Effects of Haedokgumhwa-san Water Extracts on LPS-induced Inflammatory Response in Macrophage (해독금화산 물추출물이 LPS로 유도된 대식세포의 염증반응에 미치는 영향)

  • Objectives : TheHaedokgumhwa-sanwater extract (HDKHS) is used in Korea, Japan and China as a traditional therapeutic agent to cure an infectious disease. But its study is not enough. Therefore, the present study focused on the elucidation of HDKHS to investigate the anti-inflammatory effects and to established the possible mechanisms involved in its action on LPS-stimulated immune response in murine macrophages.Methods : Inflammatory status was induced by LPS and measured by increasement of inflammatory mediators. LPS induced secretions of NO and PGE2in RAW 264.7 cells were measured using griess reagent and enzyme-linked immunosorbent assay (ELISA) kit respectively. production of IL-6 was examined using ELISA kit and expression of IL-6 mRNA was measured by RT-PCR method. To investigate the effects of HDKHS on inflammatory mediators, such as iNOS, COX-2 and MAPKs, western blot and RT-PCR were performed.Results : HDKHS significantly reduced production of NO and PGE2 which were induced by LPS. Also, activation of IL-6 was reduced both protein and mRNA levels. The expressions of inflammatory mediator include iNOS and COX-2 were decreased by pretreatment with HDKHS. futhermore The result showed HDKHS down-regulate the LPS induced phosphorylation of ERK 1/2, one of the MAPK family, which is considered as a main regulator of transmission from pathogens to nucleus of immune cells.Conclusions : Our results suggest that the anti-inflammatory properties of HDKHS may stem from the inhibition of pro-inflammatory mediators via suppression of initiation of inflammatory response by inhibiting MAPKs signaling pathways.

Effects of Rumecis Radix Water Extract on Development of Atopic Dermatitis in BALB/c Mice (Balb/c 마우스의 아토피피부염에 대한 참소리쟁이 물추출물의 효과)

  • The roots of Rumecis Radix have been used for the treatment of heat phlegm, jaundice, constipation, scabies and uterine hemorrhage. The aim of this study was to confirm whether Rumecis Radix water extract (RJWE) has a preventive effect on the development of atopic dermatitis (AD) in 2,4-dinitrochlorobenzene (DNCB)-applied BALB/c mice. Oral administration (12.5 mg/kg, 25 mg/kg) and topical application (0.5 mg/mouse, 1.0 mg/mouse) of RJWE decreased the development of AD-like skin lesions, ear swelling, spleen weight and total serum IgE. RJWE significantly also inhibited the infiltration of mast cells in the dorsal skin. Furthermore, the release of histamine from rat peritoneal mast cells (RPMCs) was suppressed significantly. These results suggest that the inhibitory effect of RJWE on AD might be associated with mast cells.

Anti-obesitic Effect of Orostachys japonicus in Rats Model fed a Hyperlipidemic Diet

  • We investigated the effect of Orostachys japonicus extracts on serum lipids, leptin and insulin level in hyperlipidemic rats. Also, diacylglycerol acyltransferase (DGAT) activity and thiobarbituric acid reactive substance (TBARS) were assessed. Inhibitory effect of DGAT related to triglyceride synthesis emerged approximately 96% in EtOAc fraction and showed 90% and 67%, respectively, in CHCl3 and BuOH fractions. Furthermore, the EtOAc and BuOH fractions inhibited 81% and 77%, respectively, in glycerol-3-phosphate acyl transferase (GPAT). Hyperlipidemia and obesity marker, contents of leptin and insulin on serum of hyperlipidemic rats, decreased 50% and 25%, respectively, compared with control group in treated EtOAc fraction. The oxidative stress marker, a concentration of TBARS, showed decrease of approximately 30% in treated EtOAc fraction. Moreover, high density lipoprotein (HDL)-cholesterol contents on serum of rats fed a hyperlipidemic diet were increased 10% and low density lipoprotein (LDL)-cholesterol decreased 50% as well as triglyceride amount of feces multiplied approximately two times more than control group in treated EtOAc fraction. The data suggest that the fractions of O. japonicus may be a potent biomaterial for treatment of hyperlipidemia or obesity.

