Background: Ecophysiological characteristics of Rosa rugosa were analyzed under different environmental factors from May to October 2022. Photosynthesis, chlorophyll fluorescence, chlorophyll content, leaf water content (LWC), osmolality, carbohydrate content, and total ion content were measured to compare the physiological characteristics of R. rugosa at two study sites (i.e., in large pots and in the Goraebul coastal sand dune area). Results: When R. rugosa was exposed to high temperatures, photosynthetic parameters including net photosynthetic rate (PN) and stomatal conductance (gs) in both experiment areas declined. In addition, severe photoinhibition occurs when R. rugosa is continuously exposed to high photosynthetically active radiation (PAR), and because of this, relatively low Y(II) (i.e., the quantum yield of photochemical energy conversion in photosystem II [PSII]) and high Y(NO) (i.e., the quantum yield of non-regulated, non-photochemical energy loss in PSII) in the R. rugosa of the pot were observed. As the high Y(NPQ) (i.e., the quantum yield of regulated non-photochemical energy loss in PSII) of R. rugosa in the coastal sand dune, they dissipated the excessed photon energy through the non-photochemical quenching (NPQ) mechanism when they were exposed to relatively low PAR and low temperature. Rosa rugosa in the coastal sand dune has higher chlorophyll a and carotenoid content. The high chlorophyll a + b and low chlorophyll a/b ratios seemed to optimize light absorption in response to low PAR. High carotenoid content played an important role in NPQ. As a part of the osmotic regulation in response to low LWCs, R. rugosa exposed to high temperatures and continuously high PAR used soluble carbohydrates and ions to maintain high osmolality. Conclusions: We found that Fv/Fm was lower in the potted plants than in the coastal sand dune plants, indicating the vulnerability of R. rugosa to high temperatures and PAR levels. We expect that the suitable habitat range for R. rugosa will shrink and move to north under climate change conditions.
This study was conducted to find out the influence of drought stress on physiological responses of Synurus deltoides. Drought stress was induced by withholding water for 25 days. Leaf water potentials were decreased of both predawn (${\Psi}_{pd}$) and mid-day (${\Psi}_{mid}$) with increasing drought stress, but water saturation dificit (WSD) was 7 times increased. ${\Psi}_{pd}-{\Psi}_{mid}$ showed the significant difference of 0.22~0.18 MPa in stressed before 10 days, and nonsignificant as treatment time became longer. A strong reduction of stomatal conductance ($gH_2O$) and stomatal transpiration rate (E) were observed after 15 days of drought stress Significant reductions of net apparent quantum yield (${\Phi}$) and maximum photosynthesis rate ($Pn_{max}$) were observed after 20 days of drought stress; However, water use efficiency (WUE) was shown the opposite trend. This implies that decrease of photosynthesis rate may be due to an inability to regulate water and $CO_2$ exchanged through the stomata. From JIP analysis, flux ratios (${\Psi}_O$ and ${\Phi}_{EO}$) and performance index on absorption basis ($PI_{ABS}$) were dramatically decreased withholding water after 15 days, which reflects the relative reduction of photosystem II activity. The leaf of S. deltoides showed osmotic adjustment of -0.35 MPa at full turgor and -0.40 MPa at zero turgor, and also cell-wall elastic adjustment of 9.4 MPa, indicating that S. deltoides tolerate drought stress through osmotic adjustment and cell-wall elastic adjustment. The degree of change in water relations parameters such as Vo/DW, Vt/DW decreased with increasing drought stress. This result showed that S. deltoides was exhibited a strong reduction of photosynthetic activity to approximately -0.93 MPa of predawn leaf water potential, and both of osmotic adjustment and cell-wall elastic adjustment in drought stress condition appears to be an important adaptation for restoration in this species.
