알코올 및 휘발성 유기산류가 뽕나무버섯의 균사속 생산에 미치는 영향 (Alcohols and Volatile Organic Acids as Stimulants of Rhizomorph Production by Armillaria mellea)

  • 천마와 뽕나무버섯의 공생관계를 검토해 보고자 천마 중에 존재할 것으로 추측되는 저분자의 휘발성 성분들로 알코올류와 휘발성 유기산들이 뽕나무버섯의 균사속 생육에 미치는 영향을 검토하였고 또한 천마 중에 함유된 휘발성 유기산의 존재를 GC로 확인하였다. 각종 알코올류와 휘발성 유기산류 중 acetic acid와 methanol을 제외한 성분들은 뽕나무버섯의 균사속 생육을 촉진하였는데 이 중, 가장 큰 효과를 보인 것은 valeric acid와 ethanol이었으며 valeric acid는 0.1%(v/v)에서 ethanol은 1%(v/v) 수준의 농도에서 효과가 있었다. 이들 화합물들은 3일 간격으로 보충할 경우는 이 농도보다 훨씬 낮은 0.01%(v/v)에서 양호한 촉진효과를 볼 수 있었다. 또한 천마 중에 존재하는 휘발성 유기산류를 GC로 분석한 결과 propionic, iso-butyric, butyric, iso-valeric, valeric, iso-caproic, caproic 및 heptanoic acid 등이 함유되어 있음을 확인하였으며, 이들 중 가장 많은 함량을 차지하는 것은 valeric acid 이었다.

Rhodopseudomonas sphaeroides에 의한 수소 생산 -Glucose 및 유기산의 영향- (Production of Hydrogen from Glucose by Rhodopseudomonas sphaeroides.)

  • R. sphaereides K7 및 E15-1은 혐기적 광합성조건에서 glucose를 탄소원으로 하여 배양초기 24시간 동안은 수소가스를 계속 생산하였으나, 그 이후에는 배양액에 축적된 acetic acid및 formic acid가 배양액의 pH를 4.2-4.8로 저하시켜 수소를 거의 생산하지 못하였다. 또한 배양 6일 후에도 R. sphaeroides K7 및 E15-1의 glucose의 이용율은 각각 43% 및 74%에 불과하였다. 그러나 배양액의 pH를 6.8-7.0으로 유지하면서 배양한 결과 R. sphaeroides K7및 E15-1 두 균주 모두의 수소생산율과 glucose의 이용율이 증가되어, 수소생산은 배양 10일까지도 계속 증가되었으며, glucose도 두 균주 각각 배양후 2.5일 및 4.5일 후에 완전 소비하였다. 뿐만 아니라 균체 배양액의 pH를 중성으로 유지하면서 R. sphaeroides K7 및 E15-1을 배양할 경우 균체의 표백현상이 제거되어 배양 7일 후에는 각각 균체의 bacteriochlorophyll 함량이 약 44배 및 9배 증가되었으며, 이때 균체의 농도는 각각 약 10배 및 2.4배 증가되었다. R. sphaeroides K7 및 E15-1은 혐기적 광합성조건에서 acetic, lactic, butyric 및 malic acid로 부터도, 비록 그 양이 glucose로 부터보다는 적으나, 수소를 생산하였다. 본 실험 결과로 미루어 혐기적 광합성 조건에서 R. sphaeroides K7 및 E15-1은 glucose로부터 수소를 생성할 때 NADH 산화 및 hydrogenase가 관여한 대사가 우선적으로 일어나고, 2차적으로는 이때 생성된 유기산을 전자 공여체로 광합성 작용에 의해 질소원이 존재하지 않을 때 nirogenase에 의해서 양성자(H$^{+}$)가 환원되어 수소(H$_2$)가 생성되는 것으로 생각된다.