Inhibitory Effects of Water Extract of Cervi parvum cornu, Carthami tinctorii fructus and Their Combination on Osteoclast Differentiation and Bone Resorption (녹용(鹿茸), 홍화자(紅花子) 단일 및 혼합 물 추출물( 抽出物)의 파골세포(破骨細胞) 분화(分化) 억제(抑制)와 골흡수(骨吸收) 억제(抑制) 효과(效果))

  • Cervi parvum cornu (Deer Antler) and Carthami tinctorii fructus (Also known as Carthami seed) are widely used for treating osteoporosis and rheumatoid arthritis. In this study, We found out that the water extract of Cervi parvum cornu(WECPC), Carthami tinctorii fructus(WECTF) and their combination have effects of suppressing the RANKL-induced osteoclast differentiation. We assayed mRNA expression levels of NFATc1, c-Fos, TRAP and GAPDHS from bone marrow macrophages(BMMs) by means of RT-PCR. Similarly, the protein expression levels of NFATc1, c-Fos, MAPKs and $\beta$-actin in cell lysates were analyzed by means of Western blotting. then we determined the anti-osteoporotic effects of WECPC, WECTF and their combination using Lipopolysaccharide (LPS)-induced bone-loss mouse. WECPC, WECTF and their combination showed remarkable inhibition on RANKL-treated osteoclast differentiation without cytotoxicity. WECPC suppressed degradation of I-${\kappa}B$. WECPC, WECTF and their combination down-regulated the induction of c-Fos and NFATc1 by RANKL. Lastly, in vivo data showed that WECPC, WECTF and their combination rescued the bone erosion by LPS treatment. Thus, these results demonstrate that WECPC, WECTF and their combination can be efficacious remedies for bone-loss diseases such as osteoporosis and rheumatoid arthritis.

Antimicrobial, Antiperspirant and Deodorant Effect of Herbal Extract Mixtures Including Schisandra chinensis (오미자를 함유한 복합물의 항균, 발한억제 및 소취효과)

  • The aim of this study was to investigate the antimicrobial, antiperspirant and deodorant effect of herb mixtures including Schisandra chinensis(SC). This herb mixtures including Schisandra chinensis was made of Astragalus membranaceus(AM), Triticum aestivum(TA), Atractylodes japonica(AJ) and peppermint essential oil extracted from Mentha piperita(MP). We examined antimicrobial activity, antiperspirant activity and L-leucine dehydrogenase activity of the herbal extracts. We found that the mixture of SC and MP have the best antimicrobial effects on B. subtilis subsp. spizizenii and mixture of SC, AJ, AM and TA have best antiperspirant effect. Moreover, the combination of SC and AM showed the most inhibitory effect on L-leucine dehydrogenase activity. These results suggested that the herb mixtures Including Schisandra chinensis have good antimicrobial and antiperspirant effects.

Hepatoprotecive Effects of Puerariae Flos Extract on Experimental Non-alcoholic Fatty Liver Disease Models (비알콜성 지방간 동물모델에서 갈화추출물의 간보호 효과)

  • Objectives : The aim of this study is to investigate the preventive effect of Puerariae Flos ethanol extract (PE) on methionin and choline deficient (MCD)-diet-induced Nonalcoholic fatty liver disease (NAFLD) in C57BL/6J mice. Methods : In the in vivo experiments, C57BL/6J mice were divided into 4 groups; Normal group, Control group, MCD+PE 100 group, and MCD+PE 300 group. After 4 weeks, body weight, liver weight, biochemical parameters for liver function test, histological changes, reverse transcription polymerase chain reaction (RT-PCR), and Western blot were assessed. Results : Mice lost body weight with the MCD-diet and the MCD+PE 100 group and MCD+PE 300 groups lost less than the control group, though showed no statistical significance. Liver weights were decreased by the MCD diet, but MCD+PE 300 groups were increased significantly. In the liver function test, all the values were decreased with the MCD-diet, MCD+PE 100 group and MCD+PE 300 groups were increased significance. In histological findings of the livers, MCD-diet induced severe fatty accumulation in the livers, but this fatty change was reduced in the MCD+PE 100 group and MCD+PE 300 groups was inhibited respectively. In lipid accumulation factors (such as SREBP-1c, $C/EBP{\alpha}$, PPAR-${\gamma}$), MCD+PE 100 group and MCD+PE 300 groups showed inhibitory effect on liver lipogenesis by reducing associated gene expressions caused by MCD diet. Conclusions : We were able to know that Puerariae Flos ethanol extract (PE) shown hepatoprotective effects via a decrease on the hepatic lipogenesis factors in the experimental NAFLD Models.