How does $CO_2$ affect on the stomatal mechanism? The mechanism of stomatal opening by $CO_2$ is not clear as it is difficult to see $CO_2$ effect on light-induced stomatal opening. Furthermore, stomata may react differently according to the concentration of $CO_2$. The significance of the possible endogenous rhythms must consider to understand on $CO_2$-related response. It is clear that $CO_2$ has an effect on the accumulation of osmotic materials which determines the degree of stomatal apertures because it is known that stomata open in the condition of the reduced $CO_2$ concentration. However, it is not fully understood how $CO_2$ leads to the stomatal opening. It has been thought that $CO_2$ can not affect on the ion fluxes which determines the increase of osmotic potential in guard cells. However, in this study, the changes of guard cell membrane permeability by $CO_2$ have been focused on. There are many reports that $CO_2$ related reactions are dominant when the leaf is exposed to certain a mount of $CO_2$. The hypothesis of the stomatal opening by light is based on the increase of osmotic materials in guard cells including $K^+$, $Cl^-$, sucrose and $malate^{2-}$. It was reported that $CO_2$ induced a big hyperpolarization indicating that $H^+$ was extruded to the cell outside. It was also found that $CO_2$ caused guard cell membrane hyperpolarization in the intact leaf up to 3 or 4 times higher than that of light induced membrane hyperpolarization. These results represent that $CO_2$ can affect on the change of physical characteristics which affects on the change of the membrane permeability.
Purpose: Regardless of the underlying diseases, the proteinuric condition demonstrates ultrastructural changes in podocytes with retraction and effacement of the highly specialized interdigitating foot processes. We examined the molecular basis for this alteration of the podocyte phenotypes, including quantitative and distributional changes of ZO-1 protein as a candidate contributing to the pathogenic changes in the barrier to protein filtration. Methods: To investigate whether high glucose and advanced glycosylation endproduct(AGE) induce podocyte cytoskeletal changes, we cultured rat GEpC under 1) normal glucose(5 mM=control) or 2) high glucose(30 mM) or 3) AGE-added or 4) high glucose plus AGE-added conditions. The distribution of ZO-1 was observed by confocal microscope and the change of ZO-1 expression was measured by Western blotting and RT-PCR. Results: By confocal microscopy, we observed that ZO-1 moves from peripheral cytoplasm to inner actin filaments complexes in both AGE-added and high glucose condition. In Western blotting, high glucose or AGE-added condition decreased the ZO-1 protein expression by 11.1%(P>0.05) and 2.3%(P>0.05), respectively compared to the normal glucose condition. High glucose plus AGE-added condition further decreased ZO-1 protein expression to statistically significant level(12%, P<0.05). No significant change was seen in the osmotic control. In RT-PCR, high glucose plus AGE-added condition significantly decreased the expression of ZO-1 mRNA by 12% compared to normal glucose condition. Conclusion: We suggest that both high glucose and AGE-added condition induce the cytoplasmic translocation and suppresses the production of ZO-1 at transcriptional level and these changes may explain the functional changes of podocytes in diabetic conditions.
Eight healthy Holstein bulls, 4-6 years old were used to study the effect of season of the year on the biophysical characteristics of semen. Semen was collected twice a week by AV (artificial vagina) over one-year period. The analyses revealed that all the basic seminal traits studied were differed significantly due to season, except the ejaculate volume and consistency and the percentage of swollen spermatozoa in a hypo-osmotic fructose-citrate solution. Ejaculates collected during hot summer season had significantly lower sperm motility, concentration and total counts, and higher percentage of dead spermatozoa than those collected during winter time. Warm spring had moderate semen quality. The temperature-humidity index was calculated and it was associated (p < 0.01) negatively with the ejaculate pH, sperm concentration and total counts, and positively with the % of dead sperms. Ejaculate volume, percentage of swollen spermatozoa, individual motilities did not correlate significantly with the change in temperature-humidity index values. The total live, motile spermatozoa per ejaculate during both the winter and spring seasons showed significant increase of about 37% and 32% respectively over the summer season. Also, rectal temperatures of the bulls were elevated during the hot summer season, while the values of blood hemoglobin and packed-cell volume were decreased.

Leptin plays an important role in energy homeostasis and reproductive function in fish, especially in reproduction. Migrating fish, such as salmonoids, are affected by external environmental factors, and salinity changes are a particularly important influence on spawning migrations. The aim of this study was to test whether changes in salinity affect the expression of leptin, estrogen receptors (ERs), and vitellogenin (VTG) in chum salmon (Oncorhynchus keta). The expression and activity of leptin, the expression of ERs and VTG, and the levels of estradiol-$17{\beta}$ and cortisol increased after the fish were transferred to FW, demonstrating that changes in salinity stimulate the HPG axis in migrating female chum salmon. These findings reveal details about the role of elevated leptin levels and sex steroid hormones in stimulating sexual maturation and reproduction in response to salinity changes in chum salmon.
