Effects of Yuklinzu on the ovariectomized rat model of postmenopausal osteoporosis (육린주가 난소적출 백서의 골다공병에 미치는 영향)

  • Aging is characterized by an age-dependent reduction in bone density. Pronounced bone loss by postmenopausal estrogen deficiency induces a bad effect on dynamic balance of bone metabolism. When the bone density is reduced to the level below the fracture threshold, the risk for fracture is greatly increased. The purpose of this study is to examine what are the effects of the Yuklinzu(YLZ: 毓麟珠) on the ovariectomized rat model of postmenopausal osteoporosis. The results of the experiment are as follows: Body weight in control group showed significant increase in comparison with sham, but that in YLZ-treated showed no change in comparison with control. The level of serum albumin in control group showed significant decrease in comparison with sham. That in YLZ-treated was slightly decreased in comparison with control. The level of serum ALP activity in control group showed significant increase in comparison with sham, but that in YLZ-treated was significantly decreased in comparison with control. The level of serum phosphorous in control group showed significant increase in comparison with sham, but that in YLZ-treated showed no change in comparison with control. The level of serum calcium in control group showed no change in comparison with sham, but that in YLZ-treated was significantly decreased in comparison with control. The level of serum GOT in control group showed no change in comparison with sham, but that in YLZ-treated was significantly decreased in comparison with control. The level of serum GPT did not significant change among the three groups. The level of serum estrogen in control group showed slightly increased in comparison with sham, but that in YLZ-treated showed no change in comparison with control. The level of serum T3 in control group showed no change in comparison with sham, and that in YLZ-treated showed no change in comparison with control. The level of serum T4 in control group showed significant increase in comparison with sham, but that in YLZ-treated was significantly decreased in comparison with control. Mechanical strength of the femur neck in control group showed significant decrease in comparison with sham. That in YLZ-treated showed slightly increased in comparison with control. Trabecular bone area as well as trabecular thickness in control group showed significant decrease in comparison with sham. Those in YLZ-treated showed significant increase in comparison with control. Trabecular number and did not significant change among the three groups. Trabecular separation only in YLZ-treated showed significant decrease in comparison with control. Osteoclast number and oseoblast surface in control group showed significant increase in comparison with sham. Those in YLZ-treated showed slightly decreased in comparison with control. According to the above these results, YLZ has shown to be capable of preventing and curing osteoporosis caused by old-aged and postmenopause.

Effects of Lycii Fructus and Lycii Folium Extracts on Osteoporosis in Ovariectomized Rats (구기자와 구기엽 추출물이 난소적출 흰쥐의 골다공증에 미치는 영향)

  • The effects of Lycii fructus and Lycii folium on osteoporosis and serum cholesterol levels were tested in ovariectomized (OVX) rats. Twenty-four female Sprague-Dawley rats were divided into four groups: Sham group (sham-operated), Control group (OVX, ovariectomized model), LCF group (Ovx+Lycii fructus extract), and LCL group (OVX+Lycium folium extract). After 8 weeks, the OVX ($330{\pm}5.39$ g), LCF ($315{\pm}2.99$ g), and LCL ($318{\pm}2.06$ g) groups showed increased body weight compared with sham group ($281{\pm}1.71$ g). The levels of serum osteocalcin (OC) also increased in the LCF ($444.6{\pm}26.9$ ng/mL) and LCL ($407{\pm}18.9$ ng/mL) groups compared with the OVX group ($107{\pm}3.52$ ng/mL). The activities of serum alkaline phosphatase (ALP) increased in the LCF ($108{\pm}2.7$ U/L) and LCL ($407{\pm}18.9$ ng/mL) groups compared with the OVX group ($95{\pm}2.9$ U/L). Stereomicroscopy found that the low bone density that developed in the OVX group was significantly reversed in the LCF and LCL groups after 8 weeks. We also obtained molecular-based in vivo evidence that supports a mechanism of action involving novel estrogen receptor ($ER{\alpha}$) modulator in the uterus. We found that expression of ER${\alpha}$ mRNA in the OVX rat uterus was elevated by Lycium chinense. These results suggest that Lycii fructus and Lycii folium administered to rats during 8 weeks after oophorectomy may partially recover postmenopausal osteoporosis or delay the progression of osteoporotic changes.

