the glutathiones in gastric mucosa. Depletion of glutathiones in the stomach favors irreversible oxidative destruction of ascorbic acid. Both persistent inflammatory burdens in the stomach by H. pylori and resultant vitamin C depletions synergistically and uninhibitedly might aggravate the hypothetical sequence of gastric carcinogenesis: atrophic gastritis${\rightarrow}$intestinal metaplasia${\rightarrow}$dysplasia${\rightarrow}$gastric adenocarcinoma. High intake of vitamin C could reverse the hypothetical sequence of the gastric carcinogenesis via direct and indirect effects on H. pylori and host-parasite relationships.

  • Corrosion has been around for all of recorded history. Cathodic protection is the electrical solution to the corrosion problem. Corrosion is not exactly a new topic. It has been around since the beginning of time. Corrosion is simply the loss of material resulting from current leaving a metal, following through a medium, and returning to the metal at a different point. Corrosion takes many forms and has various names, such as oxidation, rust, chemical, and bacteria action. Regardless of the agent, all corrosion is the result of electrical current flow. Various methods are used to treat corrosion or to try to prevent ti. Some of these include chemical treatment. coatings, and electrical current. Especially, proper impressed current can stop corrosive action on the protected surface. In this article, we introduce the Impressed Current Cathodic Protection (ICCP) Control and monitoring system developed by ourselves. The ICCP system is composed of a power supply, anode, reference electrode and controller. The main issue is to control the current flow on the desired value such that it is possible to force a metal to be more negative(cathodic) than the natural state. From the this process, we can achieve the cathodic protection. Of course, in the developed system, the necessary functions are possessed, such as remote control, monitoring of system fault detection etc. Some experimental results show the system performance and usefulness.

Mechanism of Stress-dependent Structural Change of Yeast Prx (Yeast Prx의 스트레스의존 구조적 변화의 기작)

  • Peroxiredoxins (Prxs) are a superfamily of thiol-specific antioxidant proteins present in all organism and involved in the hydroperoxide detoxification of the cell. To determine the structural organization of yeast-Prx, electron microscopic analysis was performed. The average images of yeast-Prxs revealed three different structure, i.e. spherical-shaped structure, ring-shaped structure and irregularly-shaped small particles. In order to analyze the conformational change of yeast-Prx by reduction and oxidation, Prxs were subjected to DTT and $H_2O_2$. In presence of DTT, yeast-Prx showed a high tendency to form a decamer. However, they changed into dimeric or spherical structure in the oxidized state. Here we also show ionic interaction between dimeric subunits is primarily responsible for yeast-Prx oligomerization.

  • The rapid increase in the prevalence of metabolic syndrome, which is associated with a state of elevated systemic oxidative stress and inflammation, is expected to cause future increases in the prevalence of diabetes and cardiovascular diseases. Oxidation of polyunsaturated fatty acids and sugars produces reactive carbonyl species, which, due to their electrophilic nature, react with the nucleophilic sites of certain amino acids. This leads to formation of protein adducts such as advanced glycoxidation/lipoxidation end products (AGEs/ALEs), resulting in cellular dysfunction. Therefore, an effective reactive carbonyl species and AGEs/ALEs sequestering agent may be able to prevent such cellular dysfunction. There is accumulating evidence that histidine containing dipeptides such as carnosine (${\beta}$-alanyl-L-histidine) and anserine (${\beta}$-alanyl-methyl-L-histidine) detoxify cytotoxic reactive carbonyls by forming unreactive adducts and are able to reverse glycated protein. In this review, 1) reaction mechanism of oxidative stress and certain chronic diseases, 2) interrelation between oxidative stress and inflammation, 3) effective reactive carbonyl species and AGEs/ALEs sequestering actions of histidine-dipeptides and their metabolism, 4) effects of carnosinase encoding gene on the effectiveness of histidine-dipeptides, and 5) protective effects of histidine-dipeptides against progression of metabolic syndrome are discussed. Overall, this review highlights the potential beneficial effects of histidine-dipeptides against metabolic syndrome. Randomized controlled human studies may provide essential information regarding whether histidine-dipeptides attenuate metabolic syndrome in humans.

  • Various 1-methyl-1,2,4-triazinium iodides were easily synthesized by the reaction of various 1,2,4-triazines and methyl iodide in acetone. When methyl iodide salts of 1,2,4-triazine derivatives were treated with $K_3Fd(CN)_6$ in 10% NaOH solution, 1-methyl-1,2,4-triazole derivatives as the main products were obtained by ring contraction. In addition to 1-methyl-1,2,4-triazole derivatives, 1,6-dihydro-6-oxo-1,2,4-triazine derivatives as the minor products were also obtained when there were no substitutents in $C_6$-position of 1,2,4-triazines. The formation of pseudo base by $OH^-$ and then oxidation to either 1,6-dihydro-6-oxo-1,2,4-triazines or 1-methyl-1,2,4-triazoles were suggested as the mechanism of the ring contraction. This mechanism also verified that the position of quaternization was neither $N_2$ nor $N_4$ but 1-nitrogen of 1,2,4-triazine.

Mineralogical Change of Acid Sulfate Weathering of Hydrothermally Altered Pyritic Andesite (열수변질 안산암 기원의 함황광물과 특이산성토적 풍화에 따른 광물학적 변화)

  • Oxidation of pyrite has caused a serious environmental problem such as the acidification of soil and surface water. The mineralogical change of acid sulfate weathering of hydrothermally altered andesite which contained 11.8% of pyrite and was exposed in atmosphere by lay out works for a residential area and a golf course was studied using X-ray diffraction (XRD) and electronmicroscopes. Ferrihydrite, jarosite, and an unidentified water soluble phase were observed as weathering products of the andesite. Under electronmicroscopes, showed aggregate of platy microcrystals; jarosite was platy morphology: water soluble Phase was columnar. Morphology of fresh Pyrite in the andesite changed from pyritrohedron to cubic in its frequency with increasing its particle size. The drainage water was acidic (pH 3.5) and in an equilibrium state with both ferrihydrite and jarosite.