  • From the study of Prescription on mammary diseases, the following conclusions are obtained. 1. The Lurosan (漏蘆散), Jechetang (猪蹄湯), Tongyutang (湧泉散) are often used for galactostasis in Prescription. The classification of prescription by efficacy is Chungyelyak(淸熱解毒藥), Lisuyak (利尿通淋藥), Hwalyelyak(活血祛瘀藥), etc. and the frequency of used medicines is Luro(漏蘆),Tongcho(通草),Jejo(제조), Chensangab(穿山甲),etc.. 2. The Makyajeon(麥芽煎), Sipjeondaebotang(十全大補湯) are often used for galactorrhea in Prescription. The classification of prescription by efficacy is Bogiyak(補氣藥), Bohelyak(補血藥), Sosikyak (消食藥), etc., and the frequency of used medicines is Insam (人蔘), Dangggwi(當歸), Makya(麥芽),etc.. 3. The Jogaksan(조角散), Gwarusan(瓜蔞散) are often used for mammary abscess in Prescription. The classification of prescription by efficacy is Hwalyelyak(活血祛瘀藥), Chunghwaye1damyak(淸化熱痰藥), Ligiyak(理氣藥), etc., and the frequency of used medicines is Jogakja(조角刺), Sahyang(麝香), Chongpee(靑皮),etc.. 4. The Yengyotang(連翹湯), Makyatang(麥芽湯), Lyengpayujagenbang(令敗乳自退方)are often used for distending pain of the breast due to galactostasis in Prescription. The classification of prescription by efficacy is Chungyelhaedokyak(淸熱解毒藥), Chunghwayeldamyak(淸化熱痰藥)etc., and the frequency of used medicines is Yengyo(連翹), Gwaru (瓜樓),etc.. 5. The Gwarusan (瓜蔞散), Danchungpitang(單靑皮湯), Yengyotang(連翹湯), Gamisoyosan(加味逍 遙散)are often used for mammay abscess in Prescription. The classification of prescription by efficacy is Chunghwayeldamyak(淸化熱痰藥), Hwalyelgeoyak(活血祛瘀藥), Chungyelyak(淸熱解毒藥),etc., and the frequency of used medicines is Jogakja(조角刺), Yengyo(連翹), Chongpee(靑皮),etc.. 6. The Sipyukmi1yugium(十六味流氣飮), Chungpitang(靑皮湯)are often used for breast carcinoma in Prescription. The classification of prescription by efficacy is Hwalyelyak(活血祛瘀藥), Ligiyak(理氣藥)etc., and the frequency of used medicines is Doyin(桃仁), Jogakja, Chongpee(靑皮), Jagak(枳殼),etc.

Preparation of Kyungohkgo Suspension Beverage Containing Cultivated Wild Ginseng (산양삼경옥고 현탁음료의 제조)

  • The development of healthy beverages are increasing by big concerns for well-being. The aim of this study is to develop cultivated wild ginseng-Kyungohkgo (CKOG) beverage using cultivated wild ginseng instead of natural wild ginseng for Kyungohkgo. To examine physiochemical properties of CKOG beverage, pH, degree Brix, acidity, Huntor color, particle size and sedimentation volume were measured. When compared with commercial KOG beverage, CKOG beverage was lower in pH, acidity, particle size and higher in sedimentation volume. In the sensory evaluation of CKOG beverage, the best recipe of healthy CKOG beverage was 20% CKOG, 5.7% honey, 0.2% citric acid and 74.1% of water. In stability testing, pH, degree Brix, acidity, Huntor color of CKOG beverage was not changed, microbes in CKOG beverage was not detected for 60 days. These results suggest that CKOG may have a possibility of development as healthy beverage.

Effects of Herbal treatment for In-patients with Mild Fever: Retrospective Clinical Study

  • Objectives: This study was designed to investigate the effect of herbal extract medicines for inpatients with fever symptoms in an oriental medicine hospital. Methods: Medical records of inpatients who experienced over $38^{\circ}C$ of fever from July 2010 to August 2011 has been requested with deleted personal identifiable information. The requested data were analyzed by patients' general characteristics, administrated western/herbal medicines, and changes in the body temperature of four administration groups (group 1, herbal extract medicine only; group 2, western medication(antibiotics/antipyretics) only; group 3, combination of herbal extract medicine and western medication; group 4, no medication control). The SPSS 19.0 was used for statistical analysis and p-value of less than 0.05 was regarded significant. Results: The body temperature has significantly dropped over time in the herbal extract medicine only group (p<0.001) along with the antibiotics/antipyretics only group. When both treatments were combined, stronger antipyretic effect was shown compared to the sole treatment of herbal extract medicine or antibiotics/antipyretics. Conclusion: The use of herbal extract medicines may be effective in alleviation of fever.

A Survey on Recognition of Quality Control of Oriental Medicines by People Who Work in Field of Oriental Medicine (한약재 품질관리에 관한 인식도 조사연구)

  • Objectives : The purpose of this study was to identify attitude, treatment and quality control of Oriental medicines and to find the influencing factors affecting them. Methods : We made a survey of 213 people who work in the field of Oriental medicine in Daegu. A survey on attitudes about treatment, and quality control of Oriental medicines was conducted by mail. The data was analyzed by SPSS. Results : The survey results showed that most common purchase place for Oriental medicine was a wholesale market (67.0%). Storage of Oriental medicine was mostly in warehouse (61.4%) without temperature controls. Subjects expressed necessity for quality control (100%), legal controls (96.2%), grading according to quality (95.7%), and standardization of Oriental medicine (90.6%) at a high rate. Conclusions : These results showed that subjects were interested in quality control of Oriental medicine to ensure safety.