Antiobesity Effect of Estrogen on Ovariectomized Rat (난소절제 흰쥐에서 Estrogen의 항비만 효과)

  • Obesity is a chronic disease that is increasing in prevalence and that poses a serious risk for the hypertension, osteoporosis, heart disease, diabetes mellitus and certains forms of cancer. This study was performed to develop of obesity animal model and to assess the pharmacological assay for the rats of 8 weeks or 4 days after ovariectomization treated with estradiol for 8 weeks on the body weight. fat weight and food intake. The body weight, fat weight and food intake increased in the ovriectomized rats. In the rat of 8 weeks after ovariectomization treated with estradiol (250 mg/100 g) 8 weeks, the body weight decreased significantly (p<0.05). In the rats of 4 days after ovariectomization treated with estradiol 8 weeks, the body weight decreased significantly (p<0.05). These results suggest that estrogen plays a role in regulation body weight response to food intake and fat weight.

Therapeutic Advantages of Treatment of High-Dose Curcumin in the Ovariectomized Rat

  • Objective : Although curcumin has a protective effect on bone remodeling, appropriate therapeutic concentrations of curcumin are not well known as therapeutic drugs for osteoporosis. The purpose of this study was to compare the bone sparing effect of treatment of low-dose and high-dose curcumin after ovariectomy in rats. Methods : Forty female Sprague-Dawley rats underwent either a sham operation (the sham group) or bilateral ovariectomy (OVX). The ovariectomized animals were randomly distributed among three groups; untreated OVX group, low-dose (10 mg/kg) curcumin administered group, and high-dose (50 mg/kg) curcumin group. At 4 and 8 weeks after surgery, serum biochemical markers of bone turnover were analyzed. Bone histomorphometric parameters of the 4th lumbar vertebrae were determined by micro-computed tomography (CT). In addition, mechanical strength was determined by a three-point bending test. Results : High-dose curcumin group showed significantly lower osteocalcin, alkaline phosphatase, and the telopeptide fragment of type I collagen C-terminus concentration at 4 and 8 weeks compared with the untreated OVX group as well as low-dose curcumin group. In the analyses of micro-CT scans of 4th lumbar vertebrae, the high-dose curcumin treated group showed a significant increase in bone mineral densities (p=0.028) and cortical bone mineral densities (p=0.036) compared with the low-dose curcumin treated group. Only high-dose curcumin treated group had a significant increase of mechanical strength compared with the untreated OVX group (p=0.015). Conclusion : The present study results demonstrat that a high-dose curcumin has therapeutic advantages over a low-dose curcumin of an antiresorptive effect on bone remodeling and improving bone mechanical strength.

  • The therapeutic effect of safflower seed powder on experimental osteoporosis in the rat induced by ovariectomy was evaluated. Thirty Sprague-Dawley rats were ovariectomized at the age of 12 weeks. Seven weeks postovariectomy, rats were divided into two groups: control and safflower seed powder treated group. Five animals from each group were sacrificed at the following time points: 1, 3, and 5 weeks. Scanning electron microscopic observation and morphometric analysis of the tibiae epiphysis showed that the administration of safflower seed powder significantly prevented reduction of cortical bone width and bone volume compared with the control group. In conclusion, safflower seed powder contains something that prevent bone loss due to estrogen deficiency, and was effective in preventing the osteoporotic decrease of bone mass.

  • Objectives : In this study, the author aimed to evaluate the effects of water extract of Dokwhalgisaeng-tang Gamibang (DGG) on osteoblast proliferation in murine calvarial cells. Methods : The osteoblast separated from murine calvariae was cultivated and evaluated the cell function. After the addition of DGG on the culture medium, we determined the effect of DGG on the cell proliferation, protein synthesis, alkaline phosphatase activity, collagen synthesis, and cell viability of the cultivated osteoblast in the presence of dexamethasone. Results : DGG increased the survival rate, proliferation, alkaline phosphatase (ALP) activity, protein synthesis and collagen synthesis of rat calvarial osteoblast in the presence of dexamethasone. Conclusions : DGG might improve the osteoporosis resulted from augmentation of osteoblast proliferation.