PQQ-Dependent Organic Acid Production and Effect on Common Bean Growth by Rhizobium tropici CIAT 899

  • Rhizobium tropici CIAT 899 is capable of synthesizing inactive apo-glucose dehydrogenase (GDH). To become an active holo enzyme, the GDH requires a cofactor, PQQ. When R. tropici CIAT 899 was grown in a broth culture medium containing hydroxyapatite and pyrrolo quinoline quinone (PQQ), pH decreased while the concentration of soluble P increased. The solubilization of hydroxyapatite was associated with the production of gluconic acid and 2-ketogluconic acids. The organic acid production and P solubilization were greatly enhanced when the bacterium was grown with air supply. Effect of R. tropici CIAT 899 with (CI+PQQ) and without PQQ (CI) on the common bean growth was examined. Shoot and root weight, and N and P contents in CI+PQQ treatment, were significantly higher than those in control and CI treatment. Nodule weight and acetylene reducing activities were also significantly higher in CI+PQQ treatment than in other treatments.

친환경 흑염소 사양을 위한 최적 조사료 초종 및 TMR 혼합비 비율: In vitro 반추위 발효 연구 (Optimum Forage Sources and Its Ratio in TMR for Environmently-friendly Goat Feeding: In vitro Rumen Fermentation Study)

  • This study was conducted to investigate the effects of TMR on in vitro rumen fermentation and methane production of goat with different forage sources. The experiment was arranged 4×2 factorial design. The different forage sources were rice straw (RS), Italian rye grass (IR), timothy (TI) and alfalfa (AL), respectively. There were two different forage : concentrate ratios such as 20:80 (20) and 50:50 (50), respectively. Therefore, totally 8 treatments were used: 1) RS20, 2) RS50, 3) IR20, 4) IR50, 5) TI20, 6) TI50, 7) AL20, and 8) AL50, respectively. The rumen fluid of goat was collected from the slaughterhouse. For fermentation parameters, ruminal pH, total gas, methane, hydrogen, ammonia nitrogen, and volatile fatty acid were determined. The pH values were within an optimal range across all treatments. Total gas productions at TI20 and AL50 were significantly greater than others (p<0.05). Methane production was significantly lower in TI and AL compared with other treatments (p<0.05). The relatively high dietary NDF content in treatments showed significantly lower methane production (p<0.05). Significant alterations treatments were detected at ammonia nitrogen concentration according to the ratio of forage : concentrate (p<0.05). AL treatment showed greater total volatile fatty acid production compared with other treatments (p<0.05). Therefore, the present study suggests that both Timothy and Alfalfa could be recommendable forage sources for goat based on results with volatile fatty acid as an energy source and methane as an index for energy loss and environmental issues. Also, the 50:50 (forage : concentrate) ratio would prefer to 20:80.

막걸리에서 분리한 젖산세균인 Lactobacillus casei HK-9의 특성 및 항균 활성 (Characterization and Antibacterial Activity of Lactobacillus casei HK-9 Isolated from Korean Rice Wine, Makgeolli)

  • The purpose of this work was to examine the antibacterial activity derived from a lactic acid bacterium isolated from korean rice wine, called makgeolli. Various physiological and biochemical properties of this strain were characterized. Both the BIOLOG system and phylogenetic analysis using 16S rRNA sequencing were utilized for identification, and the strain was designated as Lactobacillus casei HK-9, and registered in GenBank as [JQ951606]. Growth rate, production of organic acids (e.g., lactic acid and acetic acid), and pH changes during growth were monitored. The maximum concentrations of lactic acid and acetic acid were approximately 576 mM and 199 mM, respectively, and pH was changed from 7.00 to 3.74 after 72 h of incubation. HPLC was used to confirm the production of lactic acid and acetic acid. Significant antimicrobial activity of the concentrated supernatant was demonstrated against various food-poison causing bacteria (e.g., Bacillus cereus, Listeria monocytogenes, Staphylococcus aureus, methicillin-resistant Staphylococcus aureus, Escherichia coli, Salmonella enteritidis). Ethanol tolerance of L. casei HK-9 showed up to 12% of ethanol within the culture.