Hypochlorous acid(HOCl) is a chemical that is a safe sanitizer and disinfectant approved by the Food and Drug Administration as a food additive, exhibiting strong sterilizing power with low effective chlorine concentration of pH 5.0-6.5 and effective chlorine concentration 10-80 ppm. To apply to fishery industries, we develope the HOCl ice for store or delivery of fishery products. However when HOCl is being frozen, the contained HOCl are expelled out from the ice due to the molecular structures of ice; there is no space to contain HOCl inside. To increase chlorine containing amount in ice, we develop the alginate particles containing HOCl which is bio comparable since alginate is a natural polymer extracted from the brown algae and it is widely used for drug delivery and containing substances, etc. We produce HOCl with water as base solution suppressing osmotic flow from fishery products, and mix it with the developed alginate particles and made HOCl-alginate ice and checked the remaining amount of HOCl. We measure the change of pH and chlorine concentration optimizing the best concentration of alginate particles. Finally, we produce the alginate particle HOCl ices with respect to the alginate's optimal concentration.
The effect of the trehalose incorporation on the biological membranes was investigated with respect to the phase of the membranes using the fluorescence intensity change. Spherical phospholipid bilayers, vesicles, were prepared only with the variation in the phase of each layer via a double emulsion technique. In the aqueous inside of the vesicles, 8-Aminonaphthalene-1,3,6-trisulfonic acid disodium salt(ANTS) was encapsulated. As a quencher, p-Xylene-bis(N-pyridinium bromide)(DPX) was included in the buffer where the vesicles were dispersed. The fluorescence scale was calibrated with the fluorescence of ANTS vesicles in p-Xylene-bis(N-pyridinium bromide)(DPX)-included-buffer taken as 100% fluorescence and the mixture of ANTS and DPX in the buffer as 0% fluorescence. Trehalose injection into the vesicle solution led the distortion of the membrane. It was found that the distortion was related to the phase of each layer the vesicle up on the ratio of trehalose to lipid. In the identical measurements at glucose, the behavior of the distortion was completely different from that of trehalose. These results seem to depend on the stability of the vesicles, due to the osmotic and volumetric effects on the headgroup packing disruption.
Zinc and lead pollution are public environmental issues that have attracted lots of attention for a long time. Landfill leachate contains heavy metals, such as Zn(II) and Pb(II), which are usually related to the pollution of groundwater, especially in developing countries. Bentonite has been proven to be effective in enhancing the membrane property of clay, by which landfill liners can have better barrier performance towards the migration of contaminants. In this study, 5% sodium bentonite amended with locally available Fukakusa clay was utilized to evaluate the membrane behavior towards the heavy metals zinc and lead. The chemico-osmotic efficiency coefficient, ${\omega}$, was obtained through Zn(II) and Pb(II) solutions with different concentrations of 0.5, 1, 5, 10, and 50 mM. According to the results, ${\omega}$ continually decreased as the Zn(II) and Pb(II) concentrations increased, which is consistent with the Gouy-Chapman theory. Compared to normal inorganic ions, the membrane behavior towards heavy metal ions was lower. The migration of heavy metal ions was not observed based on experimental results, which can be attributed to the adsorption or ion exchange reaction. The mechanisms of the membrane performance change were discussed with the assistance of XRD patterns, free swelling results, XRF results, and SEM images.
Water balance has a significant impact on the overall fuel cell system performance. Proper water management should provide an adequate membrane hydration and avoidance of water flooding in the catalyst layer and gas diffusion layer. Considering the important of advanced water management in PEM fuel cell, this study proposes a simple one dimensional water transportation model of PEM fuel cell for use in a dynamic condition. The model has been created by assumption that the output is the water liquid saturation difference. The liquid saturation change is the total difference between the additional water and the removal water on the system. The water addition is obtained from fuel cell reaction and the electro osmotic drag. The water removal is obtained from capillary transport and evaporation process. The result shows that the capillary water transport of low temperature fuel cell is high because the evaporation rate is low.