The Effects of a Mineral Supplement (Aquamin $F^{(R)}$) and Its Combination with Multi-Species Lactic Acid Bacteria (LAB) on Bone Accretion in an Ovariectomized Rat Model

  • Although an adequete intake of calcium (Ca) is recommended for the treatment and prevention of osteoporosis, the intake of Ca should be restricted because of its low rate of intestinal absorption. The purpose of this experiment was to identity the effect of the combined administration of Aquamin F (AQF) (a calcium agent) and lactic acid bacteria (LAB) on osteoporosis. Thirty ovariectomized (OVX) rats and six control rats were assigned to the following six groups, with six animals per group: sham Ca-deficient diet (Ca-D), OVX, LAB, AQF, and LAB-AQF. During the experiment, the body weight was measured; and after the experiment was completed, the serum biochemical analysis, the alkaline phosphatase, calcium, and inorganic phosphorus leves were measured. The tissue of the femur was stained and then scanned via CT. The body weight of the OVX group increased more significantly than that of the control group. The results of the bone mineral content (BMC), Bone mineral density (BMD), serum biochemical analysis and histological test on the femur epiphysis showed no difference between the OVX group and the LAB group, whereas the results of the AQF group were more significant than those of the OVX group. In particular, the LAB+AQF group showed more significant increases in the aforementioned results than the AQF group. This experiment showed that the combined administration of AQF and LAB in ovariectomized rats more significantly increased bone density than did a single administration of either AQF or LAB.

  • The aim of this investigation was to examine the effects of osteocalcin, bone-specific alkaline phophatase, estrogen, insulin-like growth factor-I (IGF-I), Ca, P and bone mineral density on osteoporosis induced by ovariectomy in rats. Female Sprague-Dawley 30 rats of three-forth's birth, weighing $215{\pm}10g$, were divided into two groups including the sham operation group(5 heads) and ovariectomy group(25 heads). They were fed normal diets for 2 weeks before the experimental operation and for 8 more weeks after operation. The level of osteocalcin, TALP, BALP, estrogen, bone mineral density and IGF-I were increased in experimental group, but a little increased in sham operation group at same period. The change of rates of osteocalcin, TALP, BALP, estrogen, bone mineral density and IGF-I were significantly higher in experimental group than sham operation group. $Ca^{2+}$ was not changed between two groups and P was significantly decreased in experimental group and Ca/P ratio was higher in experimental group than sham operation group. Body weights were increased in all two groups and growth rate per day was higher in experimental group than sham operation group. However, femur weight I body weight ratio was lower in experimental group than sham operation group.

  • Objective : The purpose of this studies was to determine the effect of Carthami semen, Bogol-Tang and Rhynchosia volubilis Lour. on changes of trabecular area and physiological metabolites in the ovariectomized osteoporotic Rats. Methods : In order to induce estrogen deficient osteoporosis, ovariectomy was done on rats. Then the Carthami semen, Bogol-Tang and Rhynchosia volubilis Lour. were orally administerd: Such indexes were measured as the changes of body weight, bone mineral density, trabecular area in tibia, and levels of osteocalcin, bone alkaline phosphatase, calcium, phosphorus, cholesterol, triglyceride, aspartate aminotransferase and alanine aminotransferase in serum. Results : 1. The change of bone mineral density in Bogol-Tang group and Rhynchosia volubilis lour. group. was significantly increased compare to control group. 2. The change of trabecular area % in epiphysis of tibia in Carthami semen group was significantly increased compare to control group. 3. The change of trabecular area % in diaphysis of tibia in Bogol-Tang group was significantly increased compare to control group. 4. The change of serum osteocalcin in Bogol-Tang group was significantly decreased compare to control group. 5. The change of serum bone alkaline phosphatase in Bogol-Tang group was significantly decreased compare to control group. 5. Effect of Biochemical Makers of Bone Metabolism by Administration of Radix Concentration in Ovariectomized Rats (골다공증 쥐에서 우슬의 농도별 투여에 따른 골대사의 생화학적 마커에 미치는 영향)

  • Osteoporosis is the most prevalent metabolic bone disease and is characterized by diminished bone strength predisposing to an increased risk of fracture. We investigated the effects of the extract from Acyranthes Radix on the progress of bone loss in ovariectomized rats for 6 weeks. Female Sprague-Dawley 40 rats of 3-4 weeks, weight 200 ${\pm}$ 10g were divided into two groups including the sham operation group(8 rats) and ovariectomy group(32 rats). The dose-dependent effect of Acyranthes Radix extract on bone mineral density and biochemical testing was assessed in ovariectomized rat. Body weights were increased in all groups was higher in experimental group than sham operation group. The level of bone mineral density, GPT, and serum P concentration, ALP were increased experimental group, but a little increase in sham operation group at same period. This longitudinal study result made conclusion that Acyranthes Radix extract treatment appeared to improve the osteoporosis delaying the progression to the osteoporotic process in rats.