L-Glutamic acid 생성균에 관한 연구 2 (Studies on L-Glutamic Acid-Producing Bacteria(II))

  • Searches for the nutrition requirements of three strains of Brevibacterium ammoniagenes reported in the previous paper were carried out with an aim of achieving the striking accumulation of L-glutamic acid and the large multipication of cells. It was recognized that all three strains required both biotin and thiamine, together with amino acids such as histidine or cysteine, for their good growth and extracellular L-glutamic acid accumulation. The quantity of biotin required for remarkable growth of these microorganisms was quite different from that for the maximum production of L-glutamic acid. This result, however, did not apply in the case of thiamine. It was also confirmed that, of 18 amino acids, histidine and cysteine were the msot effective organic nitrogen sources, while the most available inorganic ammonium salt resulting in a large amount of L-glutamic acid-production and considerable cell gorwth was found to be only urea. Maximum accumulation of extracellular L-glutamic acid, more than 50%(w/w) of the initial sugar content, could be obtained from fermentation in the medium containing wheat-bran extract(Brev. ammoniagenes T-1 and Brev.ammoniagenes Y-2) or rice-bran extract(Brev. ammoniagenes YR-2), which confirmed us a possibility that these bacteria might be employed for industrial fermentation of L-glutamic acid.

Physicochemical Components and Antioxidant Activities of Daebong Persimmon (Diospyros kaki cv. Hachiya) Peel Vinegars

  • To evaluate utilization of persimmon peels as novel bio-materials, the general composition and antioxidant activities of Daebong persimmon vinegar (DPV), Daebong persimmon peel vinegar (DPPV) and commercial persimmon vinegar (CPV) were investigated. The pH of DPPV had slightly higher than that of other vinegars. The titratable acidities of vinegars were 3.24% (DPV), 2.77% (DPPV) and 7.78% (CPV), respectively. The reducing sugar contents showed that DPV had slightly higher than that of other vinegars. The browning degree of DPV was lower than CPV in contrast to the turbidity. The results of Hunter's color value have showed that overall values of CPV had significantly higher than DPPV and DPV. The total phenolic contents of DPV and CPV were 19.49 and 17.13 mg/100g GAE, respectively. The total flavonoid contents of DPPV (8.04 mg/100g CE) were two fold higher than that of DPV (3.85 mg/100g CE). The antioxidant activities, by DPPH and FRAP assays, of DPV showed stronger than those of other vinegars. Free sugars were mainly composed of fructose and glucose. Organic acids were presented in the order acetic acid, succinic acid, malic acid, tartaric acid, citric acid and oxalic acid. These results suggest that the Daebong persimmon peels could be utilized for vinegar production as a health-benefit material.

Antioxidant and Antibacterial Activity of Caprylic Acid Vanillyl Ester Produced by Lipase-Mediated Transesterification

  • Vanillyl alcohol (VA), which is abundant in Vanilla bean, has strong antioxidant activity. However, the use of VA in the food and cosmetics industries is limited, due to its low solubility in emulsion or organic solvents. Meanwhile, medium chain fatty acids and medium chain monoglycerides have antibacterial activity. We synthesized butyric acid vanillyl ester (BAVE) or caprylic acid vanillyl ester (CAVE) from VA with tributyrin or tricaprylin through transesterification reaction using immobilized lipases. BAVE and CAVE scavenged 2,2-diphenyl-1-picrylhydrazyl radicals in organic solvents. In addition, BAVE and CAVE decreased the production rate of conjugated diene and triene in the menhaden oil-in-water emulsion system. While BAVE showed no antibacterial activity, CAVE showed antibacterial activity against food spoilage bacteria, including Bacillus coagulans. In this study, the antibacterial activity of vanillyl ester with medium chain fatty acid was first revealed. Zeta potential measurements confirmed that BAVE and CAVE were inserted into B. coagulans membrane. In addition, the propidium iodide uptake assay and fluorescent microscopy showed that CAVE increased B. coagulans membrane permeability. Therefore, CAVE is expected to play an important role in the food and cosmetics industries as a bi-functional material with both antioxidant and antibacterial activities.