A Study on the Properties of Fe-Se-Te System (Fe-Se-Te계의 특성 연구)

  • The properties of Fe-Se-Te system(FeSe(sub)1-xTe(sub)x, x=0.2, 0.5, 0.8) have been studied by means of the X-ray diffraction method and Mossbauer spectroscopy. The results of X-ray diffraction patterns show that three samples have the ixed structure of tetragonal PbO and a small amount of hexagonal NiAs structure respectively. For x=0.5 the lattice parameters of tetragonal PbO structure are a=3.795$\AA$, c=5.896$\AA$ and c/a=1.55. The Mossbauer spectra were obtained with the various temperature variation and than they do not exhibit magnetic hyperfine structure but show a strong doublet. The values of observed isomer shift and quadrupole splitting suggest that the irons of all samples exist in the +2 oxidation state with a major covalent contribution. The temperature dependence of isomer shift values for x=0.8 seems to be originated from the second order Doppler effect.

Removal of High Strength Hydrogen Sulfide Gas using a Bioreactor Immobilized with Acidithiobacillus ferrooxidans and a Chemical Absorption Scrubber (Acidithiobacillus ferrooxidans를 고정화한 생물반응기와 흡수탑을 이용한 고농도 황화수소 제거)

  • To treat a waste gas containing a high strength H2S, the two-stages microbial desulfurization process that conof a bioreactor immobilized with Acidithiobacillusferrooxidans and a chemical absorption scrubber has was proposed. After 4 times repeat of batch cultures, the immobilized bioreactor has been stabilized and the rate of iron oxidation reached 0.89 kg . $m^{-3}{\cdot}m^{-1}$ at steady state. The two-stages microbial desulfurization prowas able to be operated for a long term over 54 days. The removal efficiencies of H2S were 97-99% at a space velocity of 70 h-I and a inlet concentration of 37,000 ppmv. The maximum elimination capacity of H2S was approximately 3.3 kg S . $m^{-3}{\cdot}m^{-1}$. In the bioractor, the concentrations of the $Fe^{3+}$ and the immobilzed cell were constantly maintained during the desulfurization.

Methods for Evaluation of Antioxidant Activity: Application to Taurine (항산화 활성의 평가를 위한 연구법: 타우린의 적용)

  • Although taurine (2-aminoethanesulfonic acid) can inhibit oxidative stress in both animal and epidemiological studies, it is obscure whether taurine directly scavenges oxy-radicals or indirectly regulates oxidant production and/or antioxidant defense system. The reason for this discrepancy remains unknown but may be due, in part, to the lack of a validated assay system for evaluating oxy-radical scavenging capacity. The antioxidant activities of taurine and hypotaurine (2-aminoethanesulfinic acid), a precursor of taurine, against peroxyl radicals, hydroxyl radicals and peroxynitrites were determined by the total oxy-radical scavenging capacity (TOSC) assay and cell-based assay using H4IIE cells. tert-Butylhydroperoxide or hydrogen peroxide-induced cell toxicity determined by MTT assay was markedly inhibited by 10mM taurine or hypotaurine. The tert-butylhydroperoxide- or hydrogen peroxide-induced changes in oxidative stress markers, such as cellular glutathione and malondialdehyde, were ameliorated by 10mM taurine or hypotaurine. However, specific TOSC values calculated from the slope of the linear regression for taurine against peroxyl radicals, hydroxyl radicals or peroxynitrites were all less than 1 TOSC/mM. On the other hand specific TOSC values for hypotaurine against peroxyl radicals, hydroxyl radicals or peroxynitrites were 48, 2096, or 69 TOSC/mM, respectively. These results suggest that taurine protects cells against oxidative insults, which is not ascribed to directly scavenging activity of taurine against oxy-radicals. These results support the idea that the oxidation state of sulfur in antioxidants may be a determinant of oxy-radical scavenging capacity.

Use of Chicken Meat and Processing Technologies

  • The consumption of poultry meat (chicken and turkey) grew the most during the past few decades due to several contributing factors such as low price, product research and development, favorable meat characteristics, responsive to consumer needs, vertical integration and industry consolidation, new processing equipments and technology, and aggressive marketing. The major processing technologies developed and used in chicken processing include forming/restructuring, tumbling, curing, smoking, massaging, injection, marination, emulsifying, breading, battering, shredding, dicing, and individual quick freezing. These processing technologies were applied to various parts of chicken including whole carcass. Product developments using breast, thigh, and mechanically separated chicken meat greatly increased the utilization of poultry meat. Chicken breast became the symbol of healthy food, which made chicken meat as the most frequent menu items in restaurants. However, the use of and product development for dark meat, which includes thigh, drum, and chicken wings were rather limited due to comparatively high fat content in dark meat. Majority of chicken are currently sold as further processed ready-to-cook or ready-to-eat forms. Major quality issues in chicken meat include pink color problems in uncured cooked breast, lipid oxidation and off-flavor, tenderness PSE breast, and food safety. Research and development to ensure the safety and quality of raw and cooked chicken meat using new processing technologies will be the major issues in the future as they are now. Especially, the application of irradiation in raw and cooked chicken meat products will be increased dramatically within next 5 years. The market share of ready-to-eat cooked meat products will be increased. More portion controlled finished products, dark meat products, and organic and ethnic products with various packaging approaches will also be introduced.