축산폐수의 혐기성 고정법에 있어서 암모니아성 질소의 영향 (Effect of ammonium nitrogen in anaerobic biofilter using live-stock-wastewater)

  • In this research, the synthetic livestock wastewater was prepared to study the characteristics of organic matter removal, the change of VFA production, and the amount of gas production with respect to the change of ammonium nitrogen concentration in the waste using anaerobic fixed bed process, which is an anaerobic biofilm process. The HRT and operation temperature were 1 day and $35{\pm}1^{\circ}C$, respectively. Also, the characteristics of organic matter removal and the inhibitory effect on microorganism in the anaerobic process were studied on the organic loading and ammonium nitrogen concentration. The results obtained were as follows: For COD loading of $10kg/m^3$-day and five levels of ammonium nitrogen concentration ranging from 1,000 to 5,000 mg/L, organic removal efficiencies were about 81, 74, 67, 58, and 51%, and gas productions were 3,860, 3,520, 3,240, 3,020, and 2,790 ml/l-day, respectively. Average methane contents in the gas produced on COD loading of $10kg/m^3$-day was about 76%. Throughout the whole period of experiment, remaining VFA (as COD base) in the effluent was over 90% of remaining COD. This result indicated the inhibitory effect of high concentration of ammonium nitrogen through the facts that accumulated VFA was almost COD and organic removal efficiency decreased also with the increase of ammonium nitrogen. Especially, that implys which high concentration of ammonium nitrogen not only inhibits methane forming bacteria, but also acid forming bacteria.

Effect of Wood Vinegar on the Performance, Nutrient Digestibility and Intestinal Microflora in Weanling Pigs

  • Two experiments were conducted to investigate the feeding value of wood vinegar in weanling pigs. In Experiment 1, weanling pigs (n = 224; Landrace ${\times}$Yorkshire ${\times}$Duroc, 21${\pm}$3 d-old, initial BW 6.12${\pm}$0.10 kg) were assigned to four dietary treatments. Different levels of wood vinegar were added to the diets as dietary treatments (0, 0.1, 0.2 and 0.3%). Each treatment comprised 4 replicates with 14 piglets in each. Experimental feeding was conducted for 28 d in two phases (phase I, d 0 to 14 and phase II, d 15 to 28). Feeding of wood vinegar linearly (p<0.05) improved the phase I, phase II and overall ADG and increased (linear, p<0.05) the overall and phase II ADFI. Linear improvements in the apparent fecal digestibility of dry matter (p = 0.013), gross energy (p = 0.019) and crude protein (p = 0.033) were observed as the level of wood vinegar was increased in the diet of pigs. Experiment 2 was conducted to compare dietary wood vinegar with commonly used growth promoters, organic acid (mixture of 21% phosphoric acid, 3.25% propionic acid, 2.8% formic acid, 10% calcium formate and 5% calcium propionate) and antibiotic (aparamycin). A total of 288 weanling piglets (Landrace ${\times}$Yorkshire ${\times}$Duroc, 22${\pm}$2 d-old, initial BW 6.62${\pm}$0.31 kg) were assigned to four treatments with four replicates (18 piglets/pen) for 28 days and fed in 2 phases: phase I, d 0 to 14 and phase II, d 15 to 28. The dietary treatments were control (corn-soybean meal basal diet without antibiotics) and diets containing 0.2% antibiotic, 0.2% organic acid and 0.2% wood vinegar. Pigs fed antibiotic showed higher (p<0.001) ADG and better feed efficiency followed by pigs fed wood vinegar and organic acid diets while those fed the control diet had lowest ADG and poorest feed efficiency. The overall and phase I ADFI was highest (p<0.001) in pigs fed wood vinegar and lowest in pigs fed the control diet. Apparent fecal digestibility of dry matter, gross energy and crude protein was significantly higher (p<0.05) in pigs fed the antibiotic diet when compared with pigs fed the control but comparable among pigs fed antibiotic, organic acid and wood vinegar diets. Higher populations of Lactobacillus (p = 0.004) were noted in the ileum of pigs fed the wood vinegar diet, while the population of coliforms in the ileum and cecum was higher (p<0.001) in pigs fed the control diet when compared with pigs fed antibiotic, organic acid or wood vinegar diets. These results indicated that wood vinegar could improve the performance of weanling pigs by improving the nutrient digestibility and reducing harmful intestinal coliforms; moreover performance of pigs fed wood vinegar was superior to those fed organic acid.
